Description
【発明の詳細な説明】
本発明は、粒子被覆性を改良すべく少量の高分子量ポ
リマーで処理されたスルホン酸カチオン交換樹脂粒子
と、該粒子より製せられた長期連続放出性医薬品に関す
る。米国特許第4,221,778号に記載の如く、かかる製剤
には、表面に薬理学上活性な薬剤を吸着させて薬剤−樹
脂複合体粒子を形成しているイオン交換樹脂粒子が含ま
れ、また該粒子の少くとも一部分は含浸剤で処理され且
つ拡散バリヤー被覆を付与されている。 本発明の背景 米国特許第4,221,778号には、ポリエチレングリコー
ル、プロピレングリコール、マンニツト、ラクトースお
よびメチルセルロースより選ばれる含浸剤で予め処理さ
れたイオン交換樹脂薬剤複合体粒子に拡散バリヤー被覆
を施すことにより、消化管内で遭遇される如き条件下薬
理学上活性な薬剤の制御された(すなわち選択的長期)
連続放出性を達成しうることが開示されている。 本発明の概要 本発明は、(a)スルホン酸カチオン交換樹脂粒子に
して、その被覆性を改良すべく、カルボキシポリメチレ
ン、キサンタンガムおよびアルギン酸プロピレングリコ
ールエステルより選ばれる、約1.1重量%(上記粒子と
本ポリマーとの総重量を基準とする)までの有効量の高
分子量ポリマーで処理された樹脂粒子、(b)表面に塩
基性薬剤を吸着させて薬剤−樹脂複合体粒子を形成して
いる上記樹脂粒子、(c)ポリマー処理に続いて透水性
拡散バリヤーで被覆された上記薬剤−樹脂複合体粒子お
よび(d)かかる被覆された薬剤−樹脂複合体粒子より
なる医薬品に関する。かかる製剤から、消化管内で遭遇
される条件下薬剤の長期連続放出を達成することができ
る。 被覆量を変化させまた(或は)、被覆薬剤−樹脂複合
体粒子と未被覆薬剤−樹脂複合体粒子とをブレンドする
ことにより、製剤の薬剤溶出特性を所望通り選択的に調
節することができる。 イオン交換樹脂、薬剤および被覆並びに、薬剤樹脂複
合体の製造方法、該複合体の被覆方法および、ブレンド
ないし被覆の程度による製剤溶出特性の選択的調節方法
については米国特許第4,221,778号に開示され且つ例示
されている。 詳細な説明 米国特許第4,221,778号は、樹脂の被覆性を改良する
のに通常約10〜25重量%の処理剤が必要とされると記し
ているが、本発明の上記ポリマー処理剤は、樹脂および
ポリマーの重量に対しわずか約1.1重量%(薬剤−樹脂
複合体とポリマーとの重量を基に約0.8重量%に相当)
以下の使用量で済むとわかつた。カルボキシポリメチレ
ン(例えば、分子量約3,000,000の高純度銘柄アクリル
酸ポリマーと製造業者が述べているCarbopol 934P)
は、通常約0.1〜1.1重量%(好ましくは0.4〜0.8%)量
で用いられ、キサンタンガムは通常約0.04〜0.20重量%
(好ましくは0.07〜0.16%)、そしてアルギン酸プロピ
ルグリコールエステルは通常約0.3〜0.9重量%(好まし
くは0.5〜0.7%)量で用いられる。なお、これらの%
は、樹脂とポリマーとの合計量(重量)を基準とする。
後記表Iに示す如く、本発明のポリマーにより樹脂処理
(例1〜3)は、未処理樹脂(比較例)と較べ樹脂の被
覆性を高める。これは、処理樹脂から製せられる被覆薬
剤−樹脂複合体の薬剤溶出速度が有意に遅延することか
ら立証される。 一般に、全ての薬理学上活性な塩基性薬剤特に、生物
半減期が約8時間以下程度に短い薬剤は、本製剤に含ま
せられる候補物質である。非制限的例として、フエニル
プロパノールアミン(PPA)、デキストロメトルフア
ン、コデイン、ヒドロコドン、ヒドララジン、プロプラ
ノロル、ドキセピン、メタプロテレノル、モルフイン、
エフエドリン、プソイドフエドリンおよびベラパミルが
挙げられる。例示例で用いるモデル薬剤として、人体で
の生物半減期が3.9時間、pKaが9.4の交換神経興奮剤ア
ミンPPAを選定した。樹脂粒子に対する薬剤の使用量は
約1〜90重量%としうるが、標準的実用範囲は15〜50%
である。 樹脂は、粒度において通常約25〜1000μm範囲のスル
ホン酸カチオン交換樹脂である。例示例では、アンバー
ライト(Amberlite )IRP−70樹脂を用いた。これは、
アンバーライトIR−120を100〜200メツシユ(75〜150μ
m)に破砕した粒状カチオン交換樹脂である。アンバー
ライトIRP−70および親樹脂のアンバーライトIR−120に
ついては、製造業者が、pH範囲0〜14の水中で膨潤する
ゲルタイプのジビニルベンゼンスルホン酸カチオン交換
樹脂と説示している。樹脂は先天的な薬理学的性質若し
くは毒性を有すべきでない。 イオン交換樹脂粒子に薬剤を吸着させて薬剤樹脂複合
体を形成することは、米国特許第2,990,332号および第
4,221,778号に示されるように周知技法である。一般的
には、薬剤に樹脂の水性懸濁物を混合し、次いで形成せ
る複合体を洗浄し、乾燥する。樹脂への薬剤吸着は、反
応媒体のpH変化によつて検知しうる。 透水性拡散バリヤー被覆材料は一般に拡散バリヤー性
を有ししかも先天的な薬理学的性質若しくは毒性をもた
ないものであれば慣用の合成若しくは天然フイルム形成
性物質のどれでもよい。例示例では、モデル拡散バリヤ
ー膜材料として、水不溶性フイルム形成剤エチルセルロ
ースを用いた。エチルセルロースの皮膜形成特性を改善
するのに、可塑剤Durkex 500植物油を用いた。被覆の
使用量は、所望の薬剤溶出遅延度により異なる。 粒子を被覆するのに慣用の被覆溶剤(例 エタノール
若しくは、塩化メチレン/アセトン混液又は塗料用エマ
ルジヨン)および被覆方法(例 流動層被覆)を用いる
ことができる。流動層被覆の技法については、米国特許
第3,089,824号、同第3,117,027号および同第3,253,944
号に教示されている。被覆は通常、薬剤樹脂複合体に施
されるが、別法として薬剤との複合体生成前の樹脂に施
すこともできる。 後記例では、薬剤−樹脂複合体粒子は16重量%量(塗
料の不揮発性固体と複合体とそして、在れば処理剤の乾
量として)で、約5重量%のエチルセルロース(50cp
s)、約2重量%の精製植物油Durkex500、約10重量%の
アセトンおよび残り%の塩化メチレンを含む塗料溶液と
一緒に被覆される。塗料溶液は、アセトンおよび塩化メ
チレンにDurkexとエチルセルロースを溶かすことにより
製造される。複合体粒子の被覆は、流動層被覆装置にお
いて1分当たり16〜17mlの塗料溶液量で実施される。装
置入口の空気温度は約33℃で、出口空気温度は約22〜25
℃である。 後記の対照試験(対照)に示す如く、未処理未被覆樹
脂−薬剤複合体は擬似胃液(0.1N塩酸)中でその薬剤を
迅速に溶出した(30分で86.5%)。被覆されている未処
理の複合体粒子(比較例)はこの溶出速度を部分遅延さ
せる(30分で30.9%の薬剤溶出)が;この溶出速度は、
被覆前複合体粒子をポリマー剤で処理するとき(例1〜
3)、更に遅延される(30分で16.1〜21.0%の溶出)。
ポリマーは通常、複合体に施されるが、樹脂粒子が薬剤
との複合体生成前被覆されている場合の如く複合体生成
前の樹脂粒子に施すこともできる。後記例では、十分な
脱イオン水に溶解せるポリマー剤をPPA樹脂複合体に加
え、約10分間(適当な遊星形ミキサー内で)混合し、次
いで流動層を5%未満の水分にまで乾燥した。 被覆層および(又は)被覆/未被覆複合体混合物使用
における変化を用いて溶出特性を所望通り選択的に修正
することができる。経口投与に加えて、本発明の製剤は
局所投与又は直腸ないし膣投与にも適し、用量は薬剤の
種類およびその意図せる用途によつて広範囲に変動し、
例えば約0.1〜約1000mgである。組成物は錠剤、粉末、
カプセル若しくは液状懸濁物形態又は他の慣用投与形態
を取りうる。 例では、胃腸管内で遭遇される条件に似せるために下
記の如き溶出試験装置および手順を用いた:適当な水浴
中に浸漬した丸底フラスコに500mlの溶出試験液(0.1N
塩酸)を入れ、温度を37±0.5℃に上げる。フラスコに
は、100rpmで可動する櫂が備えられている。溶出試験液
はその容器からコツトンフイルターを経てポンプ給送さ
れ、過された該試験液は、ポリエチレン管により、ぜ
ん動運動ポンプを介しベツクマン式35記録型分光光度計
(セルチエンジヤー付き)の1cm流れセルに搬送され、
そして容器に移される。流量は16ml/minに調節される。
このようにして、六つの容器の各々と一つの標準を15分
毎に或は他の適当な時間間隔でモニターすることができ
る。六つの樹脂複合体試料と一つのPPA塩酸塩標準をモ
ニターするのに、分光光度計を単一ビームモードにおけ
る257nmで作動させた。各溶出溶器には、PPA塩基90.6mg
に相当する樹脂複合体試料が入つている。標準PPA溶液
には、0.1N塩酸500ml中PPA塩基90.6mgが入つている。溶
出された薬剤を、複合体粒子中に存在する全薬剤の百分
率として表わす。 例示例 対照−未被覆,未処理複合体: ガラスライニングせる混合用かま内の脱イオン水850
にPPA塩酸塩(96.84kg)を溶かした。撹拌下のPPA塩
酸塩溶液に樹脂アンバーライトIRP−70(243.77kg)と
洗浄水素サイクルを加えた。混合を2時間続けた。次い
で、PPA樹脂スラリーを収容槽に移し、そのあと遠心器
に移した。樹脂コアを、塩化物イオンが無くなるまで少
なくとも10分間脱イオン水で洗浄した。次いで、入口空
気温度70℃の流動槽乾燥機内において水分が5%未満に
なるまで樹脂コアを乾燥した。この乾燥樹脂複合体は2
3.8%のフエニルプロパノールアミンを含むとわかつ
た。溶出結果を表1に示す。 比較例−被覆,未処理複合体 上記の如く被覆したPPA−樹脂複合体粒子に関する溶
出結果を表1に示す。 例1〜3−予備処理,被覆複合体: 被覆PPA−樹脂複合体粒子のパツチ三種を用意した。
その一つは約0.8%のCarbopol934P(例1)で予備処理
したもの、もう一つは約0.07%のキサンタンガム(例
2)で、他は約0.7%のアルギン酸プロピレングリコー
ルエステル(例3)で予備処理したものである。全ての
%は、樹脂およびポリマーの重量を基にした重量によ
る。溶出結果を表1に示す。 上記発見を応用するとき、被覆複合体粒子と未被覆複
合体粒子との適当な混合物を含むよう、ヒト若しくは獣
医用に制御された溶出投与形態を処方して、所望される
薬物の制御された溶出特性を得ることができる。投与形
態は固体(例えば、粉末、カプセルおよび錠剤)又は液
体(例えば口にあうベヒクル中の複合体粒子懸濁物)で
ありうる。

号に教示されている。被覆は通常、薬剤樹脂複合体に施
されるが、別法として薬剤との複合体生成前の樹脂に施
すこともできる。 後記例では、薬剤−樹脂複合体粒子は16重量%量(塗
料の不揮発性固体と複合体とそして、在れば処理剤の乾
量として)で、約5重量%のエチルセルロース(50cp
s)、約2重量%の精製植物油Durkex500、約10重量%の
アセトンおよび残り%の塩化メチレンを含む塗料溶液と
一緒に被覆される。塗料溶液は、アセトンおよび塩化メ
チレンにDurkexとエチルセルロースを溶かすことにより
製造される。複合体粒子の被覆は、流動層被覆装置にお
いて1分当たり16〜17mlの塗料溶液量で実施される。装
置入口の空気温度は約33℃で、出口空気温度は約22〜25
℃である。 後記の対照試験(対照)に示す如く、未処理未被覆樹
脂−薬剤複合体は擬似胃液(0.1N塩酸)中でその薬剤を
迅速に溶出した(30分で86.5%)。被覆されている未処
理の複合体粒子(比較例)はこの溶出速度を部分遅延さ
せる(30分で30.9%の薬剤溶出)が;この溶出速度は、
被覆前複合体粒子をポリマー剤で処理するとき(例1〜
3)、更に遅延される(30分で16.1〜21.0%の溶出)。
ポリマーは通常、複合体に施されるが、樹脂粒子が薬剤
との複合体生成前被覆されている場合の如く複合体生成
前の樹脂粒子に施すこともできる。後記例では、十分な
脱イオン水に溶解せるポリマー剤をPPA樹脂複合体に加
え、約10分間(適当な遊星形ミキサー内で)混合し、次
いで流動層を5%未満の水分にまで乾燥した。 被覆層および(又は)被覆/未被覆複合体混合物使用
における変化を用いて溶出特性を所望通り選択的に修正
することができる。経口投与に加えて、本発明の製剤は
局所投与又は直腸ないし膣投与にも適し、用量は薬剤の
種類およびその意図せる用途によつて広範囲に変動し、
例えば約0.1〜約1000mgである。組成物は錠剤、粉末、
カプセル若しくは液状懸濁物形態又は他の慣用投与形態
を取りうる。 例では、胃腸管内で遭遇される条件に似せるために下
記の如き溶出試験装置および手順を用いた:適当な水浴
中に浸漬した丸底フラスコに500mlの溶出試験液(0.1N
塩酸)を入れ、温度を37±0.5℃に上げる。フラスコに
は、100rpmで可動する櫂が備えられている。溶出試験液
はその容器からコツトンフイルターを経てポンプ給送さ
れ、過された該試験液は、ポリエチレン管により、ぜ
ん動運動ポンプを介しベツクマン式35記録型分光光度計
(セルチエンジヤー付き)の1cm流れセルに搬送され、
そして容器に移される。流量は16ml/minに調節される。
このようにして、六つの容器の各々と一つの標準を15分
毎に或は他の適当な時間間隔でモニターすることができ
る。六つの樹脂複合体試料と一つのPPA塩酸塩標準をモ
ニターするのに、分光光度計を単一ビームモードにおけ
る257nmで作動させた。各溶出溶器には、PPA塩基90.6mg
に相当する樹脂複合体試料が入つている。標準PPA溶液
には、0.1N塩酸500ml中PPA塩基90.6mgが入つている。溶
出された薬剤を、複合体粒子中に存在する全薬剤の百分
率として表わす。 例示例 対照−未被覆,未処理複合体: ガラスライニングせる混合用かま内の脱イオン水850
にPPA塩酸塩(96.84kg)を溶かした。撹拌下のPPA塩
酸塩溶液に樹脂アンバーライトIRP−70(243.77kg)と
洗浄水素サイクルを加えた。混合を2時間続けた。次い
で、PPA樹脂スラリーを収容槽に移し、そのあと遠心器
に移した。樹脂コアを、塩化物イオンが無くなるまで少
なくとも10分間脱イオン水で洗浄した。次いで、入口空
気温度70℃の流動槽乾燥機内において水分が5%未満に
なるまで樹脂コアを乾燥した。この乾燥樹脂複合体は2
3.8%のフエニルプロパノールアミンを含むとわかつ
た。溶出結果を表1に示す。 比較例−被覆,未処理複合体 上記の如く被覆したPPA−樹脂複合体粒子に関する溶
出結果を表1に示す。 例1〜3−予備処理,被覆複合体: 被覆PPA−樹脂複合体粒子のパツチ三種を用意した。
その一つは約0.8%のCarbopol934P(例1)で予備処理
したもの、もう一つは約0.07%のキサンタンガム(例
2)で、他は約0.7%のアルギン酸プロピレングリコー
ルエステル(例3)で予備処理したものである。全ての
%は、樹脂およびポリマーの重量を基にした重量によ
る。溶出結果を表1に示す。 上記発見を応用するとき、被覆複合体粒子と未被覆複
合体粒子との適当な混合物を含むよう、ヒト若しくは獣
医用に制御された溶出投与形態を処方して、所望される
薬物の制御された溶出特性を得ることができる。投与形
態は固体(例えば、粉末、カプセルおよび錠剤)又は液
体(例えば口にあうベヒクル中の複合体粒子懸濁物)で
ありうる。
Claims (1)
- (57)【特許請求の範囲】 1.スルホン酸カチオン交換樹脂粒子にして、その被覆
性を改良すべく、カルボキシポリメチレン、キサンタン
ガム及びアルギン酸プロピレングリコールエステルより
なる群から選ばれる高分子ポリマー約1.1重量%(該ポ
リマーと前記粒子との総重量を基準とする)以下で処理
された樹脂粒子。 2.粒子の表面に塩基性薬剤を吸着させて薬剤−樹脂複
合体粒子を形成している、特許請求の範囲第1項記載の
樹脂粒子。 3.ポリマー処理に続いて透水性拡散バリヤーで被覆さ
れた、特許請求の範囲第2項記載の薬剤−樹脂複合体粒
子。 4.スルホン酸カチオン交換樹脂粒子と、その表面に吸
着させた塩基性薬剤との、被覆された薬剤−樹脂複合体
粒子よりなる医薬品にして、前記スルホン酸カチオン交
換樹脂粒子または薬剤−樹脂複合体粒子を、その被覆性
を改良すべく、カルボキシポリメチレン、キサンタンガ
ム及びアルギン酸プロピレングリコールエステルよりな
る群から選ばれる高分子ポリマー約1.1重量%(該ポリ
マーと前記粒子との総重量を基準とする)以下で処理
し、次いで透水性拡散バリヤーで被覆した医薬品。 5.ポリマーがカルボキシポリメチレンである、特許請
求の範囲第4項記載の医薬品。 6.ポリマーがキサンタンガムである、特許請求の範囲
第4項記載の医薬品。 7.ポリマーがアルギン酸プロピレングリコールエステ
ルである、特許請求の範囲第4項記載の医薬品。
