例えば、所望の結合力を有する抗体が表面発現されたファージを固定化された抗原と結合させて、溶出してファージを再び増殖させると、ターゲット抗体をコーディングする遺伝子をファージから確保することができる(米国特許第5837500号)。このようなパニング法は、抗体のライブラリーを大量にファージに表面発現することによりターゲット抗体を選別する手段を提供することができて、次のような段階を含む連続過程からなる:(1)ライブラリーを製造する段階;(2)ライブラリーの表面発現段階;(3)固定化された抗原と結合する段階;(4)結合されたファージを溶出する段階;最終的に(5)選別されたクローンを増殖する段階。
前述の表面展示技術は、抗原またはその一部を細胞表面に展示して再組み合わせ生ワクチンの伝達手段を提供するようになる。現在までのワクチンは、弱毒化された病原性細菌やウイルスを主に使用して、特に、バクテリアの場合は、抗原を細胞内、細胞膜または細胞外に分泌発現して宿主細胞に伝達した。一方、表面展示された生ワクチンは、非常に強力な免疫反応を示し、且つ宿主細胞内に増殖しながら持続的に抗原を発現することができるため、新しいワクチン伝達手段として注目を浴びている。特に、非病原性大腸菌やサルモネラ菌の表面に病原性由来の抗原エピトープを発現し、生きている状態のまま経口投与する場合は、さらに持続的且つ強力な免疫反応を示すと知られている(Georgiou et al., 1997; and Lee et al., 2000)。
化学的反応に使用できる酵素が表面に展示された全細胞を生物触媒として使用する場合は、酵素の直接発現、分離及び安定化などの過程が要らないという長所がある。生物転換に使用される酵素を細胞内に発現させる場合は、細胞を培養して回収した後、トルエンのような化学物質を使用して基質が透過できるようにする過程が必須的である。また、持続的に使用する場合、酵素が活性を失うか、基質と産物の物質伝達に問題が生じて、工程全体の生産性が低下される問題点がある。
Martineauらは、大腸菌の表面展示技術を用いて抗ペプチド抗体を生産する、非常に簡単な方法を報告した(Martineau et al., 1991)。この文献に開示された内容をみると、MalEと細胞外膜タンパク質であるLamBの表面突出部位に所望のペプチドを発現した後、全細胞または粉砕された細胞を動物に投与して抗ペプチド抗体の生成を誘導して、このような方法による場合は、化学的にペプチドを合成するか、またはこれを伝達タンパク質に付着せずとも、抗体を生産することができるようになる。
吸着クロマトグラフィーに使用される抗体やタンパク質を適当な担体に固定化するためには、醗酵によるタンパク質の生産、純粋な状態としての分離及び精製、そして担体への固定化過程を経なければならない。しかしながら、大抵の場合、このようなバイオ吸着剤は、その生産工程が単純ではない。
バシラス胞子を構成しているコートタンパク質または構造タンパク質(morphogen)以外に、細胞内で発現されるタンパク質が胞子の表面に結合して展示されることができるという事実はいまだに報告されていない。本発明者らは、バシラス胞子の表面が疎水性を帯びているため(Wiencek, K.M. et al., Appl. Environ. Microbiol., 56:2600-2605(1990))、リパーゼのように疎水性ドメインを有するタンパク質は(Brockerhoff H., Chem. Phys. Lipids, 10:215(1973))、胞子表面に疎水性結合により表面展示が可能になるという仮説を定立した。前記仮説を立証するために、精製されたPseudomonas fluorescensのリパーゼを純粋分離されたBacillus subtilis胞子と結合させた後、酵素活性を測定した。
宿主細胞から発現された野生形リパーゼが胞子表面に展示できるかどうかを次のように調査した:胞子表面展示のための野生形リパーゼ遺伝子を含むプラスミドpBS:lipA (Bell P.J.L. et al, Biotechnol. Lett., 21:1003-1006(1999))はオーストラリアのBergquist博士から得た。前記プラスミドを鋳型として序列1のプライマーlip1と序列2のプライマーlip2を使用してPCRを行った。重合酵素はBoehringer Mannheim社から購入したTaq重合酵素を使用して、アニーリングは55℃で30秒、延長反応は72℃で1分、変性は94℃で30秒間行って、総35サイクルを行った。
カルボキシルメチルセルラーゼの胞子表面展示のために、Bacillus thurigiensis 毒素タンパク質遺伝子cry1Aaのプロモーターの後にBacillus substilis BSE616菌から分離されたカルボキシルメチルセルラーゼ遺伝子(Park S.H. et al., Agric. Biol. Chem., 55:441-448(1991))をクローニングした。
一般に、タンパク質のN-末端分泌信号は、N-末端の端が2〜3個のカチオンを帯びるアミノ残基と後続の疎水性ドメインから構成されているが、前記カチオンアミノ酸は、細胞膜の陰イオン性燐脂質と結合することにより、タンパク質の分泌を手伝うと知られている(Tjalsma H., Microbiol. Mol. Biol. Rev.,64:515-547(2000))。さらに、前記カチオンアミノ酸を中性に置換させるとタンパク質の分泌が多少減少すると知られている(Chen M. and Nagarajan V., J. Bacteriol., 176:5796-5801(1994))。従って、本発明者らは、タンパク質の分泌を減少させる場合は、細胞内に存在する該当タンパク質の量が増加して、増加されたタンパク質は、N-末端の疎水性ドメインによる胞子表面との疎水性結合を通じて胞子表面展示がさらに容易にできるという仮説を定立した。このような仮説を証明するために、次のような実施例を行った。
バシラス胞子の表面は、実施例1に記載のように疎水性を帯びるが、アニオン性も帯びるものとして知られている(Nishihara T., et al., Microbiol. Immunol., 25:763-771(1981))。従って、本発明者らは、カチオンを帯びる目的タンパク質はイオン結合により胞子表面展示が起こり、目的タンパク質がカチオン性を有しなくても、カチオン性を帯びるモチーフと融合されることにより、胞子表面展示が可能になるという仮説を定立した。前記仮説を立証するために、次のような実施例を行った。
ファージコートタンパク質と結合できる目的タンパク質はファージ表面展示が可能であるということは、前記実施例から充分予測でき、このような事実は、下記のようにして確認できる。一方、ファージコートタンパク質と自然状態で結合できない目的タンパク質は、ファージコートタンパク質と結合できるモチーフと融合されると、ファージ表面展示が可能になる。
本発明から構築されたシステム、即ち、遺伝子担体の表面との相互作用により、発現された目的タンパク質の表面展示を可能にしたシステムを用いて、目的タンパク質の方向性進化ができる。まず、カルボキシルメチルセルラーゼをコーディングする遺伝子に対してのエラー誘発PCRをCadwell, R.C. and Joyce, G.F., PCR Methods Appl., 2:28-33(1992)に開示された方法により行う。PCR反応は、目的タンパク質遺伝子が含まれた前記実施例3のpCPaC3を鋳型として、カルボキシルメチルセルラーゼ遺伝子に特異的に結合するプライマーを使用する。これらプライマー0.3μMと鋳型 DNA5ngを反応溶液(10mM Tris(pH 8.3), 50 mM KCl, 7 mM MgCl2, 0.01% (w/v) ゼラチン)に入れて、これに0.2 mM dGTP, 0.2 mM dATP, 1 mM dTTP, 1 mM dCTP, 0.15 mM MnCl2, Taq重合酵素5Uを入れて総100μlの反応液を用意する。反応条件は、変性は94℃で30秒、アニーリングは50℃で30秒、延長反応は72℃で1分として、総13サイクルを行う。
有機溶媒上でリパーゼを用いた生物転換反応は、過去数年間報告されてきた(Zaks, A. et al., Proc. Natl. Acad. Sci. USA. 82:3192(1985); and Klibanow, A.M., CHEMTECH, 16:354(1986))。このような反応界では、酵素の不活性化無しに反応を行うことが必須的である。このために一般的にリパーゼの固定化を行った(Mustranta, A. Forssell et al., Enz. Microb. Technol.,15: 133(1993); and Reetz, M.T. et al., J. Biotechnol. Biogen., 49:527(1996))。前記報告によると、リパーゼを固定化した場合、遊離されたリパーゼに比べ有機溶媒上での酵素の安定性が増加されて、且つ合成反応も増加されたことを報告している。
スライドグラス表面にアルデヒド活性基がコーティングされているタンパク質アレイ用固体表面(BMS社、ドイツ)に、特定抗原に対しての単一クローン抗体が表面展示された遺伝子担体106〜109個を、自動化アレイ装置を用いて付着する。これは、遺伝子担体表面のタンパク質に存在するアミノ基とすライドグラス表面のアルデヒド基とが反応して、シッフ塩基(Schiff base)を形成することにより、固体表面に共有結合形態して付着されるのである。例え、付着された面では表面展示された目的タンパク質が固体表面に付着されて活性を喪失しても、表面に露出された遺伝子担体の表面に展示された目的タンパク質は一定な方向性を有することができる。
生体内で免疫反応を誘導できる抗原を遺伝子担体に表面展示させる場合には、これを用いて抗体生成を誘導することができる。
結合ドメインが表面展示された遺伝子担体を用いて混合物から特定物質を分離することができる。まず、結合ドメインをコーディングする遺伝子に対してのエラー誘発PCRをCadwell, R.C. and Joyce, G.F., PCR Methods Appl., 2:28-33(1992)に開示された方法により行う。PCR反応は、目的タンパク質遺伝子が含まれたプラスミド或いは染色体を鋳型として、目的タンパク質遺伝子に特異的に結合するプライマーを使用する。これらプライマー0.3μMと鋳型DNA 5ngを反応溶液(10mM Tris(pH 8.3), 50 mM KCl, 7 mM MgCl2, 0.01% (w/v) ゼラチン)に入れて、これに0.2 mM dGTP, 0.2 mM dATP, 1 mM dTTP, 1 mM dCTP, 0.15 mM MnCl2, Taq重合酵素5Uを入れて総100μlの反応液を用意する。反応条件は、変性は94℃で30秒、アニーリングは50℃で30秒、延長反応は72℃で1分として、総13サイクルを行う。

Claims (29)
- (a)胞子を含む宿主細胞を、疎水性であって、前記宿主細胞に由来するタンパク質分解酵素に対して耐性を有する目的タンパク質をエンコーディングする遺伝子と、宿主細胞内で活性なプラスミド pCry1-CMCase から得られるプロモーターとから製造される組み換えベクターにより形質転換する段階、
(b)前記形質転換された宿主細胞を培養して、胞子が宿主細胞内で形成される時期に目的タンパク質を発現する段階、及び
(c)前記発現された目的タンパク質が前記胞子表面との非共有結合を形成し、胞子表面に展示されるようにする段階を含むことを特徴とする、胞子の外表面に結合された目的タンパク質の製造方法。 - 目的タンパク質は、ホルモン、ホルモン類似体、酵素、酵素阻害剤、信号伝達タンパク質またはその一部、抗体またはその一部、単鎖抗体、結合タンパク質またはその一部、ペプチド、抗原、付着タンパク質、構造タンパク質、調節タンパク質、毒素タンパク質、サイトカイン、転写調節因子、血液凝固因子及び植物生体防御誘導タンパク質からなる群から選択されることを特徴とする請求項1に記載の方法。
- 前記結合タンパク質またはその結合ドメインは、抗体または抗体ドメインであることを特徴とする請求項2に記載の方法。
- 前記結合タンパク質またはその結合ドメインは、タンパク質分解酵素阻害剤、クラムビン(crambin)、エンテロトキシン、コノトキシン、アパミン、リゾチーム、リボヌクレアーゼ、チャリブドトキシン(charybdotoxin)、シスタチン、エグリン、オボムコイド、アズリン(azurin)、腫瘍壊死因子及びCD4からなる群から選択されることを特徴とする請求項2に記載の方法。
- 前記結合タンパク質は、モノマーまたはマルチマーであることを特徴とする請求項2に記載の方法。
- 前記目的タンパク質は、胞子との非共有結合を改善するために変形されたものであることを特徴とする請求項1に記載の方法。
- 前記目的タンパク質は、(i)目的タンパク質のアミノ酸序列の一部を除去する方法、(ii)目的タンパク質または前記(i)の方法によりアミノ酸序列の一部が除去された目的タンパク質と胞子との非共有結合を手伝う他のオリゴペプチドまたはポリペプチドと融合させる方法、(iii)目的タンパク質を位置特異的に突然変異させる方法、及び(iv)目的タンパク質を無作為突然変異させる方法により変形されたものであることを特徴とする請求項6に記載の方法。
- 前記目的タンパク質のアミノ酸序列の一部を除去する方法は、目的タンパク質のN−末端部位の序列の中でイオン性アミノ酸序列を除去して実施されることを特徴とする請求項7に記載の方法。
- 前記融合されるオリゴペプチドは、陽イオン性ペプチドであることを特徴とする請求項7に記載の方法。
- 前記胞子は、目的タンパク質との非共有結合を増加させるために、その表面タンパク質が変形されたものであることを特徴とする請求項1に記載の方法。
- 前記胞子は、(i)胞子と目的タンパク質との非共有結合を手伝う他のオリゴペプチドまたはポリペプチドを胞子の表面タンパク質と融合させる方法、(ii) 胞子表面タンパク質を位置特異的に突然変異させる方法、及び(iii)胞子表面タンパク質を無作為突然変異させる方法により変形されたものであることを特徴とする請求項10に記載の方法。
- 前記胞子を含む宿主細胞は、胞子を形成するグラム陰性菌、胞子を形成するグラム陽性菌、胞子を形成する放線菌、胞子を形成する酵母及び胞子を形成するカビからなる群から選択されることを特徴する請求項1に記載の方法。
- 前記胞子を形成するグラム陽性菌は、クロストリジウム(Clostridium)属、パエニバシラス(Paenibacillus)属及びバシラス(Bacillus)属からなる群から選択される微生物であることを特徴とする請求項12に記載の方法。
- 前記バシラス(Bacillus)属の微生物は、Bacillus subtilis、Bacillus thuringiensis及びBacillus megateriumからなる群から選択される微生物であることを特徴とする請求項13に記載の方法。
- 前記プラスミドpCry1p-CMCaseは図5又は図7の配列地図を有することを特徴とする請求項1に記載の方法。
- 前記宿主細胞は、表面展示のために発現された目的タンパク質の分解に係わる細胞内タンパク質分解酵素または細胞外タンパク質分解酵素を生産できないように変形されたものであることを特徴とする請求項1に記載の方法。
- 前記方法は、非共有結合を通じて目的タンパク質の胞子表面展示が成された後、物理的方法、化学的方法及び生化学的方法により、目的タンパク質と胞子表面間との間に共有結合を形成して、目的タンパク質が胞子表面により安定に結合されるように処理する段階を追加的に含むことを特徴とする請求項1に記載の方法。
- 前記共有結合を形成するために処理する方法の中で、化学的方法はグルタルアルデヒドの処理であり、物理的方法は紫外線の処理であり、生化学的方法は共有結合の形成を手伝う酵素の処理であることを特徴とする請求項17に記載の方法。
- 胞子を含む宿主細胞を、目的タンパク質をエンコーディングする遺伝子と、プラスミドpCry1p-CMCaseから得られるプロモーターから製造される組み換えベクターとにより形質転換して製造されることを特徴とする形質転換微生物。
- 前記宿主細胞は、前記目的タンパク質をエンコーディングする遺伝子から発現された目的タンパク質の分解に係わる細胞内タンパク質分解酵素または細胞外タンパク質分解酵素を生産できないように変形されたものであることを特徴とする請求項19に記載の形質転換微生物。
- ホルモン、ホルモン類似体、酵素、酵素阻害剤、信号伝達タンパク質またはその一部、抗体またはその一部、単鎖抗体、結合タンパク質またはその一部、ペプチド、抗原、付着タンパク質、構造タンパク質、調節タンパク質、毒素タンパク質、サイトカイン、転写調節因子、血液凝固因子及び植物生体防御誘導タンパク質が、胞子の表面に、請求項1に記載の方法により展示されていることを特徴とする胞子−目的タンパク質の複合体。
- 前記目的タンパク質は、(i)目的タンパク質のアミノ酸序列の一部を除去する方法、(ii)目的タンパク質または前記(i)の方法によりアミノ酸序列の一部が除去された目的タンパク質と胞子との非共有結合を手伝う他のオリゴペプチドまたはポリペプチドと融合させる方法、(iii)目的タンパク質を位置特異的に突然変異させる方法、及び(iv)目的タンパク質を無作為突然変異させる方法により変形されたものであることを特徴とする請求項21に記載の胞子−目的タンパク質の複合体。
- 前記胞子−目的タンパク質複合体は、非共有結合を通じて目的タンパク質の胞子表面展示が成された後、物理的方法、化学的方法及び生化学的方法により、目的タンパク質と遺伝子担体表面との間の結合を安定させる追加的な共有結合を有することを特徴とする請求項21に記載の遺伝子担体−目的タンパク質の複合体。
- 前記プラスミドpCry1p-CMCaseが図5又は図7の配列地図を有することを特徴とする請求項21に記載の複合体。
- 前記胞子は、遺伝学的方法、化学的方法及び物理的方法からなる群から選択される一つ以上の方法により生殖不可能に変形されたものであることを特徴とする請求項21乃至23のいずれか一つの項に記載の胞子−目的タンパク質の複合体。
- 前記胞子生殖を不可能にする遺伝学的方法は、宿主細胞の胞子生殖に関与する遺伝子の欠乏により実施されることを特徴とする請求項25に記載の胞子−目的タンパク質の複合体。
- 前記胞子は、物理的方法、化学的方法及び遺伝学的方法からなる群から選択される一つ以上の方法により、凝集性が増加されるように変形されたことを特徴とする請求項24に記載の胞子−目的タンパク質の複合体。
- 前記目的タンパク質は、モノマーまたはマルチマーであることを特徴とする請求項21に記載の胞子−目的タンパク質の複合体。
- 非ヒト脊椎動物内での抗原に対しての抗体の生成方法において、前記方法は、前記請求項21乃至23のいずれか一つの項に記載の胞子−目的タンパク質複合体の免疫学的有効量を含む組成物を脊椎動物に投与する段階を含むことを特徴とする抗体の生成方法。
