Abstract
Description
そのうえ、癌による死亡の多くは、化学療法薬に耐性のできた癌によるものである。癌が薬剤耐性を獲得するしくみについては、ハウスマン(Housman)ら(Cancers 6: 1769-1792, 2014)において考察されている。そこに詳細に記載されているように、癌は、薬剤の不活性化や代謝(または代謝活性化の防止)、薬剤の標的の変異や改変、ABCトランスポータを介した薬剤排出など、様々なメカニズムにより薬剤耐性を獲得しうる。かかるメカニズムによって、癌は多剤耐性(MDR)となりうる。下記に論じられているように、薬剤耐性は、白金系化学療法剤による治療にとって特に問題である。
Anx−A1に対する抗体が、ある精神状態、特に不安、強迫性障害(OCD)や関連する疾患の治療に有用であることも示されている(WO2013/088111)が、そうなるメカニズムはわかっていない。
WO2005/027965は、Anx−A1がアポトーシス細胞の表面に局在し、抗Anx−A1抗体がアポトーシスを監視するために使用できることを示している。前記文献は、これに基づき、癌の監視および診断にかかる抗体が使用できることを教示している。前記文献はまた、アポトーシス細胞表面におけるAnx−A1の発現が、当該細胞に対する免疫応答を阻害することも教示している。これに基づき、前記文献はまた、アポトーシスが始まった細胞へのAnx−A1の免疫抑制効果をブロックして癌に対する免疫応答を刺激することにより、Anx−A1に結合する抗体が癌治療に使用できると推測している。
(i)前記特異的結合分子は、相補性決定領域(CDR)VLCDR1、VLCDR2、VLCDR3、VHCDR1、VHCDR2、およびVHCDR3を含み、前記CDRそれぞれは以下のアミノ酸配列を有する。すなわち、
VLCDR1は、配列番号1、配列番号7または配列番号8に示す配列を有し、
VLCDR2は、配列番号2に示す配列を有し、
VLCDR3は、配列番号3に示す配列を有し、
VHCDR1は、配列番号4に示す配列を有し、
VHCDR2は、配列番号5に示す配列を有し、
VHCDR3は、配列番号6に示す配列を有するか、あるいは各配列において、その配列に対する配列同一性が少なくとも85%であるアミノ酸配列であり、および/または

本発明に係る使用のための特異的結合分子は、全長ヒトAnx−A1(すなわち、ANXA1−002転写産物またはANXA1−003転写産物がコードする、346個のアミノ酸のタンパク質である、配列番号17のAnx−A1)に結合する。特異的結合分子は、ANXA1−004転写産物またはANXA1−006転写産物がコードする断片のような、全長Anx−A1の特定の断片、部分、または変異体(variant)に結合してもよい。
WO2018/146230に開示されている1つの抗体は、下記のCDR配列を有する。

実施例1―細胞の増殖に対する抗体の効果
材料
細胞株MCF7、細胞株MCF―7/TAMR7、細胞株A2780、細胞株A2780cis、細胞株A2780ADR、細胞株HCT116、細胞株Caco−2、細胞株SW480、細胞株COR−L23、細胞株COR−L23.5010、細胞株MIA−PaCa−2、細胞株PANC−1、および細胞株BxPC−3は、Public Health England Culture Collectionsから入手した。細胞株HCC1806は、ATCCから入手した。MCF7は、エストロゲン受容体とプロゲステロン受容体とに陽性であるヒト乳腺癌細胞株である。MCF−7/TAMR7は、MCF7のタモキシフェン耐性誘導体である。HCC1806は、トリプルネガティブヒト乳癌細胞株である。A2780は、ヒト卵巣癌細胞株であり、A2780cisは、シスプラチン耐性ヒト卵巣癌細胞株(A2780由来)である。A2780ADRは、アドリアマイシン耐性ヒト卵巣癌細胞株(A2780由来)である。MIA PaCa−2は、ヒト膵癌細胞株である。BxPC−3は、ヒト膵臓腺癌細胞株である。PANC−1は、ヒト膵臓類上皮癌細胞株である。Caco−2は、ヒト結腸直腸腺癌(colorectal carcinoma)細胞株である。HCT116は、ヒト結腸直腸癌細胞株である。SW480は、ヒト結腸直腸腺癌(colorectal adenocarcinoma)細胞株である。COR−L23は、ヒト肺大細胞癌細胞株である。COR−L23.5010は、COR−L23のアドリアマイシン耐性誘導体である。
L1M2H4抗Anx−A1抗体およびL2M2H2抗Anx−A1抗体は、WO2018/146230に開示され、配列は本明細書に記載されている。L1M2H4抗体は、配列番号13に示すアミノ酸配列を有する軽鎖と、配列番号14に示すアミノ酸配列を有する重鎖とを有する。L2M2H2抗体は、配列番号15に示すアミノ酸配列を有する軽鎖と、配列番号16に示すアミノ酸配列を有する重鎖とを有する。
抗Anx−A1抗体ab65844は、アブカム(Abcam)社(英国)から入手した。前記抗体は、ヒトAnx−A1アミノ酸3−24(配列番号30)に結合するポリクローナルウサギ抗体である。このエピトープ配列は、Ca2+非存在下でAnx−A1のコアのポケット内で結合する、Anx−A1のN末端領域の一部を形成する。これは上述した通りである。
細胞培養
初期増殖アッセイにおいて、細胞を下記培地で培養した。DMEM+グルタマックス、10%のFBS+ペニシリン/ストレプトマイシン(MCF7、MIA PaCa−2、HCC1806)、PRMI1640+2mMのL−glu,10%のFBS+ペニシリン/ストレプトマイシン(BxPc−3、A2780)。A2780cisおよびA2780ADRは、A2780と同一の増殖培地で培養されたが、様々な培養の段階で、薬剤耐性を維持するためにそれぞれの薬剤を含有した(すなわち、A2780cisはシスプラチンを、A2780ADRはアドリアマイシンを含有した)。
さらなる増殖アッセイにおいて、細胞を下記培地、下記条件下で培養した。
MCF7、HCC1806、MIA PaCa−2、PANC−1を、10%のFBS、1%のペニシリン/ストレプトマイシン、および1%のL−グルタミンを含有するDMEM中で培養。
MCF7/TAMR7を、1%のFBS、1%のペニシリン/ストレプトマイシン、1%のL−グルタミン、および1%のインスリンを含有するフェノールレッド不含のDMEM/F12中で培養。
A2780、COR−L23,COR−L23.5010、およびBxPC−3を、10%のFBS、1%のペニシリン/ストレプトマイシン、および1%のL−グルタミンを含有するRPMI中で培養。
HCT116を、10%のFBS、1%のペニシリン/ストレプトマイシン、および1%のL−グルタミンを含有するMcCoy’s5A中で培養。
SW480を、10%のFBS、1%のペニシリン/ストレプトマイシン、および1%のL−グルタミンを含有するL−15中で培養。
Caco−2を、10%のFBS、1%のペニシリン/ストレプトマイシン、1%のL−グルタミン、および1%の非必須アミノ酸溶液を含有するMEM中で培養。
さらに、COR−L23.5010を、アドリアマイシンの存在下で培養し、薬剤耐性を維持させた。細胞株はすべて、37℃で、5%のCO2を含む雰囲気中で培養した。
細胞の増殖は、細胞代謝活性を測定するためにMTT比色アッセイを用いて測定した。このアッセイでは、NADPH依存性細胞酸化還元酵素が、黄色テトラゾリウム色素、つまりMTTを、還元して不溶性紫色ホルマザン生成物とし、分光光度計を用いて500〜600nmでの吸光度を測定することによって、これを定量する。ホルマザンの量は、細胞増殖のレベルに比例し、急速に分裂する細胞は、より高いレベルのMTTを還元する。アッセイは3回行った。細胞は、100μLの最終体積で播種した。
初期増殖アッセイにおいて、細胞は下記の密度で播種した。1×105/mL(MCF7、MIA PaCa−2、およびn=2のA2780cis)、2×105/mL(A2780、A2780ADR、およびn=1のA2780cis)、5×104/mL(HCC1806)および2.5×104/mL(BxPC−3)。
さらなる増殖アッセイにおいて、細胞は下記の密度で播種した。
MCF7、HCC1806、A2780、A2780、A2780cis、COR−L23、およびHCT116の細胞を、5×103/ウェルずつ播種し、
MCF7/TAMR7、COR−L23.5010、SW480、Caco−2、MIA PaCa−2、BxPC−3、およびPANC−1の細胞を、1×104/ウェルずつ播種した。
そして細胞をアッセイ前に24時間培養し、その後細胞増殖を測定した。細胞増殖は、下記プロトコルのうちの一つにしたがって測定した。
(a)抗体非存在下(コントロールとして)、抗体(L1M2H4またはL2M2H2)を1μM加えて、または抗体(L1M2H4またはL2M2H2)を10μM加えて、48時間培養した。MTTアッセイは、最大3回別々に、様々な癌細胞株で行われた(初期増殖アッセイ)。または、
(b)抗体非存在下、または2.5μM、5μM、7.5μM、または10μMの濃度の抗体存在下で72時間培養した。このプロトコルで用いられた抗体は、L1M2H4(本明細書においてはMDX−124とも称される),市販の抗Anx−A1抗体ab65844、およびアイソタイプコントロールとしての非Anx−A1特異的IgG(サーモフィッシャーサイエンティフィック社、米国、カタログ番号31154)であった(さらなる増殖アッセイ)。
コントロール培養の細胞数は、増殖アッセイ用のベースラインカウントと定められた。抗体がある培養の細胞のカウントは、ベースラインを基準に標準化し、ベースライン値のパーセンテージとして提示した(“生存率”と称する)。細胞増殖アッセイ結果の統計的な分析は、マン・ホイットニーのU検定を用いて行われた。
ELISAは、標準的なELISA技術を用いてThe Antibody Company社(英国)によって行われた。25μg/mlの全長Anx−A1またはAnx−A1 N末端ペプチド(Anx−A1アミノ酸2−26、配列番号31に対応)、およびコーティング緩衝液(1mM CaCl2を添加した45mM Na2CO3、pH9.6)を用いて、4℃で17時間、ELISAプレートをコーティングした。その後、ブロッキング緩衝液(1mM CaCl2、10mM HEPES、2%w/v BSA)を用いて、室温で1.5時間、プレートのブロッキングを行った。
次いで、一次抗体(ab65844)をプレートに加えた。抗体は、プレートの全体に4倍希釈で2回塗布した。すなわち、1μg/mlの濃度から始まって、最後は2.38×10−7μg/mlの濃度とした。0.1 BSAを添加した洗浄緩衝液(10mM HEPES,150mM NaCl、0.05%(v/v) Tween−20、および1mM CaCl2)で抗体を希釈した。一次抗体を、プレートに室温で1時間添加した後、洗浄緩衝液を用いてプレートを洗浄した。
次いで、検出抗体を加えた。検出には、西洋ワサビペルオキシダーゼ(HRP)結合ヤギ抗ウサギ抗体(メルク社、ドイツ、カタログ番号AP156P)を、1:3000に希釈して用いた。これを、室温で1時間、ELISAプレートに加えた。その後、再度、洗浄緩衝液を用いてELISAプレートを洗浄した。
次に、比色用基質であるOPD(o−フェニレンジアミン二塩酸塩、シグマ・アルドリッチ社、P4664)をプレートに加えた。OPD溶液を製造者の説明書にしたがって作製し、リン酸−クエン酸緩衝液(pH5)を用いた0.4mg/ml OPD溶液を得た。使用直前に、OPD溶液100mlあたり40μlの30% H2O2を加えた。その後、得られたOPD溶液100μlを、プレートの各ウェルに加えた。
室温、暗所で20分間、プレートをインキュベートし、その後、50μlの3M H2SO4を加えて反応を停止させた。H2SO4の添加直後に、プレートの492nmにおける吸光度を読み取った。
市販の抗Anx−A1抗体ab65844をELISAによって調べ、その報告されたエピトープの結合を確認した。このアッセイは、抗体ab65844は全長Anx−A1にも、N末端Anx−A1ペプチドにも結合することを示し(データは示していない)、ヒトAnx−A1(配列番号30)のアミノ酸3−24の報告されたエピトープが正しいことを示している。
初めの増殖アッセイは、48時間にわたって増殖を測定し、2つの抗体、すなわち抗体L1M2H4と抗体L2M2H2の、当該細胞株への効果を、抗体が無い状態での(すなわち、上記のプロトコル(a)を用いた)インキュベーションと比較した。
乳癌細胞株についてのこれら増殖アッセイの結果を図1および図2に示す。図示されたように、L1M2H4抗体は、10μMで統計的に有意な効果(p<0.001)を有し、MCF7細胞の増殖を低減した。ただしこれはn=1である。HCC1806細胞株において、L2M2H2抗体もまた、10μMで、十分に有意に増殖を減少させた(p<0.05)(n=2)。
卵巣癌細胞株A2780の増殖アッセイの結果を、図3に示す。図示されたように、L1M2H4抗体とともにインキュベーションすると、10μMで、十分に有意な増殖の減少が見られた(p<0.01)(n=2)。シスプラチン耐性卵巣癌細胞株、A2780cis(図4)では、L1M2H4抗体とともにインキュベーションすると、1μM(p<0.001)および10μM(p<0.01)で、統計的に有意な増殖の減少が見られ(n=2)、L2M2H2抗体を使用すると10μMで、十分に有意な増殖の減少が見られた(p<0.05)(n=3)。アドリアマイシン耐性の卵巣癌細胞株A2780ADRの増殖アッセイの結果を、図5に示す。L2M2H2抗体は、これら細胞の増殖に対して有意な効果を有していた。
膵臓癌細胞株の増殖アッセイの結果を、図6および図7に示す。
前記増殖アッセイを繰り返し、72時間かけて増殖を測定した。これらアッセイは、本発明のある抗体(L1M2H4,MDX−124としても知られる)の効果を、非特異的IgGコントロールの増殖に対する効果と比較し、記載のある場合は市販の抗Anx−A1抗体ab65844の増殖に対する効果と比較した。抗体非存在下での増殖に対する比較も行い、ベースラインとして使用した。実験はすべて3回行った(MDX−124とIgGコントロールについて3回行い、ab65844も調べられた場合については、この抗体を用いた実験は2回行った。)
MDX−124に暴露することにより、乳癌(トリプルネガティブ細胞株、ホルモン受容体陽性細胞株、および薬剤耐性細胞株を含む)、大腸癌、卵巣癌、肺癌、および膵臓癌からの細胞株の増殖が有意に減少した。
MDX−124の癌細胞増殖への影響は、抗体特異的であり、つまり同じ標的(Anx−A1)に対してすべての抗体が同じ影響を有するとは限らない。これは、ab65844が調べられた細胞株のいずれの増殖も有意に減少させることが無く、むしろ多くの場合、増殖を増大させたという事実によって示されている。MDX−124を使用した場合に見られた増殖の有意な減少を、非特異的IgGコントロールが起こすことも無かった。
HDX解析は、L1M2H4抗体を用いて、ドイツ、チュービンゲン大学の自然科学医学研究所(Natural and Medical Sciences Institute)(NMI)にて行った。
抗体―抗原複合体形成および水素―重水素交換
抗体−抗原サンプルの5つのアリコート、および抗体を伴わない抗原の5つのアリコートを、下記のように調製した:0.8μLのAnx−A1(41μM)と、1.8μLの抗体(38.7μM)またはHEPES緩衝液(10mM HEPES,1mM CaCl2,150mM NaCl、pH7.4)をそれぞれと、1μLのHEPES緩衝液と、0.5μLのCaCl2(8mM)とを混合し、10分間20℃でインキュベートした。8.5μLのHEPES緩衝液を添加して、塩含有量を調整した。この抗体−抗原複合体を0℃で一晩、その後15℃で2時間、凍結乾燥させて、水分をできるだけ取り除いた。10個の凍結乾燥アリコートを、HDX交換とLC−MS分析まで−20℃で凍結した。各抗体−抗原複合体と、抗体を伴わない抗原とは、アリコート一つずつを12.5μlのH2O中で可溶化し、その他を12.5μLのD2Oで可溶化した。各アリコートを分析直前に別々に調製し、アリコートを下記時間インキュベートした。
0分間(H2O標準サンプル);
5分間、70分間、360分間、および24時間(D2O重水素交換動的サンプル)。
用意したて(freshly prepared)のクエンチング溶液12.5μLを加えて、前記交換をクエンチした(塩酸グアニジンを0.8M、TCEPを0.4M、100mMの蟻酸アンモニウム緩衝液、pH2.5)。
クエンチング溶液添加直後、0.35μLのペプシン(100μM)を加え、20℃で2分間消化を行った。アリコートをすぐに−20℃に予め冷却されたオートサンプラーバイアルに入れ、予め冷却された注射器を介して液体クロマトグラフィ質量分析(LC−MS)に注入した。
得られたペプチド混合物を、逆相高速液体クロマトグラフィ(HPLC、RSLC3000 LC, Thermo Scientific Dionex, Idstein, Germany)を使用して前処理せずに注入して分離した。サンプルの分離には、LCカラム(ACQUITY UPLC BEH300 C18 1.7 μm 1x50mm Thermo Scientific Dionex, Idstein, Germany)を使用した。空試験運転と、カラム洗浄運転とを、連続したサンプル運転中に行った。
クロマトグラフィによる分離は、ほぼ均一濃度の勾配を31分間使用して行った。溶離液Aは、0.1%蟻酸を含む水であり、溶離液Bは、0.1%蟻酸を含むアセトニトリルであった。最適化した20分の直線勾配を、傾斜を変えて、0℃までの温度で次のように適用した(分/%B):0/8、3/8、11.9/20、31.9/20、33/99、34/99、35/8。手動注入を行った。注入量は、体積20μLのサンプルループを使用して、25.35μLであった。流速は40μL/minであった。HPLCの溶離液は、QTOF型の質量分析計(MaXis HD、ブルカー(Bruker)社)に直接注入した。質量分析計は陽イオンモードで動作し、スプレー電圧は1.9kV、キャピラリ温度は275℃、SレンズRF電圧は55Vであった。
データは、ソフトウェアHDExaminer2.40 beta1、64ビット(SierraAnalytics、米国カリフォルニア州モデスト市)を使用して解析した。簡単に言うと、異なる交換時点(exchange time points)を含む未処理データセットと、各時点について抗体を伴うAnx−A1の分析と抗体を伴わないAnx−A1の分析とを検討した。Anx−A1配列情報と、対応する保持時間と荷電を併記した消化性ペプチド(peptic peptides)の配列リストとを使用して、前記ソフトウェアにより、重水素交換されたペプチドと、重水素交換されていないペプチドとを識別し、重水素化ペプチドと非重水素化ペプチドとの重心質量の違いとしてペプチドごとの重水素取り込みを算出する。重複するペプチド情報(重複するペプチドの個々の質量シフト)を使用して、エピトープ領域を手動でさらに限定した。
HDExaminerを使用した初期データ評価の後、統計的に妥当な重水素取り込みについて個々の消化性ペプチド(peptic peptides)を手動で検証した。いくつかの重複するペプチドの場合、エピトープ領域は、重水素取り込みのあるペプチドのN末端部分およびC末端部分をカバーする、重水素取り込み無しのHDXデータを用いて、さらに限定した。全実験は、2回繰り返した。1回目の実験では、潜在的なエピトープ領域が識別されたが、統計的に妥当な重水素取り込みもまた、N末端を含む非常に長いペプチド中に観察された。構造的な観点からは、これはややフレキシブルである。2回目の実験では、エピトープ領域を確定可能であったが、N末端は重水素取り込みを示さなかった。
配列中の下線の領域は、抗体が結合するエピトープを指す。

方法
忍容性試験
クラウン・バイオサイエンス社(米国)により、忍容性試験がなされた。マウスに2週間のあいだ、週1回、MDX−124を1mg/kg、10mg/kg、または29mg/kg、投与した。各マウスの体重を、実験期間中毎日測定した。体重の減少は、抗体のマウスに対する毒性を示すものと考えられよう。
クラウン・バイオサイエンス社(米国)により、マウスを使用した実験がなされた。使用されるマウスは、8〜9週齢の雌のBALB/cマウスであった。使用された乳癌モデルには、ルシフェラーゼ発現マウス乳癌細胞株4T1−Lucが使用された。細胞株は、ATCCから入手し、10%のFBSと、2mMのL−グルタミンと、2μg/mlのピューロマイシンを含有するRPMI培地で培養した。
マウスは毛をそり、72時間後に個々のマウスの識別のためのトランスポンダ・チップを埋め込んだ。毛をそった後すぐにと、その後は腫瘍を接種するまで毎日、ベパンテンクリームを塗った。
各マウスには、まず、100μlのPBSに懸濁された4T1−Luc細胞を5×104個接種した。接種は、0日目に、マウスにガス麻酔をしながら、乳腺皮下脂肪(左下側、下から2番目の乳腺皮下脂肪)内に行った。接種部位の皮膚は、接種前に70%エタノールで消毒した。
マウスは、グループ間で平均腫瘍体積が一定になるように、1グループに12体ずつ4グループに分けた。治療は、毎週投与することで行った。4つのマウスのグループうち、コントロールのグループには、媒質のみ(PBS)を投与した。3つ実験グループは、PBS中1mg/kg、10mg/kg、または25mg/kgのMDX−124の投与量を摂取した。各投与は、10ml/kgの体積で静脈内に投与した。治療は、最長3週間継続した。週3回の腫瘍の測定は、治療開始後も継続した。マウスの体重測定は、治療前は週3回、治療開始後は毎日行った。
MDX−124がマウスにとってもともと毒性があるかどうかを調べるために、忍容性試験を行った。マウスに抗体を投与し、その体重を監視した。明らかな体重減少はどのマウスにも見られず(データ示していない)、これは調査したどの投与量でも、抗体はマウスに対して毒性が無かったことを示している。
MDX−124の抗癌効果を、乳癌のマウスモデルで調べた。調べたマウスの各グループの平均腫瘍体積を、図20に示す。0日目の腫瘍細胞の接種の後、最初の投与量(treatment dose)を、12日目にすべてのグループに投与した。図示されたように、調査17日目までに、MDX−124で治療されたマウスには、媒質で治療されたコントロールのマウスと比べて、腫瘍体積に有意の減少が見られた。コントロールグループにおいて益々腫瘍が成長する傾向は、19日目まで継続した。MDX−124で治療したグループでは腫瘍体積の減少がみられたが、これはまた、これらのグループの相対腫瘍体積の減少と対応していた(図21参照)。
12日目の腫瘍体積を、ベースラインの腫瘍体積(つまり、相対腫瘍体積100%)と定義する。19日目までに、MDX−124で治療されたマウスの腫瘍は、約2.5倍のサイズに増大していた。コントロールのマウスの腫瘍は、約3.3倍のサイズに増大していた。これは、MDX−124での治療の結果、19日目までに、コントロールと比較して腫瘍の成長を約3分の1減少させたことを意味し、この抗体の抗癌効果を示している。異なる3つの濃度(1mg/kg、10mg/kgおよび25mg/kg)で抗体をマウスに投与したが、これらの治療投薬法のどれも、腫瘍の成長には同じような効果を有した(すなわち、投与された抗体の量を増やしても、治療の効果が上りはしなかったように見える)。
Claims (26)
- 対象における癌の治療に使用するための、ヒトAnx−A1に結合する特異的結合分子であって、
(i)前記特異的結合分子は、相補性決定領域(CDR)VLCDR1、VLCDR2、VLCDR3、VHCDR1、VHCDR2、およびVHCDR3を含み、
VLCDR1は、配列番号1、配列番号7または配列番号8に示す配列を有し、
VLCDR2は、配列番号2に示す配列を有し、
VLCDR3は、配列番号3に示す配列を有し、
VHCDR1は、配列番号4に示す配列を有し、
VHCDR2は、配列番号5に示す配列を有し、
VHCDR3は、配列番号6に示す配列を有するか、または、各配列において、その配列に対する配列同一性が少なくとも85%であるアミノ酸配列であり、および/または
(ii)前記特異的結合分子は、配列番号17の197〜206位、220〜224位、および227〜237位のアミノ酸からなる不連続エピトープでAnx−A1に結合する、特異的結合分子。 - VLCDR1は、配列番号1に示す配列を有し、
VLCDR2は、配列番号2に示す配列を有し、
VLCDR3は、配列番号3に示す配列を有し、
VHCDR1は、配列番号4に示す配列を有し、
VHCDR2は、配列番号5に示す配列を有し、
VHCDR3は、配列番号6に示す配列を有する、請求項1に記載の使用のための特異的結合分子。 - 前記特異的結合分子は抗体またはその断片である、請求項1または2に記載の使用のための特異的結合分子。
- 前記抗体またはその断片は、ヒト化されている、請求項3に記載の使用のための特異的結合分子。
- 前記抗体がモノクローナル抗体であるか、あるいは前記抗体断片が、Fab抗体断片、Fab’抗体断片、もしくはF(ab’)2抗体断片、またはscFv分子である、請求項3または4に記載の使用のための特異的結合分子。
- 前記抗体またはその断片は、
(i)配列番号9または配列番号10に示すアミノ酸配列、あるいは、これらのアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む、軽鎖可変領域と、
(ii)配列番号11または配列番号12に示すアミノ酸配列、あるいは、これらのアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む、重鎖可変領域と、を含む、請求項5に記載の使用のための特異的結合分子。
- 前記特異的結合分子はモノクローナル抗体であり、前記モノクローナル抗体は、
(i)配列番号13に示すアミノ酸配列、または、このアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む軽鎖と、
(ii)配列番号14に示すアミノ酸配列、または、このアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む重鎖とを含む、請求項6に記載の使用のための特異的結合分子。 - 前記特異的結合分子はモノクローナル抗体であり、前記モノクローナル抗体は、
(i)配列番号15に示すアミノ酸配列、または、このアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む軽鎖と、
(ii)配列番号16に示すアミノ酸配列、または、このアミノ酸配列に対して少なくとも70%の配列同一性を有するアミノ酸配列を含む重鎖とを含む、請求項6に記載の使用のための特異的結合分子。 - 前記癌はAnx−A1を発現する、請求項1〜8のいずれか一項に記載の使用のための特異的結合分子。
- Anx−A1は、前記癌の細胞の表面に発現する、請求項9に記載の使用のための特異的結合分子。
- 前記癌は1つ以上の化学療法剤に対して耐性がある、請求項1〜10のいずれか一項に記載の使用のための特異的結合分子。
- 前記癌は、多剤耐性である、請求項11に記載の使用のための特異的結合分子。
- 前記癌は、白金系化学療法剤に対して耐性がある、請求項11または12に記載の使用のための特異的結合分子。
- 前記癌は、シスプラチン、アドリアマイシン、および/または、タモキシフェンに対して耐性がある、請求項11〜13のいずれか一項に記載の使用のための特異的結合分子。
- 前記治療はさらに、前記対象に対して第2の治療薬を投与することを含む、請求項1〜14のいずれか一項に記載の使用のための特異的結合分子。
- 前記第2の治療薬は化学療法剤である、請求項15に記載の使用のための特異的結合分子。
- 前記化学療法剤は、細胞障害性薬剤である、請求項16に記載の使用のための特異的結合分子。
- 前記対象はヒトである、請求項1〜17のいずれか一項に記載の使用のための特異的結合分子。
- 前記癌は、乳癌、大腸癌、卵巣癌、肺癌、および膵臓癌から選択される、請求項1〜18のいずれか一項に記載の使用のための特異的結合分子。
- 請求項1〜8のいずれか一項で定義された特異的結合分子を前記対象に投与することを含む、対象における癌を治療する方法。
- 前記癌、前記治療、および/または前記対象は、請求項9〜19のいずれか一項で定義されるものである、請求項20に記載の方法。
- 対象における癌の治療のための薬剤の製造における特異的結合分子の使用であって、前記特異的結合分子は、請求項1〜8のいずれか一項で定義されるものである、使用。
- 前記癌、前記治療、および/または前記対象は、請求項9〜19のいずれか一項で定義されるものである、請求項22に記載の方法。
- 請求項1〜8のいずれか一項で定義される特異的結合分子と、化学療法剤とを含むキット。
- 請求項1〜8のいずれか一項で定義される特異的結合分子と、対象における癌の治療において、別々に、同時にもしくは逐次的に使用するための、第2の治療薬とを含む製品。
- 前記癌、前記第2の治療薬、および/または前記対象は、請求項9〜14、または16〜19のいずれか一項で定義されるものである、請求項25に記載の使用のための製品。
