Abstract
Description
本発明は、生体材料分野に属し、具体的にはスルフヒドリル変性高分子化合物のヒドロゲル及びその調製方法並びに用途に関する。
生体医用材料(Biomedical Materials)は生体材料(Biomaterials)とも略称され、生体に対して病理組織や臓器を診断、治療、修復又は交換したり、その機能を強化したりするために用いられる新規なハイテク材料である。組織工学の肝心な技術の一つは、生体材料で優れた生体適合性を有すると共に体に分解されて吸収される細胞足場を調製することである。ゲル状態は固体と液体の中間状態であり、ヒドロゲルとは、水中で膨潤し、溶解せずに大量の水分を保持できる、親水性架橋三次元ポリマーネットワークを指し、その水分含有量が90%以上に達することができる。ヒドロゲルは理想的な生体材料であり、それ自体又は簡単な変性によって天然の細胞外マトリックスと類似する十分な物理的及び化学的性質を有することができると同時に、酸素ガス、栄養物質、細胞代謝物及び水溶性金属イオンに対して良好な透過性を示している。ヒドロゲルを調製するための親水性高分子は、その由来に応じて天然高分子と合成高分子に分類される。その中で、天然高分子にはコラーゲン、ゼラチン、フィブリン、多糖類などが含まれ、合成高分子には合成ポリペプチド、ポリエチレングリコール(PEG)及びその誘導体、ポリメチルメタクリレート(PMMA)及びその誘導体、ポリ乳酸-グリコール酸共重合体(PLGA)及びその誘導体などが含まれる。近年、注射可能なイン・サイチュ(in situ)架橋ヒドロゲルもますます注目を集めている。注射可能なイン・サイチュ架橋ヒドロゲルは、注射前に流動可能な液体状態を呈し、標的位置に注射された後、標的位置の形状に完全に一致するコロイドを形成できるという特徴を有する。このような注射可能な特性によって、操作プロセスを簡単且つ便利にするだけでなく、患者への移植手術による痛みを回避し、手術の外傷を大幅に軽減することができる。
上記問題を解決するために、本発明の目的は、新規構造のスルフヒドリル変性高分子化合物と、アクリロイル化高分子化合物、アクリロイル基を含む低分子架橋剤の少なくとも1種とのゲル化によって生成された新規構造を有するヒドロゲルを提供することである。具体的には、本発明は、新規構造のスルフヒドリル変性高分子化合物を採用し、それをアクリロイル化高分子化合物及び/又はアクリロイル基を含む低分子架橋剤と組み合わせてヒドロゲルを形成する。前記スルフヒドリル変性高分子化合物は、生理学的条件下で前記アクリロイル化高分子化合物及び/又はアクリロイル基を含む低分子架橋剤と架橋してヒドロゲルを形成することができる。また、形成されたヒドロゲルの造形効果及び代謝耐性・分解耐性に関連する物理的性質、化学的性質も従来技術に比べて明らかな優位性を有する。具体的には、その代謝耐性・分解耐性は従来技術のヒドロゲルより明らかに優れ、更に、スルフヒドリル-ビニル架橋反応が迅速であるという特性により、前記2つの化合物によって形成されたヒドロゲルシステムは、体内に注射された後、迅速にイン・サイチュゲル化することができる。これに基づいて、本発明のヒドロゲルは、生物医学、医療美容と形成外科及び化粧品などの分野への適用に一層有利である。
前記スルフヒドリル変性高分子化合物は、下記シリーズ化合物であるシリーズスルフヒドリル変性高分子化合物のうちの少なくとも1種であり、
前記シリーズスルフヒドリル変性高分子化合物は、その変性された高分子化合物の構造に-COOH、-NH2、-OH、式aに示されるアクリレート基、式bに示されるアクリルアミド基、式cに示されるアクリロイル基のうちの少なくとも1種を含み、
前記システムには、更に、
C1.アクリロイル化高分子化合物、
C2.アクリロイル基を含む低分子架橋剤、の少なくとも1種を含む。
(i)前記スルフヒドリル変性高分子化合物、及び
(ii)物質C1と物質C2の少なくとも1種
を含むシステムをゲル化することにより、前記ヒドロゲルを調製するステップを含む。
本発明は、新規構造を有するスルフヒドリル化変性高分子化合物を原料として、架橋反応により得られた新規構造を有するヒドロゲルを提供し、かつ当該ヒドロゲルは従来技術におけるヒドロゲル(例えば、従来技術においてヒアルロン酸又は変性ヒアルロン酸を出発原料として、かつ架橋反応により得られたヒドロゲル)に比べると、物理的及び化学的性質、造形効果及び代謝耐性・分解耐性などの面において、予想外の技術的優位性を有する。

[スルフヒドリル変性高分子化合物]
前述したように、本発明のゲル化されるシステムにおいて、以下に示すシリーズ化合物であるシリーズスルフヒドリル変性高分子化合物のうちの少なくとも1種を使用する必要がある:
前記シリーズスルフヒドリル変性高分子化合物は、変性された高分子化合物の構造に-COOH、-NH2、-OH、式aに示されるアクリレート基、式bに示されるアクリルアミド基、式cに示されるアクリロイル基のうちの少なくとも1種を含み、
R1は、水素、ハロゲン、脂肪族基、芳香族基などから選ばれ、具体的には、前記ハロゲン、脂肪族基、芳香族基は後述の更なる定義を満たし、好ましくは、R1は、水素、ハロゲン、脂肪族基から選ばれ、また好ましくは、R1は、水素、ハロゲン、C1-6アルキル基(例えば、メチル基、エチル基など)から選ばれ、

前述したように、本発明は、上記スルフヒドリル変性高分子化合物の調製方法を提供し、この調製方法は、
1)構造に-COOH、-NH2、-OHのうちの少なくとも1種を含む高分子化合物のアクリロイル化ステップ、即ち、高分子化合物の構造に含まれる-COOH、-NH2、-OHの少なくとも1種を直接又は間接に下記基に結合し、
又は、構造に上記式aに示されるアクリレート基、上記式bに示されるアクリルアミド基、上記式cに示されるアクリロイル基のうちの少なくとも1種を含む高分子化合物を直接に反応原料とするステップと、
2)ステップ1)で得られた高分子化合物の少なくとも1種をポリスルフヒドリル化合物HS-R4-SHと反応させ(R4の定義は上記と同じである)、前記スルフヒドリル変性高分子化合物を調製するステップと、を含む。
1)構造に-COOH、-NH2、-OHのうちの少なくとも1種を含む高分子化合物のアクリロイル化ステップ、即ち、高分子化合物の構造に含まれる-COOH、-NH2、-OHの少なくとも1つを-R-基により下記基に結合するか、又は、直接に下記基に結合し、
又は、構造に式aに示されるアクリレート基、式bに示されるアクリルアミド基、式cに示されるアクリロイル基のうちの少なくとも1種を含む高分子化合物を直接に反応原料とするステップと、

前述したように、本発明のゲル化されるシステムにおいて、物質C1であるアクリロイル化高分子化合物を更に含んでもよく、本発明のアクリロイル化高分子化合物は、
1)構造に-COOH、-NH2、-OHのうちの少なくとも1種を含む高分子化合物のアクリロイル化化合物、即ち、前記高分子化合物の構造に含まれる-COOH、-NH2、-OHの少なくとも1種を直接又は間接に下記基に的に結合して形成されたアクリロイル化化合物、
2)構造に式aに示されるアクリレート基、式bに示されるアクリルアミド基、式cに示されるアクリロイル基のうちの少なくとも1種を含む高分子化合物、の少なくとも1種から選ばれてもよい。
前述したように、本発明のゲル化されるシステムにおいて、物質C2である、アクリロイル基を含む低分子架橋剤を含んでもよい。当該低分子架橋剤は、アクリロイル基を含む低分子化合物又はアクリロイル基を含むオリゴマーを含むが、これらに限定されず、具体的には、エチレングリコールジアクリレートEGDA、ポリエチレングリコールジアクリレートPEGDA、トリメチロールプロパントリアクリレートTMPTA、ペンタエリスリトールトリアクリレートPTA、ペンタエリスリトールテトラアクリレートPTTA、ジ(トリメチロールプロパン)テトラアクリレートDTTAなどから選ばれてもよい。
前述したように、本発明はヒドロゲルを提供し、当該ヒドロゲルは、
(i)上記スルフヒドリル変性高分子化合物、及び
(ii)物質C1と物質C2の少なくとも1種
を含むシステムのゲル化により調製して得られる。
そのうち、前記スルフヒドリル変性高分子化合物と前記アクリロイル化高分子化合物との用量比(質量部で、合計1部)は、0.01:0.99~0.99:0.01である。例えば、0.1:0.9~0.9:0.1であってもよく、例えば、0.01:0.99、0.1:0.9、0.15:0.85、0.2:0.8、0.3:0.7、0.4:0.6、0.5:0.5、0.6:0.4、0.7:0.3、0.8:0.2、0.85:0.15、0.9:0.1、0.99:0.01又は上記範囲内の任意の比率である。
前述したように、本発明は、上記ヒドロゲルの調製方法を提供し、
この調製方法は、
(i)前記スルフヒドリル変性高分子化合物、及び
(ii)物質C1と物質C2の少なくとも1種
を含むシステムをゲル化することにより、前記ヒドロゲルを調製するステップを含む。
(a)前記スルフヒドリル変性高分子化合物、
(b)C1.前記アクリロイル化高分子化合物、C2.アクリロイル基を含む低分子架橋剤、の少なくとも1種、
(c)他の生体機能材料、薬物、成長因子及び細胞懸濁液からの任意選択的な少なくとも1種
を含むシステムをゲル化することにより、前記ヒドロゲルを調製するステップを含む。
ヒドロゲルは親水性ポリマーセグメントを架橋することによって形成された水中で膨潤できる三次元空間グリッドであることが知られている。ゲル化プロセスは、ポリマーセグメントの物理的絡み合い、静電相互作用、共有化学架橋、可逆化学架橋、超分子化学架橋、及び親水性疎水性架橋などを含む様々な反応メカニズムによって実現することができる。近年、ヒドロゲル機能に関する研究の深まりに伴って、ヒドロゲルは、薬物送達システムの調製、軟組織創傷用の修復被覆材、骨修復用の足場材料、眼科手術における支持用の粘弾剤、手術後の組織癒着を防止するための材料、及び3Dバイオプリンティング用の足場材料などの医学分野で広く適用されている。これは、組織工学及び再生医療分野の研究の焦点になっている。
200 mLのビーカーに1 gのヒアルロン酸(華熙福瑞達社から購入され、その重量平均分子量は約300 kDa)、50 mLの脱イオン水、50 mLのジメチルホルムアミド、12 mLのトリエチルアミン、14 mLのグリシジルアクリレートを加える。室温で均一且つ透明になるまで撹拌した後、48時間撹拌し続ける。300 mLのアセトンを加え、大量の白色沈殿物が生成された。遠心分離により得られた沈殿物を100 mLの脱イオン水に溶解させ、無色透明溶液を得る。上記溶液を透析バッグ(分子量カットオフ8 kDa)に入れ、5 Lの脱イオン水で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に921 mgの白色綿状固体であるHA-A1を得て、収率は92.1%である。

200 mLのビーカーに1.2 gのコンドロイチン硫酸(その重量平均分子量は約80 kDa)、50 mLの脱イオン水、50 mLのジメチルホルムアミド、5.4 gの無水アクリル酸を加え、撹拌して溶解させる。1 mol/LのNaOHで溶液のpHを8±0.5に維持し、24時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ3.5 kDa、Spectrumlabs)に入れ、5 Lの脱イオン水で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に781 mgの淡黄色綿状固体であるCHS-Aを得て、収率は65.1%である。

200 mLのビーカーに1 gのグリコールキトサン(その重量平均分子量は約200 kDa)、50 mLの脱イオン水、50 mLのジメチルホルムアミド、8 mLのトリエチルアミン(Sigma)、13 mLのグリシジルメタクリレートを加える。室温で均一且つ透明になるまで撹拌した後、48時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ3.5 kDa、Spectrumlabs)に入れ、5 Lの脱イオン水で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に726 mgの淡黄色綿状固体であるCTS-MAを得て、収率は72.6%である。
200 mLのビーカーに調製例9の方法により調製された1 gのCHS-A、0.25 gのジチオスレイトール(VWR)、100 mLの脱イオン水を加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ3.5 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に629 mgの淡黄色綿状固体であるCHS-A-SH1を得て、収率は62.9%である。

200 mLのビーカーに調製例21の方法により調製された2 gのPHEMA-A、0.42 gのジチオスレイトール(VWR)、50 mLの脱イオン水、50 mLのジメチルホルムアミドを加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ2 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に1.67 gの白色固体であるPHEMA-A-SH1を得て、収率は83.5%である。
200 mLのビーカーに調製例22の方法により調製された2 gのPHEMA-MA、0.41 gのジチオスレイトール(VWR)、50 mLの脱イオン水、50 mLのジメチルホルムアミドを加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ2 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に1.62 gの白色固体であるPHEMA-MA-SH1を得て、収率は81%である。
200 mLのビーカーに調製例23の方法により調製された1 gのPVA-A、100 mLの脱イオン水を加え、PVA-Aが完全に溶解するまで溶液を加熱して撹拌する。次に溶液に0.47 gのジチオスレイトール(VWR)を加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に737 mgの白色固体であるPVA-A-SH1を得て、収率は73.7%である。
200 mLのビーカーに調製例24の方法により調製された1 gのPVA-MA、100 mLの脱イオン水を加え、PVA-MAが完全に溶解するまで溶液を加熱して撹拌する。次に溶液に0.47 gのジチオスレイトール(VWR)を加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に718 mgの白色固体であるPVA-MA-SH1を得て、収率は71.8%である。
200 mLのビーカーに5 gの超分岐PEGであるHB-PEG(Mw=20 kDa、Blafar Ltdから購入)、0.86 gのジチオスレイトール(VWR)、100 mLの脱イオン水を加え、室温で撹拌して溶解させる。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ2 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に3.84 gの無色粘性液体であるHB-PEG-SH1を得て、収率は76.8%である。

調製例35 スルフヒドリル-アクリレートにより修飾されたヒアルロン酸(HA-A2-SH5と略称)の合成
200 mLのビーカーに調製例2の方法により調製された1 gのHA-A2、0.52 gの1,3-ベンゼンジチオール(Sigma社から購入)、100 mLの脱イオン水を加え、室温で撹拌して溶解させ、透明溶液を得る。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に836 mgの白色綿状固体であるHA-A2-SH5を得て、収率は83.6%である。

200 mLのビーカーに調製例2の方法により調製された1 gのHA-A2、0.74 gのトリメチロールプロパン-トリス(3-メルカプトプロピオネート)(Sigma社から購入)、50 mLの脱イオン水及び50 mLのジメチルホルムアミドを加え、室温で撹拌して溶解させ、透明溶液を得る。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に785 mgの白色綿状固体であるHA-A2-SH8を得て、収率は78.5%である。
200 mLのビーカーに調製例3の方法により調製された1 gのHA-MA1、0.50 gの1,3-ベンゼンジチオール(Sigma社から購入)、100 mLの脱イオン水を加え、室温で撹拌して溶解させ、透明溶液を得る。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に828 mgの白色綿状固体であるHA-MA1-SH5を得て、収率は82.8%である。

200 mLのビーカーに調製例4の方法により調製された1 gのHA-MA2、0.92 gのメルカプトポリエチレングリコール(Sigma社から購入)、100 mLの脱イオン水を加え、室温で撹拌して溶解させ、透明溶液を得る。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に876 mgの白色綿状固体であるHA-MA2-SH7を得て、収率は87.6%である。
200 mLのビーカーに調製例4の方法により調製された1 gのHA-MA2、0.68 gのトリメチロールプロパン-トリス(3-メルカプトプロピオネート)(Sigma社から購入)、50 mLの脱イオン水及び50 mLのジメチルホルムアミドを加え、室温で撹拌して溶解させ、透明溶液を得る。得られた透明溶液を12時間撹拌し続ける。上記溶液を透析バッグ(分子量カットオフ8 kDa、Spectrumlabs)に入れ、5 LのpH=4の塩酸溶液で5日間透析し、1日2回水を交換する。最後に透析バッグ内の溶液を収集し、凍結乾燥後に825 mgの白色綿状固体であるHA-MA2-SH8を得て、収率は82.5%である。
調製例1、2、9、11、13で調製されたいずれか1種のアクリロイル化高分子化合物又はエチレングリコールジアクリレート(EGDA)10 mgを1 mLのリン酸塩緩衝溶液(pH=7.4)に溶解させ、質量体積濃度が1%のシリーズ溶液Aを得る。
実施例1で調製された2 mLのヒドロゲルの混合溶液を円筒型モールドに入れ、室温で24時間架橋した後に取り出して架橋サンプルの貯蔵弾性率を測定し、各群のサンプルについて3回検出する。検出機器はTA-DHR2レオメータであり、検出プローブは20 mmの平行板プローブであり、検出温度:25℃、せん断周波数:1Hzである。テスト結果は表2に示される。
本実施例1で調製されたヒドロゲルを、事前に重量が秤量された20 mLのガラス瓶に入れ、質量減算法によりヒドロゲルの質量を得て、m0として記録する。ガラス瓶を37℃のシェーカーに入れ、一定の時間間隔で質量を秤量し、ヒドロゲルのリアルタイムの質量を得て、mtとして記録する。ヒドロゲルの保水能力は、次の式により計算される:
保水率(%)=mt/m0×100%
保水率の結果は表3を参照する。
分解安定性テスト:温度37±0.1℃、相対湿度65%±5%の実験条件下で、実施例1で調製されたヒドロゲルに10 mlのPBS溶液を添加する。初期時点でのヒドロゲルの重量をm0として記録し、分解実験開始後の1、4、8、16週間でヒドロゲルの重量を秤量し、mtとして記録する。ヒドロゲルの分解率は次の式により計算される:
分解率(%)=(m0-mt)/m0×100%
ヒドロゲルの体外分解テストの結果は表4を参照する。
「GBT 16886.5-2017医療機器生物学的評価+パート5+インビトロ細胞毒性試験」の基準を参照して、本発明のHA-SHの細胞活性と生体適合性を測定する。具体的には、以下のMTT法を使用する。MTT法はMTT比色法とも呼ばれ、細胞の生存と増殖の検出方法である。検出原理は、生細胞のミトコンドリアにあるコハク酸デヒドロゲナーゼが外因性MTTを水不溶性の青紫色結晶ホルマザン(Formazan)に還元して細胞内に沈着させることができるが、死んだ細胞にはそのような機能がないことである。ジメチルスルホキシド(DMSO)は細胞内のホルマザンを溶解させることができ、酵素結合免疫測定計により490 nmの波長での光吸収値を測定することで、生細胞の数を間接的に反映することができる。細胞数の特定の範囲内で、MTT結晶形成の量は細胞の数に比例する。具体的な試験プロセスと結果は以下のとおりである。
C57BL/6マウスに麻酔をかけた後、背中の毛を取り除き、従来の方法で消毒し、120 μLの実施例1に記載の方法で調製されたHA-A1とHA-A1-SH1溶液、HA-A2とHA-A2-SH1溶液(濃度がそれぞれ10 mg/mLである)、即ち、それぞれ12 mgの各材料を、1.2 mLのPBS(リン酸塩緩衝生理食塩水)溶液に溶解させ、均一に振とうし、試験用サンプルを得て、使用に備える。注射器で当該試験用サンプルをそれぞれ120 μL吸引し、均一に混合し、得られたヒドロゲル前駆体溶液を24G針によりマウスの背中の皮下部位に注射する。同じ方法で同じ体積の生理食塩水をマウスの背中の皮下に注射する。
C57BL/6マウスに麻酔をかけた後、背中の毛を取り除き、従来の方法で消毒し、120 μLの実施例1に記載の方法で調製されたHA-A1とHA-A1-SH1溶液、HA-A2とHA-A2-SH1溶液(濃度がそれぞれ10 mg/mLである)、即ち、それぞれ12 mgの各材料を、1.2 mLのPBS溶液に溶解させ、均一に振とうし、試験用サンプルを得て、使用に備える。注射器で当該試験用サンプルをそれぞれ120 μL吸引し、均一に混合し、得られたヒドロゲル前駆体溶液を24G針によりマウスの背中の皮下部位に注射する。注射の60分後に1匹のマウスを安楽死させ、マウス皮下でのヒドロゲルのイン・サイチュ形成状況及び形態を観察する。従来の方法で残りの動物を飼育し続け、注射後の4週目、8週目、12週目に注射部位の写真を取り、ゲル体積の変化を観察して記録する。同じ方法で同じ体積の生理食塩水をマウスの背中の皮下に注射する。
Claims (10)
- スルフヒドリル変性高分子化合物を含むシステムのゲル化により調製して得られるヒドロゲルであって、
前記スルフヒドリル変性高分子化合物は、下記シリーズ化合物であるシリーズスルフヒドリル変性高分子化合物のうちの少なくとも1種であり、
前記シリーズスルフヒドリル変性高分子化合物は、その変性された高分子化合物の構造に-COOH、-NH2、-OH、式aに示されるアクリレート基、式bに示されるアクリルアミド基、式cに示されるアクリロイル基のうちの少なくとも1種を含み、
前記-COOH及び/又は-NH2及び/又は-OH及び/又はアクリレート基及び/又はアクリルアミド基及び/又はアクリロイル基の一部又は全ては、末端基が下記基である側鎖を形成するように修飾され、
上記基において、*は結合点を表し、R1は、水素、ハロゲン、脂肪族基、芳香族基などから選ばれ、R2とR3は、相同又は相異であり、互いに独立して水素、ハロゲン、脂肪族基、芳香族基などから選ばれ、R4は、ポリスルフヒドリル化合物断片であり、
前記システムには、更に、
C1.アクリロイル化高分子化合物、
C2.アクリロイル基を含む低分子架橋剤、の少なくとも1種を含む、
ことを特徴とするヒドロゲル。 - 前記物質C2である、アクリロイル基を含む低分子架橋剤は、アクリロイル基を含む低分子化合物又はアクリロイル基を含むオリゴマーを含むが、これらに限定されず、具体的には、エチレングリコールジアクリレートEGDA、ポリエチレングリコールジアクリレートPEGDA、トリメチロールプロパントリアクリレートTMPTA、ペンタエリスリトールトリアクリレートPTA、ペンタエリスリトールテトラアクリレートPTTA、ジ(トリメチロールプロパン)テトラアクリレートDTTAなどのうちの1種又は複数種から選ばれる、ことを特徴とする請求項1~6のいずれか1項に記載のヒドロゲル。
- 請求項1~8のいずれか1項に記載のヒドロゲルの調製方法であって、
この調製方法は、
(i)前記スルフヒドリル変性高分子化合物、及び
(ii)物質C1と物質C2の両方のうちの少なくとも1種
を含むシステムをゲル化することにより、前記ヒドロゲルを調製するステップを含み、
好ましくは、前記スルフヒドリル変性高分子化合物の溶液、前記アクリロイル化高分子化合物の溶液、前記低分子架橋剤の溶液及び任意選択的な他の生体機能材料、薬物、成長因子又は細胞懸濁液のうちの少なくとも1種の溶液をそれぞれ配合し、混合し、ゲル化することにより、前記ヒドロゲルを調製し、
任意選択的に、前記他の生体機能材料、薬物、成長因子又は細胞懸濁液のうちの少なくとも1種は、前記スルフヒドリル変性高分子化合物の溶液又は前記アクリロイル化高分子化合物の溶液又は前記低分子架橋剤の溶液に直接添加することによって導入されてもよく、
好ましくは、ヒドロゲル調製プロセスにおいて、前記スルフヒドリル変性高分子化合物の溶液を前記アクリロイル化高分子化合物の溶液及び/又は前記低分子架橋剤の溶液に加えてもよく、前記アクリロイル化高分子化合物の溶液及び/又は前記低分子架橋剤の溶液を前記スルフヒドリル変性高分子化合物の溶液に加えてもよく、具体的には、2つの溶液は、通常の注射器で混合されてもよく、二重針注射器で混合されてもよく、他の方法で混合されてもよい、
ことを特徴とする請求項1~8のいずれか1項に記載のヒドロゲルの調製方法。 - 請求項1~8のいずれか1項に記載のヒドロゲルの用途であって、
生物医学、医療美容と形成外科、及び化粧品などの分野に適用され、具体的には、薬物送達システムの調製、軟組織創傷用の修復被覆材、骨修復用の足場材料、眼科手術における支持用の粘弾剤、手術後の組織癒着を防止するための材料、及び3Dバイオプリンティング用の足場材料などに適用可能である、ことを特徴とする請求項1~8のいずれか1項に記載のヒドロゲルの用途。
