Abstract

I et første aspekt fremskaffer foreliggende oppfinnelse en tumorcellelinje som er stabilt transfektert med en ekspresjonsvektor inneholdende et reportergen, fortrinnsvis et fluorescent protein, operativt knyttet til en promoter som også kontrollerer ekspresjon av et protein som er assosiert med tumorregresjon, stabilisering av tumorvekst eller inhibering av metastatisk vekst, og som er særpreget ved at nevnte cellelinje er i stand til å danne en tumor når implantert eller injisert i det ikke-humane dyr. Sammenlignet med tradisjonelle in vivo-modeller er foreliggende oppfinnelse forskjellig ved at reportergenet ikke blir konstituitivt uttrykt, men kun etter eksponering til en forsøksforbindelse som resulterer i ekspresjonen av et protein eller enzym som er assosiert med tumorregresjon, stabilisering av tumorvekst eller inhibering av metastatisk vekst. Kun når en forbindelse som skal undersøkes, kommer inn i sirkulasjonen og infiltrerer tumoren, kan den generere reportersignalet, forutsatt at den fremmer ekspres3onen av et protein assosiert med tumorregresjon, og promoteren av nevnte protein er operativt knyttet til reportergenet.
