性の皮膚疾患の治療のための、細胞の分化と増殖の調節
に関するものである。

の欠損が一連の形態学的、生化学的変化に反映されてい
る。細胞学的、組織学的、組織化学的、生化学的なきわ
めて多数の変化が、この疾患の経過の原因ではなく結果
であることが知られている。

も有効な処置は、コルチコステロイドのプラスチックラ

乾癬の患者には種々のシステマチックな化学療法薬、特
にメトトレキセート(methotrexate)を用いることが必
要である;後者の薬は細胞増殖の阻害に見合う細胞機能
の阻害なしに、増殖を阻害する効力を持つ。光化学療法
は1974年に導入された、いわゆるPUVA療法である。この
療法は、主に長波長紫外線(UV−A)を放射する特別な
照明系を用いた局所的あるいは全身照明に先立つソラレ
ン(psoralen)の投与より成る。

は、1,25(OH)2D3に対する高親和性(1.0×10-10M)、
低容量の受容体様のタンパク質を含んでいる。1,25(O
H)2D3に対するレセプターを持つ腫瘍細胞に対しては、
ホルモンはその増殖を阻害し、分化を誘導する(スダ
(Suda)ら、ビタミンD:Chemical,Biochemical,Clinica
l Update(ノーマン(Norman)ら監修)Walterde Gruyt
er社 N.Y.1985 pp.187−196)。1,25(OH)2D3はヒトの
培養線維芽細胞とケラチン細胞の増殖を阻害し、ケラチ
ン細胞を最終分化に誘導する(スミス(Smith)ら、J.I
nvest.Dermatol.86:709−714,1986)。1,25(OH)2D3と
その類似体1,24−ジヒドロキシビタミンD3(1,24−dihy
droxyvitamin D3)が、経口的あるいは局所的に投与さ
れた場合、乾癬の治療に有用であることは示されていた
(モリモト(Morimoto)ら、Brit.J.Dermatol.115:421
−429,1986;カトウ(Kato)ら、Brit.J.Dermatol.115:4
31−433,1986)。

球、骨肉腫細胞を含む種々の細胞が上皮小体ホルモンに
対する受容体を持っている(ヤマモト(Yamamoto),I.
ら、J.Clin.Invest.71:404−407,1983;ゴールドリング
(Goldring),S.ら、J.Clin.Endo.and Met.46:425−43
3,1978)。上皮小体ホルモンの検定のために、種々の試
験管内(in vito)と生体内(in vito)の試験が開発
されている。これらには単離したイヌの腎膜内(ニセン
ソン(Nissenson)ら、J.Clin.Endo.Metab.52:840,198
1)、骨肉腫細胞内(リンダール(Lindall),A.W.ら、
J.Clin.Endo.Metab.57:1007,1983)とヒトの線維芽細胞
(ゴールドリング(Goldring)ら、J.Clin.Endocrinol.
and Met.46:425−433,1978)でのCAMPの産生の測定が含
まれる。モルモットの腎臓におけるグルコース−6−リ
ン酸デヒドロゲナーゼ活性を測定する細胞化学的検定法
も開発されている(ゴルツマン(Goltzman),D.J.ら、C
lin.Invest.65:1309,1980)。さらに、上皮小体ホルモ
ン類似体の作動薬(agonist)と拮抗薬(antagonist)
特性の両者を測定する多反応性上皮小体ホルモン検定法
も開発されている(ホリウチ(Horiuchi),N.ら、Am.J.
Physiol.244:E589−595,1983)。
−34アミノ末端断片がが試験管内と生体内で生物学的に
活性であるという観察は興味深かった(ナスバウム(Nu
ssbaum),S.R.ら、J.Prot.Chem.4:391−406,1985)。ラ
ットPTH(1−34)は、ヒト及びウシPTHの両者の相当す
る1−34領域と5アミノ酸の配列の相違があるが、イヌ
のアデニル酸シクラーゼ系でヒトPTH(1−34)より8
−10倍、ウシPTH(1−34)より2−4倍活性であるこ
とが見出されている(コイトマン(Keutmann),H.T.
ら、Endocrinol.117,1230,1985)。構造活性研究によ
り、PTHにより刺激される試験管内の生物学的反応は〔N
le8,Nle18,Tyr34〕ウシ(3−34)PTHアミド(bPTH−
(3−34))により阻害され得ることが明らかになっ
た。上皮小体ホルモン類似体〔Tyr34〕ウシPTH−(7−
34)−アミドは、甲状腺上皮小体切除ラットで、生体内
で血漿カルシウムのPTHによる上昇を阻害することがで
き、PTH様作動体活性を欠いている(ドッペルト(Doppe
lt),S.H.ら、Proc.Natl.Acad.Sci.USA.83,7557,198
6)。
ンパク質を産生していることが明らかになったが、これ
を本明細書では“高カルシウム血症因子(hypercalcemi
c factor)”と称する(メレンディノ(Merendino),J.
J.ら、Science 231:388−390 1986)。ケラチン細胞に
よる順化培地(keratinocyte−conditioned medium)を
集密的(confluent)な、初代継代培養(first passage
cultures)から回収した。各々10のケラチン細胞培養
由来の順化培地はクローンのROS17−2.8細胞検定でアデ
ニル酸シクラーゼ活性を刺激した。この生物学的に活性
な物質は、種々の悪性度の患者において、ホルモン性高
カルシウム血症を引き起こす原因となっている因子の一
つであると考えられる。このPTH関連タンパク質はヒト
肺癌細胞株から単離され、このペプチドをホ乳類細胞で
産生するのに用いた発現ベクターに、これをコードして
いる全長の相補DNAクローンを挿入した。このクローン
は36アミノ酸のプレプロペプチドと、上皮小体ホルモン
とアミノ末端領域で顕著な相同性を持つ141アミノ酸の
成熟タンパク質をコードしていることが明らかになった
が、この成熟タンパク質の最初の16残基のうちの8つが
ヒトPTHと同一であることが明らかになった(スバ(Suv
a).L.J.ら、Science 232;893−896,1987)。本明細書
の第1図はスバンらの第2図から転載したものである
が、これら2つのタンパク質の配列を記している。
2D3のケラチン細胞増殖抑制と最終分化誘導能に基づい
た、乾癬の治療への1,25(OH)2D3の使用と、培養ヒト
ケラチン細胞がPTH様高カルシウム血症因子を産生して
いるという観察が一緒にまとまって本発明になった。本
発明は部分的には次の発見から生まれたものである:1)
ヒト培養ケラチン細胞をヒトPTH(1−34)(本明細書
ではPTH(1−34))と10-7と10-8Mの濃度で保温した場
合、このペプチド断片はこれらの細胞の増殖を阻害し、
最終分化を誘導する;2)高カルシウム血症因子の合成断
片([Tyr34]高カルシウム血症因子断片(1−34)ア
ミド(本明細書ではCFF(1−34))も、10-10,10-8,10
-7Mの濃度で量依存的にケラチン細胞の増殖を阻害し、
最終分化を誘導する;3)[Nle8,Nle18,Try34]ウシ(3
−34)PTHアミド(本明細書ではPTH(3−34))は、PT
H(1−34)のように作動薬である、すなわちそれはケ
ラチン細胞の増殖を阻害し、最終分化を誘導する;4)
[Tyr34]ウシPTH(7−34)アミド(本明細書ではPTH
(7−34))は、PTH(1−34)とCCF(1−34)の抗増
殖性、分化誘導性作用を阻害する拮抗薬である;5)拮抗
薬PTH(7−34)は、1,25(OH)2D3のケラチン細胞増殖
及び最終分化誘導活性を阻害する。

を哺乳類細胞に接触させることによる、その細胞の増殖
の促進を含むものである。

が、老化によって低下した増殖能を持つ人々の表皮の再
成長の刺激と同様、火傷、皮膚潰瘍、外傷の患者の皮膚
成長の促進に有用である。

ペプチドから誘導することができる。例えば、以下にバ
ッチ・ケム・カタログ(Bach Chem catalog)の一節が
複写されているが、ここには、多数の入手可能なヒト、
ラット、ウシのアナログおよび断片が記載されている。

カタログも、入手可能な断片を記載している。) 本発明のいずれかの方法にによって、候補となるペプ
チドを選択する際、好ましい第一の段階は、ヒトPTHま
たは高カルシウム血症因子のアミノ末端の34アミノ酸領
域内の、8あるいはそれ以上のアミノ酸長の断片中に、
少なくとも10%、またさらに好ましくは50%以上の相同
性を有する断片を含むペプチドを選択することである。
ヒトPTHと他の種のPTH間の相同性が高いため、ヒト由来
の断片あるいはアナログと同様にヒト以外のものも用い
ることができる。さらにこの断片は、例えばカルボキシ
ル末端のアミノ酸残基を末端アミド基にすることといっ
た、種々の標準的な化学的手法のいずれによっても修飾
可能であり、また、アミノ末端残基は、例えば脂溶性を
増強するために官能基で修飾することができ、そしてペ
プチドは、溶解性の上昇あるいはin vivoでの半減期を
延長するために化学的にグリコシル化することができ、
また、D−アミノ酸をペプチド中のL−異性体と置換す
ることが可能である。
Hあるいは高カルシウム血症因子の末端6アミノ酸の一
部か、全部を含むことが一般に好ましいが、一方、一般
にこのペプチドが抗増殖促進効果および分化を阻害する
ために用いられる場合、これらのアミノ酸のわずかのも
のが用いられるかまたは全く用いられない。
ト培養細胞を用いて、細胞増殖および分化の阻害物質あ
るいは促進物質としての適正を検討されている。ケラチ
ノサイト培養細胞中で増殖を阻害し、分化を誘導するこ
れらのペフチドは、細胞増殖の抑制が求められる乾癬お
よびガンといった病気を処置する場合の治療薬として潜
在的に有用なものである一方、これらの拮抗ペプチドの
効果を妨げるペプチドあるいは1,25(OH)2D3は、細胞
増殖の維持または活性化が求められるような火傷、皮膚
の潰瘍、けがなどの病気治療における治療薬として潜在
的に有用である。

学療法薬は、分裂をしている細胞にのみ有効であり、こ
の妨害ペプチドは、他の休止中の細胞の分裂を誘導し、
化学療法の影響を受け易くする効果を持ち得る。妨害ペ
プチドは、他に、毛髪の生育を刺激および維持するため
にも用いることができる。妨害ペプチドはまた、新しい
細胞、例えば局部的に用いるスキン・クリームにおい
て、皮膚細胞の生育を促進するために有用である。分化
誘導ペプチドは、PTH受容体をもつ単球およびリンパ球
の成熟を誘導することにより免疫賦活剤として用いるこ
とができる一方、妨害ペプチドは、リンパ球の成熟を阻
害し、したがって、例えば若年性糖尿病、リューマチお
よび成熟したリンパ球が原因となる他家移植拒絶などの
自己免疫不全症を治療するために用いることができる。
NIH 3T3細胞は、35mm組織培養基当り0.5×105細胞を植
え、2日後コバルト60線源(5000ラッド)を用いて致死
量の放射線照射を行った。ケラチノサイトは、新生児の
包皮から、4℃において一夜トリプシン処理を行い、0.
02%EDTA処理を行うことにより得られた。ケラチノサイ
トは、2mlの無血清培地を含む培養基中の、致死量の放
射線を照射した3T3細胞植え移植した。各々の実験は、
異なる皮膚試料から得られた初代あるいは第二代ケラチ
ノサイト培養において行った。無血清培地は、高(1.8m
M)濃度のカルシウム(M.A.バイオプロダクツ、ウォー
カーズビル、MD)を含み、7種の増殖因子、すなわち表
皮増殖因子(25ng/ml);ヒドロコルチゾン(203ng/m
l);インシュリン(5μg/ml);プロスタグランジンE
1(50ng/ml);トランスフェリン(5μg/ml);コレラ
トキシン(0.1μg/ml);(シグマ・ケミカル会社、セ
ントルイス、MO);およびセレナス酸(2ng/ml)を含む
デュベッコの修飾イーグル培地(DMEM)からなってい
る。一週間の培養で、ヒドロコルチゾンとコレラトキシ
ンを培地から除き、培養基を全ての残存する3T3細胞を
除くために0.02%EDTAで洗った。
(1−34);PTH(3−34);PTH(7−34)の4種類のペ
プチドが、種々の濃度で検定された。これに加え、1,25
(OH)2D3が検定され、単独および、PTH(7−34)、な
らびにPTH(1−34)、PTH(7−34)との組合せでとも
に検定された。
基を前述の化合物あるいは媒体のみと保温した。一週間
の投与後、各々の培養から培地を除き、遠心して、剥離
した遊離細胞を計数するために再度懸濁した。位相差顕
微鏡下で異なる細胞型を計数するために、血球計数盤を
用いた。接着した細胞を、次に0.1 EDTA,0.1%トリプシ
ンにより30−40分間トリプシン処理し、その後培地で中
和した。このケラチノサイトを遠心して、既知の容量の
培地に再懸濁した。基底(小型,球形)および鱗状(よ
り大きい、不定形、偏平)細胞を計数するために、二連
のアリコットをとった。残りの細胞を遠心し、室温で10
分間、10mM Tris−HCl(pH7.4)、1%β−メルカプト
エタノールおよび1%ドデシル硫酸ナトリウム(SDS)
により処理した。この処理の後では、角化した包膜を持
つ細胞のみが存在する。

た。第2図、第3図、第4図および第5図に見るよう
に、1,25(OH)2D3は投与量依存的に培養中の全細胞数
および基底細胞数を減少させた。同じ様な様式で、10-8
Mと10-7MにおけるPTH(1−34)および10-10Mと10-8Mに
おけるPTH(3−34)は、10-8Mおよび10-7MにおけるCFF
(1−34)と同様に、基底細胞の総数を顕著に減少させ
た。10-8Mおよび10-7MにおけるPTH(7−34)は効果が
なかった。さらに、10-8Mにおける1,25(OH)2D3を10-7
MのbPTH(7−34)とともにインキュベーションする
と、このPTH拮抗剤は顕著に1,25(OH)2D3に対する応答
を減少させた。同様に、PTH(7−34)をhPTH(1−3
4)あるいはCFF(1−34)とともにインキュベーション
すると、PTH(7−34)は、培養中で細胞総数および基
底細胞の総数についてPTH(1−34)およびCFF(1−3
4)の効果を阻害した。

でみるように、1,25(OH)2D3は、投与量依存的に角化
した包膜の総数を増加させた。同様に、PTH(1−3
4)、PTH(3−34)およびCFF(1−34)もまた、投与
量依存的に対照培養に比べ角化した包膜の総数を増加さ
せた。10-8Mまたは10-7MのPTH(7−34)は効果がなか
った。さらに、PTH(7−34)は、角化した包膜の分化
において1,25(OH)2D3に、PTH(1−34)およびCFF
(1−34)の効果を阻害した。
の増殖阻害および最終分化の誘導において生物学的に効
果的であり、そのため、乾癬および癌などの過増殖性の
病気の処置のための治療薬として用いる候補であり; 2) CFF(1−34)およびPTH(3−34)は、hPTH(1
−34)と同様な効果を有し、したがってこれも過増殖性
の病気に対する治療薬としての候補であり; 3) PTHの作用に対する拮抗剤であるPTH(7−34)
は、それ自身では、増殖あるいは最終分化において実質
的に効果がないが、ヒトケラチノサイト培養細胞の増殖
阻害あるいは終末分化の誘導において、1,25(OH)
2D3、PTH(1−34)およびCFF(1−34)の活性を妨げ
ることができ、したがって修復治療、たとえば火傷、潰
瘍、けがなどの処置などのための候補である。
療的に効果的な量において投与される。一般に、このペ
プチドは薬学的に許容できる担体物質、たとえば炭酸マ
グネシウムあるいは乳酸と混合して投与される。この組
成物は、経口投与のためには丸薬、錠剤、カプセル、溶
液あるいは継続的放出を行う錠剤といった形をとり;鼻
腔投与のためには溶液で;静脈、皮下、非経口あるいは
腹腔内投与のために溶液状をとることができる。投与
は、受容者の年齢、健康状態および体重、同時に行う処
置の種類と、もし少しでもあれば治療の回数、ならびに
望まれる効果の性質に依存するだろう。一般に、日常的
な投与は、0.001μg/kgから10,000μg/kgであり、好ま
しくは体重kgあたり0.1から10μgである。通常、1日
あたり0.1から1000μg/kgを、1日当り1つあるいはそ
れ以上の適用法によって投与することが望ましい結果を
得るために有効である。上述のようにして投与すること
が可能であることに加え、この化合物は、水、グリセロ
ール、アルコール、プロピレングリコール、脂肪酸アル
コール、トリグリセリド、脂肪酸エステルあるいはミネ
ラルオイルのような担体を含む、ゲルからなるもの、軟
膏あるいはクリームといった薬学的に不活性の局所的な
担体中で用いられることができる。そのほかの可能な担
体は、液体ワセリン、イソプロピルパルミチン酸、ポリ
エチレングリコールエタノール95%、ポリオキシエチレ
ンモノラウレート5%水溶液、ラウリル硫酸ナトリウム
5%水溶液およびそれに類したものがある。酸化防止
剤、吸湿剤、粘度安定化剤およびそれに類したものなど
の物質は、もし必要であれば加えることができる。
リポソーム担体といった手段により供与されることもで
きる。このペプチドは、薬学的に許容される塩類の形で
供給されることもできる。好ましい塩の例としては、薬
学的に許容可能な有機酸、例えば酢酸、乳酸、マレイン
酸、クエン酸、リンゴ酸、アスコルビン酸、コハク酸、
安息香酸、サリチル酸、メタンスルホン酸、トルエンス
ルホン酸あるいはパモイン酸と同様に、タンニン酸ある
いはカルボキシメチルセルロースといった酸重合体およ
びヒドロハリン酸例えば、塩酸、硫酸あるいはリン酸と
いった無機酸塩などである。
Claims (7)
- 【請求項1】哺乳動物の非ガン性高増殖性の皮膚疾患の
治療に有用な医薬組成物であって、ヒト副甲状腺ホルモ
ン又はヒト高カルシウム血症因子のN−末端34個のアミ
ノ酸領域と10%以上のホモロジーを有し、かつインビロ
トで培養されたヒト角化細胞の分化を促進し、増殖を阻
害する活性を示す、少なくとも8個のアミノ酸長のペプ
チドを有効成分とする医薬組成物。 - 【請求項2】前記高増殖性の皮膚が乾癬であるである請
求項1記載の医薬組成物。 - 【請求項3】哺乳動物の皮膚細胞の増殖を誘導する医薬
組成物であって、ヒト副甲状腺ホルモン又はヒト高カル
シウム血症因子のN−末端34個のアミノ酸領域と10%以
上のホモロジーを有し、かつインビロトで培養されたヒ
トケラチノサイトに対するPTH(1−34)、1,25(OH)2
D3又はCCF(1−34)による分化の刺激及び増殖の抑制
作用、を阻害する活性を示す、少なくとも8個のアミノ
酸長のペプチドを有効成分とする医薬組成物。 - 【請求項4】前記ペプチドが、少なくとも18個のアミノ
酸長であり、かつ上記N−末端34個のアミノ酸領域と50
%以上のホモロジーを有する請求項3記載の医薬組成
物。 - 【請求項5】哺乳動物の皮膚外傷の治癒を促進するのに
有用な請求項3又は4記載の医薬組成物。 - 【請求項6】哺乳動物の毛髪成長を刺激し維持するのに
有用な請求項3又は4記載の医薬組成物。 - 【請求項7】哺乳動物の上皮増殖を刺激するのに有用な
請求項3又は4記載の医薬組成物。
