Abstract
Description

(i)少なくとも2.0μm.cm/volt.sec(または少なくとも1.0μm.cm/volt.sec)の電気泳動移動度によって規定される表面電位;または
(ii)少なくとも1.077g/mlの密度;または
(iii)toll様受容体の発現もしくは活性;ならびに/またはプログラム細胞死1(PD-1)受容体;CD115;CD224;CXCR1;および/もしくはCXCR2の発現の非存在もしくは不活性;ならびに
がん細胞を殺傷する能力を有する顆粒球が、細胞培養物中に含まれる全顆粒球(好ましくは、全細胞)の少なくとも70%、75%、80%、85%、90%または95%を占めることを意味する。
a.本明細書に記載の方法によって顆粒球または造血細胞を取得すること;および
b.前記顆粒球または造血細胞を対象に投与すること
を含む方法を提供する。
ドナーの採用
ドナーは、実施例2に記載されるCKAアッセイにおいて高レベルのがん殺傷活性(CKA)を示す好中球を有する確率に基づいて予備選択される。予備選択基準は、
・同意書に提供または試料採取に影響する重篤な医学的または精神状態にないこと;
・療法のために標的化されるがんの既往歴または家族歴がないこと;
・試料採取日以前の3ヶ月間に化学療法または放射線療法の履歴がないこと;
・18〜24歳の年齢であること;
・任意選択により、男性であること(理論によって束縛されることを望むものではないが、男性由来好中球は、CKAアッセイにおいて試験した場合、最も高いレベルのCKAを示すと考えられる);および
・任意選択により、血液型OまたはRh−であること
を含む。
CKAアッセイにおける抽出された顆粒球のCKAの試験
細胞を、T25フラスコ中のDMEM+10%FBS中で80%の集密度になるまで培養する。細胞株を、T75cm2細胞培養フラスコ中の、以下の成分:10%体積/体積のFBS、ペニシリン(Sigma.St. Louis、MO)、ストレプトマイシン(Sigma.St. Louis、MO)、およびL−グルタミン補給物(Sigma.St. Louis、MO)を添加したDMEM中、37℃、8%CO2で増殖させ、維持する。培養された膵がん細胞(例えば、American Type Culture Collection - United Kingdom (U.K.)、Guernsey、Ireland、Jersey and Liechtenstein、LGC Standards、Queens Road、Teddington、Middlesex TW11 0LY、UKから商業的に入手可能なCapan-2、ATCC HTB-80;Panc 10.05、ATCC CRL-2547;CFPAC-1、ATCC CRL-1918;HPAF-II、ATCC CRL-1997;SW 1990、ATCC CRL-2172;BxPC-3、ATCC CRL-1687;AsPC-1、ATCC CRL-1682;ATCC(登録商標)TCP-1026(商標);SW1990、ATCC CRL-2172;SU.86.86、ATCC CRL-1837;BXPC-3、ATCC CRL-1687;Panc 10.05、ATCC CRL-2547;MIA-PaCa-2、ATCC CRL-1420;PANC-1、ATCC CRL-1469;またはATCC(登録商標)TCP-2060(商標))を分割し、培養フラスコ中で70%の表面集密度に達するまで継代する。
造血細胞および好中球の表面電位の試験
電気泳動を使用して、電気泳動移動度を測定することによって造血細胞(例えば、造血幹細胞、および/または前駆細胞)および好中球における表面電位の変化を調査する。機械的剥離および培養基質からの収集によって、懸濁した細胞を培養物から収集する。収集された細胞を、10mM Tris−HClおよび291mMグルコースを含有する電気泳動緩衝溶液中に再分配し、方形ガラス電気泳動チャンバー中に導入する。電気泳動チャンバーにわたって200VのDCを印加する。CCDカメラを備えた顕微鏡下で、3mAで、細胞が固定した長さを通過するのに必要な時間を記録することによって、細胞の電気泳動速度、uを測定する。電気泳動移動度、μは、μ=ugS/I(式中、gは媒体の電導度であり、Sは電気泳動チャンバーの断面積であり、Iは電流である)によって算出される。それぞれの条件について、典型的には、少なくとも9回の読取りを行って、細胞の電気泳動移動度を算出する。

造血細胞の拡張および分化

・造血細胞の培養および増殖;

細胞バンクの調製
造血幹細胞、それから取得可能な顆粒球前駆細胞および顆粒球を、低温凍結し、適切な細胞バンク中で保存する。
固形腫瘍の処置のための患者における使用
保存された造血細胞(例えば、造血幹細胞またはそれから取得可能な顆粒球前駆細胞)、およびそれから分化した顆粒球(例えば、好中球)を、患者のがんの型、血液型(ABO、rhおよびHLA)、および/または遺伝学に基づいて、がん患者とマッチングする。また、患者を、ヒト白血球抗原(HLA)類似性に基づいてマッチングしてもよい。
・患者への、顆粒球−コロニー刺激因子、ヒト成長ホルモン、セロトニン、およびインターロイキンと一緒の造血細胞(造血幹細胞、および顆粒球前駆細胞を含む)のIV輸注;または
・患者への、刺激された顆粒球前駆細胞(造血幹細胞から取得可能)のIV輸注。理論によって束縛されることを望むものではないが、前記細胞は、in vivoでのCKAアッセイにおいて高いCKAを有する顆粒球(例えば、好中球)に自然に分化すると考えられる;または
・造血細胞(例えば、造血幹細胞)から分化したCKAアッセイにおいて高いCKAを有する顆粒球(例えば、好中球)の直接IV輸注。
膵がんを有する患者の処置
メアリーは、69歳で転移性膵管腺癌(PDAC)と診断される。外科手術は最早選択肢になく(切除不能)、ゲムシタビンは疾患進行の防止には不十分であり、アブラキサンまたはフォルフィリノックスは、彼女に残された時間を家族と共に楽しむことをできなくさせるであろう副作用のため、彼女の腫瘍学者の推奨によると適切ではない。メアリーの予後は、3〜6ヶ月の生存であり、彼女が、近々生まれると期待されるその孫を見るのに十分なほど長く生きるのは絶望的である。
MTT「CKAアッセイ」
MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド)は、黄色のテトラゾールであり、正に荷電し、生きている真核細胞に容易に浸透する。活発な代謝を示す生細胞は、NAD(P)H依存的オキシドリダクターゼミトコンドリア酵素により、MTTを、紫色のホルマザン生成物(1(E,Z)−5−(4,5−ジメチルチアゾール−2−イル)−1,3−ジフェニルホルマザン)に変換し、570nm近くの最大吸光度を示す。細胞が死ぬとき、それらはMTTをホルマザンに変換する能力を失い、かくして、色の形成は生細胞のみの有用かつ便利なマーカーとして役立つ。可溶化溶液を添加して、不溶性の紫色のホルマザン生成物を、着色溶液中に溶解する。この着色溶液の吸光度を、分光光度計によって570nmの波長で測定することによって定量することができる。690nmの参照波長の吸光を、570nm波長の吸光から差し引く。したがって、MTTアッセイを使用して、がん殺傷活性(CKA)、つまり、がん細胞に対する細胞傷害性を決定するための方法としてどれぐらい多くの生細胞が残存するかを測定した。
1日目:
1)HeLa細胞(頑強型の子宮頸がん細胞)を培養し、それらが対数増殖期に達した時に回収した。
2)10000個のHeLa細胞(標的細胞)を、最終容量100μLで96ウェル平底プレートの各ウェルに添加した。
3)標的細胞を静置して一晩接着させた後、白血球(エフェクター細胞)を添加し、全ての実験条件を3組設定した。
4)エフェクター細胞を、様々な比(例えば、1:1、5:1、10:1、50:1のエフェクターの標的細胞に対する比)で標的細胞に添加した。
5)細胞を静置して37℃で16〜24時間インキュベートした。
6)標的細胞のみ、およびTriton Xの存在下の標的細胞も、それぞれ、0%および100%の細胞傷害性に関する対照として3組播種した。
1)ウェルをPBSで2回洗浄して、エフェクター細胞および死滅した標的細胞を除去した。
2)培養培地を用いてキット溶液を10倍に溶解することにより、MTT溶液を調製した、すなわち、100ウェルあたり、1000μL(1mL)のMTTストック(キット中に提供される)を取り、9000μL(9mL)の培養培地(RPMI-1640)を添加した。
3)ウェルあたり100μlの工程2で調製されたMTT溶液を添加した。
4)これを4時間インキュベートした。
5)MTT溶液を全てのウェルから除去し、100μl/ウェルの溶媒を添加した(キット中に提供される)。
6)必要となった場合にピペッティングによってホルマザン結晶を溶解し、プレートを570および690nmで読み取った。690nmで測定したバックグラウンド吸光度を、570nmで測定した吸光度から差し引いた。
匿名の血液ドナーに由来する白血球コーンを選択し、好中球を、Ficoll−Hypaque分離(Oh H, Siano B, Diamond S. Neutrophil Isolation Protocol. Journal of Visualized Experiments: JoVE. 2008;(17)745)によって単離した。これらの好中球を、1:1および5:1のエフェクターの標的細胞に対する比で上記のMTTアッセイにおいて使用した。
幹細胞由来好中球のCKAの証明
CD34+幹細胞に由来する好中球の培養
本発明者らは、Timmins NE、Palfreyman E、Marturana F、Dietmair S、Luikenga S、Lopez Gら、Clinical scale ex vivo manufacture of neutrophils from hematopoietic progenitor cells. Biotechnology and bioengineering. 2009;104(4):832−40によって記載されたプロトコールを使用して、CD34タンパク質を発現する臍帯血由来幹細胞に由来する好中球を培養した。
幹細胞由来好中球(バッチ008A、709Aおよび915)を、HeLa標的細胞を使用するCKA MTTアッセイにおいてエフェクター細胞として使用した(実施例9を参照)。エフェクターの標的細胞に対する比は、CD11b+/CD15+に基づく。結果を図2にまとめるが、これは、CD34+幹細胞由来好中球が、HeLa細胞のCKAアッセイにおいて細胞傷害性を示すこと、およびその結果が異なるドナー間で異なるということを示す。バッチ008Aは、他のバッチと同時に調製され、同じように培養されるにもかかわらず、10:1までのエフェクターの標的に対する比でバッチ915および709Aよりも一貫して低い細胞傷害性を示す。
xCELLigence「CKAアッセイ」
ACEA Biosciences xCELLigence RTCA DP Analyzerシステム(登録商標)を使用し、製造業者の指示書に従った。xCELLigence Systemは、電気インピーダンスを測定することによって連続的に細胞表現型の変化を、標識を使用せずに動的にモニタリングすることができるリアルタイム細胞分析装置である。このシステムは、組織培養Eプレートの各ウェルの底部に組み込まれた互いに咬合する金微小電極を使用してインピーダンスを測定する。インピーダンス測定値は、Cell Index(CI)値として表示され、生存能力を含む、細胞の生物学的状態に関する定量的情報を提供する。細胞生存能力のインピーダンスに基づくモニタリングは、細胞数およびMTTに基づく読出し値と相関する。インピーダンスに基づく細胞生存能力の測定値の動的態様は、好中球を使用して細胞傷害効果を誘導する場合に必要な時間的情報を提供する。特に、xCELLigence Systemはまた、細胞傷害および細胞死アッセイにおいて、最も低いCI値によって示される、好中球がその最大効果を達成する場合(そのようなデータが望まれる場合)の最適な時点を正確に示すこともできる。典型的には、6,000個のがん細胞(HeLaもしくはPANC-1)または健康な、非がん性細胞(MCF-12F)を、16ウェルプレートの底部に入れる(システムは、最大3個のプレートを同時に読み取ることができる)。細胞をウェルに添加した最初の数時間については、インピーダンスが急速に増大する。これは、懸濁液から落下し、電極上に沈着し、接着斑を形成する細胞によって引き起こされる。添加される細胞の初期数が低く、ウェルの底部に空の空間がある場合、細胞は増殖し、CIの徐々にであるが、安定的な増大を引き起こすであろう。細胞が集密に達した時、CI値は、バルク媒体に接近できる電極表面積が最早変化しないという事実を反映して、プラトーに達する。「正規化点」と呼ばれるこの点で、好中球を添加する(典型的には、変化するエフェクター:標的比で)。細胞溶解のパーセンテージは、簡単な式:細胞溶解のパーセンテージ=((細胞指数エフェクターなし−細胞指数エフェクターあり)/細胞指数エフェクターなし)x100を使用して容易に算出される。
ドナー由来好中球における可変的CKAの証明
図3は、異なるドナーに関してxCELLigenceアッセイによって記録された最大細胞傷害性パーセンテージを示す。このアッセイはまた、1:1と5:1の比(好中球のHeLa細胞に対する比)でのドナー間の差異を示す。結論として、xCELLigenceアッセイも、異なるドナーに由来する好中球間でのCKAの差異を証明することができ、これが、顆粒球のがん細胞に対する様々な比にわたって一貫していたことを証明することができる。

CD34+幹細胞に由来する好中球の培養
臍帯血由来幹細胞に由来する好中球を培養したさらなる結果を、好中球を臍帯血から単離されたCD34+造血幹細胞から生成することができることを示す、表2に提示する。CD34+は、造血幹細胞マーカーである。CD11bおよびCD15は、成熟好中球マーカーである。
CD34+幹細胞由来好中球(SCDN)のCKAの証明
結果は、CD34+幹細胞由来好中球のさらなる集団を用いるxCELLigenceアッセイによって得られ(図6)、上記のMTTアッセイによって得られた結果と一致していた。SCDN(ex vivoで生成された)は再度、示差的なCKAを有することが示され、培養物5は低いCKAの好中球を示し、培養物1は高いCKAの好中球を示した。

好中球のCKAが遺伝的にコードされることの証明
3人の異なるドナーから単離された好中球(DDN)、ならびに同じドナーのCD34+幹細胞に由来するSCDNを、CKAについて試験した。
SCDNがDDNよりも迅速にがん細胞を殺傷することの証明
ドナーLC267、LC268、およびLC269のSCDNおよびDDNのCKAを、最大45時間の期間で決定した。このアッセイを、実施例11に従って行った。
高密度好中球の単離
10mlのヘパリン処理した(20U/ml)ヒト血液を、塩水中の等量の3%デキストランT500と混合し、室温で30分間インキュベートして、赤血球を沈降させる。10mlのスクロース1.077g/mlを用いて50mlの円錐ポリプロピレンチューブを調製し、1.077g/mlのスクロース層の頂部に白血球に富む上清をゆっくりと層化させた後、ブレーキを使用せずに室温で30分間、400xgで遠心分離する。高密度好中球(HDN)がペレット中に出現する。低密度好中球(LDN)は、1.077g/mlのスクロース層と血漿との間の境界面に単球およびリンパ球と共に同時精製される。
人工多能性幹細胞(iPSC)の高いCKAを有する好中球への分化
高いCKAを有する好中球を含むドナーを同定する。体細胞(例えば、線維芽細胞)をドナーから単離し、iPSCの培養物を確立するために使用する。iPSCを、例えば、Sweeney CL、Merling RK、Choi U、Priel DB、Kuhns DB、Wang HおよびMalech HL、Generation of functionally mature neutrophils from induced pluripotent stem cells. - がんの処置における使用にとって好適な造血細胞を取得するためのin vitroでの方法であって、
a.がん細胞株と、ドナーから取得可能な顆粒球とを接触させて、試験試料を形成し、前記試験試料をインキュベートすること;および
b.試験試料中で殺傷されたがん細胞の%が、同じ型のがん細胞株および異なるドナーから取得可能な顆粒球を含む対照試料中で殺傷されたがん細胞の%よりも高い場合、前記ドナーに由来する試料から造血細胞を取得すること
を含む方法。 - がんの処置における使用にとって好適な造血細胞を取得するためのin vitroでの方法であって、

- がん細胞株が、膵がん細胞株、肝臓がん細胞株、食道がん細胞株、胃がん細胞株、子宮頸がん細胞株、卵巣がん細胞株、肺がん細胞株、膀胱がん細胞株、腎臓がん細胞株、脳腫瘍細胞株、前立腺がん細胞株、骨髄腫のがん細胞株、非ホジキンリンパ腫(NHL)細胞株、喉頭がん細胞株、子宮がん細胞株、または乳がん細胞株から選択される1つまたは複数である、請求項1〜11のいずれか一項に記載のin vitroでの方法。
- がん細胞株が、膵がん細胞株である、請求項1〜12のいずれか一項に記載のin vitroでの方法。
- がん細胞株が膵管腺癌細胞株であり、好ましくは、がん細胞株がPANC−1細胞株である、請求項1〜13のいずれか一項に記載のin vitroでの方法。
- 顆粒球が好中球である、請求項1〜14のいずれか一項に記載のin vitroでの方法。
- 混合物中の5%未満のがん細胞を殺傷する顆粒球を廃棄することをさらに含む、請求項6もしくは7または12〜15のいずれか一項に記載のin vitroでの方法。
- 選択された顆粒球が取得可能である前記ドナーに由来する試料から造血細胞を取得することをさらに含む、請求項6もしくは7または12〜16のいずれか一項に記載のin vitroでの方法。
- 好ましくは、造血細胞が正に荷電した細胞表面を有する、請求項1〜5、8〜15または17のいずれか一項に記載の方法によって取得可能な造血細胞またはそのin vitroの細胞培養物。
- 好ましくは、顆粒球が正に荷電した細胞表面を有する、請求項6もしくは7もしくは12〜17のいずれか一項に記載の方法によって取得可能な、または請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物から分化した顆粒球またはそのin vitroの細胞培養物。
- 造血細胞のin vitroの細胞培養物であって、前記造血細胞が、
a.少なくとも1.0μm.cm/volt.secの電気泳動移動度によって規定される表面電位および/または1.077g/mlより高い密度;ならびに
b.がん細胞を殺傷する能力
を特徴とする顆粒球を形成するように分化する、細胞培養物。 - a.請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、または請求項20に記載の造血細胞のin vitroの細胞培養物;および
b.顆粒球−マクロファージコロニー刺激因子(GM-CSF)、顆粒球コロニー刺激因子(G-CSF)、増殖ホルモン;セロトニン、ビタミンC、ビタミンD、グルタミン(Gln)、アラキドン酸、AGE−アルブミン、インターロイキン、TNF−アルファ、Flt−3リガンド、トロンボポエチン、ウシ胎仔血清(FBS)、またはその組合せ
を含む医薬組成物。 - a.請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、請求項20に記載の造血細胞のin vitroの細胞培養物、または請求項21に記載の医薬組成物;および
b.医学におけるその使用のための指示書
を含むキット。 - がんの処置における使用のための、請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、請求項20に記載の造血細胞のin vitroの細胞培養物、請求項21に記載の医薬組成物、または請求項22に記載のキット。
- がんの処置のための薬剤の製造における、請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、請求項20に記載の造血細胞のin vitroの細胞培養物、請求項21に記載の医薬組成物、または請求項22に記載のキットの使用。
- 請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、請求項20に記載の造血細胞のin vitroの細胞培養物、または請求項21に記載の医薬組成物を、それを必要とする対象に投与することを含む、がんの処置のための方法。
- がんが固形腫瘍がんである、請求項18〜25のいずれか一項に記載の造血細胞もしくはそのin vitroの細胞培養物、顆粒球もしくはそのin vitroの細胞培養物、医薬組成物、または使用のためのキット、使用または方法。
- がんが、膵がん、肝臓がん、食道がん、胃がん、子宮頸がん、卵巣がん、肺がん、膀胱がん、腎臓がん、脳腫瘍、前立腺がん、骨髄腫のがん、非ホジキンリンパ腫(NHL)、喉頭がん、子宮がん、または乳がんのうちの1つまたは複数である、請求項18〜26のいずれか一項に記載の造血細胞もしくはそのin vitroの細胞培養物、顆粒球もしくはそのin vitroの細胞培養物、または使用のための医薬組成物、使用または方法。
- 請求項18に記載の造血細胞もしくはそのin vitroの細胞培養物、請求項19に記載の顆粒球もしくはそのin vitroの細胞培養物、請求項20に記載の造血細胞のin vitroの細胞培養物、または請求項21に記載の医薬組成物を含む細胞バンク。
