Abstract
Description
驚くべきことに、公知の抗TeNT SDAのKDよりも有意に低いKDを有し、及びin vivoで破傷風毒素中和活性を有する抗TeNT SDAが取得可能であることが今や判明した。そのような新規の抗TeNT SDAは、極めて高い親和性を伴い、TeNTに結合する能力を有するという長所を有する。これは、フリーのTeNT分子と結合したTeNT分子との間のバランスを、極端に結合したTeNT分子の側に変化させ、ひいては破傷風菌に感染した後でも、破傷風の致命的な症状を十分に抑制する。
1)重鎖のみから構成され、且つ軽鎖を本来欠損しているラクダ類及びサメから取得可能な抗体;
2)VHHドメイン又はVNAR断片と通常呼ばれ、本明細書では、まとめて単一ドメイン抗体(SDA)と呼ばれる、1)で定義する抗体の可変ドメイン;
3)ラクダ類(又はサメ)VHHドメインのフレームワーク配列が、別の供給源から得られるCDRとグラフト結合している、1)で定義する抗体、又は2)のドメインの工学的に操作された形態、例えば「ラクダ類化(camelidised)」、又は「ラクダ化(camelised)」抗体等;
4)例えば、国際公開第04/108749号に記載されているように、様々な免疫グロブリン様分子に由来するフレームワーク配列が、所与の標的分子に対して特異的なCDRと結びついている、免疫グロブリン様の可変ドメインの工学的に操作された形態
が特に挙げられるが、但しこれらに限定されない。
(i)配列番号24;
(ii)配列番号25;
(iii)配列番号20;
(iv)配列番号17、又は配列番号19;及び
(v)配列番号22、配列番号15、配列番号23、又は配列番号14;
からなる群から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、
但しCDR1、CDR2、及びCDR3領域のアミノ酸配列同一性が少なくとも75%である。
(i)配列番号15、及び配列番号17;
(ii)配列番号24、及び配列番号17;
(iii)配列番号20、及び配列番号17;
(iv)配列番号15、及び配列番号24;又は
(v)配列番号24、及び配列番号20;
と少なくとも70%の全体的なアミノ酸配列同一性を有し、但しCDR1、CDR2、及びCDR3領域のアミノ酸配列同一性が少なくとも75%である。
i)好ましくは配列番号40、37、38、39、41、及び42からなる群から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、但しCDR1、CDR2、及びCDR3のアミノ酸配列同一性が少なくとも75%である、血清アルブミンに結合する能力を有するSDAを提供するステップと、
ii)ステップi)のSDAを、テストサンプルと接触させるステップと、
iii)ステップi)のSDAと、ステップii)のサンプル中に存在するアルブミンとの間の結合の可能性を検出するステップと
を含む方法を提供する。
ラマ(Lama glama)(no.9236、及び9237)を同一の方式で免疫化した。市販の破傷風ワクチン(既製タイプ、破傷風トキソイドに基づく)を、一次免疫化後の0、21、及び42日目(DPI)において、ラマ1匹当たり1mlで、右側大腿部内に、さらなるアジュバント添加を行わずに筋肉内注射した。0.5mgのchrompureウマIgG(Jackson Immunoresearch Laboratories社、West Grove、PA)、0.5mgのchrompureイヌIgG(Jackson Immunoresearch Laboratories社)、0.5mgのウマAlb(Rockland Immunochemicals社、Limerick、PA)、0.5mgのイヌAlb(Molecular Innovations社、Novi、MI)、及び8μgの組換え破傷風毒素断片C(rTTC;Reagent Proteins社、San Diego、CA)からなる標的抗原混合物を調製した。In vitro 実験では[5]、断片C(Hc又はTTCとも命名される)が、ニューロンに対する破傷風毒素の結合に関与していることが示唆された。抗原混合物を、Stimuneアジュバント(Thermofisher Scientific社、Lelystad、the Netherlands)を用いて乳化し、そして0、及び21DPIにおいて、各ラマの左側大腿部内に筋肉内注射した。IMS1312アジュバント(Seppic社,France)を用いてアジュバント化された同一の抗原混合物を、42DPIにおいて各ラマの左側大腿部内に筋肉内注射した。ヘパリン処理された血液サンプル(150ml)を28及び49DPIにおいて取得し、そして末梢血リンパ球(PBL)を速やかに単離した。血清調製用の血液サンプルを、0、28、及び49DPIにおいて取得した。
破傷風ホロ毒素に結合するが、しかしrTTCには結合しない単一ドメイン抗体(SDA)cAb−TT2[46]を、後続するTeNT結合SDAのファージディスプレイ選択で使用するために、パン酵母菌において産生させた。酵母菌インベルターゼシグナル配列及び5’−リーダーに対する融合体としてこのSDAをコードする合成遺伝子を、SacI−BstEII断片として生成した。その合成遺伝子をプラスミドpRL188内に挿入すると、単一のシステイン及びhis6−タグを含有するラマ長鎖ヒンジ領域とリンクしたSDAの生成を引き起こした[2]。そのような発現フォーマットは、SDAの固体表面への固定化に特に適する[3]。cAb−TT2 SDAを、パン酵母菌菌株SU51内、150mlスケールにおいて産生させ、固定化金属アフィニティークロマトグラフィー(IMAC)により精製し、そして単離されたSDAの一部を、以前記載された通りにビオチン化した[3]。
下記の試薬を、ファージディスプレイ選択で使用した。酵母菌内産生cAb−TT2は、これまでの実施例に記載されている。破傷風毒素中和mAb TT10[47、48]を、国立生物製品基準規制機構(NIBSC;Potters Bar、英国)から購入した。真正破傷風ホロ毒素(TeNT)を、List Biological Laboratories社(Campbell、CA)から購入し、rTTCは実施例1に記載されている。

3つのアイソタイプ(ヒンジプライマー)のファージライブラリーをプールした。
IgG結合SDAのファージディスプレイ選択は、これまでの実施例に記載したように、直接コーティングされたイヌ又はウマ由来のIgGを使用して実質的に実施した。イヌ及びウマIgGの両方に結合するSDAを選択するために、第2ラウンドのパニングでは、同一種起源のIgGについてだけでなく、その他の種のIgGについても実施した。
Alb結合SDAのファージディスプレイ選択は、実施例1に記載するイヌ又はウマ由来の直接コーティングされたAlbを使用して、これまでの実施例に記載するように実質的に実施した。イヌ及びウマAlbの両方に結合するSDAについて選択するために、第2ラウンドのパニングを、同一種起源のAlbだけでなく、その他の種のAlbにおいても実施した。
14個のSVT SDA(表2)、8個のSVG SDA(表3)、及び6個のSVA SDA(表4)が、分泌性酵母菌発現により生成した。この目的のために、SDAコード領域を、ファージディスプレイプラスミドからPCRにより増幅し(実施例3〜5)、そしてPstI及びBstEIIを用いて裁断し、同様に裁断されたpUR4585プラスミドとライゲーションした。pUR4585は、c−myc及びhis6タグにC末端側で融合したSDAの発現に適する酵母菌−大腸菌シャトルベクターである。この方式で生成されたSDAを接尾辞「L」により示す。そのようなSDAをコードするpUR4585に由来するプラスミドを、接尾辞「L」に付加してSDAの名称及び接頭辞「p」により表す。さらに、FR4領域内に酵母菌内産生レベルを増加させる突然変異を有する3つの変異体SVT SDAを生成した(実施例3及び[11]を参照):
SDA SVT15L−3FW4Mは、突然変異K120Q、I122T、及びL123Qを含有するSVT15Lの誘導体である
SDA SVT20L−L123Qは、突然変異L123Qを含有するSVT20Lの誘導体である
SDA SVT34L−L123Qは、突然変異L123Qを含有するSVT34Lの誘導体である
抗原をプレートにコーティングし、そしてSDA検出用にmycタグを利用しながら、非標識SDAを主に用いてそれを検出することにより、異なる種に由来するAlbに対するSVA SDAの結合を、ELISAで分析した。ELISAを、実施例3に記載されるように実質的に実施した。精製された血清アルブミンを5μg/mlの濃度でコーティングした。ウマ及びイヌAlbの供給源は、実施例1に記載されている。ウシAlb、及びニワトリオバルブミンを、Sigma Aldrich社から、ヒトアルブミンをJackson Immunoresearch Laboratories社から、及びマウス、ヒツジ、及びブタアルブミンをAntibodies Online社(Beijing、CN)から入手した。ネコアルブミンを、500倍希釈された正常なネコ血清(Agrisera Antibodies社、Vannas、Sweden)から、1000倍希釈された免疫親和性吸着ヤギ抗ネコAlb IgG(Antibodies Online社)でコーティングされたプレート上で捕捉した。次にプレートを、1μg/mlのSDA濃度で開始し12ウェルにわたるSDAの2倍希釈系列と共にインキュベートした。SDAを、0.5μg/mlの抗mycクローン9E10mAbペルオキシダーゼコンジュゲート(Roche Applied Science社)を使用して検出した。吸収データを、エクセル(Excel)(登録商標)スプレッドシートテンプレート(Microsoft Corporation社、Redmond、USA)を使用して評価し、最大A450値を計算した。4パラメーターロジスティック曲線を吸収及びSDA濃度に近似させて、有効濃度(EC)を内挿すると、各ELISAにおいて各SDAについて0.2の吸収値が得られた。
種の起源が異なるIgG、及びいくつかの種の抗体断片Fab、F(ab’)2、及びFcに対する8つの酵母菌内産生SVG SDAの結合を、これまでの実施例の記載と同様に分析した。いくつかのIgG及びその断片の供給源は実施例4に記載されている。マウス、ネコ、ヒツジ、ウシ、及びモルモットのγグロブリン(GG)並びにウマF(ab’)2断片を、Jackson Immunoresearch Laboratories社から得た。イヌFc断片を、Rockland Immunochemicals社から得た。結果を表7に記載する。マウス抗myc mAb PO−コンジュゲートとコーティングされた抗原との交差反応におそらく起因して、3つのELISAには比較的高いバックグラウンドが含まれた(表7の脚注b)。従って、これらのELISAにおける吸収値について、バックグラウンドが差し引かれた(表7の脚注b)。バックグラウンドが差し引かれた吸収値が0.2を上回るとすれば、それは抗原結合を示唆すると考えられた。
様々な種に由来するIgGに対する、以前単離されたブタIgG結合SDA(VI−クローン)4例[19]のELISAにおける結合を、ほとんどの種のIgGと反応するSDA SVG13Lと比較した。ELISAは、実施例8に記載されるように実施した。結果(表8)より、SDA VI−4L、VI−8L、VI−11L、及びVI−14Lは、ウシ、ネコ、イヌ、マウス、及びヒトのIgGに結合しないことが分かる。最大吸収が、SDAを含まないバックグラウンドを上回りわずかに増加しているので、おそらくは、それらはヒツジIgGには結合する。SDA VI−11Lはさらに、ウマIgGと結合する。しかしながら、すべての事例において、SVG13Lは、ブタIgGを除くすべての種に由来するIgGにおいて、かなり高めの吸収値をもたらす。ブタIgGでは、逆に、すべてのVIクローンは、SVG13Lよりも高い吸収値をもたらす。
SVA、及びSVG SDAについて記載された方式と類似した方式で、いくつかの酵母菌内産生SDA、及び6つの抗TeNTmAbを、ELISAにおいて抗原結合について分析した。抗TeNTmAbを異なる供給業者から取得した。それらの起源、及びマウスバイオアッセイにおけるTeNTの中和又はウェスタンブロットにおける抗原結合について、供給業者より提供された情報を表9に記載する。これらのmAbの一部は、実施例6に記載されるようにビオチン化された。
6つのmAbが分析に含まれた。TeNT軽鎖と結合するので、mAb 11n185を選択した。マウスバイオアッセイにおいてTeNTを中和するので、さらなる5つのmAbを選択した。mAbは異なるELISAにおいて、以下のように結合する:
1.mAbの14F5及び6F55は、SVT02L及びSVT03Lと類似して、捕捉されたTeNTと比較してそれよりも非効率的に直接コーティングされたTeNTと結合する。
2つのSDA(SVT02L及びSVT03L)は直接コーティングされたTeNTに結合せず、従ってTeNTの直接コーティングされた断片C(rTTC)(Hc又はTTCとも命名される)に対するその結合については、粒子の特異性に関して結論に達してない(実施例10)。受動吸着が抗原結合を無効にした可能性があるからである。mAb6E7を用いて捕捉されたrTTCに対するこれらのSDAの結合が、従って実施例10の記載と同様に分析された。96ウェルプレートの6個のウェルをTeNT(0.75μg/ml)で、及び18個のウェルをmAb6E7(1μg/ml)でコーティングした。6つのmAb6E7コーティングウェルを2μg/mlのrTTCと共に、6つのさらなるウェルを0.75μg/mlのTeNTと共にその後インキュベートした一方、mAb6E7がコーティングされた残りの6つのウェルをELISAバッファー(陰性コントロール)と共にインキュベートした。その後、6つのSDAを、2μg/mlのSDA濃度で、異なる抗原がコーティングされた4つのウェル内でインキュベートした。抗mycMAb PO−コンジュゲートと共にインキュベートすることにより、結合したSDAを検出した。
結果(表11)は、下記事項を示す:
陰性コントロールのSVA12L、及び捕捉された抗原を含まないmAb6E7コーティングは、陰性である(吸収<0.07)。
TeNTの軽鎖又は重鎖に対するSVTクローンの結合を、これまでに実施したように[19]、1ゲル当たり2.5μgの真正TeNTを使用し、並びに免疫ブロッティング用として0.5μg/mlのビオチン化されたSDA(実施例6)、及び0.1μg/mlのストレプトアビジンPOコンジュゲート(Jackson Immunoresearch Laboratories社)を使用してウェスタンブロッティングにより分析した(図4)。SDA SVA12Lを、陰性コントロールとして使用した。ほとんどのSDAは、ウェスタンブロットにおいて、軽鎖又は重鎖のいずれに対しても結合を示さなかった。これは、高次構造エピトープを認識するそのようなSDAに起因し得る。しかしながら、SDA SVT06、及びSVT08は、TeNT重鎖に該当するに間違いない約100kDaのポリペプチドと結合する。これらのクローンは同一のCDR3群(群B)に属し、そして同一の抗原部位を認識する(表12)。SDAのいずれも、重鎖に由来するrTTC(表2及び表10)にも結合する。従って、TeNT重鎖に対するSDA SVT06及びSVT08のウェスタンブロットにおける結合は、以前の観察知見と整合する。
SVT SDA、及び6つの抗TeNTmAbの抗原部位を、ビオチン化されたSDA及びmAbを使用するブロッキング/競合ELISAによりマッピングした。可能な場合、各CDR3群の2つの代表例をこの目的のために使用した。SDAは、TeNT−GT1b相互作用におけるそのIC50値に基づき、主に選択した。しかしながら、酵母菌内で産生されたSVT29Lは、0.07mg/Lにすぎず、従ってSVT15L−3FW4Mに置き換えた。競合/ブロッキングELISAを、これまでの実施例に記載された手順と類似したELISA手順を使用して、但し直接コーティング用として0.5μg/mlのTeNTを使用して実施した。ほぼ最大の吸収値をもたらすビオチン化SDA又はmAb濃度を使用し、そして5μg/mlの様々な非標識SDA又はmAbを用いることで、このSDA又はmAbと競合させ、それをブロッキングした。TeNTコーティングプレートを、非標識SDA又はmAbと共に、90μl/ウェル中、30分間、最初にインキュベートした(ブロッキングステップ)。次に50μg/mlのビオチン化されたSDA又はmAb、10μlを添加し、そしてさらに30分間インキュベートした(競合ステップ)。ビオチン化されたSVT02及びSVT04の場合、これらのSDAは捕捉されたTeNTにのみ結合し、直接コーティングされたTeNTには結合しないので、cAb−TT2を用いてTeNTを捕捉した。その他のすべてのケースでは、直接コーティングされた抗原を使用した。抗原コーティングを有さないコントロール、及びビオチン化されたSDAを有さないコントロールが含まれた。SDAの競合/ブロッキングに起因する抗原結合の阻害率(%)を、次に100−100×([競合するSDA又はmAbが存在するA450]−[Agコーティングを有さないA450])/([競合するSDA又はmAbが存在しないA450]−[Agコーティングを有さないA450])として計算した。すべてのSDA及びmAbは、そのビオチン化されたカウンターパートの結合を少なくとも75%ブロッキングし、アッセイは有効であることを示唆した。
SVTクローンの結果を表12にまとめる。一般的に、結果は、SDAのAg特異性及びTeNTの抗原構造に関する一般的見解と整合する:
同一のCDR3群のSDAは、同一の抗原部位に分類される。
同一の抗原部位のSDA又はmAbは、類似した抗原結合特異性を示す:
抗原部位AのSDA又はmAbは、TeNTの直接コーティングに対して結合の低下を示すので、立体構造的に敏感なエピトープと結合する
抗原部位DのSDAは、ウェスタンブロットにおいてTeNT Hcと結合する。
SDA産生レベルを増加させるために、多量体SDAを、プラスミドpRL44を使用して[69]、酵母菌菌株SU50において[70]、安定なMIRY組み込み体を作成することにより生成した[75]。そのようなMIRY組み込み体は、2ミクロンベースのプラスミドと比較して、それより5倍増加したSDAの産生レベルを平均して有する。TeNT結合SVT−SDAとAlb結合SVA12SDA(5個のプラスミド)、IgG結合SVG06SDA(2個のプラスミド)、又はIgG結合SVG13SDA(5個のプラスミド)の融合体から構成される、多量体SDAをコードする12個のプラスミドを生成した(表14)。これらの多量体SDAが作成されたエレメントは以下の通り:

合成SacI−HindIII断片を生成し、そしてその後、SacI及びHindIII部位を使用してプラスミドpRL44にサブクローン化し[69]、プラスミドpRL482〜pRL501を得た(表14)。
様々な多量体酵母菌内産生SDAのいくつかの標的抗原結合能力を、ELISAにおいて特に評価した。これら多量体SDAの構成ブロックを形成する一価のSDAのコントロールが含まれた。ELISAを、以前の実施例に記載されるように基本的に実施した。異なる7種類のELISAを実施した(表15及び表16)。GT1b−TeNT阻害ELISAは、実施例3及び実施例10においてすでに記載されている。抗hisタグmAb及びポリクロナール抗SDA血清の両方がSDA検出用として使用された3つのさらなるELISAが、受動吸着されたTeNT(0.75μg/ml)、Alb(5μg/ml)、又はIgG(5μg/ml)に対する一価のSDAの結合を測定するのに役立った。TeNT、及びIgG若しくはAlbに対する二重特異性結合、又は二価のTeNT結合を測定するために、以前記載されたように、ビオチンに対するタンパク質の重量比として5を用い、スルホ−NHS−LC−ビオチン(Pierce社、Rockford、IL)を使用して、TeNT、及びウマAlbもビオチン化した[72]。それらを、下記事項を測定するために3つのELISAで使用した:
コーティングされたイヌIgG又はAlb(5μg/ml)、及びビオチン化されたTeNT(0.25μg/ml)に対する二重特異性結合
受動吸着されたcAb−TT2(1μg/ml)を用いて捕捉されたTeNT(0.75μg/ml)、及びビオチン化されたウマAlb(1μg/ml)に対する二重特異性結合
コーティングされたTeNT(0.75μg/ml)、及びビオチン化されたTeNT(0.25μg/ml)に対する二価の結合
様々なELISAの結果を、到達した最大吸収(TeNT−GT1b阻害ELISAの場合、最低吸収;表15)、及びSDAの有効濃度(表16)の両方について提示する。これらのELISAから、以下のように結論付けることができる:
GT1b−TeNT阻害ELISAは、SVT02含有SDA、並びに単量体のSVA、及びSVG SDAはいずれも阻害しない一方、すべてのさらなる単量体及び多量体SDAは確かに阻害することを示す。これは、単量体SDA SVT06L、SVT15L、及びSVT16Lにおける以前の観察知見と整合する(実施例10)。
SDA SVT06−GS3−SVT16−L123Q−GS2−SVA12M2−H6は、2つのTeNT−TeNT分子に対して二価結合を示す。
単量体及び多量体SDAを用いたいくつかのin vitroアッセイ(実施例4、5、7、8、9、15、及び18)では、これらのSDAは、いくつかの種(例えば、ウマ、イヌ、ネコ、及びブタ)の血液成分と結合することができることが実証された。SDAのいくつかについて、血清半減期の延長に関するそのin vivoでの特徴を評価するために、動物実験を実施した。SDAはウマ及びイヌタンパク質のみの使用により生成したので、動物実験は、真に異種間となるようにブタを対象に実施した。この目的のために、オス12匹、及びメス12匹からなる約6週齢の仔ブタ24匹を使用した。多量体SDAのイノキュレーションの前にそれらを10日間秤量し、並びにそれぞれ性別の分布が等しく、好ましくは平均体重が等しく、及び好ましくは母ブタの起源に基づき仔ブタの分布が等しい、仔ブタ6匹からなる4群に割り振った。群(表18)は、筋肉内注射される二重特異性SDAをコードするプラスミドの名称(pRL489、pRL490、pRL495、及びpRL499)によりそれぞれ示す。群pRL489の仔ブタ3匹(オス2匹及びメス1匹)は、M8ggsVI4q6eと呼ばれるプラスミドpRL276によりコードされる第2の二重特異性SDAの投与も受けた[44]。この二重特異性SDAは、半減期を測定するのに以前使用された[19]が、しかし口蹄疫ウイルス(FMDV)抗原に対して特異的である異なる融合型SDAドメインであるSDA2 K609ggsVI−4q6eと同一のIgG結合SDAドメインを含有する。
SDA(t)=SDA(0)*(0.5(t/T1/2β))、式中、tは時間(時)であり、SDA(0)は、使用した最初の時点におけるSDA濃度(半減期の計算では体重増加について補正される)であり、SDA(t)は、時間tにおけるSDA濃度であり、T1/2βは最終半減期である。
単量体及び多量体SVTに基づくSDAを用いてin vitroアッセイ(実施例3、10、11、12、13、15、及び18)を行い、これらのSDAは、いくつかの実験的なテスト構成において破傷風ホロ毒素に有効に結合することができることを実証した。これらのSDAのいくつかについてin vivoでの特徴を評価するために、その破傷風毒素中和能力を評価する動物実験を実施した。マウス毒素中和テストを使用して、6つの候補SDAサンプル(表19を参照)を、抗破傷風毒素効力についてテストした。1つの単量体SDA(SVT03L)、4つの二重特異性SDA(SVT02−GS2−SVG13M4−H6、SVT06−GS2−SVG13M4−H6、SVT153FW4M−GS2−SVG13M4−H6、SVT16L123Q−GS2−SVG13M4−H6)、及び1つの二重特異性(アルブミン及び破傷風に対する)、及び二価の(破傷風毒素に対する)SDA(SVT06−GS3−SVT16−L123Q−GS2−SVA12M2−H6)を、マウス毒素中和テスト(TNT)において評価した。破傷風抗毒素に関するヨーロッパ薬局方モノグラフ0091に記載される方法と類似した方法に従い、実施した。SDAの中和エンドポイントを正確に決定するために、Ph Eurモノグラフ0091に記載の方法よりも高い感度レベルを使用してアッセイを実施し、より低い量の毒素中和抗体の検出を可能にした。実施したアッセイの破傷風毒素(NIBSC:AWX4664、1/100に希釈)用量レベルは、Lp/200であった。各試験において、参照破傷風抗毒素TE3(最初の希釈において0.025IU/mlまで1/400事前希釈)を含め(1群マウス4匹)、テストサンプル毎に効力の決定を可能にした。毎回、0.35mlの容積が固定された毒素を、2.15mlの事前希釈されたSDAテストサンプルと混合し、そして30分間放置した後、マウスに注射した(0.5ml s.c.、左大腿部)。各事前希釈されたサンプルを、バッファーを用いて連続希釈して、2倍又は4倍希釈系列を作出した。各SDA希釈は、n=4匹のマウスを有した。動物を、破傷風麻痺の兆候について96時間観察した。各アッセイにおいて、参照TE3をバッファーで希釈した(2倍又は4倍)。各試験(1、2、及び3)において、メスNIHマウス、16〜20g、5〜6週齢を使用した。合計3回の試験を連続して実施した。
いくつかのSDAを、その標的(異なる種のTeNT、アルブミン、及び免疫グロブリン)結合特性についてテストした。破傷風毒素結合特性は、各SVT SDAが破傷風毒素をin vivoで中和する能力において特に重要である(実施例17)。SVA、及びSVG SDAが、一旦SVT SDAに融合すれば、SVT SDAの最終血清半減期を延ばすのに役立つ。従って、SVA及びSVG SDAにとって、異なる血液成分に対する結合特性[19、22、23、25]は、重要である。獣医学的目的にとって、さらに、そのような血液成分間の種の相違に対処することが重要である。特定の異種間疾患標的(例えば、破傷風)の場合、種毎に個別の治療用SDAを開発する代わりに、いくつかの種を対象とする治療用途で、単一のSDAを使用することが好ましい。
さらに2つの多量体SDA(SVT06−GS3−SVT15−3FW4M−GS2−SVA12M2−H6、及びSVT15−3FW4M−GS3−SVT06−GS2−SVG13M5−H6)を、酵母菌菌株SU50[70]において、プラスミドpRL44[69]を使用して、安定なMIRY組み込み体[75]を作成することにより生成した。これらの多量体SDAが作成されたエレメントは以下の通り:

合成SacI−HindIII断片を生成し、その後SacI及びHindIII部位を使用してプラスミドpRL44[69]中にサブクローン化し、プラスミドpRL505及びpRL506を得た(表34)。
cAb−TT1及びcAb−TT2と比較したとき[46]、Octet Red96装置(Pall Life Sciences社)を使用したとき、ストレプトアビジン(SA)バイオセンサーを装備したときに、選択されたSDAについて、その結合を試験するために、さらなるアッセイを実施した。この目的のために、破傷風毒素に対するSDA SVT06L、SVT15L、及びSVT06−GS3−SVT16−L123Q−GS2−SVA12M2−H6、cAb−TT1、及びcAb−TT2の結合親和性を評価した。
選択された二価及び二重特異性(多量体)SDAについて、ウマ、イヌ、及びヒト血清アルブミンに対するその結合親和性を決定するために、追加のアッセイを実施した。
いくつかのSDAについて、標的のTeNTに対する親和定数は、Octet Red96装置により決定したときpMの範囲内であった。この知見を確認するために、追加の技術、Creoptix(クレオプティクス)(商標)センサー専用のグレーティング−カップルドインターフェロメトリー(GCI)法[76]を使用した。GCIは、分子間相互作用をモニタリングし、及び特徴づけ、並びに相互作用するアナライトの動力学的速度、親和定数、及び濃度を決定するための導波路インターフェロメトリーに基づく方法である。その他の光学的ラベルフリー法、例えば表面プラズモン共鳴法(SPR)等と同様に、導波路インターフェロメトリーは、相互作用する分子が複合体形成することによる質量変化に起因して、センサー表面近傍のエバネッセント場内に生ずる屈折率変化を検出する。2つの非常に鋭敏な方法、すなわちインターフェロメトリー及び平面光導波路センシングを併用すれば、非常に低い検出限界を実現する。
SDA SVT06−GS3−SVT16−L123Q−GS2−SVA12M2−H6について、血清半減期の決定を、ウマにおいても実施した。SDAはウマ及びイヌアルブミンタンパク質のみの使用により生成したので、関連する標的種である。この目的のために、4.5〜6.5歳のポニー(メス)3頭を使用した。それらを筋肉内イノキュレーションの前に秤量した。体重に基づき、動物1頭当たりの多量体SDAの必要とされる量を計算した(0.17mg/kgで投薬)。フィルター滅菌処理されたSDAをPBSに溶解し、それを単一部位において後部大腿部中に約2.5mlの容積で筋肉内注射した。血清調製用の血液サンプルを、SDAイノキュレーションの直前、並びにその後の1、2、4、7、10、13、17、及び21日目において頸静脈から収集した。

Claims (18)
- 破傷風神経毒素(TeNT)に結合する能力を有する単一ドメイン抗体(SDA)であって、配列番号17、13、14、15、16、18、19、20、21、22、23、24、25、及び26からなる群から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、但しCDR1、CDR2、及びCDR3のアミノ酸配列同一性が少なくとも75%である単一ドメイン抗体(SDA)。

- CDR1、CDR2、及びCDR3のアミノ酸配列同一性が、少なくとも76%又は好ましくは100%である、請求項1又は請求項2に記載のSDA。
- 請求項1〜3のいずれか一項に記載の、TeNTに結合する能力を有する少なくとも1つのSDA、及び血清タンパク質に結合する能力を有する少なくとも1つのSDAを含むポリペプチドコンストラクト。
- 前記血清タンパク質が、血清アルブミン又は免疫グロブリンである、請求項4に記載のポリペプチドコンストラクト。
- 血清アルブミンに結合する能力を有するSDAが、配列番号40、37、38、39、41、及び42からなる群から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、但しCDR1、CDR2、及びCDR3のアミノ酸配列同一性が少なくとも75%である、請求項5に記載のポリペプチドコンストラクト。
- 前記免疫グロブリンが、免疫グロブリンG(IgG)である、請求項5に記載のポリペプチドコンストラクト。
- 前記免疫グロブリンに結合する能力を有するSDAが、配列番号30、27、28、29、31、32、33、及び34からなる群から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、但しCDR1、CDR2、及びCDR3のアミノ酸配列同一性が少なくとも75%である、請求項5又は7に記載のポリペプチドコンストラクト。
- TeNTに結合する能力を有する少なくとも2つのSDAを含み、TeNTに結合する能力を有する前記少なくとも2つのSDAのそれぞれが、選択肢A;配列番号24、又は選択肢B;配列番号25、又は選択肢C;配列番号20、又は選択肢D;配列番号17若しくは19、又は選択肢E;配列番号22、15、23、若しくは14から選択される配列と少なくとも70%の全体的なアミノ酸配列同一性を有し、但し前記少なくとも2つのSDAが、同一の選択肢に由来する配列を含まず、並びにCDR1、CDR2、及びCDR3のアミノ酸配列同一性が、少なくとも75%である、請求項4〜8のいずれか一項に記載のポリペプチドコンストラクト。
- 請求項1〜3のいずれか一項に記載の少なくとも1つのSDA、及び/又は請求項4〜9のいずれか一項に記載の少なくとも1つのポリペプチドコンストラクト、及び薬学的に許容できる担体を含む医薬組成物。
- 請求項1〜3のいずれか一項に記載のTeNTに結合する能力を有する少なくとも2つのSDA、及び/又は請求項9に記載の少なくとも1つのポリペプチドコンストラクトを含む、請求項10に記載の医薬組成物。
- 医薬としての使用のための、請求項1〜3のいずれか一項に記載のSDA、又は請求項4〜9のいずれか一項に記載のポリペプチドコンストラクト。
- 破傷風菌(Clostridium tetani)の疾患/症状の予防又は処置における使用のための、請求項1〜3のいずれか一項に記載のSDA、請求項4〜9のいずれか一項に記載のポリペプチドコンストラクト、又は請求項10若しくは11に記載の医薬組成物。
- 請求項1〜3のいずれか一項に記載のSDA、又は請求項4〜9のいずれか一項に記載のポリペプチドコンストラクトをコードするDNA断片。
- 請求項14に記載のDNA断片を含む核酸であって、請求項14に記載のDNA断片が、プロモーター、及び任意選択でその他の調節エレメントと作動可能にリンクしている核酸。
- 請求項15に記載の核酸を含む宿主細胞。
- 請求項1〜3のいずれか一項に記載のSDA、又は請求項4〜9のいずれか一項に記載のポリペプチドコンストラクトを製造する方法であって、a)SDA又はポリペプチドコンストラクトの発現を可能にする条件下で、請求項16に記載の宿主細胞を培養するステップと、任意選択でb)宿主細胞及び培養培地のうちの少なくとも1つからSDA又はポリペプチドコンストラクトを回収するステップとを含む方法。
- 請求項1〜3のいずれか一項に記載のTeNTに結合する能力を有する少なくとも1つのSDAを含む診断キット。
