Abstract
Description

以上に記載の系は、様々な方法を使用して有効に利用できる。系は、定常状態と、物理的または化学的な刺激に応答するときとの両方で、細胞シグナル伝達経路の活性を照合する方法において有用である。レポーターエレメントが特定のレポーターエレメントに結合したUMI配列を含む場合、系は多重アッセイで展開できる。
合成転写因子は、遺伝子の発現を標的して調節可能な人工タンパク質である。いくつかの合成転写因子は、複数の異なる遺伝子由来のドメインを含有するキメラタンパク質である。ある実施形態では、合成転写因子は、1つの遺伝子由来のDNA結合ドメインおよび別の遺伝子由来の転写調節ドメインを含む。
合成転写因子プロモーターヌクレオチド配列は、合成転写因子によって結合可能な核酸の配列である。ある実施形態では、前記合成転写因子ヌクレオチド配列は、内因性の転写因子によって結合されない。前記合成転写因子プロモーターヌクレオチド配列は、レポーター分子の転写を活性化するために前記合成転写因子の補充を補助する。前記レポーター分子は、前記合成転写因子プロモーターヌクレオチド配列の3’に位置する核酸にコードされる。
レポーター核酸は、合成転写因子によって結合できる調節エレメントと、レポーターをコードするヌクレオチド配列とを最低限含む。レポーターをコードする前記ヌクレオチド配列は、前記合成転写因子によって結合できる前記調節エレメントの下流である。前記合成転写因子は、前記レポーターの発現を調節する。
レポーター分子の活性化は、ルシフェラーゼタンパク質、β-ガラクトシダーゼタンパク質、β-グルクロニダーゼタンパク質、クロラムフェニコールアセチルトランスフェラーゼタンパク質、分泌型胎盤アルカリホスファターゼタンパク質を検出するために標準的なアッセイを使用して決定できる。概して、基質に対するタンパク質酵素活性に基づいて検出可能なシグナルが産生される酵素アッセイがある。例えば、ルシフェラーゼ発現は、ルシフェラーゼ基質が存在するときはルミノメーターによって測定できる。蛍光レポーターは、基質を必要とせず、シグナルは、蛍光顕微鏡法または蛍光プレートリーダによって測定できる。蛍光レポーターは、生細胞におけるレポーターの活性化を測定するのに特に有用である。

本明細書に記載の方法において有用な細胞は、概して、合成転写因子およびレポーターエレメントをコードする1つ以上の外因性の核酸をトランスジェニックに容易に付与できるものである。合成転写因子およびレポーターエレメントをコードする系の核酸は、リン酸カルシウムトランスフェクション、脂肪系トランスフェクション(例えばLipofectamine(商標)、Lipofectamine-2000(商標)、Lipofectamine-3000(商標)またはFugene(登録商標)HD)、エレクトロポレーションまたはウイルス形質導入などの当技術分野において既知の方法を使用して好適な細胞株にトランスフェクトまたは形質導入できる。細胞はまた、適切な組織培養容器でコンフルエントまたはほぼコンフルエントになるまで増殖させられた同じ種類の細胞集団であってもよい。
本発明のポリヌクレオチド配列は、細胞にトランスフェクトされると利用できる。トランスフェクションは、リポフェクチンを含むがこれらに限らない様々なトランスフェクション剤、リン酸カルシウム沈澱、ウイルス形質導入またはエレクトロポレーションによって達成できる。トランスフェクションは、一過性のものであるか、あるいは安定的なものであってもよい。トランスフェクションが安定的なものである実施形態では、安定的にトランスフェクトされた細胞は、後に使用するために凍結またはバンクできる。
この実施例では、GPCRシグナル伝達を誘導する潜在的な化合物についてスクリーニングするために、図1Aおよび図1Bに構成されている核酸を含む転写リレー系を使用する。この実施例では、図1Aの核酸は、cAMP応答エレメント(CRE)の活性化を含み、これは、結果として(Gal4 DNA結合ドメインと、キメラ活性化ドメインVP64-p65-Rtaとを含む)合成転写因子Gal4-VPRの発現をもたらす。図1Bの核酸は、Gal4-VPR合成転写因子によって結合されて活性化できるプロモーターを含み、これは、結果としてルシフェラーゼ遺伝子と、UMIをコードする遺伝子とを含むレポーターエレメントの発現をもたらす。使用される細胞は、図1Aおよび図1Bの系をコードする、安定的に組み込まれた核酸と、所与のGPCRとを含む。各UMIは、所与のGPCRに対応付けられ、CREの発現が、特定のGPCRにマップされることを可能にする。これによって、アッセイの多重化が可能になる。
この実施例では、GPCRシグナル伝達を誘導する潜在的な化合物についてスクリーニングするために、図1Aおよび図1Bに構成されている核酸を含む転写リレー系を使用する。この実施例では、図1Aの核酸は、活性化T細胞応答エレメントの核因子(NFAT)の活性化を含み、これは、結果として(Gal4 DNA結合ドメインと、キメラ活性化ドメインVP64-p65-Rtaとを含む)合成転写因子Gal4-VPRの発現をもたらす。図1Bの核酸は、Gal4-VPR合成転写因子によって結合されて活性化できるプロモーターを含み、これは、結果としてルシフェラーゼ遺伝子と、UMIをコードする遺伝子とを含むレポーターエレメントの発現をもたらす。使用される細胞は、図1Aおよび図1Bの系をコードする、安定的に組み込まれた核酸と、所与のGPCRとを含む。各UMIは、所与のGPCRに対応付けられ、CREの発現が、特定のGPCRにマップされることを可能にする。これによって、アッセイの多重化が可能になる。

この実施例の実験では、転写リレー系を使用すると、転写リレーを有さない系と比較して、ルシフェラーゼシグナルが増加し、ルシフェラーゼシグナルの変動係数が低下することを示す。半無作為に組み込まれたCRE-Gal4-VPRの複数のコピーと共に、単一で組み込まれたCRE-ルシフェラーゼを保有するHEK293由来の細胞、または、単一で組み込まれたUAS-ルシフェラーゼを保有する細胞を、白色壁のポリ-L-リシンをコートした96ウェルプレートの100μLのDMEM+10%FBSに30,000細胞/ウェルでプレーティングする。45ngのドキシサイクリンを有する50μLのOpti-memを細胞の上に加えた。24時間後に、DMSOを加えた。表示された期間にわたって、細胞をDMSOで処理した。表示されたインキュベーション時間の後に、培地を吸引して、35μLのDMEMと置き換え、そして、製造業者の説明書に従ってBright-Glo Luciferase Assayキット[Promega]を使用して、細胞をアッセイした。半無作為に組み込まれたCRE-Gal4-VPRの複数のコピーと共に、結果として生じる、単一で組み込まれたCRE-ルシフェラーゼを保有する細胞(灰色)と、単一で組み込まれたUAS-ルシフェラーゼを保有する細胞(黒色)のルシフェラーゼ活性化の発現を図2に示す。実験は技術的に3回繰り返して実施し、図3に各試料の変動係数を算出した。
この実施例の実験では、転写リレー系のGal4-VPRにデグロンタグを含むと、ルシフェラーゼシグナルの誘導倍率が増加することを示す。単一で組み込まれたTRE-CHRM3::UAS-ルシフェラーゼ二重遺伝子カセットと、多重に半無作為に組み込まれたFOS-Gal4-VPR-CP(デグロン)またはFOS-Gal4-VPR(デグロンなし)とを保有するHEK293由来の細胞を、白色壁のポリ-L-リシンをコートした96ウェルプレートの100μLのDMEM+10%FBSに30,000細胞/ウェルでプレーティングした。45ngのドキシサイクリンを有する50μLのOpti-memを細胞の上に加えた。24時間後に、8時間にわたって、細胞をDMSOまたは1μMのカルバコールで処理した。表示されたインキュベーション時間の後に、培地を吸引して、35μLのDMEMと置き換え、そして、製造業者の説明書に従ってBright-Glo Luciferase Assayキット[Promega]を使用して、細胞をアッセイした。結果としてもたらされる、カルバコールにおけるルシフェラーゼ活性対DMSOにおけるルシフェラーゼ活性の比率を、図4にプロットする。
この実施例に記載の細胞株は、組み込まれたNFAT-応答エレメント転写リレー(合成転写因子のNFATプロモーター駆動転写)のコピーを有する。これらの細胞株は、コピー数および組み込み部位の面で異種の遺伝子プールとして生成した。このプールから、単一の細胞クローンを単離して拡張させた。これらの株をさらに使用して、GPCRおよびUAS-ルシフェラーゼバーコードレポーターを組み込み、NFATシグナル伝達の多重化を検出する能力を試験した。これらの10の細胞ライブラリから、コントロールアゴニストに対して、最も大きい数の明確なGPCRヒットを検出することができたものを2つ同定したが、これらは、図5に示しているように cb29(クローンc713から構成されている)およびcb37(クローンc708から構成されている)である。
Claims (40)
- 転写リレー系であって、
a)応答エレメント調節プロモーターヌクレオチド配列と、合成転写因子をコードするヌクレオチド配列とを含む転写因子核酸であって、前記応答エレメント調節プロモーターヌクレオチド配列は、前記合成転写因子をコードする前記ヌクレオチド配列の5’である、転写因子核酸と、
b)合成転写因子プロモーターヌクレオチド配列と、レポーターをコードするヌクレオチド配列とを含むレポーター核酸であって、前記合成転写因子プロモーターヌクレオチド配列は、前記レポーターをコードする前記ヌクレオチド配列の5’であり、前記合成転写因子プロモーターヌクレオチド配列は、前記合成転写因子によって結合できる、レポーター核酸と、
を含む、転写リレー系。 - 前記応答エレメント調節プロモーターヌクレオチド配列は、cAMP応答エレメントヌクレオチド配列、NFAT転写因子応答エレメントヌクレオチド配列、FOSプロモーターヌクレオチド配列または血清応答エレメントヌクレオチド配列を含む、請求項1に記載の転写リレー系。
- 前記合成転写因子は、第1の転写因子由来のDNA結合ドメインおよび第2の転写因子由来の転写活性化ドメインを含む、請求項1または2に記載の転写リレー系。
- 前記DNA結合ドメインは、Gal4、PPR1、Lac9またはLexA由来である、請求項3に記載の転写リレー系。
- 前記DNA結合ドメインは、配列番号1に示す配列と少なくとも約90%同一であるアミノ酸配列を含む、請求項4に記載の転写リレー系。
- 前記DNA結合ドメインは、配列番号1に示す配列と少なくとも約95%同一であるアミノ酸配列を含む、請求項4に記載の転写リレー系。
- 前記DNA結合ドメインは、配列番号1に示す配列と同一であるアミノ酸配列を含む、請求項4に記載の転写リレー系。
- 前記DNA結合ドメインは、配列番号1のアミノ酸配列変異を含む、請求項5に記載の転写リレー系。
- 前記転写活性化ドメインは、VP64、p65およびRtaを含む、請求項3に記載の転写リレー系。
- 前記転写活性化ドメインは、配列番号14に示す配列と少なくとも約90%同一であるアミノ酸配列を含む、請求項9に記載の転写リレー系。
- 前記転写活性化ドメインは、配列番号14に示す配列と少なくとも約95%同一であるアミノ酸配列を含む、請求項9に記載の転写リレー系。

- 前記合成転写因子プロモーターヌクレオチド配列は、Gal4、PPR1、Lac9またはLexAによって結合できるヌクレオチド配列を含む、請求項1~18のいずれか1項に記載の転写リレー系。
- 前記レポーターは、蛍光タンパク質、ルシフェラーゼタンパク質、β-ガラクトシダーゼ、β-グルクロニダーゼ、クロラムフェニコールアセチルトランスフェラーゼ、分泌型胎盤アルカリホスファターゼまたは固有の分子識別子を含む、請求項1~19のいずれか1項に記載の転写リレー系。
- 前記レポーターは、蛍光タンパク質、ルシフェラーゼタンパク質、β-ガラクトシダーゼ、β-グルクロニダーゼ、クロラムフェニコールアセチルトランスフェラーゼまたは分泌型胎盤アルカリホスファターゼおよび固有の分子識別子を含む、請求項20に記載の転写リレー系。
- 前記固有の分子識別子は、試験ポリペプチドに特有であり、前記試験ポリペプチドは、前記レポーター核酸によってコードされる、請求項20または21に記載の転写リレー系。
- 前記転写因子核酸は、前記転写抑制因子によって結合できる、前記応答エレメント調節プロモーターヌクレオチド配列の近位にあるヌクレオチド配列を含む、請求項1~22のいずれか1項に記載の転写リレー系。
- 前記転写因子核酸は、前記合成転写因子をコードする前記ヌクレオチド配列によってコードされるmRNAの5’非翻訳領域を伸長させる、前記応答エレメント調節プロモーターヌクレオチド配列の近位にあるヌクレオチド配列を含む、請求項23に記載の転写リレー系。
- 前記合成転写因子をコードする前記ヌクレオチド配列によってコードされるmRNAの前記5’非翻訳領域は、前記合成転写因子の翻訳を低減する1つ以上の配列を含む、請求項24に記載の転写リレー系。
- 前記転写因子核酸および前記レポーター核酸は、単一の核酸の成分である、請求項1~25のいずれか1項に記載の転写リレー系。
