II.ATF5ペプチド及び組成物
ATF5ペプチド
ATF5は、N末端酸性活性化ドメイン、及びC末端塩基性ロイシンジッパー(bZIP)ドメインを有する、282アミノ酸の真核細胞転写因子である。bZIPドメインは、DNA結合領域及びロイシンジッパー領域を含有する。ロイシンジッパーは、二量体化ドメインにおいて、通常、7つのアミノ酸毎にロイシンを有する、一般構造モチーフである。bZIP転写因子は、ロイシンジッパーを介してホモ及び/またはヘテロ二量体化し、DNAに特異的に結合する。野生型ヒト、ラット、及びマウスATF5は、それぞれ、NCBI寄託番号NP_001180575、NP_758839、及びNP_109618に説明されているアミノ酸配列を有する。
定性及び阻害活性のために、強化ロイシンジッパー領域が必要とされることが、以前の研究で示された(Krylov et al.1995;Olive et al.1997;Moll et al.2000;Acharya et al.2006)。それ故、延長ロイシンジッパー領域の不存在下において、ST-3が、新生細胞を特異的に標的にして殺傷する能力を保持しているという発見は、予想されなかった。
D-アミノ酸配列
加の試薬、及び/または1つ以上の追加の活性剤を更に含有することができる。キットは通常、キットの内容物の意図される使用を示す標識を含む。用語「キット」は、キット上に、もしくはキットとともに提供される、または別様においては、キットに付属する、あらゆる、記述または記録された材料を含む。
細胞の標的化
当該技術分野において既知の方法により、本発明のATF5ペプチドを標的細胞に導入することができる。選択される導入方法は例えば、目的の用途に応じて左右される。
長さが7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29、30、31、32、33、34、35、36、37、38、または39アミノ酸、例えば、10~30アミノ酸である。CPPは、ポリペプチド、ポリヌクレオチド、及びナノ粒子などの、共有結合した、または非共有結合した分子カーゴを、細胞膜及びBBBを超えて輸送する能力を有する。転座は、転座によるエンドサイトーシス性、またはエネルギー依存性(即ち非エンドサイトーシス性)であることができる。多数のCPPが、文献に記載され、識別されている(例えば、Handbook of Cell-Penetrating Peptides(2d ed.Ulo Langel ed.,2007);Herve et al.2008;Heitz et al.2009;Munyendo et al.2012;Zou et al.2013;Krautwald et al.2016を参照のこと)。CPPのキュレーションされたデータベースは、crdd.osdd.net/raghava/cppsiteにて維持されている(Gautam et al.2012)。

III.調製方法
本発明のATF5ペプチドは例えば、固相ペプチド合成もしくは液相ペプチド合成、または両方の組み合わせを使用して、化学的に合成することができる。合成は、ペプチドの断片として行われ得、その後に化学的、または酵素的のいずれかにより組み合わせられる。本発明のATF5ポリペプチドは、組み換え法を用いて発現することができる。
ある、所望のATF5ペプチドのアミノ酸配列、及びこれらのコドンの選択に基づき設計することができる。標準的な方法を適用して、対象のATF5ペプチドをコードする核酸分子を合成することができる。
実施例1。保存的システイン置換を有するST-3多様体は、インビトロ活性を有する
細胞毒性活性に対する、アミノ酸位置21におけるST-3の1つのシステインの必要性を、HL60細胞における細胞毒性アッセイを使用して試験した。システイン残基はネイティブATF5ドメインの中で非常に保存され、DNA結合の前に、ATF5のホモ二量体化のために必要であると考えられている。

実施例2。非保存的置換を有するST-3多様体は、インビトロ活性を有する
本実施例で議論されるST-3多様体を、表4にまとめている。ST-3は、約12~20μMのEC50を有する。
実施例1に記載のHL60細胞生存能アッセイを使用して、1つの非保存的アミノ酸置換を含有するST-3多様体の細胞毒性活性を試験した(図3)。多様体ST-7は、位置28において、極性のアスパラギンを無極性のロイシンで置換しており、約3μMのEC50を有する。ST-8は、位置22において、極性のグルタミンを無極性のアラニンで置換しており、約5μMのEC50を有する。ST-7及びST-8の活性におけるこの増加は、ST-3よりも3~6倍高い。ST-6は、位置37において、セリンをアラ
ニンで保存的置換しており、ST-3に相当する活性を有する。
複数の置換
実施例1に記載のHL60細胞生存能アッセイを使用して、複数の非保存的アミノ酸置換を含有するST-3多様体の細胞毒性活性を試験した。多様体ST-9は、それぞれ、正電荷を帯びるアルギニンで置換された、負電荷を帯びる2つのグルタミン酸残基、ならびに、それぞれ、負電荷を帯びるグルタミン酸で置換された、正電荷を帯びる1つのアルギニン、及び正電荷を帯びる1つのリジンを有する。(表4を参照のこと。)これらの変化には、約18%の、ST-3のATF5ロイシンジッパー領域を伴った。ST-9の活性は、ST-3の活性よりも著しく増加した(図4)。ST-9は、ST-3の約12~20μMと比較して、約2μMのEC50を有する。
実施例3。レトロ逆転ST-3多様体はインビトロ活性を有する
実施例1に記載したHL60細胞生存能アッセイにおいて、ST-11のレトロ逆転形態の細胞毒性を試験した。レトロ逆転多様体のST-13は、ST-11に相当する活性を有し、ST-3よりも優れた活性を有した(図5)。
細胞生存能を定量化した。
ST-13(図6A~6E)、及びST-14(図7A~7E;表5)は共に、広範囲にわたる腫瘍細胞型において、著しい細胞毒性活性を示した。ST-14は、末梢血単核球細胞(PBMC)及び骨髄単核球(BMMC)においてもまた、細胞毒性であった(表5)。
実施例4。ST-3多様体はインビボ活性を有する
HL60皮下腫瘍モデルにおいて、腫瘍体積におけるST-3、及び3つの多様体の効果を試験した。簡潔に述べると、Matrigelに1:1で懸濁した、5×106のHL60細胞を、皮下注射により、NU/Jマウスの腋窩に移した。腫瘍播種の2日後に投与を開始した。平均の腫瘍体積は、144~176mm3であった。25mg/kgの用量で、腹腔内(IP)注射によりATF5ペプチドを1日2回投与した。試験した多様体は全て、レトロ逆転多様体であるST-13を含むビヒクルと比較して腫瘍体積を減少させた。このことは、最大の抗腫瘍活性を示している(図9)。
下注射により、NU/Jマウス(MCF7及びHL60細胞に関して)、またはNOD/SCIDマウス(U251及びA375細胞に関して)の腋窩に移した。

iv.E9が正電荷を帯びる残基で置換されている;
v.R11が負電荷を帯びる残基で置換されている;
vi.N12が無極性残基で置換されている;
vii.K16が負電荷を帯びる残基で置換されている;
viii.S21がアラニンで置換されている。 - 短縮ATF5ロイシンジッパー領域を含むATF5ペプチドであって、前記短縮ATF5ロイシンジッパー領域が、LEGEGQGLEARNRELKERAESV(配列番号54)、LEGEAQGLEARNRELKERAESV(配列番号55)、LEGECQGLEARNRELKERAEAV(配列番号56)、LEGECQGLEARLRELKERAESV(配列番号57)、LEGECAGLEARNRELKERAESV(配列番号58)、LEGRCQGLRAENRELEERAESV(配列番号59)、LEGRCQGLRAELRELEERAEAV(配列番号60)、及びLEGRAQGLRAELRELEERAEAV(配列番号61)からなる群から選択されるアミノ酸配列を含む、前記ATF5ペプチド。
- 細胞膜透過性領域を更に含み、前記ATF5ペプチドが細胞膜透過性ATF5ペプチドである、請求項1または請求項2に記載のATF5ペプチド。
- 前記細胞膜透過性領域が、RQIKIWFQNRRMKWKK(配列番号25)、RQLKLWFQNRRMKWKK(配列番号26)、YGRKKRRQRRR(配列番号40)、及びYGRKKRRQRR(配列番号41)からなる群から選択される、請求項3に記載の細胞膜透過性ATF5ペプチド。
- アミノ酸配列RQIKIWFQNRRMKWKKLEGECQGLEARNRELKERAESV(配列番号3)の多様体を含む細胞膜透過性ATF5ペプチドであって、前記多様体が、以下からなる群から選択される配列番号3の位置において改変されている、前記細胞膜透過性ATF5ペプチド:
i.C21G(配列番号4);
ii.C21A(配列番号5);
iii.Q22A(配列番号8);
iv.E20R、E25R、R27E、及びK32E(配列番号9);
v.N28L(配列番号7);
vi.S37A(配列番号6);
vii.E20R、E25R、R27E、N28L、K32E、及びS37A(配列番号10);ならびに
viii.E20R、C21A、E25R、R27E、N28L、K32E、及びS37A(配列番号11)。 - 短縮ATF5ロイシンジッパー領域を含むATF5ペプチドであって、前記ペプチドが、先行請求項のいずれかに記載のアミノ酸配列と比較して、逆アミノ酸配列中にD-アミ
ノ酸を含む、前記ATF5ペプチド。 - 前記短縮ATF5ロイシンジッパー領域が、D-アミノ酸配列VAEAREELERLEARLGQARGEL(配列番号65)を有する、請求項6に記載のATF5ペプチド。
- D-アミノ酸配列VAEAREELERLEARLGQARGELKKWKMRRNQFWLKLQR(配列番号14)を含み、前記ATF5ペプチドが細胞膜透過性ペプチドである、請求項7に記載のATF5ペプチド。
- 前記ペプチドが延長ロイシンジッパー領域を含まない、先行請求項のいずれかに記載のATF5ペプチド。
- 前記ペプチドが、N末端アセチル基、及び/またはC末端アミド基を含む、先行請求項のいずれかに記載のATF5ペプチド。
- 先行請求項のいずれかに記載のATF5ペプチドを含む組成物。
- 医薬組成物である、請求項11に記載の組成物。
- 請求項1~10のいずれか1項に記載のATF5ペプチドを含むキット。
- 請求項1~10のいずれか1項に記載のATF5ペプチドをコードする核酸分子。
- 新生細胞における細胞毒性の促進方法であって、前記新生細胞を、請求項1~10のいずれか1項に記載のATF5ペプチドと接触させることを含む、前記方法。
- 新生細胞の増殖の阻害方法であって、前記新生細胞を、請求項1~10のいずれか1項に記載のATF5ペプチドと接触させることを含む、前記方法。
