Description
本比較例では、従来のオリゴヌクレオチドプローブの設計方法によってオリゴヌクレオチドを設計した。
本実施例では、前記比較例で考慮された事項に、標的部位に対応するヌクレオチドの5’末端側と3’末端側とのオリゴヌクレオチドのTmを計算し、これを比較してその差ができるだけ小さなオリゴヌクレオチドプローブを選択した。具体的には、比較例のように、全体プローブと標的試料とのハイブリダイゼーションシミュレーションを実施してTmが65℃〜80℃であるプローブを選択した後、標的部位、例えば、突然変異部位を基点として右側と左側とのTmの差が5℃以下であるものを選択する過程をさらに含むことを除いては、比較例と同一である。
本実施例では、前記比較例及び実施例1で製作されたオリゴヌクレオチドプローブ、すなわち、野生型標的核酸に相補的なプローブWP1、WP2、WP3及びWP4と、突然変異型標的核酸に相補的なプローブMP1、MP2、MP3及びMP4とをマイクロアレイで固定化し、ハイブリダイゼーション反応を行った後、その結果を分析した。
Claims (1)
- 標的部位に対応するヌクレオチドから5’末端側に、最初のヌクレオチドから5’末端ヌクレオチドを含むオリゴヌクレオチド配列(但し標的部位の塩基は含まない)、及びそれと相補的なオリゴヌクレオチド配列との融解温度(Tm)と、標的部位に対応するヌクレオチドから3’末端側に、最初のヌクレオチドから3’末端ヌクレオチドを含むオリゴヌクレオチド配列(但し標的部位の塩基は含まない)、及びそれと相補的なオリゴヌクレオチド配列との融解温度(Tm)との差を計算して、その差が0℃ないし5℃となるオリゴヌクレオチドプローブを選択する段階を含む、標的部位に対する結合特異性が改善されたオリゴヌクレオチドプローブを設計する方法。
