Claims (38)
- レバウジオシドNを調製する生合成方法であって、
1,2−ラムノシトランスフェラーゼ活性を有する第1の組み換えポリペプチドの存在下で、ステビオール配糖体組成物をラムノース供与部分と反応させることを含み、前記第1の組み換えポリペプチドが、配列番号3、配列番号9、又は配列番号19と少なくとも80%の配列同一性を有するアミノ酸配列を含む、方法。 - 前記第1の組み換えポリペプチドが、配列番号3と少なくとも90%の配列同一性を有するアミノ酸配列を含む、請求項1に記載の方法。
- 前記第1の組み換えポリペプチドが、配列番号9と少なくとも90%の配列同一性を有するアミノ酸配列を含む、請求項1に記載の方法。
- 前記第1の組み換えポリペプチドが、配列番号19と少なくとも90%の配列同一性を有するアミノ酸配列を含む、請求項1に記載の方法。
- 前記第1の組み換えポリペプチドを形質転換細胞系で発現させることを含む、請求項1〜4のいずれか一項に記載の方法。
- 前記形質転換細胞系が、酵母、非ステビオール配糖体産生植物、藻類、真菌、及び細菌からなる群から選択される、請求項5に記載の方法。

- 前記第1の組み換えポリペプチドを前記形質転換細胞系から単離することを含み、前記反応工程がインビトロで行われる、請求項5〜7のいずれか一項に記載の方法。
- 前記ラムノース供与部分が、UDP−L−ラムノースである、請求項1〜9のいずれか一項に記載の方法。
- 前記ステビオール配糖体組成物がレバウジオシドAを含み、前記反応工程が、レバウジオシドJの製造をもたらす、請求項1〜10のいずれか一項に記載の方法。
- 前記レバウジオシドJを、グルコシルトランスフェラーゼ活性を有する第2の組み換えポリペプチドの存在下で、グルコース供与部分と反応させることを含む、請求項11に記載の方法。
- 前記グルコース供与部分がインサイチュで生成される、請求項12に記載の方法。
- 前記第2の組み換えポリペプチドが、グルコシルトランスフェラーゼ活性及びスクロースシンターゼ活性の両方を有する、請求項12又は13に記載の方法。
- 前記第2の組み換えポリペプチドが、配列番号7、配列番号11、配列番号13、又は配列番号15と少なくとも80%の配列同一性を有するアミノ酸配列を含む、請求項12又は13記載の方法。
- 前記レバウジオシドJを、スクロースシンターゼ活性を有する第3の組み換えポリペプチドの存在下で、グルコース供与部分と反応させることを含む、請求項12又は13に記載の方法。
- 前記ステビオール配糖体組成物がレバウジオシドIを含む、請求項1〜10のいずれか一項に記載の方法。
- 前記レバウジオシドIが、グルコシルトランスフェラーゼ活性を有する第2の組み換えポリペプチドの存在下で、レバウジオシドAを含むステビオール配糖体組成物をグルコース供与部分と反応させることによって調製される、請求項17に記載の方法。
- 前記グルコース供与部分がインサイチュで生成される、請求項18に記載の方法。
- 前記第2の組み換えポリペプチドが、グルコシルトランスフェラーゼ活性及びスクロースシンターゼ活性の両方を有する、請求項18又は19に記載の方法。
- 前記第2の組み換えポリペプチドが、配列番号7、配列番号11、配列番号13、又は配列番号15と少なくとも80%の配列同一性を有するアミノ酸配列を含む、請求項18又は19記載の方法。
- スクロースシンターゼ活性を有する前記第3の組み換えポリペプチドの存在下で、レバウジオシドAを含む前記ステビオール配糖体組成物をグルコース供与部分と反応させることを含む、請求項18又は19に記載の方法。
- レバウジオシドJを調製する生合成方法であって、
1,2−ラムノシルトランスフェラーゼ活性を有する組み換えポリペプチドの存在下で、レバウジオシドAを含むステビオール配糖体組成物をラムノース供与部分と反応させる工程を含み、前記組み換えポリペプチドが、配列番号3、配列番号9、又は配列番号19と少なくとも80%の配列同一性を有するアミノ酸配列を含む、方法。 - 前記組み換えポリペプチドが、配列番号3と少なくともの90%の配列同一性を有するアミノ酸配列を含む、請求項23に記載の方法。
- 前記組み換えポリペプチドが、配列番号9に少なくとも90%の配列同一性を有するアミノ酸配列を含む、請求項23に記載の方法。
- 前記組み換えポリペプチドが、配列番号19と少なくとも90%の配列同一性を有するアミノ酸配列を含む、請求項23に記載の方法。
- 前記組み換えポリペプチドを形質転換細胞系で発現させることを含む、請求項23〜26のいずれか一項に記載の方法。
- 前記形質転換細胞系が、酵母、非ステビオール配糖体産生植物、藻類、真菌、及び細菌からなる群から選択される、請求項27に記載の方法。
