Description
本発明は、微細システム技術と微細構造形成のための新規薄層に関し、この技術分野において本発明の薄層は種々の方法で使用することができる。

微細システム技術や微細構造形成に関する現在の技術水準にあっては、膜、接触層システム、伝導路層システム等を製作するために従来用いられてきた薄層は、SiO2、Si3N4、またはAl2O3からなる使用頻度の高い層や、金属層あるいは金属層システム等の無機層の使用に基づくものである。その場合、希望する構造を作り出すために、例えば強酸、アルカリ液、酸化剤等の健康に悪影響を及ぼす有毒な化学薬品を使用したり、反応性のイオンエッチングやプラズマエッチング等の法外なコストのかかる工程を経なければならない(B▲u▼ttgenbach,Mikromechanik,B.G.Teubner Stuttgart,1994を参照)。このような構造形成プロセスにおいて使用されている唯一の有機層がフォトレジストであるが、構造を付与すべき層に対して所望の構造を付与した時点で、このフォトレジストは原則として再び除去される。

本発明は、従来汎用されてきた層より容易かつ安価に製造でき、既存の微細構造形成技術を利用することができ、特に有利な方法で物質ライブラリーを作成するのに適した薄層を提供するという課題をその根底におくものである。

前記の課題は、本発明の代表的特徴によって解決される。その有利な実施形態については、各請求項に詳述するとおりである。

微細構造形成分野において従来使用されてきた、フォトレジスト層の様な構造付与可能マスキング層は、それが存在するだけで、例えばゼラチン、アガロース、デキストロース、及び脂質等のバイオポリマーフィルムの性質に対して局部的な影響を与え得ることが知られている。さらに驚くべきことに、このバイオポリマーフィルムは、薄層技術にとって重要な30nm〜3μmの層厚でも非常に高品質を有するものが製造可能なことが見出されている。また、特定の色素物質や、光によって活性化され得る基は、光化学的に活性化させることによって、バイオポリマー層の架橋反応を引き起こし、露光しなかった層領域よりも、バイオポリマー層の酵素分解を明らかに緩徐化するということが見出されている。

バイオポリマー層の層厚は、自体公知のスピンコーティング法を利用して、固形物質含量1〜30%の溶液から数10nm程度のものとすることができる。前記の方法で製造した層は、予期しなかったほど極めて薄い層領域においても均質であり、欠陥が認められない。また前記の層は、その後行われる微細構造形成プロセスにおいて必要になることの多い250℃までの加熱工程においても変性現象を起こすことなく、問題のない状態を保持し続ける。

しかしながら、前記バイオポリマー層の主要な長所は、酵素分解が可能であるという点にあり、高度に特異的な分解が結果的にもたらされ、一般に、室温という、またpHが特に4〜9の溶液中という緩和な条件下で分解が行われる。基本的に、バイオポリマーは他の分子や層と共有結合したり、非共有結合したりすることによって特定の機能を示すという長所を有し、例えばアミノ基、カルボキシ基、チオ基を介して、あるいは水素架橋結合によって、ゼラチン層と結合させることができる。さらに、バイオポリマーに架橋反応を行わせて(例えば遊離のケト基、アミノ基、水酸基が存在する場合には、グルタルジアルデヒドを使用して)、結果的に、製造された層の性質を適切に変化させることも問題なく可能である。例えば水溶性ゼラチンに交差架橋反応を行わせると、非水溶性物質に変化し、この非水溶性物質に分子を封入させることも可能であり、分子を共有結合または非共有結合させることも可能である。本明細書に記載する薄層は、その実施形態に応じて、予定の用途において、多彩な目的のために利用することができる。その応用の可能性の詳細については、以下の実施例で説明する。

第1の実施例においては、特異性の高い触媒としての酵素の性質が利用されるが、それは、ゼラチンからなるバイオポリマーの薄層を利用して、カンチレバータイプのノボラック構造を作り出すためである。そのためには、洗浄したシリコンウェーハの表面に、回転滴下法(スピンコーティング)を利用して、約200nm厚さのゼラチン層をコーティングする。例えば、この層は、ゼラチンを水に溶解させ(10%v/v)、グルタルジアルデヒドを5%の割合で添加したものから形成させることができる。このようにして得られる層は非水溶性で、慣用の感光性樹脂現像剤には侵されない。前記の層の表面に、同様にスピンコーティングを利用して、市販のフォト反転レジストを被覆させる。このフォトレジスト層は、規定通りに処理を行い、最終的に希望する構造に対応してマスクを取り付け、露光して、構造を付与する。前記の層状体全体を酵素槽容器内に入れる。バイオポリマー層にゼラチンを使用している場合には、主に10%のSDS、10mMのNaCl、10mMのEDTAならびにTris−HClよりなる混液に10mg/mlの割合でプロテアーゼKを添加したプロテアーゼK緩衝液が好ましい酵素浴である。この酵素浴のpHを8.5に調節する。このような酵素槽を使用することにより、室温で約8時間以内に、約200nm厚さのゼラチン層の完全分解が行われる。この実施例においてバイオポリマー層は、カンチレバータイプのノボラック構造を作成するための犠牲的な層として利用される。

第2の実施例においては、第1の実施例に基づいて作成した、ゼラチンとフォトレジストよりなる層状体から出発して、適当なインヒビターまたは競合物質によって酵素機能が阻害される、特異性の高い触媒としての酵素の性質を利用する。例えば、AZ−フォトレジストにおいて汎用されている感光性コンポーネントとしてのジアゾナフトキノンは、ゼラチンのOH基と結合して、緩衝液に添加されているプロテアーゼの分解能を阻害する。露光を行うと、ジアゾナフトキノンはカルボン酸に変換されて、塩の形でゼラチンから溶出してしまうので、露光部分においては分解が妨げられることなく行われる。したがって、この場合の分解は、従来の選択的なエッチング技術である「アニソトロープ」と同等といえる。インヒビターが相変わらず存在したままの部分においては、分解速度が明らかに低下する。このようにして、微細システム技術コンポーネントとしても、マスキング材料としても、バイオポリマーとそれぞれに見合った分解酵素または調節酵素を使用するという可能性が与えられる。

第3の実施例においては、バイオポリマー薄層に感光性の添加物(例えばジアゾナフトキノン)を混和することによって、それ自体をフォトレジストとして使用することのできる層を作り出すという可能性が存在する。そのためには、感光性の添加物自体が分解酵素に対してインヒビターとして作用するか、あるいは、分解酵素のインヒビターと結合するということが前提条件となる。このような実施形態の可能性が開けることにより、酵素によって現像することの可能なフォトレジストを作成することができる。

第4の実施例としては、約100nm厚さの脂質層の製法について言及される。ここでは、ホスフォチジルエタノールアミンのクロロホルム溶液(0.1g/ml)を5000回転/分という回転速度で、適当な基板の表面に30秒間回転滴下する。

第5の実施例としては、ゼラチンに、例えばTFPI(外因系凝固インヒビター)等の、プロテアーゼ機能に対する生物由来のインヒビターを1:1000の重量比で添加したゼラチン薄層に、純粋なゼラチンよりなる薄層を重層させるという可能性が存在する。こうすることにより、後から酵素処理を行う場合にエッチングを停止させることができる。これと同様に、アガロース、デキストロース、又は脂質よりなる層においても、後から使用する酵素に対して阻害的に作用する物質を選択することによって、分解停止を生じさせることができる。

以上、記載した実施例は、現在の技術水準に基づく既存のポジ型フォトレジストの同等物として使用することのできる薄層の実施形態に関するものであるが、下記の第6の実施例においては、ネガ型レジストと同等な、本発明に基づく薄層について記載する。

可視スペクトル領域または紫外スペクトル領域で感光性を示す光活性成分としては、水に溶解させた、約345nmで感光度が最大になる、例えばジアジドスチルベンを使用する。この場合に特に好ましいのは、4,4−ジアジドスチルベン−2,2−スルホン酸のナトリウム塩であり、この物質を1:50〜1:100の混合比で水に溶解させる。このジアジドスチルベン水溶液に、製造すべきバイオポリマー層の希望する架橋度に応じて、10:1〜100:1の混合比のもとに、例えばブタ皮膚由来の高品質Bloom60のような固形ゼラチンを加える。その際には、例えば1ミリリットルのジアジドスチルベン溶液に約40mgのゼラチンを加える。ゼラチンが溶解した時点で、200nmのミクロフィルターで溶液を濾過する。このようにして得られた溶液を用いて、ミクロリソグラフィーの際と同様に、例えば金属、重合体、シリコン、コーティングを施したシリコンまたはガラス等の基板の表面にスピンコーティングを行う。例えば、シリコン基板の表面においては、約100nm厚さの平坦で均質なゼラチン層が形成される。この薄層に、最終的に希望する構造に適合させたマスクを取り付け、波長360nmの紫外線照射を約400秒間続ける。その際には、ジアジドスチルベンのアジド基が光化学的に開裂し、窒素原子が外れてビスナイトレンラジカルが形成されて、露光部分のゼラチンにおいて、アミノ酸分子鎖の架橋化が行われる。露光ゼラチン層の現像は水の中で行うが、すでに1〜2分後にはゼラチン層の非露光部分がほぼ溶解してしまう。その後に再現像を行うが、その際には、例えば10%のナトリウムドデシルサルフェート、10mMのNaCl、10mMのEDTA、およびTris−HClよりなる緩衝液に、プロテアーゼを添加した濃度0.1mg/mlのプロテアーゼ緩衝液を用い、pHは約8.5の弱アルカリ性とする。その際に達成させることのできる分解率は例えば20〜30nm/分である。上記の方法により、下方へ1μmまでの構造サイズの達成が可能である。

本発明の枠内で達成される露光領域の交差架橋反応という性質からいって、第1〜第5の実施例に基づいて作成されるポジ構造より、むしろ安定な構造にすることが可能である。

バイオポリマーは他の分子との共有結合や非共有結合によって特定の機能を実現させるという長所を有しているために、例えばアミノ基、カルボキシル基、水酸基、またはチオ基を介して、あるいは、水素架橋結合によってゼラチン層に結合させることが可能であって、また、以下に記載するように、上記の方法に従って適切な構造に作成した薄層構造を利用する場合には特に、前記の長所を享受することができる。

上記の方法に従って作成したゼラチン層から出発して、例えばゴバン目状にしつらえたマスクを載せ、上記と同様な方法で露光と現像を行うと、基板の表面に正方形を規則正しく配列させることができる。正方形の一辺の長さを、例えば16μmとする場合には、前記の現像工程が完全に行われた後における正方形構造体の高さは18nmになる。ゼラチンや類似のコラーゲンよりなる、結合部位をあらかじめ規定しておいた前記の微細構造(パッド)は、スクリーニングテスト用として、あるいは、物質ライブラリー用として特に適しており、バイオ技術、分子生物学、薬学、または医学において、分子レベルで相互作用を現すパートナーを迅速に見つけ出すことが可能になる。微細構造のゼラチンパッドには、ペプチド結合によって、タンパク(例えば抗体)や、適切に修飾を施したオリゴヌクレオチドを容易に結合させることができるので、例えばテストアッセイを行うことができる。

構造性のゼラチンパッドによって、局所的な反応空間が規定される。コラーゲンポリマーの官能基は化学的に多彩なために、特定の活性化を行った後に、種々の分子を結合させることができる。このことはとりもなおさず、多彩な官能基を有する分子を結合させるのに、同じマトリックスを利用することができるということであり、分子を固定させるための手間のかかる修飾という工程を省略することができる。
Claims (21)
- 薄層が酵素分解可能なバイオポリマーから形成されていて、前記薄層の層厚が30nm〜3μmであ り、前記酵素分解可能なバイオポリマー薄層が、部分的 に、または全表面において、インヒビターまたは競合物 質を具備していることを特徴とする微細システム技術と微細構造形成のための新規薄層。
- 薄層が酵素分解可能なバイオポリマーから形成されていて、前記薄層の層厚が30nm〜3μmであ り、前記酵素分解の可能なバイオポリマー薄層に、容積 と合致したインヒビターまたは競合物質が浸透していることを特徴とする微細システム技術と微細構造形成のための新規薄層。
- 前記酵素分解の可能なバイオポリマー薄層に、感光性添加物が添加されていることを特徴とする請求項1又は2に記載の新規薄層。
- 前記感光性添加物がジアゾナフトキノンによって形成されていることを特徴とする請求項3に記載の新規薄層。
- 前記バイオポリマー薄層がゼラチンによって形成されていることを特徴とする請求項1〜3のいずれか1項に記載の新規薄層。
- 薄層が酵素分解可能なバイオポリマーから 形成されていて、前記薄層の層厚が30nm〜3μmであ り、前記バイオポリマー薄層がアガロースによって形成 されていることを特徴とする微細システム技術と微細構 造形成のための新規薄層。
- 薄層が酵素分解可能なバイオポリマーから 形成されていて、前記薄層の層厚が30nm〜3μmであ り、前記バイオポリマー薄層がデキストロースによって 形成されていることを特徴とする微細システム技術と微 細構造形成のための新規薄層。
- 薄層が酵素分解可能なバイオポリマーから 形成されていて、前記薄層の層厚が30nm〜3μmであ り、前記バイオポリマー薄層が脂質によって形成されて いることを特徴とする微細システム技術と微細構造形成 のための新規薄層。
- 前記バイオポリマー薄層がアガロースによって形成されていることを特徴とする請求項1〜3のいずれか1項記載の新規薄層。
- 前記バイオポリマー薄層がデキストロースによって形成されていることを特徴とする請求項1〜3のいずれか1項記載の新規薄層。
- 前記バイオポリマー薄層が脂質によって形成されていることを特徴とする請求項1〜3のいずれか1項記載の新規薄層。
- バイオポリマー原料物質としての固形物質の含量が1〜30%の溶液からなる前記バイオポリマー薄層が、スピンコーティングによって、予定されている層厚領域のもとに塗布されることを特徴とする請求項1〜11のいずれか1項記載の新規薄層。
- 前記酵素分解の可能なバイオポリマー薄層が熱活性化試薬を添加することによって架橋反応、特に交差架橋反応を示すことを特徴とする請求項1〜12のいずれか1項記載の新規薄層。
- 前記熱活性化試薬として主にグルタルジアルデヒドまたはホルムアルデヒドを使用し、そしてバイオポリマーを含む溶液に1〜5%の割合で添加することを特徴とする請求項12および13記載の新規薄層。
- 光活性化添加物または物質群として分子鎖を延長させたり架橋反応を生じさせたりする働きのあるラジカル形成物質が添加されることを特徴とする請求項3記載の新規薄層。
- 前記ラジカル形成物質が240〜550nmの波長域で強い感光性を示すという基準に基づいて選択されることを特徴とする請求項15記載の新規薄層。
- 前記ラジカル形成物質としてジアジドスチルベンを塩基とする1種類の物質が使用されることを特徴とする請求項15および16記載の新規薄層。
- 前記バイオポリマー層が希望の架橋度に応じて10:1〜100:1の混合割合のラジカル形成物質と可溶性の固形ゼラチンとの水溶液から形成されていることを特徴とする請求項15〜17記載の新規薄層。
- 前記ジアジドスチルベンとして4,4−ジアジドスチルベン−2,2−スルホン酸のナトリウム塩が使用されることを特徴とする請求項17または18記載の新規薄層。
- 基板の表面に塗布され、ジアゾナフトキ ノンまたはジアジドスチルベン色素物質が添加されてい る前記のバイオポリマー層を構造付与性の紫外光に曝露 し、且つ非露光部分を水溶液及び酵素緩衝液で除去し、 残された構造をテストアッセイとして、又は物質ライブ ラリーの作成のための使用であって、薄層が酵素分解可 能なバイオポリマーから形成されていて、前記薄層の層 厚が30nm〜3μmであることを特徴とする微細システム 技術と微細構造形成のための新規薄層の使用。
- 基板の表面に塗布され、ジアゾナフトキノンまたはジアジドスチルベン色素物質が添加されている前記のバイオポリマー層を構造付与性の紫外光に曝露し、且つ非露光部分を水溶液及び酵素緩衝液で除去し、残された構造をテストアッセイとして、又は物質ライブラリーの作成のために使用することを特徴とする請求項 1〜19のいずれか1項記載の新規薄層の使用。
