Description
−動物に移植若しくは注入される場合に腫瘍を形成する能力を保持する;
−腫瘍退縮と関連するタンパク質の内因性応答と一致するシグナルを発生する;
−in vivoでの薬物物質の速度効果のリアルタイム分析を可能にするような良好なS/N比をもつシグナルを発生する;
−動物間の低い変動性を提供するような良好な再現性を伴うシグナルを発生する;および−誘導された応答の非侵襲的画像化を可能にするシグナルを発生する
べきである。
第一の態様において、本発明は、ヒト以外の動物に移植若しくは注入される場合に腫瘍を形成することが可能であることを特徴とする、腫瘍退縮と関連するタンパク質の発現もまた制御するプロモーターに機能しうるように連結されたレポーター遺伝子、好ましくは蛍光タンパク質を含有する発現ベクターで安定にトランスフェクトされた腫瘍細胞株を提供する。
−レポーター遺伝子の発現を測定する段階
を含んでなる、抗腫瘍活性についての化合物のin vitroスクリーニング方法を提供することもまた本発明の一目的であり;
ここで、対照レベルに比較してのレポーター遺伝子発現の増大が該化合物を抗腫瘍活性を有すると同定する。特定の一実施態様において、レポーター遺伝子は蛍光タンパク質であり、そしてレポーター遺伝子の発現は発射される蛍光の量として測定される。上に説明されたとおり、本in vitroの方法についてもまた、使用されるプロモーターに依存してスクリーニングの選択性を変えることが可能である。本発明の特定の一実施態様において、該in vitroスクリーニング法はヒストンデアセチラーゼ阻害剤に対し選択的であり、そして、p21の1300bpのプロモーターフラグメントがp53応答配列を含まないことを特徴とする前記プロモーターフラグメントを含んでなる発現ベクターで安定にトランスフェクトされた腫瘍細胞を含んでなる。さらなる一実施態様において、該in vitroスクリーニング法は例えばアクチノマイシンDのようなDNA損傷剤に対し選択的であり、そして最低1個のp53応答配列を含んでなる発現ベクターで安定にトランスフェクトされた腫瘍細胞を含んでなる。ある実施態様において、発現ベクターは、好ましくはHSV−TKプロモーターのような最小プロモーターの一部として配列番号10よりなるp53応答配列を含んでなる。
−本発明の腫瘍細胞をヒト以外の動物において腫瘍を形成するのに十分な量で前記ヒト以外の動物に投与する段階;

を含んでなる、製薬学的活性についての化合物のスクリーニング方法を提供することが本発明の一目的である。製薬学的活性化合物とのインキュベーションは対照レベルに比較してレポーター遺伝子発現の増大をもたらすことができる。
[発明の詳細な記述]
ベクター
本発明は、発現レベルが生理学的状態と関連するタンパク質若しくは酵素の発現もまた制御するプロモーターに機能しうるように連結されたレポーター遺伝子を含んでなるベクターに関する。
は下述されるところのp21の1300bpのプロモーターフラグメント若しくは下述されるところのp53応答配列を含んでなるHSV−TK最小プロモーターよりなる。別の実施態様において、プロモーター配列は、腫瘍退縮と関連するタンパク質の発現もまた制御するプロモーター若しくはプロモーター活性を有するそのフラグメントであることができる。
別の態様において、本発明は、発現レベルが生理学的条件と関連するタンパク質若しくは酵素の発現もまた制御するプロモーターに機能しうるように連結されたレポーター遺伝子を含んでなる発現ベクターで安定に形質転換された標的細胞に関する。
いる安定に形質転換された腫瘍細胞株を提供することが、本発明の一目的である。とりわけ、p21活性化因子に応答性であるプロモーターに機能しうるように連結された蛍光タンパク質をコードする核酸配列を含有する発現ベクターを含んでなる、安定に形質転換された卵巣癌細胞株、好ましくはA2780細胞、好ましくは、前記プロモーターは1300bpのp21プロモーター配列(配列番号1)を含んでなり、なおより好ましくは、前記プロモーター配列は1300bpのp21プロモーター配列(配列番号1)よりなる。ここで、さらなる一実施態様において、蛍光タンパク質は、EGFP、EYFP、DsRed、ZsGreen、ZsYellow、HcRed、またはpDsRed、pHcRed1、pd2EGFP若しくはpd2EYFPのような不安定化蛍光タンパク質よりなる群から選択され、そして特定の一実施態様において蛍光タンパク質はZsGreenよりなる。好ましい一実施態様において、安定に形質転換された細胞は、それぞれの受託番号LMBP 5958CBおよびLMBP 5959CBをもつpGL3−basic−ZsGreen−1300−クローン1およびpGL3−basic−ZsGreen−1300−クローン2として2003年1月20日にベルギーの微生物協調収集物(Belgian Coordinated Collection of Microorganisms)(BCCM)に寄託されたクローンから選択される。前記クローンは、商業的に入手可能なpGL3ベクター由来かつp21活性化因子に応答性であるプロモーターに機能しうるように連結されたZsGreenをコードする核酸配列を含有する発現ベクターを含んでなり、前記プロモーターは1300bpのp21プロモーター配列(配列番号1)を含んでなり、ここで前記クローンは、それらがMitsui(別名MS−275−Shering−登録番号209783−80−2)のような細胞周期の停止および細胞分化の既知の誘発因子での誘導に際して自動全身画像化系による蛍光の検出を可能にする低い基礎蛍光発現を有することを特徴とする。

なおより好ましくは、前記プロモーター配列は1300bpのp21プロモーター配列(配列番号1)よりなる。ここで、さらなる一実施態様において、蛍光タンパク質はEGFP、EYFP、DsRed、ZsGreen、ZsYellow、HcRed、またはpDsRed、pHcRed1、pd2EGFP若しくはpd2EYFPのような不安定化蛍光タンパク質よりなる群から選択され、そして特定の一実施態様において蛍光タンパク質はZsGreen若しくはZsRedよりなる。より好ましい一実施態様において、該in vitroスクリーニング法で使用される安定に形質転換された腫瘍細胞は、受託番号LMBP 5958CBおよびLMBP 5959CBでBCCMに寄託されたクローンから選択される。前述のin vitroスクリーニング法の別の実施態様において、安定に形質転換された腫瘍細胞は、p53活性化因子に応答性であるプロモーターに機能しうるように連結された蛍光タンパク質をコードする核酸配列を含有する発現ベクターを含んでなる安定に形質転換された結腸直腸癌細胞若しくは安定に形質転換された卵巣癌細胞、好ましくはA2780細胞若しくはHCT116細胞よりなり、好ましくは、前記プロモーターはp53応答配列(配列番号10)を含んでなり、なおより好ましくは、前記プロモーターはp53応答配列を含んでなる最小HSV−TKプロモーター(配列番号13)よりなる。ここで、さらなる一実施態様において、蛍光タンパク質はEGFP、EYFP、DsRed、ZsGreen、ZsYellow、HcRedまたはpDsRed、pHcRed1、pd2EGFP若しくはpd2EYFPのような不安定化蛍光タンパク質よりなる群から選択され、そして特定の一実施態様において蛍光タンパク質はZsGreenよりなる。特定の一実施態様において、該in vitroの方法で使用される安定に形質転換された細胞は、p53RE_TK/pGL3−Basic−ZsGreenベクター(図6)でトランスフェクトしたA2780若しくはHCT116細胞よりなる。
本発明の標的細胞を含んでなるヒト以外の動物を提供することもまた本発明の一実施態様である。とりわけ、腫瘍性増殖、とりわけ癌性増殖についてのヒト以外の動物モデルを提供すること。
本発明の安定に形質転換された腫瘍細胞が典型的にはヒト以外の動物の皮下に注入される106ないし108細胞を含んでなる腫瘍細胞懸濁液として投与される、本発明の腫瘍細胞をヒト以外の動物に投与する段階;および
前記ヒト以外の動物において腫瘍を形成するための十分な時間(上で概説されたところの卵巣癌細胞を使用する特定の実施態様において典型的には7〜14日、より好ましくは9〜12日およびとりわけ12日よりなる)を腫瘍細胞に与える段階
を含んでなる、抗腫瘍活性をもつ化合物を同定するためのヒト以外のin vivo動物モデルの製造方法を提供する。
もしかすると活性のある化合物を本発明のヒト以外の動物、好ましくは上で概説されたところの卵巣癌細胞を注入されたヌードマウスに投与する段階(ここで該もしかすると活性のある化合物は静脈内、経口および腹腔内を包含する全部の臨床上適切な投与経路により投与してよい);ならびに
レポーター遺伝子の発現を測定することにより腫瘍細胞に対する前記もしかすると活性のある化合物の効果を評価する段階
を含んでなる、抗腫瘍活性をもつ化合物のin vivo同定方法でその後使用しうる。

細胞培養および試薬
A2780細胞(ATCC)は、5%CO2を含む加湿インキュベータ中、37℃で、10%FCS、2mM L−グルタミンおよびゲンタマイシンを補充したRPMI 1640培地中で培養した。HCT116細胞(ATCC)は5%CO2を含む加湿インキュベータ中、37℃で、10%FCS、2mM L−グルタミンおよびゲンタマイシンを補充したマッコイ5a培地中で培養した。全部の細胞培養溶液はGibco−BRL(メリーランド州ゲイサーズバーグ)により提供される。他の材料はNuncにより提供される。
ゲノムDNAを、増殖しているA2780細胞から抽出しかつp21プロモーターのネステッドPCR単離の鋳型として使用した。第一の増幅は、鋳型としてのゲノムDNAとともにオリゴヌクレオチド対GAGGGCGCGGTGCTTGG(配列番号2)およびTGCCGCCGCTCTCTCACC(配列番号3)を使用して55℃のアニーリング温度で20周期の間実施した。TATAボックスに関して−4551ないし+88のフラグメントを含有する、生じる4.5kbのフラグメントを、4.5kbのフラグメントをもたらす88℃でのアニーリングを伴う20周期の間オリゴヌクレオチドTCGGGTACCGAGGGCGCGGTGCTTGG(配列番号4)およびATACTCGAGTGCCGCCGCTCTCTCACC(配列番号5)を用い、そしてその後TATAボックスに関して−1300ないし+88のフラグメントを含有する1.3kbのフラグメントをもたらす88℃のアニーリングを伴う20周期の間オリゴヌクレオチド対TCGGGTACCGGTAGATGGGAGCGGATAGACACATC(配列番号6)およびATACTCGAGTGCCGCCGCTCTCTCACC(配列番号7)を用いて再増幅した。オリゴヌクレオチド中に存在する制限部位XhoIおよびKpnI(下線を付けた配列)をサブクローニングに使用した。
ルシフェラーゼレポーターをpGL3−basicから除去しかつKpnIおよびXbaI制限部位でZsGreenレポーター(pZsGreen1−N1プラスミドから)により置換した。pGL3−basic−ZsGreen−1300は、ヒトp21プロモーター領域の上で挙げられた1.3kbのフラグメントのpGL3−basic−ZsGreenへのXhoIおよびKpnI部位での挿入を介して構築した。全部の制限酵素はBoehringer Manheim(ドイツ)により提供される。
A2780細胞を2×105細胞の密度で6ウェルプレートにプレーティングし、24時間インキュベートし、そして製造元により記述されたとおりリポフェクタミン(Lipofectamine)2000(Invitrogen、ベルギー・ブリュッセル)を使用することにより2μgのpGL3−basic−ZsGreen−1300および0.2μgのpSV2neoベクターでトランスフェクトした。トランスフェクトした細胞をG418(Gibco−BRL、メリーランド州ゲイサーズバーグ)で10日間選択し、そして単一細胞懸濁液を増殖させた。3週後に80個の単一クローンを得た。
A2780のトランスフェクトしたプールおよび選択した80クローンを拡大しそしてウェルあたり10000細胞で96ウェルプレートに接種した。接種24時間後に、標的を定めた化合物(近接のp21プロモーター領域中のsp1部位に影響を及ぼす)若しく
はDNA損傷剤(p53応答配列に影響を及ぼす)で細胞を追加の24時間処理した。その後細胞を4%PFAで30分間固定し、そしてヘキスト染色で対染色した。ZsGreen産生および従って蛍光に至るp21プロモーターの活性化を、Ascent Fluoroskan(Thermo Labsystems、ベルギー・ブリュッセル)および蛍光顕微鏡(Zeiss)によりモニターした。
選択したクローンをヌードマウスの脇腹に皮下注入し(107細胞/200μl)そしてキャリパーで測定可能な腫瘍を12日後に得た。第12日以降、溶媒、20mpkのMitsui(別名MS−275−Shering−登録番号209783−80−2)若しくは40mpkのJNJX(それぞれ10、10および4動物)を6日間毎日動物に経口投与した。腫瘍は、社内で開発した自動全身画像化系(GFPフィルターを装備しかつNational Instruments(R)からのIMAQ Visionソフトウェアに基づくソフトウェアパッケージにより制御されるCCDカメラ 型式JAI(R)CV−M90に接続した蛍光立体顕微鏡 型式Olympus(R)SZX12)により蛍光について評価した。
HDAC阻害剤によるp21の1300bpのプロモーターフラグメントのin vitroでの誘導
A2780の安定なトランスフェクトしたプールおよび選択した80クローンを、材料および方法に記述されたとおり、TSA(10−7M)、ブレオマイシン(15mU)およびMitsui(10−6M)で処理した。80クローンのうち6クローンがTSAおよびMitsui処理に応答した。これらのクローンの4個は、Fluoroskanで検出可能でなかったくらい低い基礎蛍光発現を示したが、しかし蛍光顕微鏡を使用するZsGreen産生の人的評価により選択し得た。2個の他のクローンは測定が可能であり、そしてTSA(10−7)により5倍およびMitsui(10−6)により1.2〜1.5倍の誘導を示した。
マウスにクローン1、2、3および5を注入し、そして材料および方法に記述されたとおり化合物(溶媒、20mpkのMitsui若しくは40mpkのJNJ’99)を投与した。基礎のZsGreen発現および誘導は、クローン2および3において、自動全身画像化系により検出されるには低すぎた。クローン1および5の基礎蛍光は測定が可能であり、また、ZsGreenの誘導は第一の用量の投与3日後に非常に明瞭でありかつ5日後にプラトーに達した(図2)。
TSA、MitsuiおよびSAHAのような数種の既知のヒストンデアセチラーゼ阻害剤が、TATAボックスに関して−60bpから+40bpまでの領域のネズミのp21waf1,cip1プロモーターのトランス活性化を誘導することが示された。この領域はわれわれのp21waf1,cip1の1300bpのプロモーター構築物中に存在する。われわれのデータは、これらの剤がpGL3−basic−ZsGreen−1300でトランスフェクトしたA2780細胞中でin vitroでZsGreen産生
を誘導することを確認する。DNA損傷剤(カンプトテシン、ブレオマイシンおよびドキソルビシン)はp21waf1,cip1プロモーターのp53依存性の調節を介してそれらの活性を発揮する。p53応答配列は、p21waf1,cip1プロモーター中のより下流に1300bpのプロモーターフラグメント中に存在しない領域に位置している。これはDNA損傷剤に対する該系の非応答性を説明する。これらのデータから、われわれは、われわれのレポーター系がヒストン脱アセチル化に関する分子事象を検討するためのモデルを提供しかつ該レポーター系の特異性をin vitroで明らかにすると結論する。この概念はまた、とりわけDNA損傷剤を、若しくは応答配列を適合させることによりいずれかの他の薬物を試験するのにも使用し得る。
細胞培養および試薬
A2780細胞(ATCC)は5%CO2を含む加湿インキュベータ中、37℃で、10%FCS、2mM L−グルタミンおよびゲンタマイシンを補充したRPMI 1640培地中で培養した。HCT116細胞(ATCC)は5%CO2を含む加湿インキュベータ中、37℃で、10%FCS、2mM L−グルタミンおよびゲンタマイシンを補充したマッコイ5a培地中で培養した。全部の細胞培養溶液はGibco−BRL(メリーランド州ゲイサーズバーグ)により提供される。他の材料はNuncにより提供される。
以下のオリゴのp53REフォワードCCTGCCTGGACTTGCCTGGGTCGACCCTGCCTGGACTTGCCTGGC(配列番号8)およびp53REリバースTCGAGCCAGGCAAGTCCAGGCAGGGTCGACCCAGGCAAGTCCAGGCAGGGAGCT(配列番号9)をEurogentecから注文した。該オリゴヌクレオチド対を、アニーリング緩衝液(150mMトリス pH7.6、15mM MgCl2、23mM DTT)中で65℃で開始して50℃、40℃、30℃にわたり20℃の最終温度まで5分ごとのアニーリング温度の段階的低下によりアニーリングした。クローニングのためのSacI/XhoIオーバーハングをもつフラグメントが形成された(応答配列は下線を付けた配列である)(配列番号10):

ルシフェラーゼレポーターをpGL3−basicから除去しかつKpnIおよびXbaI制限部位でZsGreenレポーター(pZsGreen1−N1プラスミドから)により置換した。
A2780細胞およびHCT116細胞を4×105細胞の密度で6ウェルプレートにプレーティングし、24時間インキュベートし、そして製造元により記述されたとおりリポフェクタミン(Lipofectamine)およびプラス試薬(Plus Reagent)(Invitrogen、ベルギー・ブリュッセル)を使用することにより2μgのp53RE_TK/pGL3−basic−ZsGreenおよび0.2μgのpMC1−neo−Poly Aベクターでトランスフェクトした。トランスフェクトした細胞をG418(Gibco−BRL、メリーランド州ゲイサーズバーグ)で16日間選択し、そして単一細胞懸濁液を増殖させた。3週後に60個の単一クローンをトランスフェクトしたHCT116細胞から得、また、40個の単一クローンをトランスフェクトしたA2780細胞から得た。
A2780およびHCT116のトランスフェクトしたプールおよび選択したクローンを拡大し、そしてウェルあたり20000細胞で96ウェルプレートに接種した。接種24時間後に、細胞をDNA損傷剤(p53応答配列に影響を及ぼす)で追加の24時間処理した。ZsGreen産生および従って蛍光に至るp53RE_tkの活性化を、Ascent Fluoroskan(Thermo Labsystems、ベルギー・ブリュッセル)および蛍光顕微鏡(Zeiss)によりモニターした。
DNA損傷剤によるin vitroでのp53RE_tkの誘導
HCT116およびA2780の安定なトランスフェクトしたプールおよび選択したクローンをアクチノマイシンD(10ng/ml)で処理した。選択したクローンのうち、6個のHCT116クローンおよび2個のA2780クローンがアクチノマイシンD処理に応答した。これら8クローンについて、蛍光の最低2倍の誘導が処理に応答してFluoroskanで検出可能であった。とりわけ、A2780のクローン36ならびにHCT116のクローン5および36は、アクチノマイシン処理に応答しての強い誘導係数を伴う、検出可能な低い基礎蛍光発現を示した(図3)。
Claims (11)
- プロモーターに機能しうるように連結されたレポーター遺伝子を含有する発現ベクターでトランスフェクトされた安定に形質転換された腫瘍細胞であって、該レポーター遺伝子が蛍光タンパク質よりなり、かつ、該プロモーターが配列番号1に示されたp21プロモーターフラグメントからなる、上記の腫瘍細胞。
- 安定に形質転換された腫瘍細胞が、安定に形質転換された卵巣癌細胞若しくは結腸直腸癌細胞よりなる、請求項1に記載の腫瘍細胞。
- 蛍光タンパク質が、EGFP、EYFP、DsRed、ZsGreen、ZsYellow、HcRed、またはpDsRed、pHcRed1、pd2EGFP若しくはpd2EYFPのような不安定化蛍光タンパク質よりなる群から選択される、請求項1若しくは2に記載の腫瘍細胞。
- 受託番号LMBP 5958CBを有する2003年1月20日にpGL3−basic−ZsGreen−1300−クローン1としてベルギーの微生物協調収集物(Belgian coordinated collection of microorganisms)に寄託された、請求項1ないし3のいずれか1つに記載の腫瘍細胞。
- 受託番号LMPB 5959CBを有する2003年1月20日にpGL3−basic−ZsGreen−1300−クローン2としてベルギーの微生物協調収集物(Belgian coordinated collection of microorganisms)に寄託された、請求項1ないし3のいずれか1つに記載の腫瘍細胞。
- 化合物の製薬学的活性のスクリーニングのためのヒト以外の動物であって、該動物が請求項1ないし5のいずれか1つに記載の安定に形質転換された腫瘍細胞を含んでなり、かつ該ヒト以外の動物がげっ歯類である、上記動物。
- 安定に形質転換された腫瘍細胞がげっ歯類の皮下に外科的に移植若しくは注入される、請求項6に記載のヒト以外の動物。
- 請求項6または7に記載のヒト以外の動物の製造方法であって、ヒト以外の動物において腫瘍を形成するのに十分な量の請求項1ないし5のいずれか1つに記載の細胞を前記ヒト以外の動物に投与することを含んでなる、上記の方法。
- ヒト以外の動物が遺伝子的に免疫無防備状態であるか若しくは前記腫瘍と同系である、請求項8に記載の方法。
- 請求項1ないし5のいずれか1つに記載の腫瘍細胞を、試験されるべき化合物と接触させる段階;およびレポーター遺伝子の発現を測定する段階
を含んでなる、抗腫瘍活性についての化合物のin vitroスクリーニング方法。 - もしかすると活性のある化合物が投与された請求項6または7に記載のヒト以外の動物を用意する段階;および
レポーター遺伝子の発現を測定することにより腫瘍細胞に対する前記化合物の効果を評価する段階を含んでなる、製薬学的活性についての化合物のスクリーニング方法。
