技術分野
本発明は、ケフィア・グレインから得られる乳発酵産物を含有する皮膚外用剤に関する。より詳細には、本発明は、ケフィア・グレインによる乳発酵産物であるケフィア及び/又はケフィアホエーを含有する皮膚外用剤に関する。本発明の皮膚外用剤は、優れた細胞賦活作用、皮膚老化防止作用、美白作用、メラニン生成阻害能などを有する化粧料そ、頭皮頭髪用組成物、皮膚洗浄剤組成物、皮膚疾患治療用組成物等として極めて有用である。
背景技術
皮膚の活性は加齢等の内的環境のみならず、紫外線や各種の刺激等の外的環境からの影響も大きく受け、肌荒れ、皺、シミ、くすみ、その他のトラブルを起こしがちである。これらのトラブルの改善に、様々な皮膚活性化成分についての検討がなされてきた。中でも細胞賦活成分や美白成分については多くの研究が行われている。細胞賦活成分は、細胞の分裂を促し、新陳代謝を高め、身体を健全な状態に維持する効果を呈する。また、該成分は、皮膚においては表皮の角質細胞が活性化することにより、古い角質を剥離し、ターンオーバーと呼ばれる角化の過程を整え、肌を健康で美しい状態に改善する。
ケフィア(kefir)は、コーカサス地方の伝統的な乳発酵産物であり、ケフィア・グレインと呼ばれる種菌(ケフィア粒菌とも呼ばれる。)をスターターにして獣乳を発酵させて得られたヨーグルト状の飲料であった。このようなヨーグルト状の乳発酵産物の透明な液状部分を特にケフィアホエーと称する。ケフィアは水分をほとんど除いた乾燥状のものであってもよく、また水分を比較的多量に含むゲル状のものであってもよい。乾燥状のケフィアの製造方法としては特開昭62−83842号などに記載されており、またケフィアホエーの製造方法としては特開平2−219538号などに記載されている。本明細書では、これらの文献の記載をすべて取り込む。
さらに、最近ではケフィアの健康食品としてだけではなく、その優れた保健効果に着目して医薬用途への応用についての研究も盛んになってきている(特開平5−320069号公報、特開平6−211668号公報、特開平9−37711号公報、特開平9−191852号公報など)。これらの研究及び開発の多くは、ケフィアを含有した食品や医薬品を経口摂取することを目的としたものであった。
しかし、優れた効能にもかかわらず、元来、ケフィアは発酵乳飲料であり、独特の臭いとpH特性(約4.20〜4.40)などのため、経口摂取製品以外の用途、例えば皮膚外用剤、特に使用感が重視される化粧料などへの利用は、通常のヨーグルト製品と比べても困難なことであった。
一方、健康の概念は多様性を増し、精神及び身体のみならず、従来であれば美容の領域であった肌などの外見的な部分も含んだものとなってきている。このような広義の健康を保つために、即効性が期待される医薬品に加え、作用が穏やかでも安全性が高く日常生活に容易に取り入れることができる種々の製品が望まれている。
発明の開示
本発明者らは、こうした実情を鑑み、伝統的に健康を増強する食品として用いられてきたケフィア・グレインから得られた乳発酵産物の安全性、熱安定性と皮膚に対する細胞賦活作用などの効能に着目し、上記の欠点を克服して該乳発酵産物の特性を活かした本発明を完成するに至った。
すなわち、本発明は、ケフィア・グレインから得られた乳発酵産物、及び製薬的又は化粧学的に許容される担体を含有してなる皮膚外用剤組成物に関する。また、本発明は、さらに活性酸素除去成分、抗酸化成分、抗炎症成分、紫外線防止成分及び保湿成分よりなる群から選ばれる成分の一種又は二種以上を含有する化粧料組成物、頭皮頭髪用組成物、皮膚洗浄剤組成物などの皮膚外用剤組成物に関する。さらに、本発明は該乳発酵産物を含有してなる細胞賦活剤、皮膚老化防止剤、美白剤、又はメラニン生成阻害剤に関する。
また、本発明は、ケフィア・グレインから得られた乳発酵産物の有効量を、皮膚に適用することにより、皮膚の細胞を賦活し、皮膚老化を防止し、皮膚のメラミン生合成を阻害し、及び/又は皮膚を美白する方法に関する。本発明のこの方法の好ましい例としては、皮膚を化粧又は洗浄するための方法、頭皮及び/又は頭髪の化粧又は洗浄のための方法、皮膚疾患の患者の皮膚を治療するための方法などがある。
さらに、本発明は、ケフィア・グレインから得られた乳発酵産物の、皮膚外用剤組成物を製造するための使用に関する。
発明を実施するための最良の形態
本発明で用いられるケフィア・グレインから得られた乳発酵産物は、ケフィア・グレインという種菌で、乳酸と酵母など少なくとも5〜6種類の働きが併合している粒状の菌により、獣乳、豆乳及び脱脂粉乳等を発酵させて得られる。ケフィア・グレインを牛乳や山羊乳などの獣乳、豆乳、及び脱脂粉乳等の中に入れておくと乳酸菌による乳酸発酵と酵母によるアルコール発酵の両方が行われる。ケフィア・グレインによる乳発酵産物は、乳酸菌だけが発酵するヨーグルトと異なり、酵母の出す炭酸ガスにより、酸味と、かすかなアルコール風味に加え若干の刺激ある風味を呈する。また、該乳発酵産物には、乳糖、ペプチド、アミノ酸、乳酸、ビタミン、ミネラルなどの栄養分が豊富であり、身体に優れた効能をもたらしている。
本発明で用いられる乳発酵産物は、通常の獣乳を発酵させて得られるものでよい。獣乳としては、例えば牛、馬、羊、山羊、水牛などの乳が挙げられる。これらの獣乳から発酵産物を製造する方法は、特に限定されたものではなく、通常の牛乳を原料としてケフィア・グレインにより発酵させて製造する方法、東欧で行われている馬乳を用いて製造する方法などを採用することができるが、牛乳を原料としてケフィア・グレインにより発酵させて得られた乳発酵産物が特に好ましく用いられる(特開昭62−83842号公報参照)。
また、通常の豆乳や脱脂粉乳等を用いて、ケフィア・グレインから得られた乳発酵産物を製造することも可能である。
本発明のケフィア及びケフィアホエーは、上記した乳発酵産物のヨーグルト状のもの又はその乾燥成分をケフィアと称し、ヨーグルト状から得られる透明な液状の成分をケフィアホエーと称する。本発明で用いられるケフィア・グレインから得られた乳発酵産物の好ましい例としては、ケフィア又はケフィアホエーのどちらでもよく、また、両者の混合物であってもよい。
本発明で用いられるケフィア・グレインから得られた乳発酵産物の1種であるケフィアは、保湿効果を始めとし、紫外線防御効果、メラニン生成の抑制効果、活性酸素消去効果、細胞賦活作用などの様々な有用性を示す。熱に対しても安定性を示し、フィルター除菌などが可能なため、安定した皮膚外用剤、特に化粧料原料としての可能性を有する。但し、独特の酵母発酵臭、乳酸発酵臭を持ち、pHが4.20〜4.40となっているため、他の化粧料原料や化粧料担体との配合により、心地よい使用感及びケフィア本来の効果効能を発揮すると考えられる。
本発明で用いられるケフィアは、ヨーグルト状、あるいは該ヨーグルト状のものを凍結乾燥等で乾燥処理を施して得られる粉末状、又は該粉末状ケフィアをさらに加工成形した顆粒状のものであってよい。また、これらを水、緩衝液、アルコール類、油脂類等を用いて加工した液状のものであってもよい。
上記ケフィアを皮膚外用剤に用いる場合には、外用剤全重量中に0.01〜3.00重量%を含有させることが好ましく、特に1.00〜3.00重量%を含有させることが好ましい。
また、本発明で用いられるケフィアホエーは、前記した乳発酵産物のうちのヨーグルト状のものから得られる透明な液状の成分であり、乳清とも言われ、乳糖、ペプチド、アミノ酸、乳酸、ビタミン、ミネラルなどの栄養分に富んでおり、原乳の栄養分の約半分を含有するとされている。ケフィアホエーのうち、固形分は約6%で、残りが水分である。ケフィアホエーには、L−乳酸が通常のヨーグルトよりも多く含まれており、乳酸菌による抗菌作用が注目されている。また、ケフィアホエーは肌の角質細胞内にあるNMF(天然保湿因子)に構造が似ているため、皮膚の水分を保ち、pHを理想的な弱酸性に整えることもわかっている。さらに、保湿効果に優れているとともに抗アレルギー作用もあり、皮膚を外部からの刺激や雑菌から守るともいわれている。
だが、こうした特性にもかかわらず、ケフィアホエーは従来、廃材として取り扱われることがほとんどで、有効利用について検討されたのはごく最近になってからである。
本発明で用いられるケフィア・グレインによる乳発酵産物であるケフィアホエーは、上記ケフィアと同様に保湿効果を始めとし、紫外線防御効果、メラニン生成の抑制効果、活性酸素消去効果、細胞賦活作用など様々な有用性を示す。熱に対しても安定性を示し、フィルター除菌などが可能なため、安定した化粧料原料としての可能性を持つ。但し、独特の酵母発酵臭、乳酸発酵臭を持ち、pHが4.20〜4.40となっているため、他の化粧料原料や化粧料担体との配合により、心地よい使用感を提供することができ、本来の効果効能を発揮することができる。
上記ケフィアホエーを皮膚外用剤に用いる場合には、外用剤全重量中に0.01〜3.00重量%を含有させることが好ましく、特に0.01〜3.00重量%を含有させることが好ましい。
上記ケフィア・グレインから得られた乳発酵産物は、ケフィアであれ、ケフィアホエーであれ、上述したように、細胞賦活作用を有することから細胞賦活剤としての使用が可能であり、また、該作用及び紫外線防御効果、メラニン生成の抑制効果、活性酸素消去効果とも併せて、皮膚老化防止剤、美白剤としての使用も可能であり、当然のことながらメラニン阻害剤としての使用も可能である。
本発明の皮膚外用剤組成物は、医薬用に用いる場合には、ケフィア・グレインから得られた乳発酵産物及び製薬上許容される担体を含有してなる組成物であり、化粧料や洗浄剤用に用いられる場合いは、ケフィア・グレインから得られた乳発酵産物及び化粧学的に許容される担体を含有してなる組成物である。本発明の皮膚外用剤組成物は、本発明の効果を損なわない範囲内で、通常の化粧品、医薬部外品及び医薬品等の皮膚外用剤に用いられる他の任意の成分、例えば、活性酸素除去成分、抗酸化成分、抗炎症成分、紫外線防止成分及び保湿成分よりなる群から選ばれる成分の一種又は二種以上を含有することもできる。
本発明の皮膚外用剤組成物は、各種の担体、例えば、ワセリンやマイクロクリスタリンワックス等の炭化水素類、ホホバ油や鯨蝋等のエステル類、牛脂やオリーブ油等のトリグリセライド類、ステアリン酸やオレイン酸等の脂肪酸、グリセリンや1,3−ブタンジオール等の多価アルコール類、油分、湿潤剤、界面活性剤、防腐剤、香料、水、アルコール、その他の基剤、増粘剤等を必要に応じて、適宜配合して、常法により製造することができる。
本発明の皮膚外用剤組成物に含有される活性酸素除去成分としては、スーパーオキサイドディスムターゼ(SOD)、マンニトール、カロテノイド類、アスタキサンチン、ルチン及びその誘導体、ピリルビン、コレステロール、トリプトファン、ヒスチジン、クエルセチン、クエルシトリン、カテキン及びその誘導体、没食子酸及びその誘導体、グルタチオン及びその誘導体、オウゴン抽出物、イチョウ抽出物、ケイケットウ抽出物、サンザシ抽出物、マイカイカ抽出物、ユキノシタ抽出物、メリッサ抽出物、ゲンノショウコ抽出物、ボタンピ抽出物、パセリ抽出物、トルメンチラ抽出物、羅漢果抽出物、ヤシャジツ抽出物、ジコッピ抽出物、ステビア抽出物並びにエイジツ抽出物よりなる群から選ばれる一種または二種以上が挙げられる。
本発明の皮膚外用剤組成物に含有される抗酸化成分としては、マンニトール、ルチン及びその誘導体、クエルセチン、リン脂質、ピリドキシン類、フラボノイド類、ポリフェノール化合物、イチョウ抽出物、ユキノシタ抽出物、ビタミンA類及びそれらの誘導体、ビタミンB類及びそれらの誘導体、ビタミンC類及びそれらの誘導体、ビタミンD類及びそれらの誘導体、ビタミンE類及びそれらの誘導体、ジブチルヒドロキシトルエン並びにブチルヒドロキシアニソールよりなる群から選ばれる一種または二種以上が挙げられる。
本発明の皮膚外用剤組成物に含有される抗炎症成分としては、グリチルリチン酸及びその誘導体、グリチルレチン酸及びその誘導体、メフェナム酸及びその誘導体、フェニルブタゾン及びその誘導体、インドメタシン及びその誘導体、イブプロフェン及びその誘導体、ケトプロフェン及びその誘導体、アラントイン及びその誘導体、グアイアズレン及びその誘導体、ε−アミノカプロン酸、ジクロフェナクナトリウム、トラネキサム酸及びその誘導体、アシタバ抽出物、アルニカ抽出物、アロエ抽出物、イブキトラノオ抽出物、ウコン抽出物、オトギリソウ抽出物、オウバク抽出物、カミツレ抽出物、カンゾウ抽出物、キンギンカ抽出物、クレソン抽出物、コンフリー抽出物、ゴカヒ抽出物、サルビア抽出物、シコン抽出物、シソ抽出物、シラカバ抽出物、チャ抽出物、トウキンセンカ抽出物、ニワトコ抽出物、ホオウ抽出物、ムクロジ抽出物、ヨモギ抽出物、ユーカリ抽出物、トウキ抽出物並びにレンゲソウ抽出物、よりなる群から選ばれる一種または二種以上が挙げられる。
本発明の皮膚外用剤組成物に含有される紫外線防止成分としては、パラアミノ安息香酸(PABA)、PABAエチル、PABAグリセリル、N,N−ジメチルPABAアミル、N,N−ジメチルPABA−2−エチルヘキシル、サリチル酸−2−エチルヘキシル、サリチル酸エチレングリコール、サリチル酸ホモメンチル、4−メトキシケイ皮酸−2−エチルヘキシル、4−メトキシケイ皮酸エトキシエチル、4−メトキシケイ皮酸カリウム、4−5−ジイソプロピルケイ皮酸メチル、ジパラメトキシケイ皮酸モノ−2−エチルヘキサン酸グリセリル、2−ヒドロキシ−4−メトキシベンゾフェノン、2−ヒドロキシ−4−メトキシベンゾフェノンスルホン酸、2−ヒドロキシ−4−メトキシベンゾフェノンスルホン酸ナトリウム、2,2’−ジヒドロキシ−4,4’−ジメトキシベンゾフェノン、2,2’−ジヒドロキシ−4,4’−ジメトキシベンゾフェノン−5−スルホン酸ナトリウム、2−4−ジヒドロキシベンゾフェノン、2,2’,4,4’−テトラヒドロキシベンゾフェノン、2−(2−ヒドロキシ−5−メチルフェニル)−ベンゾトリアゾール、ウロカニン酸、ウロカニン酸エチル、4−t−ブチル−4’−メトキシ−ジベンゾイルメタン、酸化チタン、酸化亜鉛、酸化鉄、酸化セリウム並びに酸化ジルコニウムよりなる群から選ばれる一種または二種以上が挙げられる。
本発明の皮膚外用剤組成物に含有される保湿成分としては、ヒアルロン酸、コンドロイチン硫酸、デルマタン硫酸、ヘパラン硫酸、ヘパリン及びケラタン硫酸から選ばれるムコ多糖類及びそれらの誘導体;コラーゲン、エラスチン、フィブロネクチン、ケラチンから選ばれるタンパク質及びそれらの誘導体並びに加水分解物;グリシン、アラニン、バリン、イソロイシン、セリン、スレオニン、アスパラギン酸、グルタミン酸、アスパラギン、グルタミン、リジン、ヒドロキシリジン、アルギニン、システィン、シスチン、メチオニン、フェニルアラニン、チロシン、プロリン、ヒドロキシプロリン、テアニン、オルチニン、シトルリン、ピロリドンカルボン酸から選ばれるアミノ酸及びそれらの誘導体;ソルビトールエリスリトール、マルトース、マルチトール、キシリトール、キシロース、トレハロース、イノシトール、グルコース、ペンタエリスリトール、果糖、蔗糖及びそのエステル、デキストリン及びその誘導体から選ばれる糖類;ムチン、D−パンテノール及びその誘導体、尿素、リン脂質、糖脂質並びにセラミド、深層水、ハチミツ、黒砂糖抽出物、アロエ抽出物、海藻抽出物、マルメロ抽出物、ハマメリス抽出物、ヘチマ抽出物、ゼニアオイ抽出物、リンゴ抽出物、グレープフルーツ抽出物、プルーン抽出物、ライム抽出物、ユズ抽出物、ボダイジュ抽出物、キイチゴ抽出物、クララ抽出物、モッカ抽出物、小麦胚芽抽出物、インチンコウ抽出物、シラユリ抽出物、ホップ抽出物、ペパーミント抽出物、ドクダミ抽出物、シャクヤク抽出物、ヨクイニン抽出物、ラベンダー抽出物、カラスムギ抽出物、ブッチャーズブルーム抽出物、アルテア抽出物、ブクリョウ抽出物、イラクサ抽出物、ウイキョウ抽出物、キウイ抽出物、キュウリ抽出物、ブドウ抽出物、サボテン抽出物、ジオウ抽出物、スギナ抽出物、センキュウ抽出物、ソウハクヒ抽出物、タチジャコウソウ抽出物、マロニエ抽出物、モモ抽出物、バラ抽出物、アンズ抽出物、トウモロコシ抽出物、ショウガ抽出物、レモン抽出物、オレンジ抽出物、イチゴ抽出物、ゲンチアナ抽出物、ウスベニアオイ抽出物、サイシン抽出物、ゴボウ抽出物、ハウチマメ抽出物、イナゴマメ抽出物、キヅタ抽出物、マツ抽出物、ヤグルマソウ抽出物、ワレモコウ抽出物、オノニス抽出物、ゲンチアナ抽出物、アマチャ抽出物、フクタンポポ抽出物、クインスシード抽出物並びにグリセリン、1,3−ブチレングリコールよりなる群から選ばれる一種または二種以上が挙げられる。
本発明の皮膚外用剤組成物には、さらに、他の美白作用を有する成分及び細胞賦活作用を有する成分を適宜配合することもできる。
上記美白作用を有する成分としては、ビタミンC及びその誘導体並びにそれらの塩、胎盤抽出物、甘草抽出物、ヨクイニン抽出物、アスパラガス抽出物、エイジツ抽出物、キイチゴ抽出物、クジン抽出物、ケイケットウ抽出物、ゴカヒ抽出物、コメヌカ抽出物、小麦胚芽抽出物、サイシン抽出物、サンザシ抽出物、シラユリ抽出物、シャクヤク抽出物、大豆抽出物、緑茶、紅茶、ウーロン茶等の茶抽出物、糖蜜抽出物、ビャクレン抽出物、ブナの芽抽出物、ブドウ抽出物、ホップ抽出物、マイカイカ抽出物、モッカ抽出物及び羅漢果抽出物、オウゴン抽出物、海藻抽出物等が挙げられる。
また、上記細胞賦活作用を有する成分としては、ビタミンAおよびその誘導体、クエン酸、乳酸、酒石酸、リンゴ酸、グリコール酸、コハク酸、セリン、グルタミン酸、ヒドロキシプロリン、テアニン、ピロリドンカルボン酸、酵母抽出物、乳酸菌抽出物、ビフィズス菌抽出物、醗酵代謝抽出物等が挙げられる。これらの細胞賦活剤のうち、特に好ましいものとしては、ビタミンAおよびその誘導体、クエン酸、乳酸、リンゴ酸、セリン、ピロリドンカルボン酸が挙げられる。
また、本発明の皮膚外用剤組成物は、外皮に適用される化粧料、頭皮頭髪用組成物、皮膚洗浄剤組成物、医薬品、医薬部外品等に広く適用することが可能であり、その剤型も、皮膚に適用できるものであればいずれでもよく、溶液系、化粧水などの可溶化系、乳液及びクリームなどの乳化系、粉末分散系、水−油二層系、水−油−粉末三層系、分散液、軟膏、ゲル、エアゾール等、従来の皮膚外用剤に用いられている任意の剤型が適用される。これらを調製するにあたっては、使用する成分の種類及び分量は、常法に従って当業者が適宜定め得る範囲で調整することが可能である。
また、本発明の皮膚外用剤組成物の使用形態も任意であり、例えば化粧水、乳液、クリーム、パック等のフェーシャル化粧料やファンデーションの他、ファンデーション、おしろい、口紅、リップクリーム、その他のメーキャップ化粧料、特殊用途化粧料、身体用化粧料、日焼け止め又は日焼けケア用化粧料、頭皮及び毛髪用化粧料、芳香化粧料、洗顔料、頭髪用(頭皮用及び毛髪用)化粧料、身体用洗浄剤、頭髪用(頭皮用及び毛髪用)洗浄剤、髭剃り用化粧料、浴用化粧料等に用いることができる。
なお、本発明の皮膚外用剤が上記の剤型および使用形態に採り得る形態は、上記に限定されるものではない。
実施例
以下、試験例及び実施例により本発明をより具体的に説明するが、本発明はこれらにより何ら限定されるものではない。
ケフィア・グレインによる乳発酵産物が有する細胞に対する各種効果を調べるために、下記の試験を行った。各試験のために、まずサンプルの調製を行った。乳発酵産物ケフィアの固形物を遠心して沈殿物を除いた後に、0.45μmのフィルターに通し、ミニカラムNAP−25にかけ、分子量5000以上を素通り画分とし、5000以下をFr.1、2、3及び4に分画した。
試験例1:ケフィアの細胞賦活試験(1)
DNAは、DNA複製エラー及び組み換えにより常にミスマッチ変異を抱えている。このミスマッチ変異を修復するミスマッチ修復遺伝子(hMLH1、hPMS1、hPMS2、hMSH2、hMSH3、hMSH6)が知られている。これらのミスマッチ修復遺伝子が変異を起こすとゲノムが不安定化し、細胞が癌化する危険がある。
分子量5000以上とFr.3のサンプルをヒト悪性黒色種細胞株HIV−Iに添加して15時間培養した。ミスマッチ修復遺伝子であるhMLH1、hPMS1、hPMS2のmRNAの発現量をRT−PCR法によって解析したところ、添加した細胞では発現量の増加が確認された。結果を第1図〜第4図に示す。
試験例2:ケフィアの細胞賦活試験(2)
試験例1で調製したFr.3をヒト悪性黒色種細胞株HIV−Iに添加して15時間培養したミスマッチ修復遺伝子の1つhMLH1の蛋白の発現量をウェスタンブロッティング法によって解析したところ、ケフィアを添加した細胞では発現量の増加が確認された。結果を第5図及び第6図に示す。
試験例3:ケフィアのPCNA増加試験
PCNA(増殖細胞核抗原:proliferating cell nuclear antigen)は、細胞増殖や修復に大きく関わる分子であり、PCNAを欠損する細胞には修復機能に重大な欠陥があることが知られている。
Fr.3をヒト悪性黒色種細胞株HIV−Iに添加して15時間培養した。PCNAの蛋白の発現量をウェスタンブロッティング法によって解析したところ、ケフィアを添加した細胞では発現量の増加が確認された。結果を第7図及び第8図に示す。
試験例4:ミスマッチ修復遺伝子hMLH1の発現増強試験
(1)Fr.3、3mlを陰イオン交換樹脂カラムであるDEAE、3mlにかけ、吸着した物質をNaClで溶出し、脱塩してヒト悪性黒色種細胞株HIV−Iに添加して15時間培養したところ、ミスマッチ修復遺伝子hMLH1のmRNAの発現量を増強する効果がRT−PCR法によって確認された。溶出の手順は以下のとおり。

試験例5:ケフィアホエーのメラニン生成阻害能試験
ケフィア発酵液由来のケフィアホエーが持つ抗酸化作用がヒトメラノーマであるSK−MELのメラニン生成に与える影響について実験を行った。ケフィアホエーは培養細胞の系で用いるという観点から中和後に浸透圧を0.300osmol/kgに調製したサンプルを用いた。
<実験方法>
SK−MELを4×10cells/mlの細胞密度で5ml dishに播種し24時間培養した。前培養後、培地成分に対して、0%(コントロール)、0.1%、1%のケフィアホエーを含む培地に交換して更に24時間培養した。細胞はトリプシン処理によって回収し、全ての条件について総細胞数が3×10cellsとなるように希釈を行い、これを1.5ml容のチューブに回収した。これを遠心分離(5000rpm、10min)し、細胞のペレットに1M NaOH400μlを加えて、100℃で10分間煮沸した。溶解物を充分に混和させ、1条件につき100μlずつ3点を96wellプレートにのせ、プレートリーダーで405nmの吸光度を測定し、メラニン量を決定した。
<結果>
結果を表4及び第15図に示す。この結果から、ケフィアホエーを添加した培地でメラノーマを培養すると、明らかにメラニンの生成を抑えることがわかった。

試験例6 頭皮頭髪改善作用の試験
健常人20名(男性12名、女性8名)に乳発酵産物ケフィアを含んだ頭髪料を1ヶ月間試してもらった。毎日洗髪後、かゆいところを中心に地肌、髪の毛両方共に塗布して、毛髪のつや、頭皮のふけ、抜け毛、かゆみについて試験した。
結果は次の通りであった。

この結果、ケフィアを含有する頭髪料が、毛髪のつやを良くし、ふけを減らし、抜け毛を減らし、かゆみを防止できる頭皮頭髪用組成物として有効なことが判明した。
試験例7 乳発酵産物ケフィアを含有したジェル(商品名:ケフラン)を使用してもらった結果は次の通りであった。
(1)26才 女性: 今までほとんどの化粧水などは使う時に手にとり、指のアトピーの傷に付いた時、とびあがる程痛いので今回妹に勧められた時もまた痛いだろうなと心配していたんですが、全く痛くなくて、もちろん顔にも痛くなかったので、これから安心をして使っていけると思います。
(2)34才 女性: 2人目の子供を出産した後、アトピーになってしまい、肌がとても敏感になりました。年齢的にもマイルドな使い心地のものでは物足りなくなり何か良いものを探していたところに購入し使っております。とても良いです。
(3)23才 女性: 私はアトピー肌の為、なかなか自分に合った基礎化粧が見つからず、悩んでいましたけど、ケフランを使ってから肌の調子が非常に良くなりました。
(4)45才 女性: 私は、日光過敏症で、顔等はメイクや帽子等で日射しをさえ切れますが手の甲、腕は時々太陽にあててしまうと夕方頃よりトリ肌のようになり猛烈なかゆみにおそわれるのですが、気付いた時早め早めにケフランを塗っておくと回復が早く、ひどいかゆみも楽になります。

上記の試験より明らかなように、本発明による皮膚外用剤は、安全で肌の乾燥、シミ、はりに有効であり、皮膚の細胞活性を高めることがわかる。
産業上の利用可能性
ケフィア・グレインから得られた乳発酵産物は、保湿効果を始めとし、紫外線防御効果、メラニン生成の抑制効果、活性酸素消去効果など様々な有用性を示す。熱に対しても安定性を示し、フィルター除菌などが可能なため、安定した皮膚外用剤、特に化粧料原料としての可能性を有する。但し、独特の酵母発酵臭、乳酸発酵臭を持ち、pHが4.20〜4.40となっているため、他の化粧料原料との適当な配合により、心地よい使用感及びケフィア・グレインによる乳発酵産物本来の効果効能を発揮すると考えられる。

Kefir is a traditional milk fermentation product from the Caucasus region. <Preparation method>

As is clear from the above test, it can be seen that the external preparation for skin according to the present invention is safe and effective for dry skin, spots, and abscissions, and increases the cellular activity of the skin.
Industrial applicability
The fermented milk product obtained from kefir grains exhibits various usefulness such as a moisturizing effect, an ultraviolet protection effect, an inhibitory effect on melanin production, and an active oxygen scavenging effect. Since it exhibits stability against heat and can be sterilized with a filter, it has the potential as a stable external preparation for skin, particularly as a cosmetic raw material. However, since it has a unique yeast fermentation odor and lactic acid fermentation odor, and has a pH of 4.20 to 4.40, it can be comfortably used and milk with kefir grains by appropriate blending with other cosmetic ingredients. It is thought that the original effect of fermented products is demonstrated.
