最も単純な非ウイルス遺伝子送達システムは、裸の発現ベクターDNAを使用する。特定の組織、特に筋肉への遊離DNAの直接注入は、高レベルの遺伝子発現を生じさせることが分かっており、このアプローチの単純さは、多数の臨床プロトコールにおいてその採用につながった。特に、このアプローチは、癌ワクチンとして機能するかもしれない腫瘍抗原を発現させるために、DNAを腫瘍内に直接注入するかまたは筋細胞に注入することが可能な癌の遺伝子治療に適用されてきた。

カチオン性リポソーム、続くCPG非含有発現ベクターの連続注射を用いたインビボタンパク質発現
この実施例は、カチオン性リポソーム、これに時間的に密接に続くCPG非含有発現ベクターの連続注射を使用してインビボタンパク質発現を用いた様々な研究を、記載している。
リポソーム調製。凍結乾燥粉末としての純粋なDOTAP脂質は、Avanti polar lipidから購入した。純粋なDOTAPカチオン性リポソームは、凍結乾燥粉末を5%デキストロースの水溶液中に20ミリモルの脂質濃度で再懸濁させることによって調製した。その後、この溶液を15分間ボルテックスして、レーザ光散乱によって測定した、平均粒径が350nmの多層状小胞(MLV)を形成した。その後、MLVから、バスソニケーター中で超音波処理することによって小さな単層状小胞(SUV)、平均粒径75nmを形成した。
DNAは、3つのエンドヌクレアーゼ制限酵素部位、DraIII、EcoRI及びNheIを含むベースベクターとして設計した。これは、Gibson Assembly
technique(NEB、MA)を使用して、gBlockの4つのDNA断片(IDT、IA)から構築した。MAR含有プラスミドについては、βGlobin MARをDraIII-EcoRI部位で塩基ベクターに挿入した。h-GCSFのCpG非含有核酸配列を図2に示す。
連続的、カチオン性リポソームIV注射、続くDNAベクター注射によってマウスにおいて産生された血清ヒトG-CSFレベル
1グループ当たり5匹のマウスに注射した。各マウスは、800nmolの純粋なDOTAPカチオン性リポソーム(MLVまたはSUV)、2分後に、80μgのmCMV-EF1-hGCSF、hCMV-hCMV-hGCSFまたはmCMV-EF1-ルシフェラーゼ、CPG非含有、プラスミドDNAベクターの単回のIV注射を受けた。血清hG-CSFレベルは、IV注射後7日目から開始し、その後、7日間隔で評価した。
DOTAP MLVの連続IV注射、2分後のEF1-hGCSFまたはEF-1ルシフェラーゼ含有CPG非含有プラスミドDNAベクターのいずれかの単回IV注射の後の21日目に、4匹のマウスのグループから全血を採取した。次いで、各マウスからの血液を、University of California Davis獣医診断研究所による総WBC、及び絶対好中球数について、盲検様式で分析した。
800nmolの純粋なDOTAP MLVカチオン性リポソームを連続IV注射し、2分後に40μgのEF1-ルシフェラーゼ-EF1-hGCSF(2発現カセット)またはEF1-hGCSF(1発現カセット)、CPG非含有、単一プラスミドDNAベクターをIV注射したのち、6日目または14日目のいずれかにおいて4匹のマウスのグループから血清を採取した。
800nmolの純粋なDOTAP MLVカチオン性リポソームを連続IV注射し、その2分後に60μgのhG-CSF遺伝子(これらは以下のエンハンサー-プロモーターの組合せ:CPGを含まない単一のカセットであるDNAベクター中の、mCMV-EF1、hCMV-hCMV、hCMV-フェリチン(hferritin)軽鎖、hCMV-フェリチン重鎖、hCMV-グルコース調節タンパク質78またはmCMV-フェリン(hferritin)軽鎖それぞれの1つに連結している)をIV注射し、その21日目後に4匹のマウスのグループから血清を採取した。
800nmolの純粋なDOTAP MLVカチオン性リポソーム単独、または2mg/kgのL-アルギニン、0.01mg/kgのコルヒチンまたは1mg/kgのデキサメタゾンを含む上記カチオン性リポソームを連続IV注射し、その7日後に、4匹のマウスのグループから肺を採取した。いずれの場合も、カチオン性リポソーム注射の2分後、40μgのmCMV-EF1-ルシフェラーゼ、CPG非含有、単一カセットDNAベクターのIV注射がなされた。
800nmolの純粋なDOTAP MLVカチオン性リポソーム単独、または2mg/kgのL-アルギニン、1mg/kgのデキサメタゾン、0.02mg/kgのシルデナフィル、0.1mg/kgのバルプロ酸または2mg/kgのL-アルギニン+0.02mg/kgのシルデナフィル(VIAGRA)を含有する上記カチオン性リポソームをIV注射し、その7日後に、4匹のマウスのグループから肺を採取した。いずれの場合も、カチオン性リポソーム注射の2分後に、40μgのEF1-ルシフェラーゼ--EF1-hGCSF、2発現カセット、CPG非含有単一プラスミドDNAベクターのIV注射がなされた。

1:4のN:P比で22kDaの線状PEIに複合体化された12.5μgのCPG非含有EF-1-Luc DNAベクターをIV注射、または、900nmolの純粋なDOTAP MLVカチオン性リポソームの連続IV注射、その2分後の40μgの同じEF-1-Luc DNAベクターのIV注射の、1日または5日後において3匹のマウスのグループから肺を採取した。
800nmolの、純粋なDOTAP MLV、純粋なDOTAP SUV、DOTAP:コレステロール2:1MLV、DOTAP:ジオレリル(diolelyl)ホスファチジルコリン(DOPC)1:1MLV、DSTAP MLV、エチルDSPC MLVまたはDOTAP:DOBAQ1:1MLVのカチオン性リポソームの連続IV注射の1日後に、4匹のマウスのグループから肺を採取した。それぞれの場合において、カチオン性リポソーム注射の2分後に、80μgのmCMV-EF1-ルシフェラーゼ、CPG非含有単一カセットDNAベクターのIV注射がなされた。
1:4のN:P比で22kDaの線状PEIと複合体化した12.5μgのCPG非含有EF-1-Luc DNAベクターのIV注射の1日後の3匹のマウスのグループから、脾臓を採取した。マウスは、PEI:DNA複合体IV注射を受ける2時間前に、マウス1匹当たり、1mlの5%DMSO単独、または200μgのアンレキサノクス、1mgのクロロキン、200μgのSAHAまたは300μgのトファシチニブを含む上記1mlの5%DMSOのいずれかの腹腔内注射を受けた。
800nmolの純粋なDOTAP MLVカチオン性リポソーム、続く2分後の40μgのEF1-Luc--EF1-hGCSF、EF1-Luc--hCMV-hCMV-hGCSF、EF1-Luc--hCMV-hCBOX-hGCSF、EF1-Luc--hCMV-hREG1-hGCSFまたはEF1-Luc--mCMV-hCBOX-hGCSFの2発現カセット、CPG非含有単一プラスミドDNAベクターの、連続IV注射の1または7日後に、4つのマウスのグループから肺を採取した。
中性MLVリポソームは、Phospholipon 90H、Lipoid GmbHから調製された。この製品の脂肪酸含量は15%のパルミチン酸、85%のステアリン酸である。リポソームは、ロータリーエバポレーターで有機溶媒中の脂質を乾燥させ、次いで乾燥した脂質膜を5%デキストロース水溶液に50ミリモルの脂質濃度で再懸濁させるか、または脂質を水和して5%(w/v)デキストロース中の乾燥粉末とすることのいずれかにより調製した。両方とも60℃で調製した。次いで、溶液を15分間ボルテックスしてMLVを形成させた。4匹のマウスのグループから、緩衝液のみの(対照)、1000nmolの純粋なDOTAP SUVカチオン性リポソーム単独、または1000nmolの中性MLVと同時注射の連続IV注射、その2分後の120μgのEF1-ルシフェラーゼ--EF1-hGCSF(2発現カセット)、CPG非含有単一プラスミドDNAベクターまたは1400nmolの純粋なDOTAP SUVカチオン性リポソームを単独または1000nmolの中性MLVと同時注射のIV注射、その2分後の100μgのEF1-ルシフェラーゼ--EF1-hGCSF DNAのIV注射し、その1日後に血清を採取した。

SUVカチオン性リポソームを連続IV注射し、2分後に、1400nmolと中性MLVと同時注射される120μgのCPG非含有EF1-ルシフェラーゼ--EF1-hGCSF DNAまたは1000nmolの純粋なDOTAP SUVカチオン性リポソームのIV注射、2分間後に100μgのEF1-ルシフェラーゼ--EF1-hGCSF DNAのIV注射の後、7日目に4匹のマウスのグループから血清を採取した。また、緩衝液のみ(対照)、それぞれ、1000、750、500、250または100nmolの中性MLVと同時注射される800nmolの純粋なDOTAP SUVカチオン性リポソームのIV注射、続いて2分後に90μgのEF1-ルシフェラーゼ--EF1-hGCSFをIV注射の後1日目に4匹のマウスのグループから血清を採取した。
800または1000ナノモルの純粋なDOTAP SUVまたはMLVカチオン性リポソームのいずれかの連続IV注射の後7日目に、4匹のマウスのグループから血清を採取した。3つのカチオン性リポソーム製剤のそれぞれの注射の後、2分後に、100または120μgのいずれかのmCMV-EF1-h-G-CSF、CPG非含有単一カセットDNAベクターのIV注射を行った。0.1μmの押出カチオン性リポソームは、MLVから、Lipex押出装置中で高アルゴンガス圧下にてサイズ化ポリカーボネート膜を通して逐次押出しにより作製した。
長期発現
この実施例では、1グループ当たり5匹のマウスに、DOTAP MLVまたはSUVのいずれか800nmol、続いてEF1-またはhCMV-駆動hG-CSF cDNAを含有するCPG非含有プラスミドベクター90ugを連続注射した。ヒトG-CSFタンパク質の血清レベルを、注射後に続く428日間において、7日または14日間隔で評価した。ヒトG-CSFレベルのためのマウス血清の取得及び分析は以下のようにして行った。各マウスを麻酔し、顎下静脈を介して採血した。BDからの血清分離管を用いて全血から血清を単離した。ヒトG-CSFレベルは、R&D systemsのヒトG-CSF ELISAを用いて、製造業者の仕様書に従って厳密に実施されたELISAによってpg/mlで測定された。結果は図17に示されており、DOTAP SUVリポソーム、次いでEF1-huG-CSFプラスミドDNAベクターの単回IV注射を受けた後、少なくとも428日の間、完全免疫応答性マウスにおいてヒトG-CSFタンパク質の治療レベルを超えるレベルが産生されたことを示している。
ラットでのタンパク質発現
この実施例では、250gのSprague-Dawley雌ラット#22及び23に、6000nmolのDOTAP SUV、その後EF1駆動hG-CSF DNAベクターを含有する300ugのCPG非含有プラスミドベクターを連続して注射した。注射後7日間隔で、ヒトG-CSFタンパク質、WBC及び絶対好中球数(ANC)の血清レベルを評価した。血清ALT及びASTレベルは、1日目のみで評価した。すべてがUC
Davis比較病理研究所で評価された。図18及び下記の表3に示すように、EF1-huG-CSFを注射したラットでは、単回IV注射後少なくとも22日間、治療レベルを超えるレベルのhG-CSFタンパク質及び顕著に上昇したWBC及びANCが産生された。注射後1日目に測定したALT及びASTは、注射していないラットのバックグラウンド対照レベルに匹敵した。
DPTAPリポソーム
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、900nmolのDOTAP、DMTAP、またはDPTAP(1,2-ジパルミトイル3-トリメチルアンモニウムプロパン)SUVリポソームの単回IV注射を受け、その2分後にhG-CSFをコードする70ugのEF-1プラスミドDNAベクターの単回IV注射を受けた。hG-CSFの血清レベルを、注射後24時間で、ELISAによって決定した。図19は、HuG-CSFタンパク質が、DOTAPまたはDPTAPで処置されたがDMTAPで処置されていないマウスからの血清中に存在することを示している。これらのデータは、複数のカチオン性脂質がインビボでのトランスフェクションを媒介することができ、タンパク質産生のレベルは脂質キャリアの選択によって制御することができることを示している。
毒性は48時間以内に消散する
この実施例では、1グループ当たり3匹のマウスに注射した。マウスはCharles
River Labsから購入した。各マウスは、示されたようにDOTAP SUVリポソームの1000ナノモル、1200ナノモル、または1400ナノモルの単回IV注射を受け、2分後に100μg、120μgまたは140μgの、ルシフェラーゼをコードするCPG非含有EF-1駆動プラスミドDNAベクターの単回IV注射を受けた。注射後24時間または48時間に血清を採取した。ALT及びAST測定は、UC Davis比較病理研究所でアッセイした。図20に示すように、連続注射の24時間後に、ALT及びASTの血清レベルは、すべての脂質その後のDNAグループにおいて2倍から5倍に上昇した。48時間後、血清ALT及びASTレベルは対照(バックグラウンド)レベルに戻った(偽注射グループによって示される)。これらのデータは、ALT/ASTによって測定された毒性が急性(注射の24時間以内に存在する)であり、一過性(48時間で消失)であることを示している。
DexPを含むリポソーム
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、純粋なDOTAP SUVリポソームのいずれか900ナノモルの単回IV注射を受けたか、または各マウスは、純粋なDOTAP SUVリポソームまたはリポソーム二重層中に組み込まれたパルミテートに共有結合したデキサメタゾン(DexP)の示されたモル%を含むリポソームのいずれかの900ナノモルの単回IV注射を受けた。リポソーム二重層への5%コレステリルパルミテート(CholP)の組み込みが対照の役目を担う。この2分後に、hG-CSFをコードする90ugのプラスミドDNAの単回IV注射を行った。hG-CSFの血清レベルを、注射後24時間においてELISAによって決定し、ALT測定をUC Davis比較病理学研究所でアッセイした。図21に示すように、連続注射後24時間で、ALTレベルで測定した毒性は、乳酸加リンゲル液のみ(ALT対照)を偽注射した動物で見られる毒性よりも2~3倍高い。リポソーム二重層中に2.5%デキサメタゾンパルミテート(DexP)を組み込むことにより、hG-CSFのピーク発現を増加させ、且つ24時間でALTレベルをバックグラウンド(正常)レベルの1.5倍以内に減少させる二重効果を生じた。
DexPは毒性を軽減し、発現を増加させる
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、純粋なDOTAP SUVリポソーム、またはDOTAPリポソームのリポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含有するリポソームのいずれかの900ナノモルの単回IV注射を受けた。この2分後、hG-CSFをコードするEF-1駆動プラスミドDNAベクター130ug(高)または40μg(低)のいずれかの単回IV注射を行った。2つのグループは、1マイクロモルのデキサメタゾンパルミテートのIP注射で、IV注射の2時間前に処置した。hG-CSFの血清レベルを、注射後24時間のELISAによって決定し、ALT測定を、UC Davis比較病理研究所により24時間でアッセイした。図22に示すように、130ugのDNAの連続注射により、24時間後に40ugのDNAより顕著に高いhG-CSFタンパク質レベルを産生した。どちらのDNA用量でも、2.5モル%のデキサメタゾンパルミテートをリポソームに含めることにより、hG-CSFタンパク質レベルがさらに増加した。さらに、デキサメタゾンパルミテートをリポソームに組み込むことにより、ALTレベルは、はるかに高いDNA用量であっても、バックグラウンド(正常)レベルの1.5倍以内に低下した。
Dex注射の前後
この実施例では、15mg/kgのトファシチニブまたは40mg/kgのデキサメタゾンをマウスにIP前注射し、2時間後に900nmolのDOTAP SUV、その後の70ugのCPG非含有EF-1駆動hG-CSFプラスミドベクターを連続IV注射した。トファシチニブまたはデキサメタゾンの別のIP注射を、DNAの注射の2時間後に行った。図23に示すように、連続したカチオン性リポソームその後のDNA注射の前及び後のデキサメタゾンの投与は、共にHuG-CSFタンパク質レベルを顕著に増加させる一方、バックグラウンド(正常)レベルの1.5倍以内に毒性を同時に減少させた。対照的に、免疫抑制剤トファシチニブの前及び後の注射はどちらも減少させなかった。
脂質対DNA比
この実施例では、グループ当たり3匹のマウスに、DOTAPリポソームのリポソーム二重層中に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含有するDOTAP SUVリポソーム(1回の注射当たり100uLの最終容量の乳酸加リンゲル液(LR)に懸濁)900、1000、1200または1500ナノモルのIV注射を行い、その2分後に40、60または75ugのCPGを非含有EF-1駆動hG-CSFプラスミドベクターで1回の注射当たり100μLにてIV注射した。偽注射されたマウスは、脂質またはDNAなしのLRのみを受けた。24時間後、hG-CSFタンパク質及びALTの血清レベルをアッセイした。図24に示すように、DOTAP SUV脂質対プラスミドDNA(ナノモル脂質:mgDNA)比が26:1未満のものを連続注射したマウスのhG-CSFタンパク質及びALTレベルは、毒性を防止しながら顕著に高いhG-CSFタンパク質レベルを産生した。一方、脂質またはDNA注射を受けていない対照マウスにおいて、バックグラウンド(正常)レベルに対して1.5倍以内または等しいのいずれかのALTレベルを産生することが立証された。
リツキシマブ発現
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、純粋なDOTAPカチオン性リポソーム、またはDOTAPリポソームのリポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含むリポソームのいずれか1000ナノモルの単回IV注射を受けた。この2分後、リツキシマブ重鎖及び軽鎖をコードする二重カセット単一プラスミドDNAベクター100ug(図36、40、及び41の構築物を参照)の単回IV注射が行われた。血清リツキシマブレベルは、注射の24時間後、その後の7日間隔においてELISAによって決定された。マウスは採血され、G-CSFと同様に血清を単離した。Eagle Biosciencesから得られたImmunoguide ELISAを用いてリツキシマブレベルを測定し、指示書に従って実施した。図25に示すように、IPデキサメタゾン前処置+DOTAPリポソーム中の2.5モル%デキサメタゾンパルミテートの組込みは、注射後少なくとも3週間で血清リツキシマブレベルを5倍以上増加させる。
リツキシマブ発現
この実施例では、1グループ当たり3匹のマウスを注射した。各マウスは、純粋なDOTAPカチオン性リポソーム、またはDOTAPリポソームのリポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含むリポソームのいずれか1000ナノモルの単回IV注射を受けた。この2分後、リツキシマブをコードするEF-1-駆動2重カセット単一プラスミドDNAベクター(図36、40、及び41の構築物を参照)100ugの単回IV注射が行われた。1グループを、IV注射の2時間前に、40mg/kgのデキサメタゾン(Dex)及び1000ナノモルの中性脂質(NL)、DMPCのIP注射で処置した。血清リツキシマブレベルの注射マウスを、注射後24時間及びその後7日間の間隔でELISAによって測定した。図26に示すように、すべてのマウスは、1回の注射後に少なくとも12週間、顕著なレベルの血清リツキシマブタンパク質を産生した。Dex、DexP及びNLの組合せを受けたマウスは、経時的に著しく高い血清リツキシマブレベルを産生した。これらのデータは、2重カセットリツキシマブプラスミドDNAベクターの1回の連続注射により、90日を超えて、動物において顕著なレベルの血清リツキシマブタンパク質を産生することができることを示している。
2重カセット、単一プラスミドリツキシマブ発現
この実施例では、Raji細胞(百万個/サンプル)を、EDTA及び0.5%BSAを含むFACS結合緩衝液中で、マウス血清サンプルまたは組換えリツキシマブ(50ng/ml)と4Cで1時間インキュベートした。洗浄後、試料を蛍光標識二次抗体(抗ヒトIgG-PE)と共に30分間インキュベートして、洗浄し、Accuriフローサイトメーターを用いて分析した。各試料について、3500~5000の事象が記録された。実験を2回繰り返し、同様の結果を得た。
機能性リツキシマブを発現する
この実施例では、Raji細胞(5×104細胞/ウェル)をRPMI+10%FBS培地を用いて96ウェルプレートに入れた。翌日、細胞をリツキシマブ(1,10ug/ml)またはマウス血清サンプル(20μl/ウェル)と共に室温で1時間インキュベートした。その後、プールされた正常ヒト血漿(Innovative Research)20ulをすべてのウェルに添加し(リツキシマブ対照条件を除く)、プレートを37℃でさらに12時間インキュベートした。細胞生存率は、Promega Cell titer glo試薬を製造者の指示書に従って使用して測定した。図28では、値は、huG-CSF DNAプラスミドDNAベクターを注射したマウス由来の血清が使用された対照条件からの変化率として示す。個々のマウス血清を、血清収集の8~78日前に、二重カセット単一プラスミドリツキシマブDNAベクターの単一連続注射を受けた5つの異なるマウスのグループから試験した。
リツキシマブの発現増強
この実施例では、1グループ当たり250gmのSprague-Dawley雌ラット2匹に、最初に40mg/kgのデキサメタゾンを前注射し、次いで4400nmolの中性DMPC脂質を含むまたは含まないリポソーム二重層に組み込んだパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含む4400nmolのDOTAP SUVリポソーム、その後EF1駆動リツキシマブcDNAを含む360μgの二重カセット単一プラスミドDNAベクターを連続注射した(図36、40、及び41の構築物を参照)。ヒトリツキシマブタンパク質の血清レベルを、注射後7日間隔で評価した。図29は、単一IV注射後少なくとも15日間、中性脂質も投与されたラットにおいて、著しく高いレベルの血清リツキシマブタンパク質が産生されたことを示している。
コドン最適化リツキシマブ発現
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、DOTAPリポソームのリポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含有する1000ナノモルのリポソームの単回IV注射を受けた。この2分後、リツキシマブをコードする2重カセット単一プラスミドEF-1駆動DNAベクター100ugの単回IV注射を行った(図36、42及び43の構築物を参照)。番号のついたプラスミド(図30)は、コドン最適化リツキシマブDNA配列ではなくオリジナルのCpG非含有コドン最適化バージョンであった。リツキシマブの血清レベルを、注射の24時間後にELISAによって決定した。図30は、連続注射後24時間で、コドン最適化DMAベクター6が、非コドン最適化リツキシマブDNAベクターよりも著しく高いレベルの血清リツキシマブタンパク質を産生したことを示している。
コドン最適化リツキシマブ発現
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、リポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含有するDOTAPカチオン性リポソーム(示されたように900ナノモルまたは1050ナノモル)の単回IV注射を受けた。この2分後、リツキシマブをコードする二重カセット、コドン最適化単一プラスミドDNAの75ugの単回IV注射を行った(図36、42及び43の構築物を参照)。両方のグループを、IV注射の2時間前に、40mg/kgデキサメタゾンのIP注射で処置した。リツキシマブの血清レベルを、注射の24時間後及びその後の7日間の間隔でのELISAによって決定した。図31は、コドン最適化2重カセット単一プラスミドリツキシマブDNAベクターの1回の連続IV注射により、少なくとも次の60日間は拡大した血清リツキシマブレベルを産生することを示している。血清リツキシマブレベルは、単回の連続IV注射後、経時的に上昇する。
バルプロ酸とテオフィリンはタンパク質発現を増加させる
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、DOTAPリポソームのリポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含有する1050nmolのリポソームの単回IV注射を受けた。この2分後、リツキシマブをコードする二重カセットプラスミドDNA75ugの単回IV注射が行われた。全てのグループを、IV注射の2時間前に、40mg/kgのデキサメタゾンのIP注射により処置した。図32に示す場合、15mg/kgのバルプロ酸(VPA)、2mg/kgのVPA、30mg/kgのテオフィリン(Theo)または15mg/kgのTheoのIP注射によっても動物に前処置した。血清リツキシマブレベルは、注射の24時間後にELISAによって測定した。図32は、薬剤バルプロ酸またはテオフィリンによる前処置によって産生される血清リツキシマブレベルが顕著に増加したため、脂質またはDNAの用量を変えずにタンパク質レベルをさらに高めるための枠組みを提供することを示している。
2重または単一カセットリツキシマブの発現
この実施例では、40mg/kgのデキサメタゾンをマウスのIPに前注射した。2時間後、これらに、リポソーム二重層に組み込まれたパルミテートに共有結合した2.5モル%のデキサメタゾン(DexP)を含む1050または1500nmolのDOTAP
SUVリポソーム、続いて60μgまたは75μgのプラスミドDNAを連続的に注射した。注射されたプラスミドDNA構築物は、2A自己切断ペプチドDNA配列によって分離されたリツキシマブ重鎖及び軽鎖の配列を含有する、コドン最適化二重カセット単一プラスミドDNAベクター(図36、42及び43の構築物を参照)、またはコドン最適化単一カセットプラスミド(図36、37及び39の構築物を参照)のいずれかであった。血清リツキシマブレベルは、注射の24時間後にELISAによって測定した。図33は、連続注射後24時間で、リツキシマブをコードする単一のカセットベクターを含有する2Aペプチドを受けたマウスは、400ng/mlに近い血清レベルを産生し、これは二重カセットベクターによって産生されるレベルの約3分の1に当たることを示している。従って、顕著なリツキシマブ血清レベルは、二重または単一カセットの2Aペプチド含有DNAベクターのいずれかによって産生され得る。
脂質対DNA比及びリツキシマブ発現
この実施例では、グループ当たり3匹のマウスに、乳酸加リンゲル液(LR)に注射当たり100uLの最終容量で懸濁された、リポソーム二重層に組み込まれたパルミテートに共有結合したデキサメタゾン(DexP)2.5モル%を含有するDOTAP SUVリポソームの900、1050、1200、1500または1650nmolをIV注射し、その2分後にCPGを含まない二重カセット単一プラスミドリツキシマブベクター75ugを注射当たり100μLでIV注射した。偽注射されたマウスは、脂質またはDNAなしでのみLRを受けた。24時間後、リツキシマブタンパク質及びALTの血清レベルをアッセイした。図34は、DOTAP SUV脂質対プラスミドDNA(ナノモル脂質:mgDNA)比が15:1未満のものを連続的に注射されたマウスのリツキシマブタンパク質及びALTレベルが、脂質もDNAも注射されていないコントロールマウスのバックグラウンド(正常)ALTレベルの1.5倍以内の血清ALTレベルを産生しながら著しく高いリツキシマブタンパク質レベルを産生した。
バルプロ酸またはテオフィリンによる第IX因子の発現
この実施例では、1グループ当たり3匹のマウスに注射した。各マウスは、リポソーム二重層中に組み込まれたパルミテートに共有結合したデキサメタゾン(DexP)2.5モル%を含有する1500ナノモルのDOTAP SUVリポソームの単回IV注射を受けた。この2分後、ヒト第IX因子のcDNAをコードするコドン最適化EF-1駆動単一カセットDNAベクター(図38及び44の構築物を参照)の60ugの単回IV注射を行った。IV注射の2時間前に、全てのグループを、40mg/kgのデキサメタゾンのIP注射で処置した。示されている場合、動物は、2mg/kgのバルプロ酸(VPA)または30mg/kgのテオフィリン(テオ)のIP注射によっても前処置された。血清ヒト第IX因子レベルを、注射後24時間でELISAによって決定した。各マウスはG-CSFと同様に採血した。血液を、凝固を防ぐためにEDTAカリウムまたはクエン酸ナトリウムを含むチューブに採取し、遠心分離して血漿を得た。ヒト第IX因子に特異的なAssayPro ELISAを製造者の指示書に従って使用し、第IX因子発現を測定した。図35は、24時間で産生された血清ヒト第IX因子レベルが、ヒト第IX因子DNAベクターを受け、加えてバルプロ酸またはテオフィリンのいずれかを用いた前処置を受けたマウスにおいて、著しく高いことを示している。
リポソームのサイズ決定
この実施例では、種々のリポソームのサイズを決定した。特に、表5のリポソームを5%w/wのグルコースで調製し、そのサイズを、準弾性レーザ光散乱を用いて決定した。これらのDOTAPリポソームのZ-平均粒径を表5に示す。
Claims (1)
- 対象において第1の治療用タンパク質及び/または生物学的に活性な核酸分子を発現させる方法であって、
a)第1の組成物を対象に投与し、
前記対象は、疾患または病態の少なくとも1つの症状を有するか、または少なくとも変化する生理学的形質を有し、
前記第1の組成物は第1の量のポリカチオン性構造体を含み、
前記第1の組成物は核酸分子を含んでいないか、実質的に含んでおらず、
b)前記第1の組成物を投与して約300分以内に前記対象に第2の組成物を投与し、
前記第2の組成物は、治療有効量の発現ベクターを含み、
前記発現ベクターはCpG非含有またはCpGが減少されており、
前記発現ベクターは、それぞれ、i)第1の治療用タンパク質及び/またはii)第1の生物学的に活性な核酸分子をコードする第1の核酸配列を含み、
前記第1の量のポリカチオン性構造体と前記治療有効量の発現ベクターとの比が5:1~25:1であり、
前記第1の組成物の投与及び前記第2の組成物の投与の結果として、前記第1の治療用タンパク質及び/または前記生物学的に活性な核酸分子は、前記疾患または病態に関して治療レベルで、または前記生理学的または疾患形質を変化させるのに十分な有効レベルで、対象において発現されることを含む、前記方法。
