Abstract
Description
本発明は、VHドメインとVLドメインとの同族対によって形成された抗原結合部位を含むVEGFに結合する抗体のVEGF-R1へのVEGF結合阻害を改善する方法であって、該抗体がVEGF分子中のVEGF-R1結合領域およびVEGF-R2結合領域と重複するVEGFのエピトープに結合することを特徴とする方法に関する。本発明の方法は、
(a)VEGFに結合する当該抗体の第1の抗原結合部位および第2の抗原結合部位によって結合されたVEGF二量体の複合体(VEGF二量体-抗体複合体)の三次構造の分析を提供すること;
(b)当該抗体のVHドメインまたはVLドメインに位置する少なくとも1つのアミノ酸残基を同定すること、ここで第1抗原結合部位内の当該アミノ酸残基および第2抗原結合部位内の当該アミノ酸残基が、VEGF二量体-抗原複合体において空間的に近接して配置される;および
(c)工程(b)で同定された当該少なくとも1つのアミノ酸残基を、
i)より小さい側鎖体積を有するアミノ酸で;および/または
ii)異なる電荷の側鎖を有するアミノで
置換すること
を含むものである。本発明の方法では、特定の抗VEGF抗体によって媒介されるVEGF-R1へのVEGF結合の阻害は、特に抗原結合に関与していない領域において、そのアミノ酸配列のわずかな修飾によって改善することができる。
1.定義
本明細書で特に定義されていない限り、本発明に関連して使用される科学技術用語は、当業者に共通して理解される意味を持つものとする。さらに、文脈上特に必要とされない限り、単数形の用語は複数形を含み、複数形の用語は単数形を含むものとする。本開示の方法および技術は、一般に、当技術分野でよく知られている従来の方法に従って行われる。一般に、本明細書に記載されている生化学、酵素学、分子生物学、および細胞生物学、微生物学、遺伝学、およびタンパク質および核酸の化学、ならびにハイブリダイゼーションに関連して使用される命名法ならびに技術は、当技術分野でよく知られ、一般的に使用されているものである。
(1)疎水性の側鎖:ノルロイシン、Met(M)、Ala(A)、Val(V)、Leu(L)、Ile(I);
電荷を持たない親水性側鎖(当技術分野では「中性」の親水性側鎖とも呼ばれる):Cys(C)、Ser(S)、Thr(T)、Asn(N)、Gln(Q);
負に帯電した側鎖(当技術分野では「酸性」側鎖とも呼ばれる):Asp(D)、Glu(E);
正に帯電した側鎖(当技術分野では「塩基性」側鎖とも呼ばれる):His(H)、Lys(K)、Arg(R);
芳香族側鎖:Trp(W)、Tyr(Y)、Phe(F)、および鎖の配向に影響を与える残基を含む側鎖:Gly(G),Pro(P)。
本発明は、VHドメインとVLドメインとの同族対によって形成された抗原結合部位を含むVEGFに結合する抗体のVEGF-R1へのVEGF結合の阻害を改善する方法であって、該抗体がVEGF分子中のVEGF-R1結合領域およびVEGF-R2結合領域と重複するVEGFのエピトープに結合する方法に関する。本発明の方法は、
(a)VEGFに結合する当該抗体の第1の抗原結合部位および第2の抗原結合部位によって結合されたVEGF二量体の複合体(VEGF二量体-抗体複合体)の三次構造の分析を提供すること、
(b)当該抗体のVHドメインまたはVLドメインに位置する少なくとも1つのアミノ酸残基を同定すること、ここで第1抗原結合部位内の当該アミノ酸残基および第2抗原結合部位内の当該アミノ酸残基が、VEGF二量体-抗原複合体において空間的に近接して配置される;および
(c)工程(b)で同定された当該少なくとも1つのアミノ酸残基を、
i)より小さい側鎖体積を有するアミノ酸で、および/または
ii)異なる電荷の側鎖を有するアミノ酸で
置換すること、
を含む。
(a)配列番号03のアミノ酸配列を含むCDR-H1;
(b)配列番号04のアミノ酸配列を含むCDR-H2;
(c)配列番号05のアミノ酸配列を含むCDR-H3;
(d)配列番号06のアミノ酸配列を含むCDR-L1;
(e)配列番号07のアミノ酸配列を含むCDR-L2;および
(f)配列番号08のアミノ酸配列を含むCDR-L3
を含む。このセットのCDRアミノ酸配列を含む1つの例示的な抗体は、本明細書で「VEGF-0089」と呼ばれる抗体である。一実施形態では、当該抗体は、配列番号01を含むVHと、配列番号02を含むVLとを特徴とする。
生成されたヒト抗VEGF抗体(抗体VEGF-0089)の特性決定
本明細書で使用される抗体「VEGF-0089」は、EP 17211032.2に概説されているように、Rocheが知的財産権を有するトランスジェニックウサギに由来する。VEGF-0089は、配列番号01のVHドメインと配列番号02のVLドメインを含む。その後の分析のために、抗体VEGF-0089は、ヒトのVHドメインおよびVLドメインと、ウサギ由来の軽鎖(CLκ)および重鎖(CH1)の定常ドメインとを有するFab断片(本明細書では「VEGF-0089 Fab断片」または単に「VEGF-0089 Fab」と呼ぶ)として生成された。VEGF-0089 Fab断片の重鎖のアミノ酸配列は、配列番号10である。VEGF-0089 Fab断片の軽鎖のアミノ酸配列は、配列番号11である。
抗His捕捉抗体(anti-His capturing antibody :GE Healthcare 28995056)を、標準的なアミンカップリング化学を用いてSeries S Sensor Chip C1(GE Healthcare 29104990)に固定化し、500~1000共鳴単位(RU)の表面密度を得た。ランニングバッファーおよび希釈バッファーとして、HBS-P+(10mMのHEPES、150mMのNaCl pH7.4、0.05%の界面活性剤P20)を使用し、測定温度はそれぞれ25℃と37℃に設定した。hVEGF-A121を表面に捕捉した結果、捕捉レベルの範囲は5~35RUとなった。抗VEGF抗体の希釈シリーズ(0.37~30nM)を120秒間注入し、30μl/minの流速で少なくとも600秒間解離をモニターした。10mMのグリシン pH1.5を60秒間注入して表面を再生した。バルク屈折率の差は、ブランク注入を差し引くことで補正し、hVEGF-A121を捕捉していない対照フローセルから得られた応答を差し引くことで補正した。速度定数はBiacore EvaluationソフトウェアのLangmuir 1:1結合モデルを用いて算出した。
384ウェルのストレプトアビジンプレート(Nunc/Microcoat # 11974998001)に、0.25μg/mlのビオチン化VEGF-R1または0.5μg/mlのビオチン化VEGF-R2(自社製造、DPBS(1×)中、各25μl/ウェル(PAN,#P04-36500))をコーティングした。プレートを室温で1時間インキュベートした。並行して、0.7nMの濃度のVEGF-121-His(自社製造)を異なる希釈率の抗体とインキュベートした(12×1:2希釈ステップ、500nMの濃度から開始)。このプレインキュベーションステップは、384ウェルのPPプレート(Weidmann medical technology, # 23490-101)を用い、1×OSEPバッファ(ビデストウォーター、10×、Roche, # 11 666 789 001+0.5%のウシ結成アルブミンフラクションV、脂肪酸不含、Roche, # 10 735 086 001 + 0,05% Tween 20)中で行った。プレートを室温で1時間インキュベートした。VEGF-R1/VEGF-R2をコートしたストレプトアビジンプレートを90μl/ウェルのPBSTバッファー(ビデストウォーター、10×PBS Roche#11666789001 + 0.1% Tween 20)で3回洗浄した後、VEGF-抗体プレインキュベーションプレートから25μlのサンプルを、コートしたストレプトアビジンプレートに移し、その後、室温で1時間インキュベートした。90μl/ウェルのPBSTバッファーで3回洗浄した後、1xOSEPに25μl/ウェルの検出抗体(抗His POD, Bethyl, # A190-114P, 1:12000)を加えた。室温で1時間インキュベートした後、プレートを90μlのPBSTバッファーで3回洗浄した。25μlのTMB(Roche, #11 835 033 001)をすべてのウェルに同時に加えた。室温で10分間インキュベートした後、Tecan Safire 2 Readerで370nm/492nmの波長でシグナルを検出した。
抗体VEGF-0089とVEGF二量体との複合体のX線結晶構造解析およびエピトープ決定
上述のVEGF-0089 Fab断片の結晶構造を、当技術分野で知られている標準的な方法に従って解析した。
・VEGF二量体内の個々のVEGF-A121分子の1つでは、アミノ酸F17、M18、D19、Y21、Q22、R23、Y25、H27、P28、I29、E30、M55、N62、L66、N100、K101、C102、E103、C104、R105およびP106;ならびに
・VEGF二量体内の個々のVEGF-A121分子のうちのもう1つでは、アミノ酸E30、K48、M81およびQ87。
VEGF-0089のバリアントの提供
上述の理論に基づき、VEGF-0089抗体を、VEGF二量体に2つの抗体分子が同時に結合しやすくなるように改良した。
VEGF-0089の改良バリアントのVEGF結合親和性
抗体の親和性は、実施例1に記載したのと同じ方法でSPRにより測定した。示された抗体の結果を表3に示す。
Claims (16)
- VHドメインおよびVLドメインの同族対によって形成された抗原結合部位を含むVEGFに結合する抗体の、VEGF-R1へのVEGF結合阻害を改善する方法であって、ここで抗体は、VEGF分子中のVEGF-R1結合領域およびVEGF-R2結合領域と重複するVEGFのエピトープに結合するものであり、以下の工程:
a)VEGFに結合する前記抗体の第1の抗原結合部位および第2の抗原結合部位によって結合されたVEGF二量体の複合体(VEGF二量体-抗体複合体)の三次構造の分析を提供する工程;
b)前記抗体のVHドメインまたはVLドメインに位置する少なくとも1つのアミノ酸残基を同定する工程であって、第1抗原結合部位内の前記アミノ酸残基および第2抗原結合部位内の前記アミノ酸残基が、VEGF二量体-抗原複合体において空間的に近接して配置される工程;および
c)工程b)で同定された前記少なくとも1つのアミノ酸残基を、
i)より小さい側鎖体積を有するアミノ酸で;および/または
ii)異なる電荷の側鎖を有するアミノ酸で;
置換する工程、
を含む、方法。 - 抗体が、配列番号01のVHおよび配列番号02のVLにより特徴付けられる抗体と同一または重複するエピトープに結合する、請求項1に記載の方法。
- 抗体が、VEGF-A121の二量体上の構造的エピトープと同一または重複するエピトープに結合し、VEGF-A121が、配列番号21のアミノ酸配列を含み、エピトープが、
・VEGF二量体内の個々のVEGF-A121分子の一方において、アミノ酸F17、M18、D19、Y21、Q22、R23、Y25、H27、P28、I29、E30、M55、N62、L66、N100、K101、C102、E103、C104、R105およびP106;ならびに
・VEGF二量体内の個々のVEGF-A121分子のうちのもう一方において、アミノ酸E30、K48、M81およびQ87、
を含む、請求項1または2に記載の方法。 - より小さい側鎖体積を有するアミノ酸が、D、E、S、T、N、G、A、V、IおよびLから選択される、請求項1から3のいずれか一項に記載の方法。
- 正の電荷の側鎖を有するアミノ酸が、負の電荷の側鎖を有するアミノ酸で置換されるか、または電荷を持たない側鎖を有するアミノ酸で置換される、請求項1から4のいずれか一項に記載の方法。
- 負の電荷の側鎖を有するアミノ酸が、正の電荷の側鎖を有するアミノ酸で置換されるか、または電荷を持たない側鎖を有するアミノ酸で置換される、請求項1から5のいずれか一項に記載の方法。
- 少なくとも1つの置換されたアミノ酸残基が、前記抗体の重鎖可変ドメインに位置する、請求項1から6のいずれか一項に記載の方法。
- 少なくとも1つの置換されたアミノ酸残基が、前記抗体の重鎖CDR内に位置する、請求項1から7のいずれか一項に記載の方法。
- 少なくとも1つの置換されたアミノ酸残基が、前記抗体のH-CDR2内に位置する、請求項1から8のいずれか一項に記載の方法。
- 少なくとも1つの置き換えられたアミノ酸残基が、前記抗体の重鎖FR内に位置する、請求項1から9のいずれか一項に記載の方法。
- 少なくとも1つの置き換えられたアミノ酸残基が、前記抗体のH-FR3内に位置する、請求項1から10のいずれか一項に記載の方法。
- 抗体が、
(a)配列番号03のアミノ酸配列を含むCDR-H1;
(b)配列番号04のアミノ酸配列を含むCDR-H2;
(c)配列番号05のアミノ酸配列を含むCDR-H3;
(d)配列番号06のアミノ酸配列を含むCDR-L1;
(e)配列番号07のアミノ酸配列を含むCDR-L2;および
(f)配列番号08のアミノ酸配列を含むCDR-L3
を含む、請求項1から11のいずれか一項に記載の方法。 - 抗体が、配列番号01を含むVHおよび配列番号02を含むVLを含む、請求項1から12のいずれか一項に記載の方法。
