がんの抗体に基づく療法および診断は、がん患者を処置および診断するための重要な戦略になってきている。正常細胞と比較して、がん細胞によって選択的に発現される細胞表面抗原は、標的がん療法および診断ツールを開発する魅力的な手段を提供する。しかしながら、この分野における重要な課題は、がん細胞を選択的に標的とするために使用することができる抗原を同定することであった。ペプチド抗原は、がん細胞特異的抗体を開発するために一般的に使用されるが、そのような抗原の適用可能性は、ある特定の状況では、例えば、ペプチド抗原の発現が正常細胞とがん細胞において類似している場合、限定的であり得る。
タンパク質におけるがん特異的グリコシル化変化は、がん細胞と、正常細胞とを区別することができ、診断適用と治療適用の両方の開発において有用であり得る別の魅力的な抗原群である。しかしながら、がん細胞上で選択的に発現される炭水化物部分を特異的に標的とする抗体はわずかしか開発されていない。かくして、がん細胞と、正常細胞とのグリコシル化の差異を特異的に標的とする新しい抗体が当業界において依然として必要である。
別の態様では、抗原結合コンジュゲートが提供される。抗原結合コンジュゲートは、薬剤に連結された本明細書に記載の抗原結合試薬のいずれか1つを含んでもよい。
さらなる態様では、細胞が提供される。細胞は、本明細書に記載の抗原結合試薬のいずれかまたは抗原結合コンジュゲートのいずれかを含んでもよい。
別の態様では、医薬組成物が提供される。医薬組成物は、本明細書に開示される抗原結合試薬のいずれか、抗原結合コンジュゲートのいずれか、または細胞のいずれかと、薬学的担体、賦形剤、または希釈剤とを含んでもよい。
別の態様では、本発明は、対象におけるがん細胞をイメージングするための方法に関する。この方法は、対象に、本明細書に記載の抗原結合試薬のいずれか、抗原結合コンジュゲートのいずれか、または医薬組成物のいずれかを有効量で投与すること、およびイメージングモダリティを使用して対象の少なくとも一部の画像を生成することを含んでもよい。好ましくは、これらの方法の実施形態では、抗原結合試薬、抗原結合コンジュゲート、または医薬組成物に結合した細胞のイメージングは、細胞ががん細胞であることを示す。
本発明の一態様では、抗原結合試薬が提供される。本明細書で使用される場合、用語「抗原結合試薬」は、最も広い意味で使用され、抗体に基づくポリペプチド親和性薬剤を指す。例えば、抗原結合試薬は、限定されるものではないが、一本鎖抗体(例えば、一本鎖Fv(scFv)、ビオボディ、ジスルフィド結合Fv(sdFv)など)、モノクローナル抗体、またはFab、Fab’、F(ab’)2、Fv断片、ダイアボディ、線状抗体などの抗体断片、または抗体断片から形成される多重特異性抗体(例えば、二重特異性抗体)を含んでもよい。抗原結合試薬は、キメラ、ヒト化、または完全ヒトポリペプチド配列であってもよい。抗原結合試薬は、IgA、IgD、IgE、IgG、IgY、およびIgMを含む免疫グロブリンの公知の主要なクラス、免疫グロブリン分子の任意のクラス(例えば、IgG1、IgG2、IgG3、IgG4、IgA1およびIgA2)、または任意のサブクラス(例えば、IgG2aおよびIgG2b)のいずれか1つであってもよい。一部の実施形態では、抗原結合試薬は、scFv、Fab、またはIgGモノクローナル抗体であってもよい。
抗原結合試薬の抗原結合アミノ酸残基は、「相補性決定領域」または「CDR」領域として一般に公知である。これらのCDR領域は、特定の抗原決定構造に対する抗原結合試薬の基本的特異性を担う。CDRは、抗体の可変領域内のアミノ酸の非連続的ストレッチである。一部の抗体の可変重鎖および軽鎖はそれぞれ、それぞれの軽鎖(L)および重鎖(H)について他方とそれぞれ非連続的である3つのCDR領域(L1、L2、L3、H1、H2、H3)を有する。驚くべきことに、本発明者らは、実施例に開示されるC9ビオボディが、3つの重鎖CDR領域(H1、H2、H3)およびただ1つの軽鎖CDR領域(L3)を含有することを見出した。
必要に応じて、抗原結合試薬は、少なくとも10-6M、10-7M、10-8M、10-9M、10-10M、または10-11Mの親和性でヒトペリオスチン糖タンパク質に特異的に結合してもよい。抗原結合試薬の親和性を決定するための方法は、当業者には公知である。例えば、Antibodies: A Lab. Manual (Harlow et al., eds., Cold Spring Harbor Lab. Press, Cold Spring Harbor, N.Y., 1988)を参照されたい。
抗原結合試薬は、ヒトペリオスチン糖タンパク質のグリカンエピトープに特異的に結合することができる。グリカンエピトープは、がん細胞上に存在するペリオスチン糖タンパク質上に特異的に存在してもよい。一部の実施形態では、グリカンエピトープは、N結合グリカン構造を含む。
本発明の別の態様では、抗原結合コンジュゲートが提供される。抗原結合コンジュゲートは、薬剤に連結された本明細書に記載の抗原結合試薬のいずれか1つを含んでもよい。「薬剤」は、抗原結合試薬に対してさらなる機能を提供する任意の物質であってもよい。好適な薬剤としては、限定されるものではないが、検出可能なイメージング剤、治療剤、免疫タンパク質ドメイン、またはその組合せが挙げられる。
「検出可能なイメージング剤」は、イメージング技術を使用してシグナルまたはコントラストとして検出することができる任意の好適な化学物質または物質であってもよい。好適な検出可能なイメージング剤は、限定されるものではないが、フルオロフォア部分、酵素部分、光学部分、磁気部分、放射標識部分、X線部分、超音波イメージング部分、ナノ粒子ベースの部分、または列挙された部分の2つ以上の組合せであってもよい。
「光学部分」は、例えば、発光または音弾光学部分などの、光学イメージングを使用してコントラストまたはシグナルを産生するために使用することができる任意の薬剤を含んでもよい。
「磁気部分」は、例えば、磁気共鳴剤のためのキレート剤を含んでもよい。磁気共鳴剤のためのキレート剤は、Gd(III)、Dy(III)、Fe(III)、およびMn(II)などの、常磁性金属イオンと安定な複合体を形成するように選択することができる。
他の例示的な検出可能なイメージング剤は、放射標識部分を含んでもよい。例示的な放射活性標識は、限定されるものではないが、99Mo、99mTc、64Cu、67Ga、186Re、188Re、153Sm、177Lu、67Cu、123I、124I、125I、nC、X3N、15O、および18Fを含んでもよい。
「X線部分」は、例えば、ヨウ素化有機分子または重金属イオンのキレートなどのX線イメージングを使用してコントラストまたはシグナルを産生するために使用することができる任意の薬剤を含んでもよい。
超音波イメージング部分は、例えば、Levovist、Albunex、またはEchovistなどの超音波イメージングを使用してコントラストまたはシグナルを産生するために使用することができる任意の薬剤を含んでもよい。
例示的な細胞傷害化合物はまた、限定されるものではないが、186Re、188Re、153Sm、67Cu、105Rh、mAg、および192Irを含む治療的放射性医薬品を含んでもよい。
抗体−薬物コンジュゲートを包含する実施形態では、リンカーは、細胞内または細胞外条件下で切断可能であってよく、リンカーの切断は、適切な環境で抗原結合試薬から治療剤を放出する。例えば、リンカーは、限定されるものではないが、リソソームまたはエンドソームプロテアーゼを含む細胞外または細胞内プロテアーゼによって切断可能であってもよい。細胞内プロテアーゼによって切断可能な好適なリンカーは、Val−CitリンカーまたはPhe−Lysリンカーを含んでもよい。例えば、米国特許第6,214,345号を参照されたい。
リンカーは、細胞内環境(例えば、リソソームまたはエンドソームまたはカベオラ)中に存在する切断剤によって切断可能であってもよい。
一部の実施形態では、リンカーは、カテプシンBおよびDならびにプラスミンによって切断可能であってよく、これらは全て標的細胞内部での活性薬物の放出をもたらすジペプチド薬物誘導体を加水分解することが知られている。
リンカーは、pH感受性、例えば、ある特定のpH値での加水分解に感受性であってよい。典型的には、pH感受性リンカーは、酸性条件下で加水分解性である。例えば、リソソーム中で加水分解性である酸不安定リンカー(例えば、ヒドラゾン、セミカルバゾン、チオセミカルバゾン、cis−アコニックアミド、オルトエステル、アセタール、ケタール、チオエーテルなど)を使用することができる。例えば、米国特許第5,122,368号;第5,824,805号;第5,622,929号を参照されたい。そのようなリンカーは、血液中のように、中性のpH条件下では比較的安定であるが、リソソームの近似pHであるpH5.5より下では不安定である。
一部の実施形態では、リンカーは、自壊性である。例えば、WO2007059404、W006110476、W005112919、WO2010/062171、WO09/017394、WO07/089149、WO07/018431、WO04/043493およびWO02/083180を参照されたい。
本発明と共に使用することができる様々な例示的リンカーは、WO2004010957、米国特許出願公開第2006/0074008号、米国特許出願公開第20050238649号、および米国特許出願公開第2006/0024317号に記載されている。
本発明のさらなる態様では、細胞が提供される。細胞は、本明細書に記載の抗原結合試薬のいずれか1つまたは抗原結合コンジュゲートのいずれか1つを含んでもよい。細胞は、限定されるものではないが、ヒト細胞などの、哺乳動物細胞であってもよい。

本明細書に開示される組成物を使用する様々なin vitroおよびin vivoでの診断および/または治療方法が企図される。
さらなる態様では、本発明は、対象におけるがん細胞をイメージングするための方法に関する。この方法は、対象に、本明細書に記載の抗原結合試薬のいずれか、抗原結合コンジュゲートのいずれか、または医薬組成物のいずれかを有効量で投与すること、およびイメージングモダリティを使用して対象の少なくとも一部の画像を生成することを含んでもよい。好ましくは、これらの方法の実施形態では、抗原結合試薬、抗原結合コンジュゲート、または医薬組成物に結合した細胞のイメージングは、細胞ががん細胞であることを示す。
本明細書で使用される場合、「イメージングモダリティ」は、対象の画像を生成することができる任意の技術を含んでもよい。一部の実施形態では、イメージングモダリティを、超音波、ポジトロン放出断層撮影(PET)、光子放出コンピューター断層撮影(SPECT)、核磁気共鳴イメージング(NMRI)、光学イメージング(OI)およびコンピューター断層撮影(CT)からなる群から選択することができる。例えば、本発明の方法の一部の実施形態では、本発明者らは、本明細書に開示される一部の組成物を超音波技術と共に使用して、対象における骨盤内腫瘤をイメージングし、そのような腫瘤が良性であるか、またはがん性であるかどうかを決定できることを企図する。骨盤内腫瘤の除去後の対象の予後は、外科手術を実施する外科医の型と直接関連するため、そのような診断イメージング法は、骨盤内腫瘤の除去前に有用であろう。イメージング法が、骨盤内腫瘤ががん性であることを示す場合、対象はがん性組織を除去するのを専門とする外科医に方向付けられてもよい。他方で、イメージング法が、骨盤内腫瘤が良性であることを示す場合、対象は、腫瘤を除去することができ、がん性組織の除去には特定の経験を有しなくてもよい一般外科医に方向付けられてもよい。そのような方法の一部の実施形態では、対象に、1つまたは複数の超音波イメージング部分を含む本明細書に開示される抗原結合コンジュゲートを投与した後、例えば、経膣または他の超音波イメージング技術を使用して、対象の骨盤領域の超音波画像を生成することができる。超音波画像が、骨盤内腫瘤における、またはその周囲の超音波イメージング部分からの有意な検出可能シグナルを示す場合、これは、骨盤内腫瘤ががん性であることを示すであろう。
本明細書で使用される場合、「接触」は、限定されるものではないが、組成物の試料への単純な添加を含む、組成物を、当業者には明らかであろう試料に適用するために使用される様々な手順のうちのいずれかによって実行することができる。
試料中での細胞に対する抗原結合試薬または抗原結合コンジュゲートの結合を「検出する」のに好適な方法は、当業者には公知であり、限定されるものではないが、ELISA、免疫蛍光、FACS分析、ウェスタンブロット、磁気イムノアッセイ、および抗体に基づくマイクロアレイを含んでもよい。過去では、血液中の細胞の検出のための判断基準は、ELISAの使用であった;しかしながら、液体生検技術は、細胞分析のための魅力的な代替手法を提供する。
本発明の方法の一部の実施形態では、方法は、対象に有効量の抗がん治療剤を投与することをさらに含んでもよい。
対象のための(例えば、抗原結合試薬、抗原結合コンジュゲート、細胞、医薬組成物または抗がん治療剤)の最大用量は、望ましくない、または許容できない副作用を引き起こさない最も高い用量である。個々の処置レジメンに関する変数の数は大きく、かなりの用量範囲が予想される。投与の経路も、用量の要件に影響するであろう。組成物の用量は、例えば、処置しない場合と比較して、少なくとも10%、20%、30%、40%、50%、60%、70%、80%、90%、100%以上、腫瘍サイズを減少させるか、または腫瘍の増殖速度を低下させることによって、がんを処置するであろうと予測される。

腫瘍特異的N結合グリカンを標的とする新規scFv抗体の選択および特性評価
材料および方法
細胞株
レトロウイルスベクター中にクローニングされたペリオスチンcDNAは、Dr.Xiao−Fan Wang(Duke University、Durham、NC)からの贈り物であった。ウイルスを、293−GP2パッケージング細胞およびレシピエント細胞(Lec4、Pro5、OVCAR3)への形質導入前のVSV−Gエンベロープを使用して産生させ、枯渇および濃縮のために使用されるペリオスチン(PN)発現細胞株を作出した。CHO細胞株Lec4およびPro5は、Dr.Pamela Stanley(Albert Einstein College of Medicine、Bronx、NY)からの贈り物であった。OVCAR3およびOVCA26対照ならびにGnT−III shRNA細胞株は、以前に記載されている3、29。ヒトメソテリンA1847、C30、およびヒトメソテリンLuc−ID8細胞株は、Dr.Scholler(SRI International、Menlo Park、CA)により生成された。
細胞培養上清(50mL)を、プロテアーゼ阻害剤を添加して、OVCAR3−PN、Pro5−PN、およびLec4−PN細胞から収集した。ペリオスチンを、製造業者の説明書に従って抗Flag樹脂(Sigma−Aldrich)上で精製した。PVDF膜に転移させる前に1X MES緩衝剤を使用して、NuPage 4〜12%ビストリスゲル上でタンパク質を分離した。ブロットを3%BSA/1X TBST中でブロックした後、ビオチン標識されたE−PHA(Vector Labs)の(1:5000)希釈液およびストレプトアビジンHRP(Vector Labs)の(1:10,000)希釈液を使用して二分岐グリカンを検出し、次いで、増強化学発光により検出した。ブロットをPierce(Thermo)ストリッピング緩衝剤中で剥がし、5%無脂肪ミルク1XTBST中でブロックし、ペリオスチンに対する抗体の(1:250)希釈液(Santa Cruz Biotechnologies)を使用して検出した。
11人の卵巣がん患者に由来する浸潤性B細胞およびPBMCから単離されたscFvの酵母ディスプレイライブラリーが、以前に記載されている30。このライブラリーを、SD−CAA(0.67%酵母窒素塩基、および0.5%カザミノ酸)中で増殖させ、scFvの細胞表面ディスプレイの誘導を、以前に記載されたように誘導した31。複数回のライブラリー枯渇を、以下のように実施した:リン酸緩衝食塩水(PBS)中の1x108の誘導酵母ディスプレイscFvを、PBSでリンスした付着性Lec4−PN細胞(T175フラスコ中で95%集密)に添加した。30分のインキュベーション後に非付着酵母を、Lec4−PN細胞の別のT175フラスコに採取し、このプロセスを合計6個のフラスコについて繰り返した。このプロセスを、Pro5−PNフラスコを使用して繰り返した。次に、この新しい枯渇したサブライブラリーを増殖させ、再度誘導し、これを使用して、OVCAR3−PN細胞を使用してペリオスチン上の腫瘍特異的グリコシル化に結合するscFvについて濃縮した。6回の濃縮と共に、細胞選択用プローブを使用する結合した酵母の手動による選択の後、以下のように酵母−細胞ELISAを使用して、濃縮のレベルをモニタリングした:scFvが濃縮されたプール中の酵母を、SD−CAAプレート上に塗布し、2〜3日間増殖させて、コロニーを生じさせた。個々のコロニーを、別々のSD−CAAプレート上に画線を作り、SGR−CAAで誘導して、酵母細胞表面上にscFvを発現させた。酵母scFvを、H2O/NaOH中、1mg/mLの蛍光増白剤28(Sigma−Aldrich、カルコフロール)を使用して標識した。簡単に述べると、細胞表面上にscFvを有する酵母を、室温で5分間、カルコフロール溶液(最終10%)中に1×107個で再懸濁した後、PBS中で洗浄した。標識された酵母を、室温で30分間、24ウェルプレート上、90%の集密度でLec4−PN/Pro5−PN/OVCAR3−PN細胞上でパンニングした。細胞への示差的酵母結合を、穏やかに撹拌しながらそれぞれ5分間洗浄する前後に(Ex355/Em405)でEnvision 2104マルチラベルリーダーを用いて測定した。洗浄緩衝剤の除去および新鮮なPBSの添加後、洗浄後の読取りを行った。
scFv DNAを、溶解した酵母からPCR増幅させた。簡単に述べると、飽和で増殖させた酵母5μLを、20μLの20mM NaOH中に懸濁し、3分間マイクロ波を照射して、酵母を溶解させた。scFv断片に対応するDNAを、Phire DNAポリメラーゼを使用するPCRによって増幅し、ゲル精製した後、エレクトロポレーションによってVYH10酵母株に線状化p416BCCPベクターを同時形質転換した17。酵母を、トリプトファン(TRP)を添加したSD CAA培地中で一晩増殖させ、以前の報告の通り6、1mLのSGR CAA/TRP中でさらに誘導した。検出のためのHISおよびV5タグを使用するELISAアッセイを使用して、可溶性scFvを確認した。可溶性scFvクローンを、以前の報告の通り32、部位特異的ビオチン化可溶性抗体(ビオボディ)に変換した。
OVCAR5細胞(条件あたり3個のウェル)を、0.8x104細胞/ウェルで播種した48時間後、scFvC9抗体のみ、抗myc抗体のみ、または抗myc抗体と混合したscFvC9の連続希釈液を添加した。0.5mg/mLのscFvC9および1mg/mLの抗myc抗体との複合体を、4℃で30分間形成させた後、細胞に添加した。複合体および対照scFvC9のみ(0.5mg/mL)または抗myc抗体のみ(1mg/mL)の連続希釈液を、48時間にわたって細胞に添加した。等量のCellTiter−Glo試薬(Promega)を、各ウェルに添加した。プレートを、オービタルシェーカー上で2分間振とうし、室温に10分間置いた後、発光を記録した。得られる細胞溶解は、生細胞数に存在するATPに比例する発光シグナルを生成する。3回の独立実験を行った。
卵巣がん細胞を、ポリL−リシンカバースリップ上に塗布し、50%集密まで増殖させた後、免疫蛍光染色を行った。PBS中のscFvC9ビオボディ抗体(50μg/mL)を、5分または30分の時点で細胞に添加した。細胞をPBSで洗浄した後、氷冷メタノール中で5分間固定した。細胞を、PBS/1%BSAで10分間ブロックした後、ストレプトアビジンコンジュゲート化Alexa Fluor 594を使用してscFvC9ビオボディを検出した。核を、DAPIの1:10,000溶液で10秒間対抗染色した後、Vectashield培地中にマウントした。
免疫不全のNSG雌マウスに、1.0×106個のA1847ヒト卵巣がん細胞を皮下注射し、6週間後にイメージング試験を行った。免疫能力があるC57Bl/6雌マウスに、1.0×106個のルシフェラーゼ形質導入ID8マウス卵巣がん細胞を卵巣内または腹腔内注射し、8週間後にイメージングを行った33。Luc−ID8腫瘍を、ルシフェリン注射を用いてモニタリングした後、イメージング試験を行った。マウスを、イソフルランで麻酔し、ベースラインのために抗体注射の前にイメージングし、次いで、抗体複合体の注射後、2分、5分、30分、60分、4時間、24時間、および48時間の時点でイメージングした。scFvC9複合体は、複合体を形成するために4℃で30分間、1:1の蛍光標識されたストレプトアビジンIRB680Wと共に予備インキュベートした30μgのscFvC9ビオボディを含んでいた。複合体のIV注射を、全てのマウスについて眼窩後方で実施した。
組織中でのscFvC9の免疫蛍光局在化
皮下A1847腫瘍を担持するNSGマウスに、30μgのscFvC9ビオボディを注射し、24時間後に犠牲にして、腫瘍、腎臓、脾臓、肺、および肝臓を収獲した。全ての組織を、ホルマリン中ですぐに固定し、組織切片にするまで70%エタノール中で保存した。キシレンおよび段階的エタノール系列に逐次的に浸すことにより、スライドを脱パラフィン化した。暗室中、室温で1時間にわたって、PBS中のストレプトアビジン−Qdot800(1:50希釈)と共に組織をインキュベートした。スライドを、PBS/0.05%tween20中で3回洗浄し、1:10,000のDAPIで15分間、対抗染色を行った。スライドを、PBS中で2回洗浄し、FluorSave試薬を使用して、スライドにマウントした。
in vitro分析−MRIイメージングを、1.5T MRシステム(Bruker PharmaScan 70/16)上で実施した。抗flag磁気ビーズに結合した25μg/mLまたは50μg/mLのscFvC9と共にインキュベートする前または後に、アガロースゲルのスペーサー間に積み重ねたA1847、C30、またはID8細胞(0.4〜1x106細胞)を用いて、Phantomチューブを生成した。scFvC9/磁気ビーズ複合体を、4℃で30分間、細胞と共にインキュベートした後、洗浄し、4℃で20分間、2%パラホルムアルデヒドで固定した。次いで、固定された細胞およびscFvC9/磁気複合体を、1%アガロースゲル中の100μLに再懸濁し、最後に、2%超低温アガロースゲルのスペーサー間に積み重ねて、ファントムチューブを生成した。最適なT1およびT2加重配列を決定し、それぞれの細胞層に関する目的の領域を、C9/磁気ビーズと共にインキュベートした細胞との比較のために、対照細胞のみおよび対照磁気ビーズのみについて測定した。3つの別々の実験の結果を算出し、正規化されたシグナル強度について±SEMを算出した。
全ての動物試験および手順を、SRI International Institutional Animal Care and Use Committeeによって認可されたプロトコールの下で行った。全ての方法を、SRI Internationalの指針および規則に従って実施した。SRI Internationalは、Association for Assessment and Accreditation of Laboratory Animal Care International(国際実験動物管理公認協会)(AAALAC)によって公認された、米国農務省(USDA)に登録された一元管理された動物管理および使用プログラムを維持し、Office of Laboratory Animal Welfare(実験動物福祉局)(OLAW)のファイル上に保証を有する。
腫瘍特異的グリカンを有するヒトscFv結合の選択
ペリオスチンは、タンパク質のC末端近くの最後のFAS1ドメイン中に位置する1つの高度に保存されたN結合グリコシル化部位を有する(図1A)。ペリオスチン上に存在するグリコシル化の機能は不明であるが、この部位は、配列が高度に保存されており、その潜在的な重要性を暗示し、この部位は、ペリオスチンの全ての既知のアイソフォーム中に存在する。体系化されていない、溶媒露出した領域中のN−グリコシル化部位の保存および位置34、35(図1B)により、本発明者らは、ペリオスチンの糖型に対する特異的scFv抗体のサブトラクションおよび濃縮を可能にする異なる糖型のペリオスチンを提示するための足場としてペリオスチンタンパク質を使用することができるとの仮説に導かれた。上の画像は、ヒトペリオスチンの最後のFAS1ドメインのNMR構造を示し(図1B)、アスパラギン599(グリコシル化されたアミノ酸)が非構造ループ中に位置することを示す。全てのFAS1ドメインの結晶構造内でのこの領域の位置は、N−グリコシル化部位の露出をさらに検証する下の画像に示される。3つの主要な形態のN−グリカンが存在する:高マンノース型、ハイブリッド型、または複合体型。典型的な糖タンパク質は、これらの3つの形態のいずれかからなってもよいいくつかのN−グリコシル化部位を有する。ある特定の部位が高マンノースである傾向を有し、他の部位がハイブリッドまたは複合体である理由はまだよく理解されていない。しかしながら、以前の研究は、糖タンパク質内のこれらのグリカン型に対する部位特異性が存在することを示している36。本発明者らは、ペリオスチン中の単一のN−グリコシル化部位が、異なるがんにおけるグリコシル化パターンの変化のため複雑なN−グリカンを示すことを決定した。乳がん組織の本発明者らの以前のグリコプロテオミック分析は、ペリオスチンが4分枝(antennary)シアル化された複合N結合グリカンを示すことを示している37。卵巣がん組織では、本発明者らの以前の研究は、ペリオスチンがコアフコースを有する、または有しない、トランケートされた、無ガラクトシル化された、無シアル化されたN−グリカン構造を示すことを示している3、24。ヒトがん組織中での高い発現にも拘わらず、付着増殖条件下で増殖させたヒトがん細胞株は、ペリオスチンを発現しない。非付着条件下で増殖させた細胞株は、ペリオスチンを発現し始めるスフェロイドの形成を可能にする。本発明者らは、卵巣がん細胞株OVCAR3ならびに非悪性チャイニーズハムスター卵巣(CHO)細胞株Pro5(親)およびLec4(GnT−V発現を欠く)中で安定なペリオスチン発現を作出して、付着増殖条件下での発現を可能にした。図1Cに示されるように、Flagタグ抗体を使用して単離されたペリオスチンは、OVCAR3−PN細胞株についてのみ、二分岐N−グリカンを認識することが知られるレクチン38であるレクチンE−PHAに結合したが、これは二分岐グリカンの存在を示している。E−PHAによって結合した反応するさらにより高分子量のバンドが存在するが、これは、他の糖タンパク質が、卵巣がん細胞中でこの形態のグリコシル化も担持するペリオスチンと共に単離されたことを示している。Pro5−PNおよびLec4−PN flagタグプルダウンは、E−PHA結合について陰性であり、これらの細胞株中に二分岐グリコシル化が存在しないことを反映している(図1C)。Pro5およびLec4細胞株から単離された糖タンパク質上に見出されるN−グリコシル化糖型の以前に公開された質量分析は、4分枝および3分枝複合型N−グリカンがこれらの細胞株において顕著であることを示唆している39。全ての細胞株が、同様のレベルのペリオスチンタンパク質を発現する(図1C)。これらの結果は、ペリオスチンが、異なる形態の複合型N−グリカンと共にこれらの細胞株中で発現されることを確認するものであり、本発明者らは、scFv抗体のサブトラクションおよび選択のためにこれらのものを使用することができる。
クローン#9は、最大収率の可溶性ビオチン標識scFv抗体産生を有し、これを、OVCAR3細胞を使用して二分岐N−グリカンに対する結合特異性についてさらに分析した。本発明者らは、いかなる遺伝子も標的としない対照ShRNAまたはGnT−III(Mgat3遺伝子)3、29を標的とするShRNAの安定な発現を有するOVCAR3細胞中で安定なペリオスチン発現を確立した。図4Aに示されるフローサイトメトリーデータは、scFvC9が、GnT−III ShRNA OVCAR3−PN細胞と比較して、対照OVCAR3−PN細胞への結合が増加していることを示し、二分岐N−グリカンに対する結合特異性を示している。OVCAR3−PN対照ShRNAと、OVCAR3−PN GnT−III ShRNAは両方とも、ペリオスチンタンパク質を発現するが、これは、結合が二分岐N−グリカンに特異的であって、タンパク質に特異的ではないことを示している。次に、scFvC9の潜在的ターゲティングおよび内在化を評価するために、本発明者らは、顕微鏡観察を使用して、以前に記載された患者由来細胞株OVCA2629を使用して、卵巣がん細胞中でのscFvC9の結合および分布を追跡した。5分でのOVCA26対照ShRNA細胞の細胞染色は、細胞表面でのscFvC9の蓄積を示している(図4B)。抗体は、30分の時点で完全に内在化される。本発明者らは、scFvC9のOVCA26 GnT−III ShRNA細胞への結合を観察しなかったが、これは二分岐N−グリカンに対する特異性をさらに検証するものである。本発明者らは、グリア芽腫細胞もGnT−IIIの発現レベルが上昇するため、この腫瘍型に対するscFvC9の結合をさらに評価した。図4Dおよび図4Eに示される本発明者らの30分の結合データは、scFvC9が、二分岐グリカンを示す対照LN18細胞に結合し、Mgat3のLN18 Crispr/Cas9 KOへの結合がないことを示す(図4E)。これらのデータは、scFvC9が、結合のために二分岐グリカンを必要とすること、および他のがんに由来する二分岐構造を標的とすることができることを確認するものである。細胞表面でのscFvC9の蓄積は、scFvC9が抗体依存性細胞傷害を開始するように機能することができることを示唆する。scFvC9ビオボディは、mycタグを含有し(図2)、本発明者らは、細胞をscFvC9/抗myc ab複合体に曝露して、細胞傷害性を評価することができる。卵巣がん細胞株OVCAR5を、48時間にわたって抗体複合体の連続希釈液と予め混合した後、発光生存能力アッセイを使用して、細胞生存能力を測定した。結果は、scFvC9のみ、または抗myc abのみでは細胞傷害を誘導しなかったことを示している(図4C)。しかしながら、複合体の最初の2つの連続希釈液(2.5μg/mLおよび1.25μg/mL)への細胞の曝露は、細胞傷害活性を有していた。
本発明者らは、in vivoでのイメージング(IVIS)を使用して、ヒト異種移植モデルおよび同系マウスモデルの両方を使用してin vivoで腫瘍を標的とするscFvC9の能力を評価した。図5の上パネルは、6週間前に確立されたヒトA1847卵巣がん皮下異種移植腫瘍を有するNOD/Scidマウスにおける、蛍光標識されたストレプトアビジンと1:1で複合体化したscFvC9の局在化および蓄積を示す。scFvC9抗体は、腫瘍を標的とし、24時間のピークで腫瘍中に蓄積し、徐々の減少が48時間で始まる。次に、本発明者らは、免疫能力があるC57Bl/6雌マウスにおいてルシフェラーゼを形質導入したID8マウス卵巣がん細胞(Luc−ID8)を標的とするscFvC9の能力を評価した。IVISイメージングの8週前に、細胞を卵巣内または腹腔内に注射した。蛍光標識されたストレプトアビジンと1:1で複合体化したscFvC9抗体を、イメージングの前の示された時間で、眼窩後方に注射した。ヒト皮下異種移植注射について観察されたように(図5、上パネル)、卵巣内注射(図5の中央パネル)は、24時間の時点で最大に蓄積した。しかしながら、48時間での減少は、より実質的であった。同系腹腔内モデルは、4時間の時点でscFvC9/蛍光ストレプトアビジン複合体の最大蓄積に達した。これらの結果は、scFvC9が眼窩後方注射によりin vivoでヒトとマウスの両方の卵巣腫瘍を標的とすることができることを確認するものである。
scFvC9による腫瘍に対するターゲティングが成功したため、本発明者らは、scFvC9ビオボディが磁気共鳴イメージング試験によるシグナルの増幅のために腫瘍に対して磁気ビーズをターゲティングすることができるかどうかを試験した。標的MRイメージングプローブとしてのscFvC9の開発の成功には、特異性、蓄積の規模、および安定性が必要である。本発明者らは、ファントムチューブを使用するMRIによりin vitroで卵巣がん細胞中のscFvC9磁気ビーズ複合体を検出する能力を測定することによって、MRイメージングプローブとしてのscFvC9の評価を開始した。A1847、ID8、およびC30細胞を、アガロースに埋め込み、積み重ねた。細胞のみ(洗浄および固定した)、抗flagタグ磁気ビーズのみ、または細胞(scFvC9/抗flagタグ磁気ビーズと共にインキュベートした後、洗浄および固定した)の層を、MRIを使用して測定した。その結果、細胞のみに対してscFvC9/磁気ビーズと共に細胞を含有する層において、正規化されたシグナル強度の有意な減少が検出可能であることが示された(図7A)。これらの結果は、シグナル増幅の蓄積を示している。
本発明者らの結果は、顕著な腫瘍特異的グリコシル化変化を標的とする完全ヒトscFv抗体であるscFvC9の単離および精製をもたらした有効なスクリーニングプラットフォームの開発成功を示している。本発明者らは、卵巣がんに対するこの抗体の結合特異性およびターゲティングを特徴付けたところ、LN18グリア芽腫細胞株を使用した本発明者らの初期顕微鏡観察データは、scFvC9がMgat3遺伝子の増幅を示す他の腫瘍に結合するはずであることを示している23。本発明者らは、ビオボディ中のscFvC9クローンを開発し、大規模精製を可能にし、in vitroおよびin vivoでの腫瘍グリカンに対するscFvC9ビオボディの特異性を証明した。in vivoでの安定性が増強された抗体の細胞表面結合および内在化は、多様なイメージングおよび治療適用の将来の開発を可能にするはずである品質である。scFvC9ビオボディを、免疫コンジュゲート、毒素、または薬物コンジュゲートなどの多様な治療分子にコンジュゲートすることができる。これらの潜在的な治療的革新に加えて、ビオボディは、腫瘍イメージングおよびイメージングと治療選択肢との潜在的な対形成にとって有用であり得る。
本発明者らは、ビオボディ#C9の2つのクローン(E11およびF11)を配列決定した。E11およびF11クローンに由来するBb#C9をコードするDNA断片を、PCR増幅し、配列決定した。クローンは、予想通りタンパク質およびDNAレベルで同一であった。
in vivoでのイメージング試験
方法
モデル系
標識された抗メソテリンナノボディ(MN)または抗N−グリカンscFv(C9)抗体を使用して、いくつかの肺がんモデルをイメージングした。モデルは、
・腹腔内マウスメソテリンintLuc−ID8マウス卵巣がんを有するWT C57Bl/6マウス(図8〜9)
・同所性ヒトメソテリンloA549ヒト肺がんを有するNSGマウス(図10)
・同所性ヒトメソテリンIntH460ヒト肺がんを有するNSGマウス(図11)
・同所性ヒトメソテリンhiEKVXヒト肺がんを有するNSGマウス(図12)
を含んでいた。
1)標識されたストレプトアビジンIRB680Wに結合したビオチン化抗メソテリンナノボディ(MN、30μg)、2)標識されたストレプトアビジンIRB680Wに結合したビオチン化抗N−グリカンscFv(C9、30μg)、および3)標識されたストレプトアビジンIRB680Wのみ(陰性対照)の眼後方注射を、上記モデルにおいて行った。

抗メソテリンナノボディは、卵巣内、IV、IPまたはSCで注射された、卵巣および肺がんによって発現されるヒトとマウスの両方のメソテリンを検出した。感度は高かった。ルシフェリン(図8)によってほとんど可視化されない、または全く視認できない(図10)腫瘍が、抗メソテリンナノボディによって検出されたが、これは、がんの早期検出に適していることを支持する。特異度は高かった。メソテリンを発現しない大きい肺腫瘍は検出されなかった(図12)。
抗N−グリカンペリオスチンscFvは、卵巣内、IP、またはSCで注射された、ヒトとマウスの両方の卵巣がんを検出した。C9生体分布は、メソナノ生体分布よりも速く、試験した全部で3つの卵巣がんモデルにおいて注射後にシグナルのより早い増加および減少を示した。
