Description
を含む組換えDNA配列及びそのような配列を含む発現ベ
クターまたはプラスミド、並びにそのようなDNA配列を
含有するかまたは示されたようなベクター若しくはプラ
スミドにより形質転換されるグラム陽性菌細胞に関す
る。本発明はさらに、グラム陽性菌細胞の同定または選
択的単離方法も包含する。
造体の使用を含む全くの新規概念に基づく新規方法を包
含する。これらの新規方法により多くの興味深い用途が
発見された。本発明の2つの主態様の一つ目は治癒的ま
たは防御免疫であり、二つ目はグラム陽性菌細胞の同定
または選択的単離の実際的な方法である。
サブユニットワクチン調製物に優れた免疫原性を示すこ
ともあるため、組換え免疫原の生送達系(live deliver
y system)の開発が非常に注目されてきた。多くの生組
換え弱毒化ウイルスが外来エピトープのキャリヤとして
試されて来た。これらの例としては、ワクシニアウイル
ス[Mossら,Nature 311,67−69(1984)]、アデノウイ
ルス[Ballayら,EMBO J.4,3861−3865(1985)]、ポリ
オウイルス[Evansら,Nature 339,385−388(1989)]
及びヘルペスウイルス[Shihら,Proc.Natl.Acad.Sci.US
A 81,5867−5870(1984)]が挙げられる。例えば、Sal
monella[Hosieth及びStocker,Nature 291,238−239(1
981)]、非定型ミコバクテリア[Jacobsら,Nature 32
7,532−535(1987)]及びE.coli[O'Callaghanら,Res.
Microbiol.141,963−969(1990)]などの生組換え細菌
を使用する細菌系も開発されており、この系では細菌全
体を組換え免疫原のキャリヤとして使用している。
物またはin vitro免疫感作したリンパ球から直接抗体遺
伝子を単離及びクローニングできるようになった(Huse
ら,Science,1989,246,1275−1280)[Borrebaeckら,Pro
c.Natl.Acad.Sci.USA 1988,85,3995−3999]。抗体能力
(repertoire)の遺伝子ライブラリーは細菌ベクター系
で確立され得、単離した免疫グロブリン遺伝子のin vit
ro操作が容易になった。
菌宿主中で次の発現をコードする遺伝子の『ランダム』
結合(combination)により、結合特性をスクリーニン
グし得るVH/VL対が新しく形成される[Huseら,Science,
1989,246,1275−1280]。しかしながら、この方法を使
用して産生した多くのクローンには、実際の方法で関連
クローンを単離し得る効率的なスクリーニング法が必要
である。近年、表面に露出した免疫グロブリンフラグメ
ントのキャリヤとして細菌ファージを使用する方法が記
載され、これにより所望の抗原を結合し得るVH/VLドメ
インの結合を保持する単一ファージ粒子を選択し得るよ
うになった(McCaffertyら,1990,Nature,348,552−55
4)。
異的に単離するための新規方法の重要性は、ホルモン−
ホルモンレセプター認識[Bassら,Proteins:Structure,
Function and Genetics,8,309−314(1990)]及び酵素
−基質混和性[Carterら,Proteins:Structure,Function
and Genetics,6,240−248(1989)]などの分野にも関
連する。
リンセグメントを含ませるための構造的強制により、負
の生物学的選択、続いてVH/VL結合の理論的能力が欠損
し得る。さらに、各ファージ粒子に露出した少数、1〜
約5個の分子の免疫グロブリン分子により、ファージ粒
子の低い親和性による温和な結合能を持つ結合の回収に
関して克服できない問題が発生し得る。

B領域と強く反応したが、M3ペプチドに対する反応性は
示され得なかった。従って、抗血清R102はブドウ球菌の
表面上でBBの検出に好適であった。
アッセイ 異なる構築物を含有する細胞を、クロラムフェニコー
ル(20mg/l)を補ったブイヨン中で37℃で一晩生長させ
た。細胞をPBS中で2回洗浄した。15−ウエルマルチテ
ストスライド(Flow laboratories)を、室温で30分湿
潤チャンバ内でコーティング緩衝液(15mM Na2CO3,35mM
NaHCO3,pH9.6)とインキュベートした。コーティング
緩衝液はPBS中細菌1滴(107細菌/ml)でディスプレイ
し、スライドを室温で30分、湿潤チャンバ内でインキュ
ベートした。未結合細菌をPBSで洗浄し、細胞1層をPBS
中1%グルタルアルデヒドで2秒間固定した。最後にス
ライドを蒸留水中で洗浄し、風乾し、−20℃で貯蔵し
た。ウサギ抗血清をPBS中1:1000に希釈し、各ウエルに
1滴ずつ加え、室温で30分湿潤チャンバ内でインキュベ
ートした。PBSで4回洗浄後、スライドをビオチン化抗
ウサギIgG−分子(15μg)と30分インキュベートし、P
BS中で再び洗浄後、30分のインキュベーション間にアビ
ジン−結合フルオレセインイソチオシアネート(FITC)
(50μg/ml)(Vector,USA)を添加した。最後に、スラ
イドを洗浄し、臭化エチジウムを添加して細菌DNAを可
視化し、UV−顕微鏡下で調べた。

−7500(1986)]により、ブドウ球菌タンパク質A(SP
A)に関する遺伝子を、SPAの転写、翻訳及び分泌シグナ
ルにより先行された、合成二価IgG−結合ドメイン、ZZ
をコードするプラスミドpEZZ318T[Nygrenら,J.Molec.R
ecogn.1,69−74(1988)]から制限されたNot I−Nde I
遺伝子フラグメントと置き換えた。得られたプラスミド
pSZZmp18Tは、E.coli及びStaphylococcus aureusの両方
に関して複製のオリジンを含んでいた。SPAのIgG−結合
領域A,B及びCと、細胞壁拡張領域X及び膜固定領域M
(図1)をコードする遺伝子フラグメントは、Hind III
及びEcoR Vを使用してプラスミドpSpA8[Uhlnら,J.B
iol.chem.259,1695−1702(1984)]から制限し、同一
酵素で予め制限したプラスミドpSZZmp18T中のmp18マル
チクローニング部位[Yanisch−Perronら,Gene33,103−
119(1985)]の下流に挿入した。得られたベクターをp
SZZmp18ABCXMと命名した。このプラスミドはHind III及
びPst Iで消化し、SPAの領域Mの半分と、領域A、B、
C及びXをコードする遺伝子フラグメント欠失する。領
域X及びMの完全配列は、ポリメラーゼ鎖反応(PCR)
法を適用することにより修復され得る。上流プライマー
としてSTST34、下流プライマーとしてSTST33及びDNA鋳
型としてプラスミドpSpA8を使用してPCR増幅を実施し
た。上流プライマーは、その非アニーリング5'配列によ
りHind III認識配列を産生し、下流プライマーはSPAの
M領域中の元のPst I認識配列に重複した。従って、PCR
で増幅したフラグメントはHind III及びPst Iで制限さ
れ得、同一酵素で予め制限されたプラスミドpRIT28[Hu
ltmanら,Nucleos.and Nucleot.7,629−638(1988)]に
サブクローンし、プラスミドpRIT28XMを産生した。PCR
でサブクローンしたフラグメントのヌクレオチド配列
を、固相DNA配列決定により確認した[Hultmanら,Nucl.
and Res.17,4937−4946(1989)]。pRIT28XMのHind II
I−Pst I制限により、SPAの領域Mの半分と領域Xをコ
ードする遺伝子フラグメントを単離し得、Hind III−Ps
t Iで消化したプラスミドpSZZmp18ABCXM(上述)に融合
すると、mp18マルチクローニング部位のM下流とin fra
me且つ完全領域XをもつプラスミドpSZZmp18XMが得られ
た。プラスミドpSZZmp18XMのNot I−EcoR I消化によ
り、ZZコード遺伝子フラグメントをプラスミドpB1B2mp1
8から制限されたNot I−EcoR Iフラグメントで置き換え
得 J.Immunol.Meth.124.,43−52(1989)]、SPAの転写、
翻訳及び分泌シグナルにより先行されたBBと命名され
た、連鎖球菌タンパク質Gの血清アルブミン結合領域を
コードする。得られたベクターpSBBmp18XM(図2A)はE.
coli及びStaphylococcus aureusの両方に関する複製の
オリジンを含んでいた。通常の発現ベクターpSBBmp18XM
中のmp18マルチクローニング部位をEcoR I−Hind III制
限により除去した。反復性の高いペプチドM3をコードす
る遺伝子フラグメント Gene 89,187−193(1990)]をプラスミドpRIT28M3 Gene 89,187−193(1990)]から切り出した。まず、M3
配列を停止する停止コドンを特定部位の固相in vivo突
然変異[Hultmanら,Nucl.Acids Res.18,5107−5112(19
90)]により除去した。M3をコードする、EcoR I−Hind
IIIで制限された遺伝子フラグメントを同様に消化した
pSBBmp18XMに連結させて、プラスミドpSBBM3XMを産生し
た(図2A)。M3ポリペプチドは、マラリヤ血液段階抗体
Pf155/RESA[Berzinsら,Proc.Natl.Acad.Sci.USA 83.10
65−1069(1986)]の免疫原性の高いC−末端部分から
誘導する。
ド、連鎖球菌タンパク質Gから誘導した血清結合BB部分
及びSPA由来の細胞壁結合XM領域を含む3部分からなる
誘導タンパク質をコードする。細胞膜を介する分泌時
に、シグナルペプチドが切断される。プラスミドpSBBM3
XMは、マラリヤ抗原ペプチドM3がBBとXM領域の間に配置
される3部分からなる融合タンパク質をコードする(図
2B)。
al xylosus(詳細は、以下の「出発物質」参照)から製
造したプロトプラストに形質転換される。表1に示され
ているように、BBまたはM3の各々に特異的なポリクロー
ナルウサギ抗血清を使用するイムノアッセイから、プラ
スミドpSBBmp18XMを含有するS.xylosus細胞は細胞表面
にBBを発現し(図3参照)、pSBBM3XMを含む細胞は細胞
表面にBBとM3の両方を発現し(図4)、この方法により
組換え融合タンパク質を発現する際に分泌シグナルと細
胞壁結合部分、XMの両方が機能的であることを示してい
る。プラスミドを含まないS.xylosus細胞は、BB及びM3
特異的抗血清の両方に各々陰性であり、前免疫血清は全
ての場合で陰性であった(図5)。
48,552−554]のH[pHERAT]及びL[pLERAT]鎖の変
異性ドメインを含有するプラスミド鋳型pHERAT及びpLER
AT上のオリゴヌクレオチドプライマー対KS1/2及びKS3/4
を各々使用するPCR増幅により、2種類の変異性ドメイ
ンをコードする遺伝子フラグメントを単離し得た。プラ
イマーを含んだ好適な制限酵素認識部位Eco R I及びBam
H Iを使用することにより、フラグメントをpRIT28に挿
入し、固相配列決定用に改造した。
及びpRIT28−VLを別個に、オリゴヌクレオチドプライマ
ー対KS6/2[pRIT28−VH]及びKS3/5[pRIT28−VL]各々
を使用する続くPCR増幅中の鋳型として使用した。得ら
れたPCR産生物の約5ナノグラムを混合し、85℃に加熱
し、その後室温に冷却した。Taqポリメラーゼ[Perkin
Elmer corp.]0.5単位、PCR緩衝液を添加後、標準PCRを
2回実施し、二重鎖DNAを得た。この方法により、KS5及
びKS6オリゴヌクレオチドによる2回目のPCRの際に取り
込まれた重複配列によって2個の免疫グロブリンコード
遺伝子フラグメントが結合した。結合DNA配列は、2個
の免疫グロブリンドメイン間に柔軟性の高い15アミノ酸
残基結合ペプチドをコードする。得られた730塩基対フ
ラグメントは、式: NH2−VL−リンカー−VH−COOH に記載の如く抗リゾチーム抗体D1.3[図6]の単一鎖Fv
[ScFv]をコードし、scFvコードフラグメントをさらに
クローニングするために十分な量を得、外部プライマー
KS3及びKS2を使用してさらにPCRを20回実施した。得ら
れたPCR産物を制限酵素Eco R I及びBam H Iで制限し、
続いてクローニングベクターpUC19に連結した。配列を
確認後、正しく組み立てられたscFv遺伝子フラグメント
を含むクローンをEco R I及びBam H I制限し、730塩基
対フラグメントをE.coli発現ベクターpRIT24のEco R I
及びBam H I部位に挿入した[Hammerbergら,Proc.Natl.
Acad.Sciences,USA,86,4367−4371]。得られた構築物p
RIT24−scFcは3部分からなる融合ZZ−scFv−BBをコー
ドする。pRIT24−scFvで形質転換したE.coli細胞を、ア
ンピシリン[100ml/l]を補ったトリプシン大豆ブイヨ
ン+酵母抽出物中、30℃で一晩生長させた。
学的活性を調査するために、一晩発酵させた培地を、ヒ
ト血清アルブミン[HSA]及び鶏卵卵白リゾチーム[HE
L]セファロースカラムを各々通過させた。0.5M HAc/NH
4Ac pH2.8で溶出したタンパク質を凍結乾燥し、SDS−PA
GEで分析した。HSA−及びHEL−親和性精製物質の両方に
関する主要バンドは、完全長であることが知見された。
HELを用いるZZ−scFv−BBの好結果のアフィニティー精
製から、scFv免疫グロブリンフラグメントは2個の親和
性末端(tail)ZZ及びBBによりフランクされるが、自然
の生物学的に活性な構造に折り畳み(fold)得ることが
示唆されている。
球菌細胞の両方に複製し得るシャトルベクターpSBBmp18
XMの構築である。scFvフラグメントの挿入に関してこの
ベクターを適用するために、mp18リンカーをM13mp8から
誘導した短いmp8リンカー[Messingら,1982,Gene 19,26
9−276]と置換して、pSBBmp8XMを産生した。scFcをコ
ードする遺伝子フラグメントをEco R I及びBam H I制限
によりUC19−scFcプラスミドから放出し、続いてpSBBmp
8XMベクターに連結した。

製用にクロラムフェニコール[20mg/l]を補ったTBS中
で37℃で一晩生長させた。形質転換したブドウ球菌細胞
から調製した、pSBB−scFv−XM構築物の制限酵素マッピ
ングは、予想した結果と一致した[図8]。このこと
は、pSBB−scFv−XM構築物がブドウ球菌宿主内で遺伝子
的に安定であることを示している。

−末端繰り返し配列、VSICSNNPTCWAISKNの3個のコピー
を含むペプチドG3をコードする遺伝子を、実施例1に記
載のM3ペプチドの構築に関して記載された重合概念に従
って、オリゴヌクレオチド: を使用して構築し、pRIT28Eに挿入したpRIT28EG3を産生
した。G3をコードする遺伝子のヌクレオチド配列を、固
相DNA配列決定[Hultmanら,(1989),Nucl.Acids Rec.
17:4937−4946]により確認した。G3遺伝子フラグメン
トをEcoR I及びHind IIIでpRIT28EG3から切り出し、同
様に消化したpBB2mp18 (1989),J.Imm.Meth.124:43−52]に連結した。得られ
たベクター、pBBG3[5153bp]は、連鎖球菌タンパク質
G[SPG]由来の血清アルブミン結合領域とトリペプチ
ド繰り返し部分からなる、BBG3[30.9kDa]の融合タン
パク質をコードする。pBBG3プラスミドを含有するE.col
i細胞をアンピシリン[100mg/l]を補った500mlトリプ
シン大豆ブイヨン[30g/l]中、37℃で一晩生長させ
た。融合タンパク質を、Nygrenら[J.Mol.Recognit.1:6
9−74]に従ったHSA−セファロース上のアフィニティー
クロマトグラフィーによりpariplasmicスペース及び培
地から精製した。
遺伝子に関する記載通り、固相部位特異的突然変異によ
りG3配列を停止する停止コドンを除去後、EcoR I及びHi
nd IIIで制限したpRIT28EG3から回収した。制限フラグ
メントを同様に制限したpSBBmp18XMに連結すると、プラ
スミドpSBBG3XM[図10]が得られた。プラスミドpSBBG3
XMは、SPA由来のシグナルペプチド、SPGから誘導した血
清アルブミン結合BB領域、RSV抗原性ペプチドG3及びSPA
由来の細胞壁結合XM領域を含むテトラペプチド誘導タン
パク質をコードする。

ら調製したプロトプラストに形質転換し、細胞を一晩生
長させた。実験の開始時に、6週齢のメスのネズミOFI4
匹[IFFACREDO,France]に各々、経口的に、pSBBG3XMプ
ラスミドを含有する一晩成長させた培地由来の1010S.xy
losus細菌[改良Neubauer counting chamberを使用して
顕微鏡で計測]を、3週間、続いて43日後に第2期の3
週間に、各火曜日、水曜日、木曜日及び金曜日に投与し
た。21、28、35及び63日目に血液を個々に採集し、ElIS
Aアッセイでコーティング抗原として精製BBG3タンパク
質を用いて抗BBG3抗体の存在を試験した。ミクロ滴定プ
レートをBBG3の1.25μg/ml溶液で一晩被覆し、次いでPB
S中の1%スキムミルクで2時間飽和させた。免疫感作
したネズミからの血液サンプルを装填し、インキュベー
ション後に濯ぎ、結合抗体を色素産生細菌アルカリ性フ
ォスターゼ基質と一緒に抗マウスIgG−アルカリ性フォ
スターゼ結合体[Sigma Inc.試薬No.A1902]を使用して
検出し、405nmでモニターした。試験は、負の対照とし
て0日に採取した血清で3回実施し、ウサギ抗BBG3ポリ
クローナル血清を正の対照として使用した。図11に示さ
れた結果は、処置63日間に4匹全部の動物でBBG3特異的
免疫応答が発達したことを示している。
Claims (12)
- 【請求項1】グラム陽性宿主内において作動可能なシグ
ナルペプチドとして機能する第1のペプチドもしくはタ
ンパク質をコードする第1のDNAフラグメントを含む組
換えDNAであって、該第1DNAフラグメントが、グラム陽
性菌の表面上に天然には認められない第2のペプチドも
しくはタンパク質をコードする第2のDNAフラグメント
に機能的に結合しており、該第2DNAフラグメントが、細
胞壁スパンニング(cell wall spanning)および膜アン
カリング(membrane anchoring)配列としてグラム陽性
宿主内で作動可能な第3のペプチドもしくはタンパク質
をコードする第3のDNAフラグメントに機能的に結合し
ており、該第2ペプチドもしくはタンパク質が抗原とし
て機能し得るか、または組換え表面レセプター構造体と
成り得ることを特徴とする前記組換えDNA。 - 【請求項2】前記第2DNAフラグメントが、ワクチンとし
て機能し得るかまたは抗体産生を誘導し得るアミノ酸配
列をコードする、請求項1に記載の組換えDNA。 - 【請求項3】前記第3DNAフラグメントが、ブドウ球菌タ
ンパク質Aまたはブドウ球菌タンパク質Gの細胞壁スパ
ンニングおよび膜アンカリング領域をコードする、請求
項1または2に記載の組換えDNA。 - 【請求項4】前記第1DNAフラグメントがグラム陽性菌細
胞由来である、請求項1乃至3のいずれか1項に記載の
組換えDNA。 - 【請求項5】前記第3DNAフラグメントが、ブドウ球菌タ
ンパク質Aの細胞壁スパンニングおよび膜アンカリング
領域をコードする、請求項1乃至3のいずれか1項に記
載の組換えDNA。 - 【請求項6】前記第1DNAフラグメントがブドウ球菌タン
パク質Aのシグナルペプチドをコードする、請求項5に
記載の組換えDNA。 - 【請求項7】請求項1乃至6のいずれかに記載の組換え
DNAを含み、グラム陽性菌宿主内で複製することができ
る発現ベクターまたはプラスミド。 - 【請求項8】請求項1乃至6のいずれかに記載の組換え
DNAを有するか、または、請求項7に記載のベクターも
しくはプラスミドによって形質転換されたグラム陽性菌
細胞。 - 【請求項9】異なった組換え表面レセプター構造体を有
する細胞の異種集団から請求項8に記載のグラム陽性菌
細胞を選択的に単離または同定する方法であって、前記
異種細胞集団を、1種類の特異的組換え表面レセプター
構造体を有する細胞の同定および/または単離が可能な
特異的に相互作用する相手と相互作用させる工程を含む
方法。 - 【請求項10】前記レセプター構造体が抗体またはその
活性フラグメントで構成されている請求項9に記載の方
法。 - 【請求項11】固定形態にある前記の相互作用する相手
を使用して前記特異的構造体を有する細胞を単離する請
求項9または10に記載の方法。 - 【請求項12】前記の相互作用する相手が固体支持体上
に固定されている請求項11に記載の方法。
