Description
(a) 磁性材料およびマトリクス材料を含み、磁性材料が磁場への曝露に際して残留性である、標的物質を結合する磁性粒子を提供すること;
(b) 標的物質を含む試料を含む液相を提供すること;
(c) 標的物質を粒子に結合させるように試料を磁性粒子と共に分散すること;および
(d) 磁場を粒子に加え、かつ粒子を液相から分離することによって、粒子を試料から単離すること。
この実施例では、フェリ磁性磁鉄鉱粒子のケイ酸ナトリウム溶液(水ガラス)の水系分散液を油相と混合し、磁鉄鉱が水相に存在する油中水型エマルジョンを形成する。酸の存在下で縮重合を実施し、無機ポリマーを用いた磁性粒子を生じる。
本実施例では、フェリ磁性磁鉄鉱粒子を有機モノマー(EGDMA)に分散し、粒子懸濁液を水相と混合することによって水中油型エマルジョンを形成した。モノマーを重合させて有機ポリマー磁性粒子を生じた。
本実施例では、フェリ磁性粒子を有機溶媒中にモノマーと共に分散し、モノマーを次いで重合して粒子を形成する。
本実施例では、 フェリ磁性 磁性粒子を有機 溶媒中に、予備重合したポリマーと共に分散し、粒子を形成する。
Claims (41)
- 磁性材料およびマトリクス材料を含み、前記磁性材料は磁場への曝露後に磁場の不存在下において液相で懸濁した場合に粒子が凝集塊を形成する程度に磁気の残留性があり、かつ前記マトリクス材料は液相の存在下で粒子の解離を促進する官能基を含む表面を有する、標的物質を結合することができる磁性粒子であって、前記磁性材料は鉄酸化物を含むフェリ磁性材料であり、前記マトリクス材料はシリカベースのポリマーを含み、及び前記官能基は親水性である、磁性粒子。
- 鉄酸化物を含むフェリ磁性材料が、任意にその第一鉄のすべてまたは一部が、カドミウム、クロム、コバルト、銅、マグネシウム、マンガン、ニッケル、バナジウム、および/または亜鉛から選択された二価遷移金属で置換されている、請求項1に記載の磁性粒子。
- 鉄酸化物を含むフェリ磁性材料がフェリ磁性磁鉄鉱を含む、請求項1に記載の磁性粒子。
- 長さまたは直径が、0.1ないし5000μmの範囲内である、前記の請求項のいずれかに記載の磁性粒子。
- 実質的に球形である、前記の請求項のいずれかに記載の磁性粒子。
- マトリクス材料が標的物質を結合するための親和性物質をさらに含む、前記の請求項のいずれかに記載の磁性粒子。
- 標的物質が核酸である、前記の請求項のいずれかに記載の磁性粒子。
- 親和性物質が、細胞、タンパク質、ウイルス、またはプリオンを結合することができる、請求項6に記載の磁性粒子。
- 親和性物質が、抗体、結合タンパク質、抗体または結合タンパク質の断片、またはリガンドを含む、請求項8に記載の磁性粒子。
- 親和性物質が、ビオチニル化された標的物質への結合のためのアビジンを含む結合タンパク質を含むか、または親和性物質がビオチンを含み、かつ標的物質がアビジン化された、請求項9に記載の磁性粒子。
- 親和性物質が、標的物質に特異的なオリゴヌクレオチドまたは金属キレートを含むリガンドを含む、請求項9に記載の磁性粒子。
- 細胞またはタンパク質が微生物起源である、請求項8から11のいずれかに記載の磁性粒子。
- 標的物質が金属を含み、かつ親和性物質が金属に対するキレート剤を含む、請求項6に記載の磁性粒子。
- 磁化されていない磁性材料を提供すること、および磁性粒子を形成するようにマトリクス材料を提供することを含み、前記磁性材料は磁場への曝露後に磁場の不存在下において液相で懸濁した場合に粒子が凝集塊を形成する程度に磁気の残留性があり、かつ前記マトリクス材料は液相の存在下で粒子の解離を促進する官能基を含む表面を有する、標的物質を結合することができる磁性粒子の調製のための方法であって、前記磁性材料は鉄酸化物を含むフェリ磁性材料であり、前記マトリクス材料はシリカベースのポリマーを含み、及び前記官能基は親水性である、磁性粒子の調製のための方法。
- マトリクス材料が予備成型されて提供され、磁性材料に添加される、請求項14に記載の方法。
- シリカベースのポリマーが、磁化されていない磁性材料の存在下でのモノマーの重合によって提供される、請求項14に記載の方法。
- 重合の段階が逐次縮合および/またはラジカル反応を含む、請求項16に記載の方法。
- 重合の段階がエマルジョン中で起こり、磁化されていない磁性材料がエマルジョンの不連続相に存在する、請求項16から17のいずれかに記載の方法。
- 重合の段階がエマルジョンの不連続相で起こる、請求項18に記載の方法。
- モノマーが重合の前にエマルジョンの連続相に存在する、請求項18または請求項19に記載の方法。
- エマルジョンが水中油型エマルジョンである、請求項20に記載の方法。
- 重合の段階が溶液中で起こる、請求項16から17のいずれかに記載の方法。
- 磁性材料が、長さまたは直径が100ないし300nmの範囲内である粒子を含む、請求項14から22のいずれかに記載の方法。
- 磁性粒子が請求項1から13のいずれかで定義される通りである、請求項14から23のいずれかに記載の方法。
- 標的物質を、そのような標的物質を含む試料から分離するために用いられる、請求項1から13のいずれかに記載の、または請求項14から24のいずれかに記載の方法によって入手可能な、磁性粒子。
- 細胞、微生物、またはタンパク質を含む標的物質を、そのような標的物質を含む試料から分離するために用いられる、請求項1から6、または8から12のいずれかに記載の磁性粒子。
- 金属を含む標的物質を、そのような標的物質を含む試料から分離するために用いられる、請求項1から6、または13のいずれかに記載の磁性粒子。
- 有機化合物を含む標的物質を、そのような標的物質を含む試料から分離するために用いられる、請求項1から6のいずれかに記載の磁性粒子。
- 核酸を含む標的物質を、そのような標的物質を含む試料から分離するために用いられる、請求項1から7のいずれかに記載の磁性粒子。
- 標的物質が試料から単離される、請求項25から29のいずれかに記載の磁性粒子。
- 標的物質が試料から除去される、請求項25から29のいずれかに記載の磁性粒子。
- セルソーティング装置に用いられる、請求項1から6、または8から12のいずれかに記載の磁性粒子。
- (a)磁性材料およびマトリクス材料を含み、前記磁性材料は磁場への曝露後に磁場の不存在下において液相で懸濁した場合に粒子が凝集塊を形成する程度に磁気の残留性がある、標的物質を結合する磁性粒子であって、前記磁性材料は鉄酸化物を含むフェリ磁性材料であり、前記マトリクス材料はシリカベースのポリマーを含み、及び前記官能基は親水性である、磁性粒子を提供すること;
(b)標的物質を含む試料を含む液相を提供すること;
(c)標的物質を粒子に結合させるように試料を磁性粒子と共に分散し、磁性粒子を破壊に供して粒子を解離させること;および
(d)磁場を粒子に加え、かつ粒子を液相から分離することによって、粒子を試料から単離すること
