Wntシグナル伝達経路作用物質を含む、皮膚外用剤。
[2]
Wntシグナル伝達経路作用物質がGSK3β阻害薬である、[1]に記載の皮膚外用剤。

GSK3β阻害薬がリチウム塩である、[2]に記載の皮膚外用剤。

リチウム塩が、塩化リチウム、炭酸リチウム、クエン酸リチウム、オロチン酸リチウム、フッ化リチウム、臭化リチウム、酢酸リチウム、コハク酸リチウム、グルコン酸リチウム、ヨウ化リチウム、硫酸リチウム、硝酸リチウム及びスルホン酸リチウムからなる群から選択される、[3]に記載の皮膚外用剤。

リチウム塩が、塩化リチウム、炭酸リチウム、クエン酸リチウム又はオロチン酸リチウムである、[4]に記載の皮膚外用剤。

本発明は、Wntシグナル伝達経路作用物質を含む、皮膚外用剤に関する。本明細書において、「Wntシグナル伝達経路作用物質」とは、Wntにより媒介されるシグナル伝達を増強し得る物質を意味する。Wntシグナル伝達経路作用物質としては、医薬として許容されるものであれば特に限定されず、例えば、Wntファミリーに属するタンパク質(例:Wnt1、Wnt3、Wnt3a、Wnt3b、Wnt4、Wnt7、Wnt7a、Wnt7b、Wnt10b)、Wnt受容体、Wnt受容体アゴニスト、抗Wnt受容体抗体、Wnt部分ペプチド、β-カテニンシグナル伝達物質、GSK3β阻害薬(例:リチウム塩、6-Bromoindirubin-3'-oxime(BIO)、CHIR99021、Kenpaullone、)等を挙げることができる。
心電図取得用クリーム(フクダ電子株式会社製 #OJE-01D)を基材として用い、試験薬を作製した。上記クリームに対して、5M CaCl2(富士フィルム、#038-24985)溶液もしくは、5M LiCl(ナカライテスク 20623-85)溶液を容量/重量比で10%となるように加え、良く撹拌し混合した。この試験薬を雄ヌードマウス8週令(N=4)(日本クレア)の後足先端部に毎日1回1カ月間塗布した。
心電図取得用クリーム(フクダ電子株式会社製 #OJE-01D)を基材として用い、試験薬を作製した。このクリームに対して、5M CaCl2(富士フィルム、#038-24985)溶液もしくは、5M LiCl(ナカライテスク 20623-85)溶液を容量/重量比で10%となるように加え、良く撹拌し混合した。このクリームを雄ヌードマウス8週令(N=4)の背部に刷毛を用いて毎日1回1カ月間塗布した。犠牲死処理後に切除した背部皮膚は4%パラフォルムアルデヒドを用いて固定し、凍結切片を作製した。凍結切片に対して、核染色:DAPI(同人化学社製 #NN280)用いた。観察は、顕微鏡(NIKON社製Ti2及び解析ソフトNiS)を用いた。それぞれのサンプルに対し、毛の長さを測定し、グラフ化した。
ヒトiPS細胞(253G1株:RIKEN細胞バンク# HPS0002)に対して恒常的に緑色蛍光タンパク質:EGFPを発現させるように遺伝子導入した細胞亜株に対し未分化を維持しながら培養した。上記のように培養した細胞亜株の2つの培養皿の一方に対し、終濃度10 mMとなるようにフィルター滅菌したLiCl(ナカライテスク 20623-85)を添加し2時間培養した。続いて、4%パラフォルムアルデヒドを用いて固定化を実施し、続いてマウス抗beta-Catenin抗体(Becton Dickinson #610154)を1次抗体、ロバ抗マウスIgG抗体(蛍光物質Alexa546を共有結合: Invitrogen社製 #A1003b)、核染色にDAPI(同人化学社製 #NN280)用いて免疫染色を行った。

- Wntシグナル伝達経路作用物質を含む、皮膚外用剤。
- Wntシグナル伝達経路作用物質がGSK3β阻害薬である、請求項1に記載の皮膚外用剤。
- GSK3β阻害薬がリチウム塩である、請求項2に記載の皮膚外用剤。
- リチウム塩が、塩化リチウム、炭酸リチウム、クエン酸リチウム、オロチン酸リチウム、フッ化リチウム、臭化リチウム、酢酸リチウム、コハク酸リチウム、グルコン酸リチウム、ヨウ化リチウム、硫酸リチウム、硝酸リチウム及びスルホン酸リチウムからなる群から選択される、請求項3に記載の皮膚外用剤。
- リチウム塩が、塩化リチウム、炭酸リチウム、クエン酸リチウム又はオロチン酸リチウムである、請求項4に記載の皮膚外用剤。
- リチウム塩が塩化リチウムである、請求項5に記載の皮膚外用剤。
- 爪幹細胞の増殖促進用、又は毛髪の伸長促進用である、請求項1~6のいずれか一項に記載の皮膚外用剤。
- 爪幹細胞の増殖促進用である、請求項7に記載の皮膚外用剤。
- 爪幹細胞の増殖促進が爪及び甘皮の伸長促進である、請求項8に記載の皮膚外用剤。
