【発明の詳細な説明】 【産業上の利用分野】 本発明は、食用えのき茸の新品種の栽培方法に関する
もので、長年の通常白色品種から突然変異により生じた
菌株を、組織栽培により分離し、反復培養により固定し
た、暗室栽培により工業的生産可能な、ほんえのきブラ
ウンJN−B−001の菌種を用いた、ほんえのきの栽培方
法に関する。 【従来の技術とその問題点】 従来の天然自生えのき茸は、自然光の下に生長するの
でその色はやや褐色であり、その形も人工栽培のものに
比べてやや大きなものである。 しかしながら、えのき茸を人工栽培する場合は、暗室
低温での菌床栽培であり、その色調は白色で茎の細い傘
の小さいものであることは、周知のとおりである。 一方、自生のえのき茸に近い商品の開発もなされ、褐
色えのき茸も効率良く生産する技術が開発されている
が、その菌種は白色えのき茸と同じもので、ただ、培養
条件を変更することにより色調を変えており、えのき茸
の形状を変えることのできないものであった。 【課題を解決するための手段】 本発明の栽培方法に係るほんえのきブラウンJN−B−
001の原種は、白色えのき茸の膨大な数の栽培瓶のうち
から発明者の25年の栽培歴のなかで、今回はじめて通常
の白色えのき茸の培養床の片側約半分が褐色に変色し、
かつ数倍〜数十倍にも及ぶ大きさの傘に生長するものを
見出した新しい菌種によるものである。 このほんえのきブラウンJN−B−001の菌種は、天然
えのき茸の菌種と異なり、本来着色しない暗所培養によ
っても、なおかつ、褐色大形化するもので、3回の反復
培養を経た後においても、その形態の変更がみられず安
定した反復再現性が認められ、新品種の固定に成功し
た。その菌を定着新種菌として昭和56年通商産業省告示
第178号の規程に基づき、「工業技術院微生物工業技術
研究所、特許微生物寄託センター」へ寄託したのである
(寄託番号,微工研菌寄第9688号,FERM P−9688)。 すなわち、このほんえのきブラウンJN−B−001は、
通常の白色品種から突然変異により生じた白色えのき茸
を親とする菌株を、組織培養により分離し、反復培養に
より固定した、暗室栽培において茶褐色を呈し、茎が太
く、中心が凹んだ大きい傘を有するものである。 本発明者はこの寄託者識別表示を上記のように、「ほ
んえのきブラウンJN−B−001」と定め、新菌種寄託を
したのであるが、この寄託菌によるほか、本発明に係る
栽培方法以外では、白色えのき茸の菌種から暗室におい
て、着色えのき茸を得ることができないのが現状であ
る。 【実施例】 本発明に係る栽培方法によるほんえのきブラウンJN−
B−001に反復培養による品種固定の過程を、次に詳し
く説明する。 本発明者は、昭和38年以来、えのき茸の栽培、製造販
売業を営み、現在その生産量は月間約26000kgにも達し
ている。その間、えのき茸の品種改良に取組み、市場の
要求に適合する茎の太くて白い日持ちのするえのき茸と
して良品質のものを市場に供している。 このような改良研究の過程において、本発明のほんえ
のきブラウンJN−B−001は、栽培開始以来約3000万本
という膨大なえのき茸栽培瓶の一つの、しかも、その約
1/2の広さに発生したもので、この褐色を呈するえのき
茸から反復再現性の確認に至る時間経過は第1表のとお
りであり、確認時の状態は第1図のとおりである。 本発明のほんえのきブラウンJN−B−001の栽培方法
を確定する過程の概要を説明すると、次のようになる。 瓶の口径70mm、瓶容積1400ccのポリエチレン製瓶にオ
ガ培地として広葉樹のオガ屑3,米糠1の容積比で混ぜ
て、石灰粉末と水を添加混合してPH7.2〜7.4,水分約65
%に調整したものを詰め、紙の蓋をして120℃で蒸気殺
菌をし、15℃位に冷却した後植菌をする。その後、15℃
の暗室内で菌糸の培養を25日間行ない、表面の菌かきを
して発芽室に移し、約12℃、10日間で発芽をする。次い
で、抑制室に移す。暗所で10日間をかけて約20万本にも
のぼる瓶口の紙掛け作業をするが、この時点で多数の白
色えのきの中にたった1本の瓶だけが、その表面の一部
を褐色を呈するえのき茸の異常発生をみたのである。 第1図はそのときのスケッチであるが、瓶口(1)の
約半分は通常の白色えのき茸(2)であり、他の半分が
茶褐色をした約1円アルミ貨の大きさの傘、鉛筆の太さ
位の茎の褐色えのき茸(3)であった。 本発明の栽培方法は、この褐色えのき茸(3)の組織
培養を次の方法で行なった。すなわち、純粋培養のでき
る寒天培地を、玉葱を千切りにしたもの50gに水300ccを
加え、煮詰めてろ過し約100ccの煮汁とし、これに砂糖2
0g、寒天15g、水420cc、醤油15gを加え、加熱溶解して5
0cc容試験管に15cc宛入れ、100℃常圧間断殺菌3日間行
なって用意した。 上記試験管に形成した寒天培地に、褐色えのき茸
(3)の組織を移植し、綿栓をして15℃で15日間培養し
たところ試験管一杯に菌糸がまわったので、次の中種培
地へ移した。 中種培地は、前述のオガ培地を入れた500ccガラス瓶
で綿栓をして15℃恒温室で25日放置して培養し、これを
1400cc容ポリエチレン製栽培瓶中のオガ培地へ接種し、
30日間経過後菌かきをして発生試験に入った。発芽の時
点で、既に床全体が着色し、そして傘が大きく、従来の
白色小粒のものとは全然、状態を異にするものであっ
た。天然自生のえのき茸は別として、えのき茸を茶褐色
に着色させるには太陽光線に当てるか、白熱電灯、又は
蛍光灯に当てる必要があるのに、暗室培養でこのように
着色したのは、はじめての経験であった。 現在、通常のえのき茸は、白色の細いものが良質とさ
れているために、人工栽培はすべて、暗室で光を一切当
てずに行ない、発芽、抑制生育をするのが一般的であ
る。このような暗室栽培にかかわらずはげしく茶褐色に
着色し、かつ、従来の数倍にも達する大きさの傘及び茎
のものが得られたのである。そして、傘の中心が凹み、
形状においても著しく相違がみられた。この凹みは自然
のえのき茸にも見られない本品種特有のものである(第
4,5図)。なお、昭和62年11月2日第3回目の着色確認
をし、第2回目と同様の経過をたどっている。したがっ
て、本発明の栽培方法によって褐色えのき茸の完全な品
種固定が達成でき、ここに、「ほんえのきブラウンJN−
B−001」と命名した。 【発明の効果】 本発明の栽培方法に係る「ほんえのきブラウンJN−B
−001」は、何度となく試食をした結果、通常の白色え
のき茸と味は変らず美味であるうえ、歯ごたえ、歯切れ
が良好で、食用えのき茸として有望な新品種褐色えのき
茸であり、工業的生産に適したものである。
