Description
DNAを固体表面に吸着させる方法(Nikiforov and Rogers,Anal Biochem.1995)
−[CR3R3−CR1R3−X]n− [化学式1]
前記式で、R1はエポキシ基を有するエステルであり、R3は水素またはアルキル基であり、Xは粘性の高いアクリレート系化合物である。

前記式で、R1はエポキシ基を有するエステルであり、R2はアルキルエステルであり、R3は水素またはアルキル基である。

W標的配列:5’Cy3−AGCCAG*GAGAAA−3’[配列番号5]

Claims (13)
- エポキシ基を有する重合体がコーティングされたプラスチック基板上に、所望のDNA配列を含むプローブPNAが固定化され、前記エポキシ基を有する重合体は、エポキシ基を有するアクリレートモノマーと、エポキシ基を有さないアクリレートモノマーとの共重合体であり、前記エポキシ基を有する重合体中のエポキシ基の含有量が、10−40重量%であることを特徴とするPNAチップ。
- プラスチック基板は、ポリメチルメタクリレート(PMMA)、ポリカーボネート(PC)、ポリノルボルネン(polynorbornene)、COC(Cyclic Olefin Copolymer)、フッ素化ポリイミド、ポリスチレン(PS)、SBC(Styrene Butadiene Copolymer)、ABS(Acrylonitrile Butadiene Styrene)、SAN(Styrene AcryloNitrile)、及びポリスルホンからなる群から選択される材質により製造された透明なプラスチック基板であることを特徴とする請求項1に記載のPNAチップ。
- 前記エポキシ基を有する重合体は、下記化学式1で示される、エポキシ基を有するアクリレートモノマーと、25℃における粘度が8〜6,000cpの範囲であるアクリレートモノマーとの共重合体であることを特徴とする請求項1に記載のPNAチップ:
−[CR3R3−CR1R3−X]n− [化学式1]
前記式で、R1はエポキシ基を有するエステルであり、R3は水素またはアルキル基であり、Xは粘性の高いアクリレート化合物である。 - 前記25℃における粘度が8〜6,000cpの範囲であるアクリレートモノマーは、ジペンタエリスリトールヒドロキシペンタアクリレート(DPHA)、9−エチレングリコールジアクリレート(9−EGDA)、ペンタエリスリトールトリ−テトラアクリレート(PETA)、ポリエチレングリコール400ジアクリレート、トリプロピレングリコールジアクリレート、トリメチロールプロパントリアクリレート、及びジペンタエリスリトールヘキサアクリレートからなる群から選択されることを特徴とする請求項1に記載のPNAチップ。
- 前記エポキシ基を有する重合体は、下記化学式2で示される、エポキシ基を有するアクリレートモノマーとアクリレート誘導体との共重合体であって、前記アクリレート誘導体は前記プラスチック基板を構成するモノマーの種類に類似しており、そのため前記アクリレート誘導体からなる重合体が前記プラスチック基板に類似する物性を有しており、前記プラスチック基板に対して前記重合体が容易に接着することを確実にすることを特徴とする請求項1に記載のPNAチップ:
−[CR3R3−CR1R3−CR3R3−CR2R3]n− [化学式2]
前記式で、R1はエポキシ基を有するエステルであり、R2はアルキルエステルであり、R3は水素またはアルキル基である。 - 前記アクリレート誘導体は、メチルメタクリレート(MMA)、エチルアクリレート、エチルメタクリレート(EMA)、n−プロピルアクリレート、n−プロピルメタクリレート、イソプロピルアクリレート、及びイソプロピルメタクリレートからなる群から選択されることを特徴とする請求項5に記載のPNAチップ。
- 前記エポキシ基を有する重合体で、エポキシ基の含有量は、20−30重量%であることを特徴とする請求項1に記載のPNAチップ。
- アミン作用基を有し、エーテル含有または非含有のC5−C8のカルボン酸リンカーがプローブPNAのアミン末端に付着されたことを特徴とする請求項1に記載のPNAチップ。
- エポキシ基を有するアクリレートモノマー、25℃における粘度が8〜6,000cpの範囲であるアクリレートモノマー、及び光開始剤を混合する段階と、前記混合液をプラスチック基板上にコーティングする段階と、紫外線を利用して前記混合液を硬化させる段階と、及びプローブPNAプリンティング溶液を前記プラスチック基板上にスポッティングする段階とを含む、請求項1に記載のPNAチップの製造方法。
- エポキシ基を有するアクリレートモノマー、プラスチック基板と類似物性を有するアクリレート誘導体、及びラジカル開始剤を混合する段階と、前記混合液をラジカル重合させる段階と、前記重合体を溶解剤に溶解させた溶液をプラスチック基板上にコーティングする段階と、プローブPNAプリンティング溶液を前記プラスチック基板上にスポッティングする段階とを含む、請求項1に記載のPNAチップの製造方法。
- 前記プローブPNAプリンティング溶液に、0.01〜1.0M濃度の塩基を添加してPNA固定化を助けることを特徴とする請求項9または請求項10に記載のPNAチップの製造方法。
- スポッティングする前に、重合体がコーティングされたプラスチック基板を30%以上の湿度が維持されるところに4時間以上保管する段階をさらに含むことを特徴とする請求項10に記載のPNAチップの製造方法。
- 請求項1から請求項8のうちいずれか1項に記載のPNAチップに標的DNAを含んだ反応試料を適用する段階と、プローブPNAと標的DNAとを混成化反応させる段階と、非特異的反応産物を除去するために、前記PNAチップを洗浄する段階と、PNA/DNA混成化による蛍光シグナルを検出する段階とを含むSNP(Single Nucleotide Polymorphism)検出方法。
