的には、F(ab′)2フラグメントの還元切断によって

ロブリンのペプシン消化によって作製できる。Fab抗体
フラグメントは、還元条件下での無傷の免疫グロブリン
のパパイン消化、または完全な免疫グロブリンの注意深

療的に有効な量の毒素または薬剤と複合されている抗体
または抗体フラグメントを非経口的にそのような腫瘍ま

明にしたがって作成された毒素もしくは薬剤の複合抗体
もしくは抗体フラグメントを使用することは明らかな改
善をもたらすであろう。
オール基をもつ複合体の毒素部分の免疫原性の減少を伴
う。実施例は、デキストラン、多糖類、ポリエチレング
リコール(PEG)などを含む。

−F(ab′)2−ペプチド−PEGをジチオエリトリトー
ルで還元し、生じたPEG−ペプチド−LL−2−Fab′SHを
精製する。
(mPE)を大腸菌で発現させ、ペリプラズムから抽出
し、イオン交換クロマトグラフィーで精製する。mPEを
活性化処理用のSMCCで処理して、活性化(Activate)す
る(AmPE)。従って、AmPEは、スクシンイミジル4−N
−マレイミドメチル−1−カルボキシレートで活性化処
理した活性化mPEの意味である。
温し、治療剤であるLL−2−PEG−Fab′−SdPEを得る。
なお、ここにいうSdPEは、P(シュードモナス外毒素)
の誘導体(dPE:derivatized PE)と抗体のFc部位のフリ
ーなスルフヒドリル基との結合体を意味する。これをゲ
ルろ過クロマトグラフィーで精製する。
よる広範囲の骨髄浸潤のために、放射性免疫療法による
治療にはふさわしくないように思われる。患者を静脈輸
液によって実施例1の複合体で処置し、リンパ腫の完全
な寛解が認められる。
ロープタンパク質、gp120のアミノ末端に存在するアミ
ノ酸構造を有する合成ペプチドで免疫する。gp120と反
応する単クローン性抗体を産生するハイブリドーマは、
通常の方法を用いる免疫マウスの脾臓リンパ球由来であ
る。この単クローン性抗体を異なるHIV分離物に対する
反応性について調べる。全分離物に対する高い親和性を
示すMAb(mB1)を抗HIVピューロマイシン複合体の作製
に使用するために選択する。mB1を産生するハイブリド
ーマクローンのmRNAを分離し、重鎖および軽鎖可変領域
のCDRの配列を決定する。その後組換え体DNA技術を用い
て、この重鎖および軽鎖の高可変領域をM13ベクターで
クローニングしたヒトIgG重鎖および軽鎖のcDNAにCDR移
植する。可変領域をコードする遺伝子をM13ベクターか
ら切り出し、pSV2gpt(重鎖プラスミド)およびpSV2neo
(プロモーターとエンハンサーエレメントを含む軽鎖プ
ラスミド)において再クローニングする。SP2/0ネズミ
ミエローマをこのプラスミドを用いてDNA感染(トラン
スフェクト)させ、ヒト化抗体(hB1)を産生するハイ
ブリドーマをクローニングによって選別する。hB1生産
ハイブリドーマをバイオリアクターで増殖させ、hB1を
イオン交換クロマトグラフィーによって培養液から分離
する。
ク質Aにおけるサブトラクションクロマトグラフィーお
よび透析ろ過によって精製する。hB1−IgGをシステイン
を用いて還元によってFab′に変換し、Fab′はゲルろ過
クロマトグラフィーによって精製する。全工程は、アル
ゴン下で実施し、緩衝液はアルゴンで浄化し、さらに、
Fab′−SHは、治療用複合体を調製するために使用する
までアルゴン雰囲気下で維持する。

味である。

弱した発熱男性患者を、静注により週2回hB1−Fab′−
S−APPで処置する。治療後2か月で患者の血液は、p24
HIV抗原について調べたとき陰性で、健康状態は顕著に
改善される。

明の範囲を外れること無く利用するために多くの変更お
よび修飾が当業者には実施可能である。

Claims (5)
- 【請求項1】標的細胞または病原性微生物に関連する物
質と反応するタンパク質を含む複合体であって、該タン
パク質は、薬剤、または機能的レセプター結合ドメイン
を欠く修飾毒素に、第一のチオール結合リンカーを通し
て複合され、さらに、少なくとも一つの多糖またはポリ
オール基に、少なくとも第二のチオール結合リンカーを
通して複合され、該リンカーに結合するタンパク質チオ
ール基は、少なくとも一つのジスルフィド結合の還元か
ら誘導され、該タンパク質は、該標的細胞または病原性
微生物の細胞質内に、該複合体を内在化させる、治療用
タンパク質複合体。 - 【請求項2】癌疾患のまたは病原性微生物に感染した患
者が、細胞毒性量の上記複合体を投与され、上記タンパ
ク質が、上記複合体を癌細胞または病原性微生物に内在
化させる、該患者の治療方法で用いられる請求項1に記
載の治療用複合体。 - 【請求項3】請求項1に記載の治療用複合体、及び約製
薬的に許容できる注入媒体を含む無菌の注入可能な組成
物。 - 【請求項4】(a)少なくとも一つのジスルフィド結合
を有するタンパク質前駆体を、ジスルフィド還元剤と反
応させて、ジメルカプトタンパク質を生成し、 (b)第一のチオール結合リンカーを通して、ジメルカ
プトタンパク質に毒素または薬剤を結合させ、 (c)第二のチオール結合リンカーを通して、ジメルカ
プトタンパク質に多糖またはポリオールを結合させるこ
とを含む、 請求項1に記載の治療用複合体の調製方法。 - 【請求項5】(a)標的細胞または病原性微生物に関連
する抗原に結合した無傷の抗体を、複数の基部遊離スル
フヒドリル基を生じるために十分な量であるが、該抗体
を免疫学的に不活性にするには不十分な量の、ジスルフ
ィド結合を切断するための還元剤で、部分的に還元し、
該部分的に還元された抗体を回収し、 (b)該部分的に還元した抗体を切断して、還元された
F(ab′)2フラグメントを生じさせ、 (c)第一のチオール結合リンカーを通して、該部分的
に還元されたF(ab′)2フラグメントと、機能的結合
ドメインを欠く毒素または薬剤を結合させ、 (d)該部分的に還元されたF(ab′)2フラグメント
の残余の重鎖ジスルフィド結合を、ジスルフィド結合を
切断するための還元剤で、さらに還元し、 (e)第二のチオール結合リンカーを通して、多糖また
はポリオールに、生じたスルフヒドリル基を結合させ
て、Fab′複合体を生成させることを含む、 請求項4に記載の治療用複合体の調製方法。
