[000210] それぞれの側部に緑色蛍光タンパク質(GFP)レポーターが隣接している2つの相同アームを有するプラスミドドナー(400ng)を、Cas9とガイドRNAとをコードするベクターと共に、猫接合体に導入する。相同アームの各々は、約800bpである。Fel d1プロモーター並びに鎖1及び鎖2開始コドンの代わりにGFPレポーターを挿入するように、sgRNA及びプラスミドドナーを設計する。プラスミドドナーは、DNAにおける二本鎖切断を塞ぐためのHDRプロセスにおいて鋳型として使用することになる。このようにして、相同アーム間のGFPのコード配列は、切断が修復されるときにプロモーター領域及び開始コドンに取って代わることになる。GFP配列のみを発現させるために、終止コドンをプラスミドドナー内のGFPレポーター遺伝子配列の後に含める。接合体へのベクターの注入後、in vitroで分化胚盤胞を培養物に外植して、胚性幹細胞を誘導する。PCR遺伝子型判定及びシークエンシングを使用して、正しく標的化された挿入物を担持する胚性幹細胞株を同定する。適切な励起及び発光フィルター設定を使用してマイクロプレート蛍光リーダーによってGFP発現を確認する。その後、正しく挿入された猫接合体を胚に発達させ、それらの胚をレシピエント雌猫に移植し、満期まで発達させる。生きて生まれてきた仔猫が、GFPを発現すること、及びまた前の実施例で説明したようにFel d1発現を欠くことを確認する。
[000212] 確認されたヒトFel d1アレルギー罹患者群に対してアレルギー検査を行って、Fel d1低アレルゲン性表現型を産生する際のFel d1ノックアウト手順の成功を確認することができる。例えば、猫の毛又は唾液をFel d1ノックアウト猫から採取し、毛又は唾液の抽出物を調製し、皮膚検査手順(皮膚プリック、皮内、又は皮膚パッチ検査)の際に接種物として使用する。結果を、GreerのStandardized Cat Hair(GREER(登録商標)、Lenoir、NC)などの皮膚検査診断で指示される、標準化猫毛抽出物と比較することができる。アレルギー反応(例えば、腫脹、発赤、じんま疹、くしゃみ、呼吸困難、喘息、皮膚のかゆみ、鼻水、鼻詰まり、咳嗽、及び/又は眼刺激)がないことで、特定のFel d1ノックアウトが低アレルゲン性表現型をもたらすことを確認することができる。
Claims (84)
- Fel d1ゲノム配列の一部分又は隣接領域と配列が同一なcrRNAと、tracrRNAと、を含むキメラガイドRNAであって、前記一部分が、NGGからなるPAM配列の5’であり、式中、N=A、T、C又はGである、キメラガイドRNA。
- 前記crRNAが、Fel d1鎖1ゲノム配列の一部分、若しくは長さ1kbの3’隣接領域と配列が同一であるか、又はFel d1鎖2の一部分、若しくは長さ1kbの5’隣接領域と配列が同一である、請求項1に記載のキメラガイドRNA。
- 前記crRNAが、配列番号1226又は配列番号1227に記載の配列の一部分と配列が同一である、請求項1に記載のキメラガイドRNA。
- 前記crRNAが、配列番号1226又は配列番号1227に記載の配列の一部分と実質的に相補的である、請求項1に記載のキメラガイドRNA。
- 前記crRNAの長さが、18~22ヌクレオチド、例えば、20ヌクレオチドである、請求項1に記載のキメラガイドRNA。
- 前記crRNAが、配列番号1~1225に記載のDNA配列と配列が同一である、請求項1に記載のキメラガイドRNA。

- 第1のポリヌクレオチドと第2のポリヌクレオチドとを含むキメラDNA分子であって、前記第1のポリヌクレオチドが、Fel d1ゲノム配列の一部分又は隣接領域と同一であり、前記一部分が、NGG(式中、N=A、T、C又はG)からなるPAM配列の5’であり、前記第2のポリヌクレオチドが、tracrRNAをコードする、キメラDNA分子。
- 前記第1のポリヌクレオチドが、Fel d1鎖1ゲノム配列の一部分、若しくは長さ1kbの3’隣接領域と同一であるか、又はFel d1鎖2の一部分、若しくは長さ1kbの5’隣接領域と同一である、請求項10に記載のキメラDNA分子。
- 前記第1のポリヌクレオチドが、配列番号1226又は配列番号1227に記載の配列の一部分と同一である、請求項10に記載のキメラDNA分子。
- 前記第1のポリヌクレオチドが、配列番号1226又は配列番号1227に記載の配列の一部分と実質的に相補的である、請求項10に記載のキメラDNA分子。
- 前記第1のポリヌクレオチドの長さが、18~22ヌクレオチド、例えば、20ヌクレオチドである、請求項10に記載のキメラDNA分子。

を含む方法。 - Fel d1タンパク質発現が損なわれているか又は存在しない猫を産生する方法であって、
猫の卵母細胞を培養すること、
5’又は3’隣接領域のFel d1ゲノム配列を標的とするように設計されたものであるガイドRNAのペアと、Cas9タンパク質とを、前記猫の卵母細胞に導入すること、
前記卵母細胞に猫の精子を受精させて胚を作出すること、
前記胚をin vitroで培養すること、
前記胚をレシピエント雌猫に移植すること、及び
前記胚を成熟させて猫にすること
を含む方法。 - Fel d1ゲノム配列の一部分を欠き、その結果、Fel d1タンパク質の発現が損なわれているか又は存在しない、猫。
- 前記Fel d1タンパク質又はその一部分が、発現されるがヒトにおいて低アレルゲン性である、請求項47に記載の猫。

前記第1のキメラガイドRNA及び第2のキメラガイドRNAが、Fel d1ゲノム配列の一部分又は隣接領域において第1及び第2の二本鎖切断を生じさせるように前記Cas9タンパク質に指示し、その結果、前記第1及び第2の二本鎖切断の修復時に、Fel d1ゲノム配列を含む介在部分が前記ゲノムから除去される、方法。 - 前記sgRNA標的配列が、配列番号1~1225に記載の配列から選択される、請求項49に記載の方法。
