8(CAR-長ヒンジ)および配列番号29(CAR-短ヒンジ)に記載される最終的なヌクレオチド配列を得た。5’および3’の配列を含めることにより、ギブソンクローニングを使用して抗nf P2X7CARをBLIVベクターに組み込むことが可能となった。
BLIV-CARベクターのクローニングおよび評価
[0216]New England Biolabsの5-アルファコンピテント大腸菌細胞(ギブソンアセンブリクローニングキットで提供される)に、生成したBLIV-CARベクターを、製造業者の説明書に従って形質転換した。概要:
- NEBの5-アルファコンピテント大腸菌細胞のチューブを、10分間氷上で解凍した。
- 1pg~100ngのBLIV-CARプラスミドDNAを含有する1~5μlを、細胞混合物に添加し、チューブを4から5回左右に揺らすことによって、混合した。
- 大腸菌およびプラスミドの混合物を、氷上に30分間、混合することなく静置した。- 細胞およびプラスミドの混合物に、42℃で30秒間熱ショックを与えた後、氷上に5分間、混合することなく静置した。
- 950μlのSOCを混合物に添加した後、60分間37℃に加熱し、激しく振盪させた。
- 選択プレートを調製し、37℃に加熱した。
- 細胞の10倍連続希釈物を、SOC溶液中に調製した。
- 各希釈物50~100μlを、選択プレートに撒き、37℃で終夜インキュベートした。
[0219]以下の方法に従って、293T細胞を使用して、3プラスミドプロトコールによりレンチウイルスをパッケージングした。
1日目:293T細胞が、翌日に90~95%コンフルエントになるように、細胞を、T-225フラスコに10%血清を有する35mlのDMEM培地に播種した。
2日目:生成したBLIV-CARプラスミド(または未改変のBLIVプラスミド)のうちのいずれか30ug、30ugのgag-polプラスミドデルタ8.2、および15ugのVSV-Gプラスミド(pMD2.G)を、OptiMEM培地に添加して、最終体積750ulにし、混合した。300ulのPEI溶液を添加し、室温で少なくとも20分間インキュベートした。混合物を、次いで、コンフルエントな293T細胞に添加した後、37℃でインキュベートした。
3日目:プラスミド混合物を添加した24時間後に、上清を293T細胞からデカンテーションし、4℃で保管した。デカンテーションした混合物を、35mlの新しい培地と置き換えた後、37℃でさらにインキュベートした。
4日目:プラスミド混合物を添加した48時間後に、培地を除去し、24時間で採取した上清と合わせた。合わせた上清を、15分間、1500gで回転処理して、残っている細胞残屑をすべて除去した。上清を0.45umのフィルターに通して濾過した後、WX超遠心分離器において17,000rpmで1時間回転処理した。遠心分離した後、上清を、手作業で、チューブに50~200ulを残してデカンテーションした。遠心チューブを、混入および蒸発を防ぐために50mlのネジ蓋のチューブに入れ、ウイルスを4℃で終夜再懸濁させた。
5日目:ウイルスを、遠心チューブの底部から再懸濁させ、新しい1.5mlのチューブに移した。再懸濁したウイルスを、微小遠心チューブにおいて5000rpmで5分間回転処理して、残っている残屑をすべて除去した。
[0221]108個のCD8 T細胞を、RosetteSep(商標)ヒトCD8+T細胞単離キット(Stemcell technologies、Vancouver、Canada)を製造業者の説明書に従って使用して、50mlのヒト血液から単離した。純度の分析により、図7に図示されるように、精製された細胞の76.6%が、CD8+であったことが示された。
- BT549細胞を、単一細胞懸濁液として調製し、PBS中で2回洗浄して、残留血清をすべて除去した。
- 細胞を、室温のPBS中に再懸濁させた。
- 細胞増殖色素eFluor(登録商標)670の10μM溶液を、室温のPBS中に調製した。
- 等量の10μM色素溶液を、調製したBT549細胞に添加して、最終濃度5μMの色素溶液を得た。
- 色素溶液中のBT549細胞を、暗所において37℃で10分間インキュベートした後、10%血清を含有する低温培養培地を4倍量添加することによって標識化を停止した後、暗所において氷上で5分間インキュベートした。
- 最後に、細胞を培養培地中で3回洗浄した後、所望される濃度で培養培地中に再懸濁させた。

代替的な抗nfP2X7キメラ抗原受容体の設計

- 0.1~50ngの各pCDH-CARベクターを、細胞アリコートに添加した。
- アリコートを緩徐に混合した後、氷上で30分間インキュベートした。
- チューブを、水浴において30秒間42℃で熱パルス処理した後、氷上で2分間インキュベートした。
- 0.9mlの予備加熱した(42℃)NZY+培養液を、各チューブに添加した後、225~250rpmで激しく混ぜながら37℃で1時間インキュベートした。
- 最大200μlの形質転換混合物を、抗生物質を含有するLB寒天プレートに入れた後、37℃で終夜インキュベートした。
- コロニーを採取し、さらに終夜培養した。
- プラスミドDNAを、Quicklyseミニプレップキット(QIAGEN)を用いて培養クローンから単離し、EcoRI/Not I消化により消化させて、適切なサイズのCAR-pCDHベクターを有するクローンを特定した。

- 12.5ugのレンチウイルスベクターDNAを、T75cmフラスコにおいて、トランスフェクション1回当たり、3.75ugのpMD2.g(VSV-Gエンベロープ発現ベクター)、6.25ugのpRSV-Rev、および7.5ugのpCMVdelta8.2と、75ulのリポフェクチンを製造業者のプロトコールに従って使用して混合し、終夜インキュベートした。
- 翌朝、培地を取り替え、ウイルスを含む上清を、48時間後に採取した。
- 採取した上清を、300×gで5分間遠心分離した後、0.45umのフィルターを通して濾過した。
- 濾過した上清からのウイルス粒子を、超遠心分離(68,000×gで90分間、4℃、Beckman SW32ローター)によって濃縮した。上清を除去し、ウイルスペレットを、氷上でDMEM中に緩徐に再懸濁させた。
- 100ulのウイルスアリコートを、必要になるまで-70℃で保管した。
抗nf-P2X7CARを発現するCD8 T細胞の産生
[0249]以下の方法に従って、ヒトCD8細胞を精製し、形質導入した。

nf-P2X7受容体または野生型(WT)P2X7受容体を発現する標的細胞の産生
[0253]抗nf-P2X7-CARを発現するCD8細胞の有効性を評価するために、非機能性P2X7受容体(K193A変異を有する)または野生型のP2X7受容体の細胞外ドメインのいずれかを細胞表面に過剰発現するHek293T細胞を、調製した。
- まず、attB隣接DNAフラグメント(EXD2_K193A、配列番号47、およびEXD2_WT、配列番号48)と、attP含有pDONR-107ベクターとの間でBP組換え反応を行って、エントリークローンを生成した。BP組換え反応を使用して、化学的にコンピテントなE.cloni(登録商標)10G細胞(Lucigen(登録商標))を製造業者のプロトコールに従って形質転換した。
- 形質転換した細胞を、50ug/mlのカナマイシン(Sigma)を含有するLB寒天プレートに播種し、37℃で終夜インキュベートした。
- 各プレートから2つのクローンを選択して、カナマイシン(SIGMA)(50ug/ml)を有するLB培養液中で微量培養物(2mL)を調製した。37℃で終夜インキュベーションの後、撹拌を行った。
- 翌日、QIAGEN QuickLyseミニプレップキットを使用して、微量培養物からプラスミドDNAを抽出した。
- 診断用Bam H1-HF(NEB)およびPmel(NEB)消化を行って、組換えクローンを特定した。Bam H1およびBam H1/PmeI消化した後、ゲル電気泳動(図18)によって、EXD2_K193AおよびEXD2_WTクローンの両方が、正しく消化されたことを確認した。
- 形質転換した細胞を、100ug/mlのアンピシリン(SIGMA)を含有するLB寒天プレートに播種し、37℃で終夜インキュベートした。
- 各プレートから6個のクローンを選択して、アンピシリン(50ug/ml)を有するLB培養液中に微量培養物(2mL)を調製し、これを、撹拌しながら37℃で終夜イ
ンキュベートした。
- 翌日、プラスミドDNAを単離し、Bam H1消化を行って、組換えクローンを特定した。組換えクローンは、適切なサイズの3つのバンド(3431、1056、および5844bp、図19を参照されたい)の存在によって特定した。図19において見ることができるように、各プレートから選択された6つすべてのクローンで、適切なサイズの制限酵素処理フラグメントが得られた。
- EXD2_K193AまたはEXD2_WTを含有するpLV-416コンストラクトを形質導入した2つのクローンを、表7に記載されるプライマーで配列決定して、コンストラクトが適切であったことを確認した。
- HEK293細胞を、トランスフェクションの前日に播種した(フラスコ1つ当たり7×106個の細胞)。
- HEK293T細胞に、レンチウイルスパッケージングベクター、ならびにpLV-416-EXD2およびpLV-416-EXD2_WTのいずれかをトランスフェクトした。トランスフェクション効率をモニタリングするために、GFP発現プラスミド(1ug)も含んだ。
- 終夜インキュベートした後、トランスフェクション試薬を含有する培地を除去し、10mlの新しい培地(10%FCSを含むDMEM)と置き換えた。10mlの培地を、24時間後に採取し、必要になるまで-80℃において2mlのアリコートで保管した。別の10mlの新しい培地(10%FCSを含むDMEM)をフラスコに添加し、これを、さらに24時間後に採取した。
- ウイルス粒子を、1200rpmで培地を遠心分離することによって、採取した培地から単離した後、0.45umのフィルターに通して濾過した。ウイルス粒子とともに、濾過した培地を、HEK293細胞のトランスフェクションに使用した。
- HEK293細胞を、形質導入の前日に、T25フラスコに播種した(フラスコ1つ
当たり7×105個の細胞)。
- 翌日に、培地を各フラスコから除去し、上述のプロトコールに従って生成したウイルス粒子を含有する新しい培地を、表8に記載される比に従って添加した。
- 8ug/mLの最終濃度まで、各フラスコにポリブレンを添加した。
- 形質導入していない細胞は、すべてが、G418補充培地で培養した4日後に死滅した。形質導入した細胞株は、G418を含む培地で正常に成長を続けた。

- 機能障害性P2X7受容体が、野生型(機能性)P2X7受容体のATP結合能力と比較して低減されたATPに結合する能力を有する、請求項1または2に記載のキメラ抗原受容体。
- 機能障害性P2X7受容体が、受容体を機能障害性にする立体配置の変化を有する、請求項1から3のいずれか一項に記載のキメラ抗原受容体。
- 立体配置の変化が、アミノ酸のトランス配置からシス配置への変化である、請求項4に記載のキメラ抗原受容体。
- トランス配置からシス配置へ変化したアミノ酸が、機能障害性P2X7受容体のアミノ酸210位のプロリンである、請求項5に記載のキメラ抗原受容体。
