Abstract
Description

[0040]ペプチドは特有な形で免疫療法に適合する。ペプチドは、設計に柔軟性があり、容易に大規模に合成され、水溶性で、比較的安定である。ワクチンとしての使用は長期にわたり成功を収めてきた歴史を有するが、高いアビディティでレクチン型受容体に選択的に結合するペプチドもまた設計されている[1〜3、およびそれらの参照文献]。受容体に媒介される免疫療法におけるペプチドの使用は、ペプチドが、炭水化物リガンドを模倣し、内因性リガンドよりも桁違いに大きいアビディティで、免疫系の細胞によって発現される細胞表面の制御性レクチン型受容体に結合する能力に基づいている。多価リガンドと多量体受容体との相互作用は、結合アビディティの劇的な増加をもたらし、最大半量結合濃度を低ナノモル範囲に減少させる。
[0047]GalNAcを含有するCLEC10Aの天然リガンドが、低い親和性で結合し、かつ抗原性であることを考慮して、CLEC10Aに結合し得るGalNAcの多価ペプチド模倣物を設計した。ファージディスプレイライブラリのスクリーニングから現れた四価の12アミノ酸のペプチド配列であるsvL4は、以前に特性決定された[60]。svL4を、sv6Dと称される、svL4のC末端の6マーの配列を有する四価構造としてさらに洗練した(図1B〜1C)。sv6Dは、svL4の結合活性を保持し、より強力な免疫細胞の刺激因子である。sv6DおよびsvL4は、GalNAcに特異的なレクチンに高いアビディティで結合する。MHC結合データベースを用いたペプチドの配列の分析は[61、62]、ペプチドがヒトにおけるMHCクラスI分子またはMHCクラスII分子によって提示される可能性も、抗体の産生を誘導する可能性も高くないと予測した。実際、抗体は、3か月にわたる隔日の皮下注射後のマウス血清において検出されなかった。重要なことに、ペプチドの皮下注射は、腹膜腔における免疫細胞の増殖および成熟を刺激した。
[0051]インターフェロン−β(IFN−β)は、疾患に対する内因性防御機構によって産生される最も重要な抗ウイルスおよび抗癌サイトカインである。IFN−βの役割の解明につながる最初の発見は、1976年に、哺乳動物細胞のインターフェロン処理が二本鎖RNA(dsRNA)の存在下でタンパク質合成の阻害を引き起こすという観察によってなされた[70]。この知見の直後に、2’−5’オリゴアデニレート(2〜5A)、2〜5A合成酵素(OAS)、および潜在性リボヌクレアーゼであるRNアーゼLの発見が続いた[70〜72]。これらの役者達は、感染に対する最も重要な細胞の抗ウイルス応答として認識されるようになった[73、74]。RNA含有ウイルスが細胞質基質において脱外被する場合、dsRNAは、ATPを2〜5Aに変換するOASを活性化させる。RNアーゼLは、2〜5A結合の際に生じる二量体化によって活性化する。活性化したヌクレアーゼはdsRNAを切断し、ウイルスの複製を阻止する。加えて、細胞のRNAもまた分解され、これによりタンパク質合成を阻害し、最終的にアポトーシスを引き起こす[75]。しかしながら、この過程において、短鎖RNA断片は、RNAヘリカーゼであるRIG−1およびMDA5、アダプターであるIPS−1、ならびに転写因子であるIRF−3およびNF−κBを介してIFN−βの発現を誘導する[76、77]。IFN−βは感染した細胞によって分泌され、隣接する細胞にて受容体に結合して、OASの発現の上方制御をもたらし、近接する細胞のウイルス複製に対する感受性を増強する。感染に対する増強した感受性は、結果としてより急速なアポトーシスを生じ、それによって感染の拡大を阻止する。
[0054]1980年代半ば以来、光線力学療法は有望な抗癌手法として発展してきた[88、89]。ヘマトポルフィリンの誘導体は、癌細胞によって選択的に取り込まれ、癌細胞を光感受性にすることが見出された。この技術の制限は、ヘマトポルフィリン誘導体が600nmより長い波長で光を弱く吸収することであった。結果として、組織の強い吸光度のために、体の表面またはその付近に生じる腫瘍のみが接触可能である。Hooberら[90]は、クロロフィルからプルプリン−18を調製し、その化合物の無水物形態が695nmで強い最大吸光度を有することを示した。無水物基が加水分解によって開環してクロリン−p6を生成した場合、最大吸光度は662nmにシフトしたが、依然として光線力学療法を支持し得る波長であった。これらの生成物は、細胞死滅に関して約10−8MのIC50濃度を有した[90]。

[0096]ペプチドの合成。svL4の機能配列を、カタツムリHelix pomatia由来のGalNAc特異的レクチンを用いた12マーのファージディスプレイライブラリ(New England BioLabs、Ipswich、MA)のスクリーニングにより同定した[60、111]。コンセンサス配列を、受容体への結合に関する、リガンド密度とエントロピー因子との関数としてのアビディティの概念に基づいて四価構造に組み込んだ[44、49、112〜114]。多価ペプチドを、CBL Biopharma LLC(Patras、Greece)によるFmoc(9−フルオレニルメトキシカルボニル)−保護アミノ酸を利用した標準的な固相化学によって合成した。トリリジン「コア」を固相レジン上で合成し、配列GGSで伸長した。C末端がGである複数の「アーム」を標準的な化学によって別個に合成し、コアを有する溶液中で縮合した[115]。C末端における修飾は、アミド基(タグ無し)、またはε−ビオチニル−リジニル−アミドを有する伸長鎖からなった。配列GGGS(配列番号3)を、模倣物配列とトリリジンコアとの間のリンカー配列として構造に含めた。svL4の誘導体もまた合成して、配列のサブセットが異なる活性を有するかを決定した。ペプチドを、0.1%トリフルオロ酢酸水溶液中5%〜25%アセトニトリルの勾配を使用した60℃のカラム温度でXDB−C8カラムでのHPLCによって95%超に精製した。正確な合成および純度を含む合成産物の品質を、MALDIおよびESI質量分析法によって評価した。凍結乾燥ペプチドを100mM NaClに溶解し、pH5に中和し、CM−Sephadex C−25カラム(2.5×10cm)に吸着させた。カラムを200mM NaClで洗浄して、確実にリポ多糖(LPS)を除去した後に、svL4を500mM NaClで溶出した。LPSは、CM−Sephadexカラムから溶出したペプチド溶液において検出されなかった(<0.01EU/10mgペプチド)。ペプチドの濃度を、210nmにおける実験的に決定された消散係数である22OD単位/mg/mlを用いて決定した。

[0126]ペプチドsv6Dは、全生存を延ばすための免疫療法薬の有効性を増強するのにもきわめて効果的である。モノクローナル抗マウスPD−1の腹腔内注射は、卵巣癌モデルのマウスにおいて適度に効果的である[102、103]。本発明者らは、sv6Dが抗PD−1の効力を延長し得るかを試験した。5用量の抗PD−1(PBS中200μg)を41から49日目の間、隔日で腹腔内注射した。sv6Dを、抗PD−1を用いた処置の直後に皮下注射し、研究の終了まで隔日で継続した(図9A、群3)。抗PD−1を用いて処置した群へのsv6Dの投与を、動物の重量が増加し始めた後、87日目まで遅延させた場合(群5)、腹水形成の本質的に完全な抑制が数週間持続した。
[0129]重量増加は、腹膜腔に播種した卵巣癌の本モデルにおける疾患進行の良好な尺度であるが、平均群重量の考察は複雑である。これはそのままの生物学的モデルであるため、固有のばらつきが存在する。処置群内の個々の動物の疾患が進行するにつれ、処置群内の重量のばらつきは大きくなる。個々の動物が末期の疾患に達し、安楽死させる場合、平均群重量は低下することがあり、群内のばらつきは小さくなる。この複雑性を考慮すると、処置群内の疾患の進行の低減が統計的有意性を達成した重量の差に反映されているいくつかの事例が残っていた。
