Abstract

本開示は特に、4−アルキル−5−ヘテロアリール−3H−1,2−ジチオール−3−チオンの回転異性体(rotomeric isomer)、個々の回転異性型が安定化により得られているそのような異性体の複合体、及び安定化された複合体として個々の回転異性体(回転異性体過剰)を過剰に含む組成物に関する。本開示はまた、そのような回転異性体;個々の回転異性体が安定化により得られていて、かつ回転異性体過剰で存在する複合体;上記回転異性体またはその複合体を含む組成物(回転異性体医薬組成物を含む)を作製及び使用する方法;そのような回転異性体またはその複合体を使用することによりヒト酸化防御遺伝子の機能を制御する方法;ならびにそのような回転異性体またはその複合体を用いてヒトまたは動物患者を処置する方法(例えば、粘膜炎などの様々な障害の症状を予防する、処置する、または減少させる、及び/または医学的目的で様々な器官及び組織において酸化的損傷に対する防御を与えるために)に関する。
したがって、オルチプラズでは2つの別個の回転異性体が可能である。2つの回転異性体はそれぞれ、相互に直交している2つの環として定義することができる;(+)−回転異性体は、頂部にチオン及びメチル基を、かつ右側にピリミジン環のアルファ窒素を配向することにより定義される一方で、(−)−回転異性体は、左側にピリミジン環のアルファ窒素を有する(図4A及び4Bを参照されたい)。
上記回転異性体組成物は、医薬組成物として、(i)回転異性体過剰の(+)−回転異性体を含む1種または複数のオルチプラズ複合体(例えば、塩)、(ii)回転異性体過剰の(−)−回転異性体を含む1種または複数のオルチプラズ複合体(例えば、実施例2及び7〜11の複合体)、または(iii)(i)及び(ii)の混合物のいずれかとして調製することができる。上の医薬組成物(i)〜(iii)は本明細書において、「回転異性体医薬組成物」と称される。そのような回転異性体医薬組成物を、乾燥または液体組成物で投与することができる。経口投与用の上記回転異性体医薬組成物の製剤を口内洗浄薬、または炭酸液、または口腔スプレーもしくはエアロゾル、または口腔軟膏剤、ゲル、またはクリームとして提示することもできる。
一部の態様では、上記回転異性体医薬組成物を局所投与に適し得る製剤に組み込むことができ、かつこれは、局所投与用の保湿剤、湿潤剤、及び他の既知の添加剤を含んでよい。上記局所組成物はまた、皮膚上に施与される貼付剤を介して経皮送達され得、そのような貼付剤は当技術分野で周知である。当業者は、使用することができる他の局所送達組成物及びビヒクルが分かるであろう。
ある特定の態様では、上記回転異性体医薬組成物を、公知の方法及び組成物を使用して直腸投与のために製剤化して、結腸特異的送達を得ることができる。一般的に述べて、直腸投与経路を介した回転異性体医薬組成物の送達は、坐剤、浣腸剤、軟膏剤、クリーム剤またはフォーム剤を使用することにより達成することができる。坐剤は最も一般的な直腸剤形のものであり、基材は一般に、本質的に脂肪質であるが、水溶性または水混和性基材も利用することができる。望ましい生物学的利用能を達成するために、活性成分は、直腸または結腸粘膜と接触すべきである。
他の態様では、上記回転異性体医薬組成物を、吸入可能な形態で、例えば、吸入デバイスで、乾燥粉末形態または液体担体中のいずれかで上記組成物を提供することにより、気道を介して送達することができる。例えば、吸入可能な組成物は、乾燥粉末吸入器に供給される乾燥粉末組成物中に活性な回転異性体医薬組成物を含むことができる。例えば、WO2014177519及びUS20140065219を参照されたい。別法では、吸入可能な組成物は、エタノールなどの液体担体中に活性成分を含み得る。例えば、EP2536412A2を参照されたい。
ある特定の態様では、経口投与用の上記回転異性体医薬組成物の液体製剤を、回転異性体医薬組成物の単回用量の投与を容易にするデバイスを用いて調製及び投与することができる。当技術分野で公知のそのようなデバイスはキャビティーまたはレザバーを含み得て、そこで、乾燥組成物及び液体、例えば、水及び/または非水性溶媒を混合し、次いで、そのデバイスの開口部を介して患者に投与することができる。典型的には、そのようなデバイスは、キャビティーと、キャビティーから区分されているコンパートメントとを含み、そのコンパートメント内に、乾燥粉末が存在し得る。投与時に、上記粉末をコンパートメントからキャビティーまたはまたはレザバーへと放出する。一部のデバイスでは、これは、コンパートメントをキャビティーまたはレザバーから区分するバリアーの破壊によって達成される。その後、粉末を、典型的には振盪により、前もって添加されていたか、またはその時に添加されてよいキャビティー内の液体と混合することができる。キャビティーは、乾燥回転異性体医薬組成物と、乾燥回転異性体医薬組成物を混合して液体組成物を形成することを可能にするのに十分な量の水及び/または非水性溶媒を含む液体量との両方を保持するために十分なサイズのものである。上記液体をパッケージング時に容器に添加して、投与時に一緒に混合することができる乾燥組成物及び液体の両方を含む自給型製品を作製することができる。別法では、上記容器は乾燥回転異性体医薬組成物のみを含有することができ、その後、上記液体を投与前に添加する。上記液体は、下で論述するとおりの香味添加剤を含有してもよい。別法では、乾燥及び液体成分を分離する他の種類のパッケージングを使用することもできる。例えば、上記粉末及び上記液体を、並んでいるか、または重なっていて、破断可能なシールにより分離されている2つのフォームフィルシールパウチ内に密閉することができる。その後、薬物の投与者は、シールを破断し、溶解または懸濁するまで、内容物を2つのコンパートメント間で相互に混合することとなる。
ある特定の態様では、上記回転異性体医薬組成物及びデバイスを、それを必要とするヒトまたは非ヒト動物患者を処置するために使用することができる。上記患者は典型的には、ヒト患者であるが、本開示の回転異性体医薬組成物を、非ヒト動物を処置するために、例えば、獣医学用途で使用することができる。本開示の組成物を、オルチプラズでの処置が公知である疾患及び状態を含む広範囲の様々な疾患及び状態を予防または処置するために使用することができる。そのような疾患及び状態の例には、粘膜炎、HIV、がん、肝炎(HBV及びHCVを含む)、皮膚水疱形成及び単純型表皮水疱症及び関連疾患を含むケラチンベースの皮膚疾患、皮膚炎、炎症性障害または疾患(内皮機能障害及び心臓血管疾患を含む)、悪液質、体重減少、敗血症、造影誘導性腎障害、糖尿病、肥満、PCOS、脂肪症、高脂血症、及び高血圧、慢性腎疾患、肺線維症、低酸素状態、化学物質誘導性肺損傷、呼吸困難障害、アニオンギャップアシドーシス、腎炎、狼瘡、間質性肺疾患、移植片機能障害、肝炎、急性腎損傷、騒音性聴力障害、毒物摂取、網膜障害、神経毒性、がん誘導性損傷、例えば、耳毒性、呼吸器感染症、自閉症、血管痙攣を伴う状態、ならびにn−アセチルシステイン、注射用還元型グルタチオン、または既知の細胞内グルタチオン増強薬の提供により処置可能と考えられている状態が含まれる。
合成オルチプラズ粉末20mg(ST−617−API、下に論述)を、アセトニトリル:水を使用して再結晶化させ、ゴム隔膜を備えた5mlガラスバイアルに添加する。バイアルに、無水クロロホルム1mlを室温で添加し、溶液が透明になるまで、バイアルを振動台の上に置く。ガスシリンジを用いて、無水塩酸ガス5mlを5分かけて、バイアルを間欠的に振盪しながら溶液にゆっくり吹き込む。すべてのHClガスを添加したら、バイアルを5分間にわたって振盪し、隔膜を取り外し、窒素ガスのゆっくりとした定常流下で溶媒を穏やかに蒸発させて、オルチプラズの塩酸塩複合体をオレンジ/赤色の固体として得る。FTIR:C=Sピーク1203/1211(相対強度3:7)及び433/421(相対強度2:8)。
実施例2:オルチプラズ:(−)−回転異性体のヒドロキシマレイン酸無水物複合体を作製する方法
合成オルチプラズ粉末20mgを、アセトニトリル:水を使用して再結晶化させ、ゴム隔膜を備えた10mlガラスバイアルに添加する。すべての水分を、ガスシリンジを使用して窒素ガスによりバイアルから除去する。バイアルを氷浴に入れ、これに、予め冷却(10℃)しておいた超脱水アセトン5ml中の(+)−ジアセチル−L−酒石酸無水物22mgの溶液を添加し、振動テーブル上で5分間にわたって振盪し、5分後に、氷酢酸0.5mlを30秒かけて、シリンジを使用して添加する。間欠的に振盪しながら0〜10℃での4時間後に、バイアルを室温にする。隔膜を取り外し、窒素ガスのゆっくりとした定常流下で溶媒を穏やかに蒸発させて、オルチプラズのヒドロキシマレイン酸無水物複合体(図5を参照されたい)をオレンジ色の固体として得る。上記複合体をエチルエーテルで3回洗浄して、痕跡量の酢酸を除去する。FTIR:C=Sピーク1201/1214(相対強度8:2)及び434/423(相対強度8:2)。上記オルチプラズ複合体のサンプル2mgをKBRと共に摩砕し、Perkin Elmer1600シリーズのFTIRでのFTIR分析のための標準的なペレットに作製する。
合成オルチプラズ粉末20mgを、アセトニトリル:水を使用して再結晶化させ、ゴム隔膜を備えたガラスバイアルに添加する。バイアルに、無水クロロホルム10mlを室温で添加し、溶液が透明になるまで、バイアルを振動台の上に置いた。シリンジを用いて、無水塩化アセチル5mlを、バイアルを間欠的に振盪しながら溶液にゆっくり添加する。すべての塩化アセチルを添加したら、バイアルを15分間にわたって振盪し、隔膜を取り外し、窒素ガスのゆっくりとした定常流下で溶媒及び過剰の試薬を穏やかに蒸発させて、オルチプラズの塩化アセチル複合体をオレンジ色の固体として得る。FTIR:C=Sピーク1202/1212(相対強度3:7)及び432/422(相対強度2:8)。
合成オルチプラズ粉末20mgを、アセトン:水を使用して再結晶化させ、ゴム隔膜を備えたガラスバイアルに添加する。バイアルに、無水クロロホルム10mlを室温で添加し、得られる溶液が透明になるまで、バイアルを振動台の上に置いた。シリンジを用いて、塩化n−ブチル5mlを、バイアルを間欠的に振盪しながら溶液にゆっくり添加する。すべての塩化n−ブチルを添加したら、バイアルを65℃に加温し、30分間にわたって振盪し、隔膜を取り外し、窒素ガスのゆっくりとした定常流下で溶媒及び過剰の試薬を穏やかに蒸発させて、オルチプラズの塩化n−ブチル複合体を赤色の固体として得る。FTIR:C=Sピーク1201/1213(相対強度2:8)及び433/421(相対強度2:8)。
合成オルチプラズ粉末50mgを、アセトン:水を使用して再結晶化させ、スクリューシールを備えた圧力定格ガラスボトルに添加する。バイアルに、無水クロロホルム20mlを室温で添加し、得られる溶液が透明になるまで、バイアルを振動台の上に置いた。シリンジを用いて、ヨウ化メチル10mlを、バイアルを間欠的に振盪しながら溶液にゆっくり添加する。すべてのヨウ化メチルを添加したら、バイアルを密閉し、圧力下で120℃に加熱し、3時間(「hrs」)にわたって振盪し、バイアルを冷却し、開放し、窒素ガスのゆっくりとした定常流下で溶媒及び過剰の試薬を穏やかに蒸発させて、オルチプラズのヨウ化メチル複合体を赤/茶色の固体として得る。FTIR:C=Sピーク1197/1214(相対強度2:8)及び431/419(相対強度2:8)。
粗製のオルチプラズ粉末20mgを、ゴム隔膜を備えたガラスバイアルに添加する。バイアルに、無水クロロホルム10mlを室温で添加し、得られる溶液が透明になるまで、バイアルを振動台の上に置いた。シリンジを用いて、無水塩化ベンゾイル1mlを、バイアルを間欠的に振盪しながら溶液にゆっくり添加する。すべての塩化ベンゾイルを添加したら、バイアルを15分間にわたって振盪し、隔膜を取り外し、10mmHg真空下で溶媒及び過剰の試薬を穏やかに蒸発させて、オルチプラズの塩化ベンゾイル複合体を赤色の固体として得る。FTIR:C=Sピーク1200/1213(相対強度3:7)及び431/419(相対強度3:7)。
合成オルチプラズ粉末22mgを、アセトン:水を用いて再結晶化させ、電磁式撹拌機及び冷水凝縮器を備えたガラス丸底フラスコに添加する。フラスコにアセトン10mlを室温で添加し、撹拌を継続しながら、ジチオリン酸ジエチルアンモニウム塩20mgを溶液にゆっくり添加する。すべてのジチオリン酸ジエチルを添加したら、温度を上昇させ、3時間にわたって還流する。放出されたアンモニアを凝縮器の頂部から、希塩酸トラップに捕捉する。3時間後に、溶媒を除去して、オルチプラズのジチオリン酸ジエチル複合体をオレンジ/赤色の固体として得る。FTIR:C=Sピーク1201/1220(相対強度9:1)及び472/444(相対強度8:2)。
合成オルチプラズ粉末22mgを、アセトン:水を用いて再結晶化させ、電磁式撹拌機及び冷水凝縮器を備えたガラス丸底フラスコに添加する。フラスコにアセトン10mlを室温で添加し、撹拌を継続しながら、チオリン酸ジエチルアンモニウム塩19mgを溶液にゆっくり添加する。すべてのチオリン酸ジエチルを添加したら、温度を上昇させ、3時間にわたって還流する。放出されたアンモニアを凝縮器の頂部から、希塩酸トラップに捕捉する。3時間後に、溶媒を除去して、オルチプラズのジチオリン酸ジエチル複合体をオレンジ/赤色の固体として得る。FTIR:C=Sピーク1208/1221(相対強度9:1)及び479/442(相対強度9:1)。
リン酸トリメチル175mgを粗製オルチプラズ100mg(純度95%)と共に、凝縮器及び撹拌機を備えた丸底フラスコに装入し、72時間にわたって80℃で加熱した。72時間後に、混合物を室温に冷却し、トルエン50mlで5回洗浄する。オレンジ/赤色のN−メチルオルチプラズのリン酸ジメチル複合体255gを下の層から収集し、真空下で60℃で乾燥させる。FTIR:C=Sピーク1210/1227(相対強度9:1)及び489/453(相対強度9:1)。
粗製オルチプラズ粉末22mgを、電磁式撹拌機及び冷水凝縮器を備えたガラス丸底フラスコに添加する。フラスコにアセトン10mlを室温で添加し、撹拌を継続しながら、イセチオン酸アンモニウム塩30mgを溶液にゆっくり添加する。すべてのイセチオン酸アンモニウム塩を添加したら、温度を上昇させ、6時間にわたって還流する。放出されたアンモニアを凝縮器の頂部から、希塩酸トラップに捕捉する。6時間後に、反応物を、最小量の濃塩酸でpH6.8にし、得られた混合物を真空下で蒸発乾固させて、オルチプラズのイセチオン酸複合体を赤/茶色の固体として得る。FTIR:C=Sピーク1216/1228(相対強度9:1)及び464/435(相対強度9:1)。
粗製オルチプラズ粉末20mgを、ゴム隔膜を備えたガラスバイアルに添加する。バイアルに、無水クロロホルム10mlを室温で添加し、得られる溶液が透明になるまで、バイアルを振動台の上に置く。シリンジを用いて、チオ安息香酸アンモニウム塩1mlを、バイアルを間欠的に振盪しながら溶液にゆっくり添加する。すべてのチオ安息香酸アンモニウム塩を添加したら、温度を上昇させ、1時間にわたって還流する。放出されたアンモニアを凝縮器の頂部から、希塩酸トラップに捕捉する。1時間後に、内容物を真空下で除去して、オルチプラズのチオ安息香酸複合体をオレンジ色の固体として得る。FTIR:C=Sピーク1210/1223(相対強度9:1)及び474/438(相対強度9:1)

ST−617−DPI及び実施例1〜11の回転異性体複合体での上記アッセイの結果を下の表1に示す。下の表1に報告されているST−617−API(WO2016207914に開示のプロセスに従って調製した再結晶化オルチプラズ)での結果は、PCT/IB2017/001312の図9(100μg/mLで前処理)から得ている。

Nrf2システム(上で論述)は、phase II解毒酵素及び抗酸化酵素をコードする遺伝子を活性化することによる、酸化的損傷に対する主な細胞防御機構であると考えられている。そのような酸化的損傷の1つが、化学療法処置中及びその後に、特に頭頚部癌処置において生じる口腔粘膜炎である。ヒト酸化ストレスPCRアレイを使用して、HGEPp細胞内で100ug/mlの、PCT/IB2017/001312の実施例1に記載のプロセスにおおむね従って調製した製剤化オルチプラズ組成物(約350nm未満のMHD)、上の実施例1〜11に記載の回転異性体複合体、及び陰性対照(オルチプラズを含まない製剤化オルチプラズ組成物)で前処理した後の84のストレス遺伝子の相対発現を評価した。
HGEPp細胞は、CellnTec Advanced Cell Systems AGから購入した。RNEasy Plus Micro Kitは、Qiagen N.V.、USAから購入した。RNase−free DNase SetはQiagen N.V.、USAから購入した。RT2 Easy First Strand Kit(DNA generator)及びRT2 SYBR(登録商標)Green fluor qPCR mastermixは、Qiagen N.V.、USAから購入した。Oxidative Stress RT2 profiler PCRアレイ(84の防御遺伝子)はQiagen N.V.、USAから購入した。Bio−Rad Inc.、USAからのiCycler PCRシステムをRT−PCRのために使用した。
細胞培養:
貯留した初代HGEPp細胞を、CellnTecが提供するCnT−Prime上皮培養培地中で、ダルベッコリン酸緩衝生理食塩水(DPBS)中で希釈した30mg/mlのI型ラット尾コラーゲン(BD Biosciences)でコーティングした100mmペトリ皿上で成長させた。HGEPp細胞を12×60mm Nunc(商標)Cell Cultureディッシュ中で、2.5×104細胞/cm2密度濃度で成長させた。
製剤化オルチプラズ結晶(ST−617−DPI)を、PCT/IB2017/001312の実施例1に記載のプロセスにおおむね従って調製した。再結晶化オルチプラズを、WO2016207914 ST−617−APIに開示のプロセスに従って調製した。ST−617−DPI、ST−617−API及び実施例1〜11の回転異性体複合体をDMSOに溶解させて、100μg/mlの最終濃度を達成した。すべての投与溶液は、HGEPp細胞では0.8%の最大許容DMSOパーセントよりも十分に低いDMSO0.3%を含有した。
ST−617−DPI及び回転異性体複合体溶液でのHGEPp細胞の前処理:
1. 対照曲線の直線部内のOD吸光度を得るために、ディッシュ細胞濃度を、2.5×105/mlであるように選択した。
2. ディッシュ上でHGEPp細胞を培養し、用意ができたら(コンフルエンスに達したら)、培地を除去し、廃棄した。細胞をDPBSで2〜3回穏やかに洗浄し、最後の洗浄液を除去し、廃棄した。
3. 次いで、HGEPp細胞培養皿を24時間にわたって、100μg/mL濃度のST−617−API、ST−617−DPI及び回転異性体複合体で(2連で)、37℃で前処理した。
HGEPp細胞からのmRNAの調製:
RNeasy UCP Micro Kitを使用して、HGEPp細胞からmRNAを精製した。
1. 処理剤を廃棄及び洗浄した後に、細胞を遠心管内で1000RPMで5分間にわたって遠心することによりペレット化した。すべての上清を吸引により慎重に除去して、すべての細胞培地が完全に除去されていることを確実にした。
2. 細胞をバッファーRULT350μlを添加することにより破壊して、細胞ペレットを管から注意して離し、ボルテックス処理して十分に混合し、溶解産物をRNアーゼ非含有シリンジに装着した20ゲージ針に5回通過させることにより混合物を均質化した。
3. 70%エタノール350μlを溶解産物に添加し、再びピペット処理により混合した(一部の沈澱物が見えるが、アッセイには問題ない)。
4. 形成されていてもよい何らかの沈澱物を含むサンプルを、2ml捕集管内に設置されたRNeasy UCP MinEluteスピンカラムに移し、10,000rpmで15秒間にわたって遠心した。通過画分を廃棄した。
5. バッファーRUWT350μlをスピンカラムに添加し、10,000rpmで15秒間にわたって遠心して、膜を洗浄した。通過画分を廃棄した。
6. DNase Iストック溶液10μlをバッファーRDD70μlに添加した。穏やかに混合し、DNアーゼIミックス80ulを直接、スピンカラム膜に添加し、15分間にわたってボルテックス処理した。
7. バッファーRUWT350μlをスピンカラムに添加し、10,000rpmで15秒間にわたって遠心して、これを洗浄し、通過画分を廃棄した。
8. スピンカラムを新たな2ml捕集管に設置し、バッファーRUPE500μlを添加し、10,000rpmで15秒間にわたって遠心した。通過画分を廃棄した。
9. スピンカラムを再び80%エタノール500μlで洗浄し、これを新たな2ml捕集管内に設置し、蓋をあけたままで全速で5分間にわたって遠心して、すべてのエタノールを確実に除去する。
10. スピンカラムを新たな1.5ml捕集管に設置し、超純水14μlを直接、スピンカラム膜の中心に添加し、蓋を閉め、全速で1分間にわたって遠心して、RNAを溶離した。スピンカラムの死容積は4μlである。超純水16μlで溶離して、RNA溶離液20μl(4μg)を得た。
RT−PCRのためのcDNAの調製:
1. 上の処理のそれぞれからのRNAサンプルをバッファーGE2(gDNA除去バッファー)40μl及びRNアーゼ非含有H2Oに添加して、60μlの最終体積にした。
2. 37℃で5分間にわたってインキュベートし、直ちに、2分間にわたって氷上に置いた。
3. BC5 Reverse Transcriptase Mix62μlを各RNAサンプル60μlに、102μlの最終体積で添加した。
4. 42℃で正確に15分間にわたってインキュベートし、次いで直ちに、95℃で5分間にわたって加熱することにより反応を停止させた(必要ならば、qPCRまで氷上で保持)。
RT−PCRは、遺伝子発現分析のための高感度かつ信頼可能な方法である。上記アッセイは、24時間にわたって100μg/mlのST−617−API、ST−617−DPI及び回転異性体複合体で前処理されたHGEPp細胞において、酸化ストレスに関連する84の遺伝子の発現レベルを分析するために、信頼して使用することができた。上記からのcDNAをRT2 SYBR Green fluor Mastermixと混合し、RT2 profiler PCRアレイのウェルにアリコットした。RT−PCRをiCyclerで行った。遺伝子発現を、Ct値を使用して比較し、結果を、ΔΔCt法を使用して計算し、5つの共通遺伝子(ACTB、B2M、GAPDH、HPRT、及びRPL13A)の平均発現レベルに対して正規化した。
ST−617−DPI及び実施例1〜11の回転異性体複合体でのアッセイの結果を下の表2に示す。下の表2に報告されているST−617−API(WO2016207914に開示のプロセスに従って調製された再結晶化オルチプラズ)での結果は、上記のとおりに行われた先行アッセイからの結果である。

態様の列挙
1. 回転異性体過剰の4−メチル−5−(ピラジン−2−イル)−3H−1,2−ジチオール−3−チオンの(+)回転異性体(オルチプラズ、図4Aを参照されたい)を含む組成物。
2. 回転異性体過剰の上記(+)回転異性体を有する1種または複数のオルチプラズ複合体(例えば、オルチプラズの1種または複数の塩)を含む、態様1に記載の組成物。
3. 上記1種または複数のオルチプラズ複合体が、N置換ピラジニウムイオン及び会合アニオンを生成する、オルチプラズのピラジニル環のアルファ窒素への基の付加(例えば、HCl、アルキルハロゲン化物またはアシルハロゲン化物の可逆的付加)から生じる、N置換ピラジニウムイオン及び会合アニオンを生成する、オルチプラズのピラジニル環のアルファ窒素への基の付加から生じる塩を含む、態様2に記載の組成物。
4. 上記1種または複数のオルチプラズ複合体が、アルキルハロゲン化物、アルケニルハロゲン化物、アルキニルハロゲン化物、アシルハロゲン化物、ベンゾイルハロゲン化物、及びその組合せからなる群から選択される1種または複数の反応物(例えば、HCl、塩化メチル、置換または非置換メチル、置換または非置換ホルミル、置換または非置換アセチル、置換または非置換ベンゾイル、置換及び非置換ブチルからなる群から選択される1種または複数の反応物)のオルチプラズのピラジニル環のアルファ窒素への付加により形成される少なくとも1種のオルチプラズ複合体を含む、態様3に記載の組成物。そのようなオルチプラズ複合体は、オルチプラズを例えば、それぞれ塩化ホルミル、塩化アセチル、塩化ベンゾイル、及びヨウ化n−ブチル、ヨウ化メチル、及びそれらの組合せと反応させることにより形成することができる。そのような態様では、上記ホルミル、アセチル、ベンゾイル及びブチル基は、ピラジン環の2または4位の窒素原子と会合してよく、例えば、ハロゲン(例えば、FまたはCl)及び/または低級アルキル(C1〜C6アルキル)から選択される1個または複数の置換基を有してよい。
5. 上記(+)回転異性体の回転異性体過剰が、30〜40%の過剰、40〜50%の過剰、50〜60%の過剰、及び60〜70%の過剰、70〜80%の過剰、及び80%超の過剰からなる群から選択される範囲である、態様1〜4のいずれかに記載の組成物。
6. オルチプラズ複合体を含む組成物であって、上記オルチプラズ複合体が、過剰の4−メチル−5−(ピラジン−2−イル)−3H−1,2−ジチオール−3−チオン(オルチプラズ)の(−)回転異性体を有する前記組成物。
7. 回転異性体過剰の上記(−)回転異性体を有する1種または複数のオルチプラズ複合体を含む、態様6に記載の組成物。
8. 上記1種または複数のオルチプラズ複合体の少なくとも1種が、ピラジニル環のアルファ窒素と電気陰性原子との間の直接結合を含む、態様7に記載の組成物。
9. 上記電気陰性原子が、酸素、硫黄、及びリンからなる群から選択される、態様8に記載の組成物。
10. 上記1種または複数のオルチプラズ複合体が、ヒドロキシマレアートオルチプラズ複合体を含む、実施形態6〜9のいずれかに記載の組成物。
11. 上記1種または複数のオルチプラズ複合体が、オルチプラズを、カルボン酸またはその塩、リン酸またはその塩、アルキルホスファート、アリールホスファート、アルキルスルホン酸またはその塩、アリールスルホン酸またはその塩、置換及び非置換チオカルボン酸またはその塩、ならびに上述の1種または複数の組合せ、例えば、乳酸、タルトロン酸、イソチオン酸、イソセリン、2−メルカプトエタンスルホン酸、タウリン、プロピルホスホン酸、2−アミノエチルホスホン酸、ジチオリン酸ジエチル、チオリン酸ジエチル、リン酸ジメチルからなる群から選択される1種または複数の反応物と反応させることにより形成される1種または複数の複合体を含む、態様6〜9のいずれかに記載の組成物。
12. 上記1種または複数のオルチプラズ複合体が、オルチプラズを、乳酸、タルトロン酸、イソチオン酸、イソセリン、2−メルカプトエタンスルホン酸、タウリン、プロピルホスホン酸、2−アミノエチルホスホン酸、ジチオリン酸ジエチル、チオリン酸ジエチル、リン酸ジメチル、及びそれらの組合せからなる群から選択される1種または複数の反応物と反応させることにより形成される1種または複数の複合体を含む、態様6〜9のいずれかに記載の組成物。
13. 上記(−)回転異性体の回転異性体過剰が、30〜40%の過剰、40〜50%の過剰、50〜60%の過剰、60〜65%の過剰、60〜70%の過剰、70〜80%の過剰、80〜90%の過剰、及び90〜100%の過剰からなる群から選択される範囲である、態様6〜12のいずれかに記載の組成物。
14. ヒトへの投与に適している、態様1〜13のいずれかに記載の少なくとも1つの組成物を含む医薬組成物。
15. 局所投与に適した形態である、態様14に記載の医薬組成物。
16. 直腸投与または吸入による投与に適した形態である、態様15に記載の医薬組成物。
17. 皮下、筋肉内、胸骨内、または静脈内注射に適した形態である、態様14に記載の医薬組成物。
18. 経口投与に適した形態である、態様14に記載の医薬組成物。
19. 散剤、丸剤、錠剤の形態であるか、またはカプセル剤の中に含まれている、態様18に記載の医薬組成物。
20. 液体形態である、態様18に記載の医薬組成物。
21. 処置を必要とするヒトまたは非ヒト動物患者を処置するためのプロセスであって、上記患者に、態様13〜20のいずれかに記載の医薬組成物を投与することを含む上記プロセス。
22. 前記投与が経口投与を含む、態様21に記載のプロセス。
23. 前記投与が頬側投与を含む、態様21に記載のプロセス。
24. 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、態様18〜20のいずれかに記載の医薬組成物を経口投与することを含む上記プロセス。
25. 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、態様18〜20のいずれかに従って調製された医薬組成物を経口投与することを含む上記プロセス。
26. 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、上記患者に、態様18〜20のいずれかに記載の医薬組成物を経口投与することを含む上記プロセス。
27. 上記粘膜炎が口腔粘膜炎である、態様24〜26のいずれかに記載のプロセス。
28. 上記粘膜炎が消化管の粘膜炎である、態様24〜26のいずれかに記載のプロセス。
29. 粘膜炎または皮膚炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、態様15に記載の組成物を局所投与することを含む上記プロセス。
30. 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、態様16に記載の組成物を直腸投与することを含む上記プロセス。
31. 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、態様16に記載の組成物を吸入により投与することを含む上記プロセス。
32. 上記患者が放射線療法を受けている、態様21〜31のいずれかに記載のプロセス。
33. オルチプラズのヒドロキシマレイン酸無水物(−)回転異性体を作製するプロセスであって、
(i)無水極性溶媒の存在下でオルチプラズを(+)−ジアセチル−L−酒石酸無水物と反応させて、反応混合物を形成するステップと;
(ii)酢酸を上記反応混合物に添加するステップと
を含む上記プロセス。
Claims (33)
- 回転異性体過剰の4−メチル−5−(ピラジン−2−イル)−3H−1,2−ジチオール−3−チオン(オルチプラズ)の(+)回転異性体を含む組成物。
- 回転異性体過剰の前記(+)回転異性体を有する1種または複数のオルチプラズ複合体(例えば、オルチプラズの1種または複数の塩)を含む、請求項1に記載の組成物。
- 前記1種または複数のオルチプラズ複合体が、N置換ピラジニウムイオン及び会合アニオンを生成する、オルチプラズのピラジニル環のアルファ窒素への基の付加から生じる塩を含む、請求項2に記載の組成物。
- 前記1種または複数のオルチプラズ複合体が、オルチプラズと、アルキルハロゲン化物、アルケニルハロゲン化物、アルキニルハロゲン化物、アシルハロゲン化物、ベンゾイルハロゲン化物、及びその組合せからなる群から選択される1種または複数の反応物との反応から形成される少なくとも1種のオルチプラズ複合体を含む、請求項3に記載の組成物。
- 前記(+)回転異性体の回転異性体過剰が、30〜40%の過剰、40〜50%の過剰、50〜60%の過剰、及び60〜70%の過剰、70〜80%の過剰、及び80%超の過剰からなる群から選択される範囲である、請求項1〜4のいずれかに記載の組成物。
- オルチプラズ複合体を含む組成物であって、前記オルチプラズ複合体が、過剰の4−メチル−5−(ピラジン−2−イル)−3H−1,2−ジチオール−3−チオン(オルチプラズ)の(−)回転異性体を有する前記組成物。
- 回転異性体過剰の前記(−)回転異性体を有する1種または複数のオルチプラズ複合体を含む、請求項6に記載の組成物。
- 前記1種または複数のオルチプラズ複合体の少なくとも1種が、ピラジニル環のアルファ窒素と電気陰性原子との間の直接結合を含む、請求項7に記載の組成物。
- 前記電気陰性原子が、酸素、硫黄、及びリンからなる群から選択される、請求項8に記載の組成物。
- 前記1種または複数のオルチプラズ複合体が、ヒドロキシマレアートオルチプラズ複合体を含む、請求項6〜9のいずれかに記載の組成物。

- 処置を必要とするヒトまたは非ヒト動物患者を処置するためのプロセスであって、前記患者に、請求項14〜20のいずれかに記載の医薬組成物を投与することを含む前記プロセス。
- 前記投与が経口投与を含む、請求項21に記載のプロセス。
- 前記投与が頬側投与を含む、請求項22に記載のプロセス。
- 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、請求項18〜20のいずれかに記載の医薬組成物を経口投与することを含む前記プロセス。
- 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、請求項18〜20のいずれかに従って調製された医薬組成物を経口投与することを含む前記プロセス。
- 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、前記患者に、請求項18〜20のいずれかに記載の医薬組成物を経口投与することを含む前記プロセス。
- 前記粘膜炎が口腔粘膜炎である、請求項24〜26のいずれかに記載のプロセス。
- 前記粘膜炎が消化管の粘膜炎である、請求項24〜26のいずれかに記載のプロセス。
- 粘膜炎または皮膚炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、請求項15に記載の組成物を局所投与することを含む前記プロセス。
- 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、請求項16に記載の組成物を直腸投与することを含む前記プロセス。
- 粘膜炎を予防する、処置する、寛解させる、その重症度を軽減する、及び/またはその持続期間を短縮することを、それを必要とするヒトまたは非ヒト動物患者について行うプロセスであって、請求項16に記載の組成物を吸入により投与することを含む前記プロセス。
- 前記患者が放射線療法を受けている、請求項21〜31のいずれかに記載のプロセス。
- オルチプラズのヒドロキシマレイン酸無水物(−)回転異性体を作製するプロセスであって、
(iii)無水極性溶媒の存在下でオルチプラズを(+)−ジアセチル−L−酒石酸無水物と反応させて、反応混合物を形成するステップと;
(iv)酢酸を前記反応混合物に添加するステップと
を含む前記プロセス。
