Description
特定の分子に対する反応を引き出させる免疫系の能力は、抗原の存在を認識する能力に決定的に依存する。従来より、抗原という用語は、B細胞の誘導により抗体発生元となる分子の能力と関わりがあるとされていた。しかし、現在は、T細胞も抗原を認識する能力を所有することが知られている。T細胞抗原認識には、主要組織適合遺伝子複合体(MHC)の分子に関連して抗原フラグメント(ペプチド)を細胞表面に提示する抗原提示細胞(APC)が必要である。T細胞は、抗原特異的受容体(TCR)を用いて、APCによって提示される抗原フラグメントを認識する。このような認識は、認識された抗原を根絶する一定の反応を引き起こすための免疫系に対するトリガーとして作用する。
個体T細胞を誘発するために、必要最小限の数のTCRが、APCに提示されたペプチド/MHC複合体と結合されなければならない。このために足りる十分な数の、APCの表面に到達するペプチドは、他のトリガーペプチド(triggering peptide)と比べた場合のT細胞活性化を誘発する能力によって「優性(dominant)」または「亜優性(subdominant)」と称することができる。正常な事象の過程においては(即ち生理学上は)、任意のタンパク質は、T細胞反応を誘発することのできるペプチドを2つ以上生成する。つまり、「優性」という用語は、最も有力な反応または最も頻繁な反応を誘発するペプチドに適用される。任意のタンパク質配列においては、優性および亜優性エピトープに加えて、反応を誘発するのに足りる十分な数でAPC表面に到達しない(T細胞が特異的な)潜在的なT細胞ペプチドエピトープがある。換言すると、APC内における抗原プロセシングのメカニズムにより、特定のペプチドは分解されてTリンパ球に有効に提示され、そのためT細胞反応を刺激するが、他方で他のペプチドは不完全に分解されてTリンパ球に提示される。これら後者のペプチドは、潜在的に反応性のTリンパ球を刺激するに足りる十分な数でAPC表面において提示されないため「潜在ペプチドエピトープ(crypticpeptide epitope)」と称されている。
潜在ペプチドエピトープは、通常体内に存在するタンパク質(自己タンパク質)および非自己(すなわち外来)タンパク質の両方に存在する。正常な生理機能においては、潜在エピトープと反応する能力を有するT細胞はin vitro一次刺激アッセイにおいて検出されない、即ち、血液から単離されたばかりのT細胞は潜在ペプチドと共に培養された場合に明らかな増殖を示さない。対照的に、有効に分解されてAPCによりTリンパ球に提示される他のペプチドは、一次培養において増殖応答を刺激し得る。これらが「優性」および「亜優性」エピトープである。
「アトピー性アレルギー」という用語は、高濃度のイムノグロブリンE(IgE)によって特徴づけられるアレルギー群に適用される。これらには、アレルギー性喘息、花粉症、慢性(perennial)アレルギー性鼻炎、蕁麻疹(urticaria、hives)および湿疹等の種類、アレルギー性結膜炎、並びに特定の食物アレルギー(特に食物過敏症)が含まれる。このようなアトピー症状の病理発生メカニズムは、抗原/アレルゲン特異的IgEの合成だけでなく、マスト細胞および好酸球などのエフェクター細胞の分化および成長を伴う。
アレルギー性IgE仲介疾患は、現在、アレルゲン成分または抽出物の周期的な注射を伴う脱感作処置によって治療されている。脱感作治療は、アレルゲンに対してIgEと競合するIgG応答を誘発するか、または、アレルゲンに対するIgEの合成をブロックする特異的サプレッサーT細胞を誘発し得る。この治療形態は、必ずしも有効ではなく特に一般的な過敏症ショックなどの重度の副作用を引き起こす危険性がある。これは、すぐに認識して、アドレナリンで治療しなければ生死に関わる場合もある。他の外来抗原に対する免疫反応性を変えることなく、またアレルギー反応自体を誘発することなく、特定のアレルゲンに対する望ましくないアレルギー性免疫反応を低下または除去する治療処置が、アレルギーをもつ個体にとって大いに有利となる。
時には、自己および非自己が免疫学的に区別される正常なメカニズムが壊れ、正常な体組織に存在する自己抗原に対して免疫反応が誘引され得る。この「自己免疫」は、自己免疫甲状腺炎、リウマチ性関節炎、および紅斑性狼瘡等の病理状態を発症する。治療養生法は、比較的非特異的かつ多くの望ましくない副作用を有する抗炎症剤または免疫抑制剤の使用にほぼ制限されている。
WO92/11859は、APCの特異的MHCクラスII分子に結合する非アレルゲン由来の非刺激性ペプチドを特定のアレルゲンに対するT細胞反応を阻害するために使用する、アレルゲンに対する免疫反応を低下する方法を記載している。
WO91/06571は、ネコアレルギーの診断、治療または予防に使用することのできるヒトT細胞反応性ネコタンパク質に由来するペプチドの開示を主旨としている。
WO94/24281は、主要ハウスダストダニアレルゲンのペプチドおよび改変ペプチドに関する。改変ペプチドは、脱感作治療に伴う望ましくない副作用のレベルを低下することを目的とする。
G.F. Hoyneらは、Immunology 83、190〜195頁(1994)において、主要アレルゲンDerpIをコードするcDNAから作成したペプチドを用いてハウスダストダニアレルギーを試験した。彼らは、主要エピトープを含むペプチドが、マウスにおいて全アレルゲンに対する経口寛容を誘導できること、また他のペプチドでも寛容を誘導できることを示すことを主旨としている。この方法において潜在ペプチドが役割を果たすことが示唆されているが、それらの同定方法または治療上の使用については開示されていない。
上記開示のいずれも、潜在ペプチドが、喘息または他のアレルギー疾患等のアトピー症状の病理において役割を果たし得ることを示唆していない。本発明者らは、潜在ペプチドの同定方法を見いだし、喘息または他のアレルギーに基づく病理を有する個体が、一次培養において、関係する病状を起こすアレルゲンに由来する潜在エピトープにより刺激され得るTリンパ球集団をもつことを認めた。上記したように、健康な個体から単離されたTリンパ球は、一次培養において潜在エピトープによって刺激されるとは考えられない。自己免疫病理においては、同様に、免疫系に対して通常潜在性である自己ペプチドが認識されるようになり、免疫反応を誘出する。
従って、本発明により、タンパク質のペプチドが潜在ペプチドであるか否かを決定する方法が提供される。該方法は、i)一次攻撃誘発においてT細胞をペプチドに暴露する工程と、ii)該工程iの該一次攻撃誘発における該ペプチドと該T細胞の反応性を測定する工程と、iii)T細胞をタンパク質に暴露して得られる予備攻撃誘発された(pre-challenged)T細胞を、二次攻撃誘発においてペプチドに暴露する工程と、二次攻撃誘発における該ペプチドと該予備攻撃誘発T細胞の反応性を測定する工程とを含む。
予備攻撃誘発により、APC表面上の優性および亜優性エピトープの発現のみならず、全ての潜在決定基の発現が可能になる。その後のこれらの細胞のペプチド再攻撃誘発により、優性、亜優性および潜在エピトープに対するT細胞反応性が明らかになる。ペプチドでの一次攻撃誘発は、正常に発現された優性および亜優性エピトープの反応だけを引き出すだろう。従って、ペプチド一次攻撃誘発だけの後に観察されたペプチドではなくて、全抗原一次攻撃誘発の後のペプチド二次攻撃誘発の後に観察されうるペプチドが潜在エピトープであると定義される。T細胞反応性が上記二次攻撃誘発においては認められるが一次攻撃誘発では認められない場合、そのペプチドが潜在ペプチドである。
本発明は、以下のように例示的に説明することができる。T細胞を含む末梢血単核細胞(PBMC)を個体から直接採取し、タンパク質由来の重複する合成ペプチドのセットと共に短時間の間(例えば、3〜7日間)インキュベートした場合、正常な事象の過程においてT細胞が認識するペプチド(即ち優性および亜優性エピトープ)に対する増殖応答が見られるだろう。しかし、PBMCが高い用量の全分子(または全てのペプチドの混合物)と共にまず1〜2週間の間培養された場合、後続するペプチドでの「二次」攻撃誘発によって、可能性のある優性、亜優性および潜在T細胞エピトープが検出されるだろう。これは、全抗原による高い用量での「一次」攻撃誘発の結果、いずれかのペプチドに対してなんらかの特異性を有するT細胞集団が誘発され増殖(数が増加)するからである。一次アッセイおよび二次アッセイにおいてPBMCをペプチドで攻撃誘発することにより、集団中の潜在エピトープを同定できる。従って、正常集団において潜在エピトープとして作用するペプチドを除き、喘息患者または関連する病理を有する個体において優性エピトープとして作用する全てのペプチドがこの一次および二次アッセイの方法を用いて検出できる。
本発明の好適な実施態様においては、予備攻撃誘発された細胞は、T細胞をタンパク質に暴露することによって、またはT細胞を集団培養(bulkculture)でタンパク質に暴露することによって得られる。
T細胞は、多数の個体(例えば、>20)を含む集団または一個体から得られることができる。集団から得た場合、健康な個体中において潜在的であると同定されたペプチドはいずれも、一般に集団中でも潜在的であると考えられるが、一方、一個体から単離されたT細胞を用いると、その個体において潜在的である(または潜在的でない)が集団においては潜在でないかもしれないペプチドしか同定されないだろう。
上記方法の工程は、記載したような順序で、または本質的に等価な結果を得るのに適した当業者に知られるそれに代わる順序で実施され得る。
本発明の特に好適な実施態様においては、ペプチドが由来するタンパク質は、Fel dI(飼育猫Felis domesticusのネコ皮膚および唾液腺アレルゲン-このアミノ酸配列はWO 91/06571に開示されている)、Der pI、Der pII、Der fIまたはDer fII(ハウスダストダニのヒョウヒダニに由来する主要タンパク質アレルゲン-これらのアミノ酸配列はWO 94/24281に開示されている)、並びに、草、木、雑草(ブタクサを含む)花粉、菌類およびカビ、食物(例えば、魚、貝類、カニ、ウミザリガニ、ピーナッツ、ナッツ、卵、および牛乳)、刺毛昆虫(例えば、ミツバチ(bee)、ジガバチ(wasp)、スズメバチ(hornet))およびユスリカ(非ヌカカ(non-biting midges))、クモおよびダニ、イヌ、ウマ、ラット、モルモット、マウス、およびアレチネズミ等の哺乳動物、ラテックス、生物学的洗浄添加剤、薬剤(例えば、ペニシリン、他の抗生物質および麻酔剤)のいずれかに存在するアレルゲンを含む一覧より選択される。
特に、ペプチドが由来し得る昆虫タンパク質は、イエバエ、ミバエ、羊クロバエまたはニクバエ(sheep blow fly)、ラセンウジバエ、穀類ゾウムシ(grain weevel)、蚕、ミツバチ、非ヌカカの幼虫、ハチミツガの幼虫、コメノゴミムシダマシの幼虫、ゴキブリ、およびテニブリオ・モリター(Tenibrio molitor)甲虫の幼虫を含む一覧より選択し得る。これらは全て、職場で起きるアレルギー問題に特に関連する昆虫アレルゲンである。
本発明の方法で使用するのに特に好適なペプチドには、図1、図2および図3に示すペプチドが含まれる。本方法はまた、配列番号25〜56のいずれか1つに示されるペプチドを使用し得る。本発明はまた、これらのペプチドを潜在ペプチドとして使用することを包含する。
本発明のさらなる態様は、本発明の方法によりスクリーニングされた場合に潜在的であると同定された全てのペプチドである。
MHCクラスII分子に結合する能力を保持するペプチドは、最高約40、好ましくは31の残基を有し得る。つまり、有用なペプチドは配列番号2〜10および12〜56のいずれか1つに示される配列を含み得ることが理解されるであろう。従って、好適な実施例の1つにおいては、連続する14merの配列は、さらに大きいペプチドの1部(好ましくは最高約31の残基)を構成する。この実施例においては、14merは、前記大きいペプチド(またはポリペプチド)の約45%を構成する。好ましくは、この配列は前記大きいペプチドの約50%以上、より好ましくは約60%以上、さらに好ましくは約70%以上、または約80%以上を構成する。特に好適な実施態様において、この配列は、前記大きい配列の約90%または約95%以上を構成する。
また、本発明の更なる態様は、医薬または診断薬として使用される好ましくは本発明の方法によってスクリーニングされた潜在ペプチドである本発明のペプチド、好ましくは本発明の方法によってスクリーニングされた潜在ペプチドである本発明のペプチドの喘息などのアトピー症状の治療医薬の製造における使用、好ましくは本発明の方法によってスクリーニングされた潜在ペプチドである本発明のペプチドの喘息などのアトピー症状の診断のための診断薬の製造における使用、好ましくは本発明の方法によってスクリーニングされた潜在ペプチドである本発明のペプチドを、適切な担体、希釈剤、または賦形剤と混合することを含む医薬または診断薬の製造方法、そのような使用および方法から製造された組成物、好ましくは潜在ペプチドである本発明のペプチドを用いてヒトの身体に対して実施される治療および/または診断の方法を含む。
本発明のペプチドは未加工のペプチド(raw peptide)として投与することが可能であるが、それらを医薬組成物として提供することが好ましい。更なる態様によれば、本発明は、潜在ペプチドと、それの医薬的に許容可能な1つ以上の担体と、任意に1つ以上の他の治療成分とを含む医薬組成物を提供する。担体は、該組成物の他の成分と適合しうるという意味で「許容可能」であり、その受容者に対して有害でないことが必要である。
前記組成物には、経口(特に吸入)および非経口(皮下、経皮、皮内、筋内、静脈内、および直腸を含む)投与に適したものが含まれるが、最も適切な経路は例えば受容者の症状および障害に依存し得る。組成物は、都合よく単位用量形態で提供されてもよいし、製薬分野の当業者に周知のあらゆる方法で製造され得る。全ての方法は、本明細書において定義される本発明の化合物あるいは薬理学的に受容可能なその塩または溶媒化合物(「有効成分」)を、1つ以上の補助成分を構成する担体と組み合わせる工程を含む。
経口投与に適した本発明の組成物は、それぞれが所定量の有効成分を含むカプセル、カシェ剤、または錠剤などの別個単位、粉末または顆粒、溶液または水性液体もしくは非水性液体の懸濁液、あるいは水中油型液体乳濁液または油中水型液体乳濁液として提供され得る。有効成分はまた、丸薬(bolus)、舐剤、またはパスタとして提供され得る。吸入のための組成物は、有効であることが知られているあらゆる手法(例えば、計量用量吸入器(metered dose inhalers))で提供され得る。
非経口投与のための組成物には、抗酸化剤、緩衝液、静菌薬、対象とする受容者の血液と等張にする溶質を含有し得る水溶性および非水溶性滅菌注射溶液、並びに、懸濁剤および増量剤を含み得る水溶液および非水溶液滅菌懸濁液を含有する。組成物は、例えば、封止されたアンプルおよび水薬瓶等の単回用量または多数回用量容器に入れられて提供されてもよく、また使用直前に例えば注射用水(water-for-injection)等の滅菌液体担体を加えるだけでよい凍結乾燥された状態で保存されてもよい。即席の注射溶液および懸濁剤は、先に記載したような種類の滅菌粉末、顆粒および錠剤から製造し得る。
直腸投与のための組成物は、カカオ脂またはポリエチレングリコール等の通常の担体と一緒にされて座薬として提供され得る。
好ましい単位用量組成物は、有効成分を、以下に説明するような有効な用量、またはその適切な割合で含む。
本発明の化合物は、典型的に1日あたり0.001〜1mg/kgの用量を経口的にまたは注射によって投与され得る。
本発明はまた、本発明の潜在ペプチドを含む診断キットを提供する。各キットは、本発明の方法によって同定された潜在ペプチドに対応する凍結乾燥されたペプチドを含む微量培養プレートからなる。各ペプチドは、(統計的な評価のために)最低3つのウェルにおいて存在し得る。スクリーニングされるペプチドの数によって更に多くの複製体(replicates)を用いることができる。凍結乾燥されたペプチドを用いることにより、キットを使用前に室温にて数ヶ月保存することができる。
アッセイを実施するための好適な実施態様によれば、末梢血単核細胞(PBMC)を標準方法によって患者の血液から単離し得る。約20mlの血液が必要となる。微量培養プレートのウェルの105細胞を含む200μlの容量の培地中にPBMCを添加する。空気中5%のCO2ガスで処理された加湿インキュベータ中で37℃にて6日間培養した後、3H-チミジン等の同位体標識、またはブロモデオキシウリジン等の非同位体標識を各ウェルに所定の時間(約12〜24時間)添加する。存在するペプチドに対する細胞増殖は、あらゆる適切な手法(例えば、DNA合成分析、液体シンチレーション分光分析または比色定量)によってアッセイすることができる。
本発明によるペプチド攻撃誘発に対する細胞反応性の測定は、上記したように細胞増殖の測定によって実施されることが好ましい。しかし、そのような反応性はまた、他の細胞反応性、例えばアレルギー反応を示す放出プロフィールを示し得るサイトカインおよびケモカイン(chemokines)等の可溶性仲介物の放出についてのアッセイ、を測定することにより測定され得る。
ペプチド免疫治療の望ましくない特定の副作用を回避しおよび/またはその効果を向上するために、治療に使用される本発明の潜在ペプチドを点突然変異させてもよい。
ペプチド免疫治療は、特異的T細胞を無能にするペプチドの投与を伴う。Allervax Catの治験から得たデータ[J. Allergy Clin. Immunol. 1996年1月、要約、815]は、一部の患者において、投与されたペプチドに対する後期(late-phase)応答が生じることを示している。この反応は本質的には喘息発作であり、この種の治療の重度な副作用の代表的なものである。「改変ペプチドリガンド」の使用によって、後期応答を発症することなく、同じ治療目的(即ち、反応性T細胞の機能的消失)が達成され得る。治療上の使用のための潜在ペプチドを同定した後、密接に関連したペプチドのパネルを作るために残基に対して一連の点突然変異を行い得る。これらは、a)元のペプチドと同様の親和性を有して適切なMHC分子に結合する能力、およびb)元のペプチドに対して特異的なT細胞をアネルギーにする(anergise)もしくは死滅させる能力についてスクリーニングされ得る。このような改変または合成ペプチドもまた本発明に包含される。
本発明による特に好適な合成ペプチド、また本発明により使用される特に好適な合成ペプチドには、図3に示すペプチドが含まれる。このようなペプチドは、本発明により個別にまたは対や複数に組み合わされて使用し得る。
予備攻撃誘発されたT細胞は、上記したように生成され得るか、または、1つの代替としてはT細胞を目的とするタンパク質に由来する複数のペプチドもしくは集団培養に暴露することによって生成され得る。
本発明による特に好適な実施態様において、タンパク質のペプチドが潜在ペプチドであるか否かを決定する方法は以下のように実施され得る。確立された技術を利用してT細胞を末梢血液から単離する[Cellular Immunology LabFax., P.J. Delves(Ed), 1994, Tissue and Cell Culture、第4章、45頁、Blackwell Scientific Publications]。細胞の一部をin vitroにて目的とするタンパク質と6〜12日間培養する。残りの細胞を、培養に小分けし、タンパク質に由来する目的とするペプチドとインキュベートする。3〜6日後、培養中のDNA合成の量を一定時間後に(例えば、8〜16時間後に)定量し得ることを可能にする標識を培養に添加する。培養中のDNAの量を、ペプチドを含まない対照培養と比較して分析することにより、この「一次」攻撃誘発においてT細胞を刺激するタンパク質に由来するペプチド配列を同定することが可能になる。全タンパク質と共に6〜12日間の間培養した細胞群をこの培養の終了後に回収し、培地中で数回洗浄した。ただし、これらの細胞は、初めに全タンパク質と共に培養されたため(即ち、「予備攻撃誘発」されたため)、後続するペプチド攻撃誘発は「二次」攻撃誘発と称する。二次攻撃誘発におけるDNA合成の量についての後の分析では、個体内のT細胞によって認識される可能性のあるタンパク質に由来する「全ての」ペプチドを同定する。
要約すると、一次培養は疾患のある個体または疾患のない個体に由来するTリンパ球により認識されたペプチドを同定し、二次培養は疾患のある個体または疾患のない個体においてT細胞が反応し得る全てのペプチドを同定する。集団レベルにおいて潜在的なエピトープは、一次培養において、疾患のある個体には認識されるが疾患のない個体には認識されない。
本発明によるT細胞は、ペプチド攻撃誘発における反応性を検出できるのに十分な純度でTリンパ球を含む1人以上の個体から得られる単核細胞の調製物として採取することができる。上記した全ての技術的方法は、その方法の所望の目的を達成可能にするために当業者に公知の他の代替方法によって実施してもよい。
本発明の更なる実施態様において、特にアトピー症状、より特定すると喘息に対する治療活性を所有する化合物を同定するための化合物スクリーニング(compound screen)における潜在ペプチドの使用が提供される。本発明のこの更なる実施態様においては、潜在ペプチドは、好ましくは本発明の方法を用いて同定されたものである。
本発明の更なる実施態様によれば、治療化合物を同定するためのスクリーニング方法、並びにこのようなスクリーニング方法を実施するのに適したキットが提供される。本発明のスクリーニング方法は、好ましくはin vitroで実施するが、in vivoで実施してもよい。好適な実施態様において、本発明のスクリーニング方法は、in vitroで以下のように実施し得る。単離された末梢血単核細胞または培養されたTリンパ球をin vitroにて、好ましくはこれらの細胞において増殖および/または他の反応を誘発するのに最適となるように予め決定された濃度の1つ以上の潜在ペプチドと共に培養する。潜在ペプチドに対するこれらの細胞の増殖応答を低下する可能性についてスクリーニングされる化合物を特定の培養ウェルに添加する。このような化合物は、好ましくは、様々な広範囲の濃度の培養に添加する。例えば、細胞の増殖応答の最大の低下をもたらす用量を確定するために、化合物を10-12M〜10-2Mの範囲で10倍希釈で培養に添加し得る。細胞増殖は、当該分野において公知のあらゆる技術を用いて測定することができる。例えば、トリチウム化されたチミジン等の化合物で細胞を標識化すると、DNA合成即ち細胞増殖の定量が可能になり得る。潜在ペプチドによって誘発された細胞増殖を低下させることができる化合物は、その潜在ペプチドに関連する医学的症状に対する治療薬であると同定することができる。in vivoで実施された場合、スクリーニング方法は、潜在ペプチドを好ましくはヒト哺乳動物へ投与してその潜在ペプチドに関連する症状を誘発し、好ましくはそれに続いてスクリーニングされる化合物を投与することを含む。誘発された症状に関わる症候を緩和できる化合物は、その症状に対する治療薬であると同定することができる。
好ましくは、in vitroにしろin vivoにしろ、スクリーニングは、喘息等のアトピー症状に関わる潜在ペプチドを使用すること、そして最も好ましくは本明細書において同定される1つ以上のFC1Pペプチドを使用することを含む。
以下に、本発明の限定を意図しない実施例を添付の図面を参照しながら記載する。図面において、
図1は、一文字表記のアミノ酸コードで表した主要ネコアレルゲンFel dIの鎖1の天然配列、およびそれに由来する9個のペプチドを示す。
図2は、一文字表記のアミノ酸コードで表した主要ネコアレルゲンFel dIの鎖2の天然配列、およびそれに由来する11個のペプチドを示す。
図3は、Fel dIの配列に由来する3個の合成ペプチドを示す。3個のペプチドは集合的にFC1Pと称する。
図4は、PBMCについて実施例に記載する、FC1PでのPBMCの一次および二次攻撃誘発に反応した細胞増殖レベル(Δcpmとして表示)についての実施例2の結果を示す。
図5は、実施例に記載する、FC1PでのPBMCの一次攻撃誘発に反応した細胞増殖レベル(Δcpmとして表示)についての実施例3の結果を示す。
図6は、実施例に記載する要因の結果として、時間の経過に対する喘息患者の肺機能(リットル毎のFEV1として表示)についての実施例4の結果を示す。
図7は、デキサメタゾンおよびフルチカゾン(fluticazone)による、FC1Pペプチドに対するT細胞増殖の阻害(トリチウム化されたチミジンの取り込み反応により測定)についての実施例5の結果を示す。
実施例1
タンパク質のペプチドが潜在ペプチドか否かを決定する方法は、以下のように実施され得る。
1-T細胞を末梢血液から単離する。
2-細胞の一部をin vitroにて目的のタンパク質と共に6〜12日間培養する。
3-残りの細胞を培養に小分けし、タンパク質に由来する目的とするペプチドとインキュベートする。
4-3〜6日後、培養に標識を添加し、培養中のDNA合成量を定量することを可能にする。
5-培養中のDNA合成の量を、ペプチドを含まない対照培養と比較して分析する。これにより、この「一次」攻撃誘発においてT細胞を刺激するタンパク質由来のペプチド配列を同定する。
6-全タンパク質と共に6〜12日間培養した細胞群を、この培養終了後に回収し、培地中で数回洗浄する。
7-二次攻撃誘発におけるDNA合成量を分析することによって、個体内のT細胞により認識され得るタンパク質に由来する「全ての」ペプチドを同定する。
上記方法により同定した潜在ペプチドは、二次攻撃誘発においてT細胞反応性を刺激するが、一次攻撃誘発においては刺激しないものである。
実施例2
末梢血単核細胞を密度勾配遠心法によって単離した。細胞の一部を一次培養においてペプチドで攻撃誘発し、残りの細胞を100μg/mlネコ鱗屑抽出物(Fel dIを含有)と共に10日間培養した。その後、ネコ鱗屑と共に培養した細胞を回収し、培地中で洗浄し、更に(本明細書の図3に示すように)全体でFC1Pを構成する3個のペプチド(濃度36μg/ml)、並びに、同数の照射された自己由来の支持細胞と共に3日間培養した。細胞増殖(Δcpmとして表示)を、新たに合成したDNAへのトリチウム化されたチミジンの取り込み反応により測定することにより定量した。これは、図4に図示する。
実施例3
新しく単離した末梢血単核細胞を、細胞増殖を測定するために、トリチウム化されたチミジンで標識する前に、1ウェル当たり105細胞で6日間培養した。ネコアレルギー喘息患者(CAA)は、非ネコアレルギー喘息患者(NCA)よりも、FC1P(36μm/ml)に対してより高い一次増殖応答を示した。結果を図5に示す。
実施例4
ネコアレルギー喘息をもつボランティアのそれぞれの前腕に対して40μgの高度に精製したFC1Pペプチドを皮内注射した。これにより、FC1Pの投与から約3時間後に単離された後期喘息反応が誘発された。肺機能(FEV1として定量)は低下し、その後数時間の間低いままであった。FEV1の低下は、8時間目のβ2アゴニストの投与、およびコルチコステロイドの吸入により逆転した。この結果を図6に図式化した。
実施例5
新しく単離したTリンパ球を、これらの細胞における増殖および/または他の反応を誘発するのに最適となるように予め決定された濃度のFC1Pペプチドと共にin vitroにて培養した。特定の培養ウェルに、グルココルチコステロイドのデキサメタゾンまたはフルチカゾン(fluticazone)を10-6Mまたは10-9Mの希釈で培養に添加した。培養終了後、トリチウム化されたチミジンで細胞を標識化し、DNA合成即ち細胞増殖の定量を可能にした。結果を図7に示す。
