本発明は皮膚老化防止効果(表皮細胞の増殖、分化の促進、角質層のターンオーバー促進、角質改善、保湿効果、美肌効果など)に優れた化粧料に関する。

【従来の技術及び課題】
老化皮膚では、表皮及び真皮の細胞活性の低下が見られ、そのため角質層の機能低下による乾燥肌や真皮繊維芽細胞の機能低下による弾力性のない肌になる。このような、老化皮膚の活性を回復させるために、細胞賦活剤の探索が行われ、その結果、カバノキ科のカバノキ属に属するシラカンバ、ハンノキ属に属するハンノキの抽出エキスに表皮細胞の増殖を顕著に促進し、角質層の機能を改善する作用があることが開示されている。(特開平6−263627号公報)
また、特開平5−139947号公報には、α−ヒドロキシ酸類も表皮のターンオーバーを促進したり、皮膚柔軟性を向上させるなど老化症状の防止、改善や肌荒れ症状の防止、改善する作用があることが開示されているが、α−ヒドロキシ酸類の配合量を多くすると安定な製剤化が困難になったり、皮膚刺激性が出現するなどの問題がある。 従ってより有効で安全性の高い皮膚老化防止用化粧料の開発が望まれていた。

本発明者らは、皮膚老化防止効果を改善、向上させるべく鋭意研究を重ねたところ、遂に、カバノキ科のカバノキ属に属する植物、又はハンノキ属に属する植物の抽出エキスと炭素数が2〜4のα−ヒドロキシ酸およびその誘導体から選ばれる1種以上を配合することにより、皮膚老化防止効果(表皮細胞の増殖、分化の促進、角質層のターンオーバー促進、角質改善、保湿効果、美肌効果など)が相乗的に改善、向上することを見出し、本発明の完成に至った。

【発明の実施の形態】
本発明に用いるカバノキ科カバノキ属植物抽出物は、例えばシラカンバ(Betula platyphylla var.japonica)、コウアンシラカンバ(Betula platyphylla var.platyphylla)、シセンカンバ(Betula platyphylla var.szechanica)、ヨーロッパシラカンバ(Betula pendula)、ケカンバ(Betula pubescens)、トルトーサカンバ(Betula pubescens var.tortuosa)、イベリアカンバ(Betula certiberica)、べトゥラ・パピリフィラ(Betula papyrifera)、べトゥラ・ケナイカ(Betula kenaica)、べトゥラ・ポプリフォリア(Betula populifplia)等からの抽出物である。また、同科のハンノキ属植物抽出物は、例えばハンノキ(Alnus japonica)、エゾハンノキ(ヤチハンノキ)(Alnus japonica var.arguta)、サクラバハンノキ(Alnus trabeculosa)、ネパールハンノキ(Alnus nepalensis)、ケヤマハンノキ(Alnus hirsuta)、ヤマハンノキ(Alnus hirsuta var.sibirica)、ヤハズハンノキ(Alnus matsumurae)、コバノヤマハンノキ(タニガワハンノキ)(Alnus inokumae)、カワラハンノキ(Alnus serrulatoides)等の抽出物である。これらの植物の内、特にシラカンバ(Betula platyphylla var.japonica)、コウアンシラカンバ(Betula platyphylla var.platyphylla)、ヨーロッパシラカンバ(Betula pendula)が好ましい。

これらの抽出物は、該植物の葉、木部、樹皮等いずれの部位でも使用でき、中でも樹皮が好ましい。抽出溶媒は特に限定されないが、ベンゼン、エチルエーテル、クロロホルム、酢酸エチル、酢酸ブチル、アセトン、メタノール、エタノール又は1、3−ブチレングリコール、及び水等が例示でき、特にエタノール、水が好ましい。これらの抽出溶媒は1種又は2種以上の混合溶媒を用いることもできる。抽出操作は常法により行われ、抽出液をそのままで用いることができ、また抽出液を濃縮した軟エキス、さらに蒸発乾固した抽出末などの形状で配合できる。

これらの抽出物の配合量は、化粧料全量に対し0.001〜10重量%、特に0.01〜5重量%が好ましい。配合量が0.001重量%未満であると皮膚老化防止効果が充分に発揮できない。また、10重量%を越えて配合しても、それ以上の皮膚老化防止効果の向上が認められない。

本発明に用いる炭素数2〜4のα−ヒドロキシ酸類は、グリコール酸、乳酸、リンゴ酸、酒石酸などが挙げられる。これらのαーヒドロキシ酸類は、その1種以上配合することができ、その配合量は化粧料全量に対して0.01〜15重量%である。配合量が0.01重量%に満たないと老化防止効果が期待できず、また、15重量%を超えて配合しても、それ以上の効果が期待出来ないばかりか、皮膚激が生じ好ましくない。

本発明の皮膚化粧料は、常法によりクリーム、化粧水、美容液、パック、洗浄料など通常の化粧料の形態で供することができ、特に化粧水、美容液などの非乳化製剤が好ましい。さらに、本発明では、それぞれの形態に応じて、本発明の効果を損なわない範囲において適宜公知の基剤、色素、香料、防腐剤、界面活性剤、抗酸化剤等を配合することができる。

2.美肌効果
荒れ肌、子じわ、乾燥肌等を訴える女子被験者(30〜50代)20人に実施例1〜2、比較例1〜3のローションを1日2回(朝、夕)連続3ヶ月塗布して、3ヶ月後の効果を測定した。測定には、適用部位のレプリカを用いた画像解析法を用いた。
測定方法を示す。それぞれの被験者について、実施例(比較例)適用部位のレプリカを作成、これらをマイクロスコープ(HI−SCOPE MODEL KH−2200 MD2:HIROX製)により観察し、画像解析装置(AVIO EXCEL:NIPPON AVIONICS製)に取り込んだ画像を統計解析した。
美肌効果は、一定面積内のキメの数をラベリングしてあらわした。ここでは、一定面積内のキメ(画像上で皮溝に囲まれた領域)の増加率(%)が大きいほど美肌効果がある。
