Description
本発明は、組織学的試験用の組織サンプルを包埋および提供するための方法および装置、より詳細には、顕微鏡試験用の組織サンプルを調製する方法に関する。
パラフィン中に組織を包埋し、ミクロトームを用いてパラフィン包埋組織を薄層に切り、スライド上に乗せることによって顕微鏡試験用の組織サンプルを調製する手順は、当該分野で周知である。包埋に先立って、組織は、実施される特定の試験に従って選択される種々の溶液中で前処理される。典型的には、パラフィン包埋前に、組織サンプルは、固定、脱水、透明化(cleared )、溶融パラフィンでの浸透、試験に依存して染色される。
本発明の第一の目的は、スライド上に乗せるための組織サンプルを処理する方法を提供することである。本発明のさらなる目的は、切断するための組織を調製する方法であって、組織を構成する細胞が実質的に均等に組織調製物およびそれ由来の組織切片中に分布される方法を提供することである。
貯蔵および切断のためにサンプリングした組織を調製する方法は、図1に図式で示される。組織サンプル10は、組織を好適な流動性培地、すなわち一連の流動性培地11に浸して組織を脱水および/または透明化することにより固体または半固体支持体(例えば、アガロースまたはパラフィン)に包埋するために調製される。次いで、脱水された組織は、ある体積の溶融パラフィン12に移動され、細胞株を含み得る組織は、細胞分散手段13(例えば、超音波器または機械的攪拌器)により該体積の溶融物中で断片化され、分配される。
Claims (4)
- (a)組織の一部をある体積の流動性包埋培地に置く工程、次いで
(b)該一部を構成する細胞が該体積の流動性包埋培地中で実質的に均一に分布されるまで該一部を粉砕する工程、次いで
(c)該体積の流動性包埋培地の一部を管状部材の軸ボアに導入する工程、次いで
(d)該軸ボア中の該流動性包埋培地を凝固させて円柱状のプラグを形成する工程;次いで
(e)該円柱状のプラグの一部を該軸ボアから押出し、該円柱状プラグの一部を切り離して、切り離し部分を形成する工程、次いで
(f)該円柱状プラグの切り離し部分を硬い支持体基材に乗せる工程
を含む、顕微鏡試験用の細胞を含む組織の一部を調製するための方法。 - 前記組織が細胞株である請求項1記載の方法。
- 同じおよび/または異なる前記流動性包埋培地を少なくとも2つのボアホールへ導入し、少なくとも2つの形成されたプラグを押出し、該少なくとも2つのプラグを含む新しいプラグを形成する、請求項1または2記載の方法。
- (a)組織の一部をある体積の流動性包埋培地に置く工程、次いで
(b)該一部を構成する細胞が該体積の流動性包埋培地中で実質的に均一に分布されるまで該一部を粉砕する工程、次いで
(c)該体積の流動性包埋培地の一部を管状部材の軸ボアに導入する工程、次いで
(d)該軸ボア中の該流動性包埋培地を凝固させて円柱状のプラグを形成する工程;次いで
(e)該円柱状のプラグの一部を含む該管状部材の一部を該管状部材から切り離し、切り離し部分を形成する工程、次いで
(f)該管状部材および円柱状プラグの切り離し部分を硬い支持体基材に乗せる工程
を含む、顕微鏡試験用の細胞を含む組織の一部を調製するための方法。
