Description
(a) 試料をAT結合パートナーを含む第1の試薬R1とAT結合パートナーが本質的にATとは相互作用しないが、阻害因子とは相互作用をする条件下で、接触させる
(b) AT結合パートナーの遊離のフラクションの1回目の測定のための第2の試薬R2を添加する
(c) AT結合パートナーがATと相互作用する条件を変化させる第3の試薬R3を添加し、AT結合パートナーの遊離のフラクションの2回目の測定を実施する、そして
(d) AT結合パートナーの遊離のフラクションの1回目と2回目の測定の相違から、試料中のAT量を決定する。
(a) AT結合パートナーを含む第1の試薬R1
(b) 遊離のAT結合パートナーを測定するための第2の試薬R2、及び
(c) ATとAT結合パートナー間の相互作用促進剤を含む第3の試薬R3であって、該第3の試薬R3が第1の試薬R1と隔離されている、前記第3の試薬R3。
Lepirudinの存在下におけるアンチトロンビンIIIの測定
1. 反応スキーム
3 μlの試料溶液を測定キュベット内にピペッティングした。175 μlの試薬R1をピペットで加えた。試薬R1は100 mM Tris-HCl, 270 mM NaCl, 12mM EDTA, 10 g/l ポリエチレングリコール6000、1g/l ウシ血清アルブミン、0.5 NIH/ml ウシトロンビンからなり、適当量のフィブリン重合阻害剤、例えば、GPAP, pH 8.10を含む。次に、5分間インキュベートし、75 μlの試薬R2(Chromozym TH 1.9 mM)を加えた。次に、415 及び 700 nm (一次及び二次波長) における連続二色測定(continuous bichromatic measurement)による速度論的試験で1回目のトロンビン活性を測定した。
Claims (3)
- 以下の工程を含む、阻害因子を含む可能性がある試料中のアンチトロンビンIII(AT)を検出する方法:
(a) 試料をAT結合パートナーを含む第1の試薬R1とAT結合パートナーが本質的にATとは相互作用しないが、阻害因子とは相互作用をする条件下で、接触させる
(b) AT結合パートナーの遊離のフラクションの1回目の測定のための第2の試薬R2を添加する
(c) AT結合パートナーがATと相互作用する条件を変化させる第3の試薬R3を添加し、AT結合パートナーの遊離のフラクションの2回目の測定を実施する、そして
(d) AT結合パートナーの遊離のフラクションの1回目と2回目の測定の相違から、試料中のAT量を決定する。 - アンチトロンビンIII(AT)結合パートナーとAT間の相互作用の測定に基づいて試料中のATを検出する方法であって、AT結合パートナーの1回目の測定がAT相互作用なしに実施され、それに続く2回目の測定がAT相互作用のもとで実施され、試料中のAT量が1回目と2回目の測定の相違から決定されることを特徴とする、前記方法。
- 以下を含む、請求項1又は2記載の方法の使用による試料中のアンチトロンビンIII(AT)の定量的検出用試薬キット:
(a) AT結合パートナーを含む第1の試薬R1
(b) 遊離のAT結合パートナーを測定するための第2の試薬R2、及び
(c) ATとAT結合パートナー間の相互作用促進剤を含む第3の試薬R3であって、該第3の試薬R3が第1の試薬R1と隔離されている、前記第3の試薬R3:であって、
試薬R1が、試料と、AT結合パートナーが本質的にATとは相互作用しないが、阻害因子とは相互作用をする条件下で、接触せしめられ、
試薬R2が、AT結合パートナーの遊離のフラクションの1回目の測定のために添加され、かつ、
試薬R3が、AT結合パートナーの遊離のフラクションの2回目の測定を実施する前に、添加されることを特徴とする、前記キット。
