Abstract
Description
本出願は、2018年5月22日出願の米国特許仮出願第62/674,843号、2018年11月20日出願の同第62/769,743号、及び2019年5月17日出願の同第62/849,719号の利益を主張する。これらの出願の内容はそれぞれ、参照により本明細書に組み込まれる。
本発明は、TRPV1モジュレーションに使用するためのカンナビノイド及び/又はテルペンに関する。
特許請求の範囲及び明細書において使用される用語は、別段の指定がない限り以下に記載するように定義される。
明細書及び添付の特許請求の範囲において使用されるように、単数形「a」、「an」及び「the」は、文脈上からそうでないことがはっきりしていない限り、複数の指示物を含むことに留意しなければならない。
本発明者らは、様々なカンナビノイド及びテルペンがTRPV1にアゴニスト効果を
有すること、各化合物のアゴニスト効果を、TRPV1発現細胞の電気生理学的分析により、互いに区別できることも、現在の治療用途において主要なTRPV1アゴニストであるカプサイシンの効果と区別できることも発見した。したがって、これらの化合物を単独で又は互いに組み合わせて、カプサイシン又は他の適切な化合物と使用して、TRPV1透過性をモジュレートすることができる。本発明者らのデータは、望ましい治療特性をもつTRPV1アゴニスト及びアゴニスト混合物の選択のための多変数モデルを支持する。方法により、電流振幅、イオン透過特性、及び活性化/不活性化反応速度の所望の範囲によって特徴づけられる製剤のオーダーメードの設計が可能になる。これらのアゴニスト又はアゴニスト混合物は、それらの所定のTRPV1状態関連アゴニスト特性に基づいて、TRPV1の急性的活性化、慢性適用によるTRPV1の脱感作、又はその両方が治療的効果で行われるように選択することができる。
少なくとも1つの化合物は、カンナビノイドでも、テルペンでもよい。
別の態様において、医薬組成物が提供される。医薬組成物は、所定のTRPV1状態関連アゴニスト特性を有する少なくとも1つの化合物及び薬学的に許容される担体又は希釈剤を含む。医薬組成物の文脈の中で、所定のTRPV1状態関連アゴニスト特性を有する少なくとも1つの化合物は、組成物の医薬品活性成分である。
典型的な実施形態において、活性成分は、医薬組成物中に少なくとも0.01mg/ml、少なくとも0.1mg/ml、少なくとも0.5mg/ml、又は少なくとも1mg/mlの濃度で存在する。ある特定の実施形態において、活性成分は、医薬組成物中に少なくとも1mg/ml、2mg/ml、3mg/ml、4mg/ml、5mg/ml、10mg/ml、15mg/ml、20mg/ml、又は25mg/mlの濃度で存在する。ある特定の実施形態において、活性成分は、医薬組成物中に少なくとも30mg/ml、35mg/ml、40mg/ml、45mg/ml又は50mg/mlの濃度で存在する。
医薬組成物は、液剤、油剤、乳剤、ゲル剤、コロイド剤、エアロゾル剤又は固形剤を含めて、ヒト又は動物用医薬に適しているいかなる形もとることができる。
いくつかの実施形態において、気化器、ネブライザー、又はエアロゾル発生器による医薬組成物の投与に適応する、本明細書に記載されている医薬組成物の単位剤形が提供される。いくつかの実施形態において、剤形は、バイアル剤、アンプル剤であり、使用者による開封を可能にするために場合によって割線が付されている。特定の実施形態において、ネブライザーは、ジェットネブライザー又は超音波ネブライザーである。
経口、口腔又は舌下投与用の製剤は、カプセル剤、カシェ剤、丸剤、錠剤、ロゼンジ剤(フレーバー付き基剤、通常スクロース及びアカシア又はトラガカントを使用)、散剤、顆粒剤の形態であり、又は水性若しくは非水性液体中の液剤若しくは懸濁剤として、又は水中油型若しくは油中水型の液体乳剤として、又はエリキシル剤若しくはシロップ剤として、又はパステル剤(不活性な基剤、例えばゼラチン及びグリセリン、又はスクロース及びアカシアを使用)として、及び/又は洗口剤としてなどであり得、それぞれが、活性成分として対象のポリペプチド治療剤の所定量を含有する。懸濁剤は、活性化合物に加えて、懸濁化剤、例えばエトキシ化イソステアリルアルコール、ポリオキシエチレンソルビトール、及びソルビタンエステル、微結晶セルロース、メタ水酸化アルミニウム、ベントナイト、寒天及びトラガカント、並びにその混合物を含有し得る。
静脈内、筋肉内、若しくは皮下注射、又は苦痛の部位における注射の場合、活性成分は、パイロジェンフリーであり、好適なpH、等張性及び安定性を有する非経口的に許容される水性溶液の形態である。当業者は、例えば、等張ビヒクル、例えば塩化ナトリウム注射液、リンゲル注射液、乳酸リンゲル注射液を使用して好適な溶液を調製することがよくできる。保存剤、安定剤、緩衝剤、抗酸化剤及び/又は他の添加剤を必要に応じて含めることができる。
局所投与用の医薬組成物及び製剤としては、経皮パッチ剤、軟膏剤、ローション剤、クリーム剤、ゲル剤、ドロップ剤、坐剤、スプレー剤、液剤及び粉末剤を挙げ得る。通常の医薬用担体、水性、粉末又は油性基剤、増粘剤などが必要であり、又は望ましいことがある。コーティングされたコンドーム、手袋なども有用であり得る。好適な局所製剤としては、本発明において特色をなすカンナビノイド含有複合混合物が局所送達剤、例えば脂質、リポソーム、脂肪酸、脂肪酸エステル、ステロイド、キレート剤及び界面活性剤と混合されているものが挙げられる。好適な脂質及びリポソームは、中性(例えば、ジオレオイルホスファチジルDOPEエタノールアミン、ジミリストイルホスファチジルコリンDMPC、ジステアロイルホスファチジルコリン)、陰性(例えば、ジミリストイルホスファチジルグリセロールDMPG)及びカチオン性(例えば、ジオレオイルテトラメチルアミノプロピルDOTAP及びジオレオイルホスファチジルエタノールアミンDOTMA)を含む。本発明において特色をなすカンナビノイド含有複合混合物はリポソーム内にカプセル化されてもよく、又は複合体をそのリポソーム、特にカチオン性リポソームに形成してもよい。或いは、カンナビノイド含有複合混合物を複合体化して、脂質、特にカチオン性脂質にしてもよい。好適な脂肪酸及びエステルとしては、アラキドン酸、オレイン酸、エイコサン酸、ラウリン酸、カプリル酸、カプリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、リノール酸、リノレン酸、ジカプレート、トリカプレート、モノオレイン、ジラウリン、グリセリル1-モノカプレート、1-ドデシルアザシクロヘプタン-2-オン、アシルカルニチン、アシルコリン、又はC1〜10アルキルエステル(例えば、イソプロピルミリステートIPM)、モノグリセリド、ジグリセリド、又はその薬学的に許容される塩が挙げられるが、これらに限定されない。
インビボ及び/又はインビトロアッセイを、使用の最適用量範囲を特定する助けとなるように場合によって使用してもよい。製剤中で使用される正確な用量は、投与経路及び状態の重篤度にも依存することになり、開業医の判断及び対象それぞれの状況に従って決定されるべきである。有効用量は、インビトロ又は動物モデル試験系に由来する用量応答曲線から外挿によって求めてもよい。
医薬組成物は、好都合なことに単位剤形で提示してもよい。
他の態様において、処置方法が提供される。
以下は、本発明を実施する具体的な実施形態の例である。実施例は、例示の目的で提示されているのすぎず、決して本発明の範囲を限定するものではない。使用される数(例えば、量、温度など)に関して正確度を確実にする試みが行われているが、もちろんいくらかの実験誤差及び偏差を考慮するべきである。
細胞培養:
HEK TRexTRPV1を、DMEM、10%ウシ胎仔血清、2mM Lグルタミン、10μg/mlブラストサイジン(Calbiochem社、San Diego CA)、400μg/mlゼオシン(InvivoGen社、San Diego CA)中で16〜24時間培養し、1μg/mlテトラサイクリンを使用して、指示された導入遺伝子発現が誘導された。別段の指示がない限り、誘導なしのTRPV1の基底発現がこれらの研究に十分であり、必要なら、トランスフェクトされていないHEKと比較を行った。ヒトTRPV2、ヒトTRPA1及びヒトTRPM8を過剰発現するHEKTRex293をSB Drug Discovery社(Glasgow, Scotland)から入手し、上記のように培養した。カンナビノイド及びテルペンは、TRPV1イオンチャネルタンパク質をトランスフェクトされたHEK細胞においてアッセイした。これらの細胞をHEK293-V1細胞と呼ぶ。
一般的な化学物質は、VWR社(West Chester, PA)及びSigma Aldrich社(St. Louis, MO)から入手した。PMA及びイオノマイシンは、Calbiochem社(Gibbstown, NJ)から入手した。IgE抗DNPは、Sigma社から、KLH-DNPは、Calbiochem社から入手した。カプサイシン及びカプサゼピンは、Sigma Aldrich社から入手した。カンナビジバリン(CBDV)、カンナビクロメン(CBC)、カンナビジオール(CBD)、カンナビジオール酸(CBDA)、カンナビゲロール(CBG)、カンナビゲロール酸(CBGA)、カンナビノール(CBN)は、Sigma Aldrich社から入手した。
細胞を洗浄し、以下の組成(単位mM)の標準改変リンゲル液:NaCl 145、KCl 2.8、CsCl 10、CaCl2 10、MgCl2 2、グルコース 10、Hepes・NaOH 10、pH7.4、330mOsm中で、0.2マイクロモル濃度のFluo-4と共に37℃で30分間インキュベートした。細胞を96ウェルのプレートに50,000細胞/ウェルで移し、指示の通り刺激した。Flexstation 3(Molecular Devices社、Sunnydale, USA)を使用して、カルシウムシグナルを取得した。SoftMax(登録商標) Pro 5(Molecular Devices社)を使用して、データを分析した。指示されている場合、名目上カルシウム不含の外部状態を、1mM EGTAを含有する0mM CaCl2リンゲル液の調製によって達成した。
この技法を使用して、HEK293-V1からのイオン電流を研究する。細胞膜の両端間電圧は制御され、電圧の変化に伴うイオン電流が測定される。マイクロピペットチップを直径数マイクロメートル引き、次にチップを加熱して、平滑面を創出する。その平滑面は、細胞膜と高抵抗性シールを形成することを支援する。この高抵抗性シールを得るために、マイクロピペットを細胞膜に押しつけ、吸引する。次いで、膜はガラス表面に溶融し、ギガオームの範囲の高抵抗性を生じる。これにより、膜パッチの両端又は細胞全体の両端で測定されたイオン電流を電子的に単離することが可能になる。
パッチクランプ実験を21〜25℃にてホールセル構成で行った。パッチピペットは、2〜3MΩの抵抗を有した。PatchMasterソフトウェア(HEKA社、Lambrecht, Germany)を用いて、EPC-9増幅器を制御して、データを取得した。-100〜100mVの電圧範囲に及ぶ50msの電圧ランプは、500ms〜180秒(3分間)にわたって0.5Hzの速度で0mVの保持電位から送達された。電圧を10mVの液間電位に対して補正した。電流を2.9kHzでフィルタリングし、100μsの間隔でデジタル化した。容量性電流を決定し、各電圧ランプ前に補正した。所与の電位に対する電流の発生は、-80mV〜+80mVの電圧で電流振幅を測定することによって個々のランプ電流の記録から抽出された。FitMaster(HEKA社、Lambrecht, Germany)及びIgorPro(WaveMetrics社、Lake Oswego, OR, USA)を用いて、データを分析した。適用可能な場合、平均データの統計誤差が、n決定子で平均値±s.e.mとして与えられる。活性化電流振幅は、pA/pFではなくnAにより分析される。この決定は、チャネルのTRPV1トランスフェクションに起因する高電流振幅(1〜6nA)のために行われた。また、パッチするために選択される細胞サイズは、9〜12pFの範囲であった。nA及びpA/pFにおける分析を比較し、有意差は、主としてnA範囲における一貫した細胞サイズ選択及び高電流振幅によって見出されなかった。
ラットTRPV1を過剰発現させる単一のHEK293細胞におけるパッチクランプ実験を介して、チャネル電流を評価した。140mM NaCl、1mM CaCl2、2mM MgCl2、2.8mM KCl、11mM グルコース、及び10mM HEPES-NaOHを含有するpH7.2及び容量オスモル濃度300mOsmolのナトリウムベース細胞外リンゲル液中で、HEK293細胞を維持した。細胞のサイトゾルを、140mM Cs-グルタメート、8mM NaCl、1mM MgCl2、3mM MgATP、及び10mM HEPES-CsOHを含有する細胞内パッチピペット溶液で灌流した。標準内部Ca2+濃度を、4mM Ca2+及び10mM BAPTAで180nMに緩衝した。WebMaxCを備えた計算機(http://www.stanford.edu/~cpatton/webmaxcS.htm)を使用して、遊離非緩衝Ca2+のレベルを調整した。最終溶液のpHをpH7.2に調整し、容量オスモル濃度を300mOsmolで測定した。カンナビノイド、テルペン、又はカプサイシンを上記の外部溶液に添加することによって、TRPV1電流を活性化した。
結果を一般に平均値+標準偏差として示す。電気生理の結果のエラーバーは、標準誤差(SEM)を示す。スチューデントのt検定又はANOVAに基づいて、統計学的有意差を決定した。各グラフにおいてデータ点に隣接して、有意差を以下の通り記録した:シングルアスタリスク、p<0.05;ダブルアスタリスク、p<0.01;トリプルアスタリスク、p<0.001;符号なし、p>0.05。実験はすべて少なくとも3のnである。
カプサイシンのTRPV1状態関連アゴニスト特性
いくつかの先行研究から、TRPV1は、疼痛過敏の機序に寄与し得るチャネル特性の動的変化を起こすことができるということが示唆される。チャネル特性の動的変化により、本発明者らは、TRPV1イオンチャネルのイオン選択性を指す。生物物理学的には、イオン選択性は、選択されたイオン種に対して高透過を有するチャネルの能力を指す。標準的なイオンチャネルのイオン選択性は、様々な生理的条件下で変化すると考えられていない。この理解は、TRPV1に関する最近の研究によって検証され、特定のイオンに対するその選択性は開口細孔(状態1)から拡張細孔(状態2)に変化することができる。状態1において、チャネルは、Na+及びCa2+の両方に対してイオン非選択的及び透過性である。アゴニストであるカプサイシンへの曝露によって、状態2に拡張する細孔が生じ、細孔は、大きなカチオン、例えばN-メチル-D-グルカミン(NMDG)の透過を可能にすることに加えて、Ca2+及びNa+イオンの高フラックスを可能にする。
カンナビノイド及びミルセンのTRPV1状態関連アゴニスト特性
次に、様々なカンナビノイド又はテルペンがイオン選択TRPV1活性化を誘導する能力、及び状態1又は状態2の状態の得られたTRPV1活性化を評価した。様々な濃度のカンナビノイド又はテルペンをパッチクランプしたHEK293-V1細胞の細胞外液にデータ点60で添加することによって、TRPV1チャネルを活性化した。カンナビノイド又はテルペン溶液を、TRPV1活性化の陽性対照として1μMカプサイシンを含有する緩衝剤によってデータ点121で置き換えた。内向き及び外向き電流発達を時間と共に示す。各データ点(DP)は約2秒に相当する。
カンナビジオール(CBD)は、カプサイシンの適用によって誘導される7〜8nAの持続電流と比べて、4〜6nAの持続内向き電流を誘導した(図3A)。CBD及びカプサイシンは両方とも、内向き電流より小さい振幅で外向き電流も誘導した。
カンナビノール(CBN)は、カプサイシンの適用によって誘導される7〜8nA持続電流と比べて、0.5〜2nAの内向き電流を誘導した(図4A)。CBNは、内向き電流より小さい振幅で外向き電流も誘導した。CBN誘導電流は、カプサイシン及びCBDによって誘導されるTRPV1持続電流とは対照的に急速に不活性化していた。
カンナビジバリン(CBDV)は、カプサイシンの適用によって誘導される7〜8nAの持続電流と比べて、2〜3nAの持続内向き電流を誘導した(図6A)。CBDVは、内向き電流より小さい振幅で外向き電流も誘導した。CBD及びカプサイシンのように、CBDV誘導電流は持続電流であり、CBNによって誘導される電流より大きかった。
カンナビゲロール(CBG)は、カプサイシンの適用によって誘導される7〜8nAの持続電流と比べて、4〜5nAの持続内向き電流を誘導した(図7A)。CBGは、内向き電流より小さい振幅で外向き電流も誘導した。CBD、CBDV、及びカプサイシンのように、CBG誘導電流は持続電流であり、CBNによって誘導される電流より大きかった。
最初に、様々な濃度のミルセンをTRPV1活性化について評価した。図8A〜8Cに示すように、ミルセンはTRPV1活性化において用量依存的応答を誘導した。この図において、各データ点(DP)は約1秒に相当する。5μM(図8A)、10μM(図8B)、及び150μM(図8C)ミルセンは、1μMカプサイシンの適用によって誘導される4〜10nAの電流(図示せず)と比べて、0.5〜2.2nAの電流を誘導した。ミルセンの用量を増加することによって、内向き整流性非選択的カチオン電流が生じ、活性化電流振幅(図8A〜8C)及びカルシウムフラックス(図12A〜12I)に依存した形式で不活性化した。50μMミルセン、次に1μMカプサイシンを用いたTRPV1活性化の誘導を図10A〜Dに示す。150μMミルセンを使用した同様の実験を図11A〜11Cに示す。
カプサイシンは、TRPV1活性化及び疼痛脱感作の治療標準である。しかし、局所疼痛処置としてカプサイシンの治療適用によって、脱感作より前に高レベルの初期不快感が生じることは公知である。これらの研究において、カプサイシンへの曝露により、TRPV1チャネルが数秒以内に状態2の拡張チャネルに活性化される。一方、TRPV1のカンナビノイド及びテルペンであるミルセンへの曝露により、チャネルがまず状態1に活性化される。興味深いことに、CBD、CBN、CBDV、CBG、及びミルセン誘導TRPV1活性化及び不活性化プロファイルには反応速度の差異がある。これらの差は、所定のカンナビノイド若しくはテルペン、又はカンナビノイド若しくはテルペンの組合せをTRPV1発現細胞に適用することによって、特定のTRPV1活性化又は不活性化プロファイルを選択的に誘導し得ることを示唆する。さらに、カンナビノイド及びテルペンのみ、非選択的状態1のTRPV1活性化を誘導したが、依然としてカプサイシンの電流振幅応答と同様の電流振幅応答を誘導した。したがって、これらの化合物は、TRPV1を拡張状態2の形式で活性化する現行のカプサイシン処置レジメンとは対照的に、TRPV1を非拡張状態1の形式で活性化する治療代替物をより多く提供し得る。
内部Ca2+濃度のTRPV1活性化に対する役割
TRPV1の活性化及び不活性化動態に対する内部カルシウムの役割を調査するために、本発明者らは、次にHEK239-V1細胞の内部Ca2+を変更し、ミルセン(図12A〜12I)及びカンナビノール(CBN、図13A〜13F)の適用後のTRPV1活性化を測定した。0nM、180nM、又は620nM Ca2+及び10mM BAPTAを補充した上記の細胞内パッチピペット溶液で、細胞を灌流した。
10μMミルセン(図12A〜12I)を、データ点60で細胞に添加し、データ点120で1μMカプサイシンを含有する緩衝剤で置き換えた。サイトゾルCa2+の濃度を増加すると、10μMミルセンの外部適用及びその後のカプサイシンの適用によって活性化されるTRPV1V1電流ピーク振幅が低減された。さらに、内部カルシウムレベルを高めると、ミルセンによる下降相又は不活性化反応速度に対する効果の方が小さく、カプサイシンによって誘導される不活性化に対する効果の方が強い。
50μM CBN(図13A〜13F)を、データ点60で細胞に添加し、データ点120で50μM CBN及び1μMカプサイシンを含有する緩衝剤を補充し、データ点150で1μMカプサイシンを含有する緩衝剤で置き換えた。CBNは、ピーク電流振幅を低減したが、不活性化又は下降相反応速度に及ぼす効果が強くなかった。CBN及びカプサイシンの混合適用と比べて、0nM Ca2+内部適用により、下降相反応速度又は不活性化が除去された。対照的に、620nMの高い内部Ca2+は、電流振幅の著しい低減及び不活性化反応速度の加速を示した。
テルペンに応答してのTRPV1を介したカルシウムフラックス
米国特許出願第15/986,316号に記載されている細胞培養系を使用して、様々なテルペンに対してのTRPV1を介したカルシウム応答を試験した。HEK293細胞系に、pcDNA6TR(Invitrogen社、CA)プラスミド(テトラサイクリン感受性TRExリプレッサータンパク質をコードする)を安定的にトランスフェクトし、DMEM+10%ウシ胎仔血清(55℃で1時間不活性化)+2mMグルタミン中で、加湿された5%CO2雰囲気中、37℃で維持した。10μg/mlブラストサイジン(Sigma社、St Louis, MO)中での連続培養によって、TRex 293細胞にかかる選択圧力を維持した。
ミルセンのTRPチャネルに及ぼす効果
次に、様々なTRPチャネルを活性化するミルセンの効果を検出した。先に記載したように、HEK293細胞に、TRPA1、TRPM8、及びTRPV2を安定的にトランスフェクトした。これらのTRPチャネルはTRPV1に加えて、感覚ニューロン束に関連している。
カンナビノイドによって惹起されるHEK-TRPV1を過剰発現する細胞におけるカルシウムフラックス
HEK-TRPV1を過剰発現する細胞において、様々なカンナビノイドに応答してのカルシウムフラックスを研究した。
カプサゼピンの存在下にカプサイシン(Capsaisin)及びCBDによって誘発されるTRPV1コンダクタンス
カプサイシン及びCBDに応答してのTRPV1コンダクタンスを、カプサゼピンの存在下に試験した。
CBD、CBN、CBDV、及びCBGによって惹起される多様なTRPV1活性化応答
カンナビノイド、すなわちCBD、CBN、CBDV、及びCBGの異なる濃度に応答してのTRPV1の活性化を研究した。
TRPV1のカンナビノイド活性化のEC50測定
CBD、CBDV、CBN、及びCBGについて、TRPV1の活性化のEC50を計算した。
TRPV1のカンナビノイド調節の外部及び内部カルシウム濃度の依存
最初に、図22A〜Cに示すように、内部カルシウムレベルを(休止サイトゾルレベルに近い)一定濃度180nM、濃度0nMに緩衝し、又は内部カルシウムレベルを非緩衝状態(Fca)にしておきながら、CBD応答の外部カルシウムレベルへの依存性を研究した。また、3つの内部緩衝条件のそれぞれの下に0、1又は3mMにおける外部カルシウムを用いて電流Imaxの時間経過も示す。いかなる一定の内部カルシウム条件下でも、外部カルシウム濃度は、TRPV1応答の活性化時間及び不活性化反応速度に影響した。例えば、図22Aは、外部カルシウム濃度が低下すると、カルシウムイオンの通電寄与のために活性化反応速度が遅くなり、最大電流が一旦得られると不活性化がはるかに遅くなることを示す。対照的に、外部カルシウム濃度を1mMから3mMに増加させると、活性化反応速度に及ぼす効果は示されなかったが、不活性化反応速度の加速が生じた。さらに、図22Bは、一定の内部カルシウム濃度180nM、及び各パネルについて一定の外部カルシウムの、Imaxに正規化したデータを示す。これらのデータは、ナトリウム/カルシウムイオンの混合物ではなくナトリウム(0 Ca外部、0 Ca内部)がTRPV1の非選択的カチオンチャネルを流れている場合の電流発達における差を強調している。
TRPV1のカンナビノイド調節及びカプサイシン応答の差
TRPV1は、2つの状態のチャネルである。カプサイシン活性化下では、TRPV1が整流性状態を急速に通過し、次に小さいカチオン、例えばナトリウムから大きいカチオン、例えばN-メチル-D-グルカミン(NMDG)までを含めて線形I/V関係及び高透過性レベルによって特徴づけられる非整流性細孔拡張状態が達成される。この細孔拡張は、チャネルのカプサイシンに対する持続的及び高透過特性をもたらし、これらは、拘束されないカルシウム及びナトリウムの流入によるニューロンの活性化及びニューロンの最終的脱感作を御する重要な特性である。したがって、カンナビジオールに応答してのTRPV1の2つの状態の挙動を試験した。最初に、カプサイシンを用いてTRPV1の2つの状態の性質を決定した。図29A〜C及び図30に示すように、低(30nM)及び中(100nM)用量のカプサイシンによって、整流性電流が生じたが、より高い濃度(500nM)では生じなかった。一方、図29A〜Cに示すように、電流の拡張及び非拡張状態は両方とも、カプサゼピンに感受性を示した。さらに、図31A〜C及び図32は、チャネルの整流性が減少し、それによって拡張状態を達成するTRPV1に導かれたときのN-メチル-D-グルカミン(NMDG)の透過の増加を示した。これは、線形化I/V関係が実は拡張又はNMDG浸透性状態のマーカーであることをさらに示す。
異なるTRPチャネルでのカンナビノイド応答
この実施例は、様々なカンナビノイドのTRPV2、TRPM8、及びTRPA1チャネルにおける異なる応答を示す。

- TRPV1チャネル透過性をモジュレートする方法であって、
TRPV1発現細胞を所定のTRPV1状態関連アゴニスト特性を有する少なくとも1つの化合物と外部接触させるステップを含む、
前記方法。 - 所定のTRPV1状態関連アゴニスト特性を有する少なくとも1つの化合物を選択する先行ステップをさらに含む、請求項1に記載の方法。
- 前記少なくとも1つの化合物が、複数の化合物であり、複数の化合物のそれぞれが、異なるTRPV1状態関連アゴニスト特性を有する、請求項1又は2に記載の方法。
