本発明は、概して、ハイブリッドキメラアンチセンスオリゴヌクレオチドに関し、より特には、Foxp3遺伝子の発現レベルを低減し、抗腫瘍活性を増加させ、およびがんを処置するための、デオキシリボヌクレオチドおよび2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチドを含むアンチセンスオリゴヌクレオチドの使用に関する。
がんは、著しい罹病率および死亡率に繋がる、抑制のない細胞増殖および転移により特徴付けられる不均一状態である。男性および女性の両方におけるがん関連死の4分の1超が肺がんからのものであり、これは2018年に米国において154,050人であると推定される。肺がんの全てのステージについての現行の5年生存率は、外科的介入、放射線、化学療法、および他の処置の進歩にもかかわらず、18.6%である。
本発明は、Foxp3 mRNAに結合する、デオキシリボヌクレオチドおよび2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチドを含むハイブリッドキメラアンチセンスオリゴヌクレオチドが、Foxp3遺伝子の発現レベルを低減するため、抗腫瘍活性を増加させるため、およびがんを処置するために使用され得るという独創的な発見に基づく。
A1−D1−A2−D2−A3−D3 ... Az−Dz
のように表されてもよく、ここで、A1、A2などのそれぞれは、1つまたは複数(例えば、1つまたは2つ)の糖修飾ヌクレオチド残基(例えば、2’−FANA)の単位を表し、D1、D2などのそれぞれは、1つまたは複数(例えば、1つまたは2つ)のDNA残基の単位を表す。各単位内の残基の数は、同じであるか、または単位毎に可変であってもよい。オリゴヌクレオチドは、奇数または偶数の単位を有してもよい。オリゴヌクレオチドは、糖修飾ヌクレオチド含有単位(例えば、2’−FANA含有単位)またはDNA含有単位のいずれかで開始(すなわち、その5’末端にある)してもよい。オリゴヌクレオチドは、糖修飾ヌクレオチド含有単位またはDNA含有単位のいずれかで終了(すなわち、その3’末端にある)してもよい。単位の総数は、わずか4(すなわち、各種類について少なくとも2つ)であってもよい。

からなる群から選択される構造を有し、ここで、m、xおよびyのそれぞれは、それぞれ独立して、1以上の整数であり、nは、2以上の数であり、Aは糖修飾ヌクレオチドであり、Dは2’−デオキシリボヌクレオチドである。
材料および方法
抗体およびフローサイトメトリー
商業的に入手可能なコンジュゲートモノクローナル抗体(mAb)をフローサイトメトリーのために使用した(BD Pharmingen)。抗Foxp3 mAbはFJK−16s(eBioscience)であり、βアクチン抗体はウサギmAb(Cell Signaling)であった。フローサイトメトリーはCyanフローサイトメーター(Beckman Coulter)で行い、FlowJo 8ソフトウェア(Tree−Star)を用いてデータを解析した。CD4+YFP+(Foxp3+)細胞およびCD4+YFP(Foxp3−)細胞をFACS Ariaセルソーター(BD Bioscience、UPenn Cell Sorting Facility)を使用して年齢および性別をマッチさせたFoxp3YFP−creマウスから選別した。
PrimeFlowは、フローサイトメトリーによるmRNAおよびタンパク質の同時の測定を可能とした。FOXP3 mAbと細胞とを、推奨される30分の代わりに1時間インキュベートしたことを除いて、製造者の説明書にしたがってPrimeFlow RNA Assay(Affymetrix)を使用した。
プールされたリンパ節および脾臓試料から新たに単離された、またはCD3/28 mAbコーティングビーズ(Invitrogen)を用いて単離および活性化されたFoxp3+ TregまたはFoxp3− TE細胞からのRNAをRNeasy Kits(Qiagen)を使用して得た。TaqMan逆転写試薬(Applied Biosystems)を用いてcDNAを合成した。TaqMan Universal PCR Master Mix(Applied Biosystems)、およびApplied Biosystemsからの特異的プライマーを使用してqPCRを行い、遺伝子発現データを18S RNAに対して正規化した。
CD4+ CD25− Tエフェクター(TE)およびCD4+ CD25+ Treg細胞をCD4+ CD25+ Treg isolation kit(130−091−041、Miltenyi Biotec)を使用してFoxp3YFP−creマウスから単離した。Cell Trace Violet標識またはCFSE標識Teff細胞(5×105)を、5×105個の照射同系T細胞枯渇脾臓細胞(130−049−101、Miltenyi Biotec)および様々な比のTregの存在下でCD3 mAb(5μg/ml)を用いて刺激した。72時間後に、TE細胞の増殖をCell Trace Violet希釈またはCFSE希釈の分析により決定した。
3つの異なる種類のインビボ実験を行った。一部の実験では、マウスを10mg/kgのFANAを用いて3回処置した。他の実験では、マウスを50mg/kgのFANAを用いて1日毎に1週間処置した。さらなる一部の他の実験では、TC1腫瘍モデルを使用して、マウスに50mg/kgのFANAを1日毎に2週間与えた。
TC1細胞は、HPV−16 E6およびE7を用いて不死化され、c−Ha−RASがん遺伝子を用いて形質転換されたマウス肺上皮細胞に由来した。腫瘍研究のために、各マウスの右脇腹を剃毛し、皮下に1.2×106個のTC1腫瘍細胞を注射した。腫瘍体積を式:(3.14×長軸×短軸×短軸)/6により決定した。
C57BL/6マウス(The Jackson Laboratory)にTC1腫瘍細胞を接種し、マウスを7日目に3つの群(10匹/群)に分けた。群1:スクランブル50mg/kg;群2:AUM−FANA−5 50mg/kg;群3:AUM−FANA−6 50mg/kg。オリゴヌクレオチドをPBS(10mg/ml)に溶解し、0.1ml(1mg)を14日間毎日腹腔内に与えた。腫瘍サイズをキャリパーを使用して測定し、腫瘍体積を週に2回算出し、実験の終わりに、腫瘍、灌流リンパ節および脾臓をさらなる分析のために回収した。
Foxp3発現細胞の数に対するFoxp3 FANAの効果の評価
Foxp3発現細胞の数に対するFoxp3 FANAの効果をフローサイトメトリーにより評価した。
FANAオリゴヌクレオチドの細胞内取込みの評価
FANAのインビボ取込みを、10mg/kgの蛍光(APC)標識スクランブル化オリゴヌクレオチドのマウスへの注射の24時間後に評価した。細胞を脾臓、リンパ節、および血液から回収し、フローサイトメトリーにより分析した。
Foxp3タンパク質発現レベルに対するFoxp3 FANAの効果の評価
Foxp3タンパク質発現レベルに対するFoxp3 FANAの効果をウエスタンブロットを介して評価して、Foxp3発現をタンパク質レベルで阻害するFoxp3 FANAの能力を評価した。
Treg免疫抑制機能に対するFoxp3 FANAの効果の評価
Treg細胞の免疫抑制機能をフローサイトメトリーにより評価し、該評価では、Tエフェクター細胞に対するFoxp3 FANA処理Tregの効果をTreg免疫抑制アッセイにおいて測定した。
50mg/kgのFoxp3 FANAオリゴヌクレオチドの効果のインビボ評価
50mg/kgという高用量のFoxp3 FANAオリゴヌクレオチドの効果を、Treg細胞の免疫抑制機能に対する影響力、およびFoxp3のタンパク質発現レベルに対する影響力などのいくつかのパラメーターを評価することによりインビボで評価した。
Foxp3 FANAオリゴヌクレオチドを用いたTC1マウスの処置のインビボ評価
選択されたFoxp3 FANAであるAUM−FANA−5(SEQ ID NO:25)およびAUM−FANA−6(SEQ ID NO:26)の腫瘍成長に対する効果を、実施例1に記載されるようにTC1腫瘍モデルを使用してインビボで評価した。さらに、腫瘍内および脾臓内Foxp3+ Tregに対するAUM−FANA−5およびAUM−FANA−6の効果をフローサイトメトリーにより評価した。
低用量のFoxp3 FANA6−Bオリゴヌクレオチドを用いたTC1マウスの処置のインビボ評価
選択されたFoxp3 FANAの効果を、より低い用量のオリゴヌクレオチドを使用してインビトロおよびインビボで評価した。
Claims (38)
- 少なくとも1つの2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチド(2’−FANA修飾ヌクレオチド)を含む修飾アンチセンスオリゴヌクレオチド(AON)であって、Foxp3 mRNAに結合する、AON。
- 少なくとも1つの2’−FANA修飾ヌクレオチドおよび少なくとも1つの非修飾デオキシリボヌクレオチドを含むハイブリッドキメラAONであり、前記AONが2’−FANA AONである、請求項1に記載のAON。
- 前記2’−FANA修飾ヌクレオチドが、式1〜16のいずれかにしたがって配置されている、請求項2に記載のAON。
- 前記2’−FANA修飾ヌクレオチドが、式6にしたがって配置されている、請求項3に記載のAON。
- 前記2’−FANA修飾ヌクレオチドのヌクレオチドの間のヌクレオチド間連結が、ホスホジエステル結合、ホスホトリエステル結合、ホスホロチオエート結合(5’O−P(S)O−3O−、5’S−P(O)O−3’−O−、および5’O−P(O)O−3’S−)、ホスホロジチオエート結合、Rp−ホスホロチオエート結合、Sp−ホスホロチオエート結合、ボラノホスフェート結合、メチレン結合(メチルイミノ)、アミド結合(3’−CH2−CO−NH−5’および3’−CH2−NH−CO−5’)、メチルホスホネート結合、3’−チオホルムアセタール結合、(3’S−CH2−O5’)、アミド結合(3’CH2−C(O)NH−5’)、ホスホロアミデート基、およびこれらの組合せからなる群から選択される、請求項2に記載のAON。

- 少なくとも1つの2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチド(2’−FANA修飾ヌクレオチド)を含む修飾アンチセンスオリゴヌクレオチド(AON)および薬学的に許容される担体を含む医薬組成物であって、前記AONがFoxp3 mRNAに結合する、医薬組成物。
- 前記AONが、少なくとも1つの2’−FANA修飾ヌクレオチドおよび少なくとも1つの非修飾デオキシリボヌクレオチドを含むハイブリッドキメラAONであり、かつ前記AONが2’−FANA AONである、請求項8に記載の組成物。
- 前記2’−FANA AONが、
約0〜約20個のデオキシリボヌクレオチド残基を含む配列に隣接している、5’末端における約0〜約20個の2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチドおよび3’末端における約0〜約20個の2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチド
を含む、請求項9に記載の組成物。 - 前記2’−FANA AONが、SEQ ID NO:1〜9、SEQ ID NO:11〜19、SEQ ID NO:21〜29、SEQ ID NO:31〜138、SEQ ID NO:139〜192、SEQ ID NO:193〜302、またはこれらに相補的な配列の、少なくとも5、少なくとも6、少なくとも7、少なくとも8、少なくとも9、少なくとも10、少なくとも11、少なくとも12、少なくとも13、少なくとも14、少なくとも15、少なくとも16、少なくとも17、少なくとも18、少なくとも19、少なくとも20、少なくとも21、少なくとも22、少なくとも23、少なくとも24、または少なくとも25個の連続的なヌクレオチドを含む、請求項10に記載の組成物。
- 細胞においてFoxp3遺伝子の発現レベルを低減する方法であって、
前記細胞を、少なくとも1つのアンチセンスオリゴヌクレオチド(AON)と接触させる工程を含み、
前記AONがFoxp3 mRNAに結合し、かつ
前記AONが少なくとも1つの2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチド(2’−FANA修飾ヌクレオチド)を含み、
それにより、Foxp3の発現レベルを低減する、方法。 - 前記細胞が制御性T細胞(Treg)である、請求項12に記載の方法。
- 前記Tregが細胞マーカーCD4およびCD25を発現する、請求項13に記載の方法。
- 前記AONが、少なくとも1つの2’−FANA修飾ヌクレオチドおよび少なくとも1つの非修飾デオキシリボヌクレオチドを含むハイブリッドキメラAONであり、かつ前記AONが2’−FANA AONである、請求項12に記載の方法。

前記AONがFoxp3 mRNAに結合し、かつ
前記AONが少なくとも1つの2’−デオキシ−2’−フルオロ−β−D−アラビノヌクレオチド(2’−FANA修飾ヌクレオチド)を含み、
それにより、抗腫瘍免疫を増加させる、方法。 - 前記AONが、制御性T細胞(Treg)の活性を減少させる、請求項17に記載の方法。
- 前記Tregが細胞マーカーCD4およびCD25を発現する、請求項18に記載の方法。
- 前記AONがTregのアポトーシスを誘導する、請求項17に記載の方法。
- 前記AONが免疫細胞の活性を増加させ、それにより、抗腫瘍免疫を増加させる、請求項17に記載の方法。
