Abstract
Description
[1]肝芽細胞をプレグネノロンまたはアドレナリン作動薬を含む培地中で培養する工程を含み、当該肝芽細胞がAFP陽性である、肝細胞を製造する方法。
[2]前記アドレナリン作動薬が、アドレナリン、ノルアドレナリン、エチレフリン、ナファゾリン、フェニレフリン、メトキサミンおよびミドドリンから成る群より選択される試薬である、[1]に記載の方法。
[3]前記アドレナリン作動薬が、エチレフリン、フェニレフリンおよびメトキサミンから選択される試薬である、[2]に記載の方法。
[4]前記肝芽細胞が、内胚葉細胞をDMSOを含む培地中で培養する工程を含む方法で製造された細胞であり、当該内胚葉細胞がSOX17陽性細胞である、[1]から[3]のいずれか1項に記載の方法。
[5]前記内胚葉細胞が、多能性幹細胞を、Activin AおよびGSK-3β阻害剤を含む培地中で培養する工程を含む方法で製造された細胞である、[4]に記載の方法。
[6]前記多能性幹細胞から内胚葉細胞を製造する工程が、Activin A、GSK-3β阻害剤およびHDAC阻害剤を含む培地で培養する工程をさらに含む、[5]に記載の方法。

本発明の別の態様においては、肝細胞はグリコーゲンの蓄積、低比重リポタンパク質(LDL)の取り込み、アルブミンの分泌、アンモニア代謝と尿素合成、シトクロムP450活性、脂質代謝、薬物代謝などの肝細胞の機能として公知の機能のいずれかひとつ、あるいは複数の組み合わせによって特徴づけられる細胞と定義してもよい。肝細胞の判定は、かかる機能に係わるタンパク質の発現によって確認することができる。本発明において、肝細胞は、他の細胞種が含まれる細胞集団として提供されてもよく、純化された集団であってもよい。好ましくは、20%以上、30%以上、40%以上または50%以上の肝細胞が含まれる細胞集団である。
(1)多能性幹細胞をActivin AおよびGSK-3β阻害剤を含む培地で培養する工程;
(2)(1)で得られた培養細胞をActivin A、GSK-3β阻害剤およびHDAC阻害剤を含む培地で培養する工程;および
(3)(2)で得られた培養細胞をActivin AおよびGSK-3β阻害剤を含む培地で培養する工程。
(i)多能性幹細胞を、Activin AおよびGSK-3β阻害剤を含む培地中で培養する工程、
(ii)工程(i)で得られた細胞をDMSOを含む培地中で培養する工程、および
(iii)工程(ii)で得られた細胞をプレグネノロンおよびアドレナリン作動薬から成る群より選択された薬剤を含む培地中で培養する工程。
患者への治療剤の投与方法としては、例えば、得られた肝細胞をシート化して、患者の肝臓に貼付する方法、得られた肝細胞を生理食塩水等に懸濁させ、患者の肝臓に直接移植する方法、マトリゲル等から構成されたスキャフォールド上で三次元培養し、得られた肝細胞塊を移植する方法などが挙げられる。
ヒトiPS細胞(201B6、648A1、648B1、604A1、604B1、604A3、606A1、606B1および610B1)は、京都大学の山中教授より受領し、従来の方法で培養した(Takahashi K, et al. Cell. 131:861-72)。201B6は、Cell. 131:861-872, 2007に記載の方法で作製し、648A1、648B1、604A1、604B1、604A3、606A1、606B1および610B1は、Okita K, et al. Stem Cells. 31:458-466, 2013に記載の方法で製造された。本パラグラフに記載の文献は引用により本願の一部を構成する。
<Stage 1>
201B6は、Accutase(Innovative Cell Technologies、AT-104)を加えて単一細胞へ分離し、2×105cells/wellでMatrigel basement Membrance Matrix Growth Factor Reduced(BD)をコートした24 wellプレートへ播種し、1μM CHIR99021(Stemgent)、100 ng/ml ActivinA(R&D systems)、2% B27(Invitrogen)および0.5% PenStrep(Invitrogen)を添加したRPMI1640中で1日間培養した(Stage 1-1)(day1)。
Stage 1の後、培地を除去し、1%DMSO、20%KSR(Invitrogen)、1 mM L-グルタミン酸、1% NEAA(Invitrogen)、0.1 mM βメルカプトエタノールおよび0.5% PenStrepを添加したKnockoutTM DMEM(Invitrogen)へ培地を交換し、7日間培養し、得られた細胞をAFPの抗体で染色し、AFP陽性細胞の含有率を調べたところ、AFP陽性細胞(肝芽細胞)を40%から60%含む細胞群を得た(day16)。
Stage 2の後、培地を除去し、150 nMまたは1 μMのプレグネノロン(東京化成)またはメトキサミン塩酸塩(Sigma)を添加したHepatocyte Culture Medium (HCM SingleQuots Kit)(LONZA)へ培地を交換し、4日間、8日間または12日間培養した。このとき、対照として、1% DMSO、または20 ng/ml HGFおよび20 ng/ml オンコスタチンM(HGF+OsM)を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)にて同様に細胞を培養した。
Stage 3にて得られた細胞群を抗アルブミン抗体(BETHYL, A80-229A)を用いて免疫染色した(図2)。また、アルブミンのmRNAの発現量を定量PCRを用いて測定した(図3)。使用したプライマーは、Foward:5'-CGCTATTAGTTCGTTACACCA-3'(配列番号:1)およびReverse:5'-TTTACAACATTTGCTGCCCA-3' (配列番号:2)である。この結果、Stage 3において8日間培養した場合に最も効率よくアルブミンを発現する細胞が得られることが確認された。アルブミン産生細胞の製造効率は、核染色(DAPI)陽性の細胞数に対してアルブミン陽性細胞の割合をアルブミン陽性細胞率として、In cell analyzerで解析した(図2B)。
Stage 1のNaBを含有する培地中で培養する期間(Stage 1-2)を1日とした場合でも最終的に得られるアルブミン陽性細胞の収率として同様の効果が得られること、さらにNaBを除いた培地で培養する期間(Stage 1-3)を2日とした場合でも同様の効果が得られることが確認された。従って、Stage 1は、4日間から9日間の範囲において行うことで肝細胞の誘導が可能であることが確認された。
メトキサミン塩酸塩の代替物として、類似化合物(アドレナリン受容体作動薬、特にα受容体刺激作用を有する薬剤)の効果を検討した。上述のStage 3において、1 μMのエチレフリン、フェニレフリンまたはメトキサミン塩酸塩を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)を用いて8日間培養し、アルブミン陽性細胞率およびアルブミンのmRNAの発現量を前記と同様の方法で測定したところ、エチレフリンおよびフェニレフリンは、共にメトキサミン塩酸塩と同等の効果を示すことが確認された(図4および5)。さらに、得られた細胞集団から培養液中へ分泌されたアルブミン量をELISA法を用いて測定したところ、Stage 3においてエチレフリン、フェニレフリンおよびメトキサミン塩酸塩のいずれかを培養液へ添加することで、アルブミンの分泌能を有する細胞が誘導できることが示された(図6)。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間とした。Stage 2にて得られた細胞集団を20 ng/ml HGFおよび20 ng/ml OncostatinオンコスタチンM(HGF+OsM)を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団、Stage 2にて得られた細胞集団を1 μMのメトキサミン塩酸塩を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団、Panc1(ヒト膵癌由来細胞株:理研バイオリソースセンター)およびHepG2(ヒト肝癌由来細胞株)を用いた。各細胞集団を、100 μMのオメプラゾール(Sigma)を添加または添加しないHepatocyte Culture Medium(HCM SingleQuots Kit)中で2日間培養した後、CYP1A1のmRNAの発現量を定量PCRにて測定した。使用したプライマーは、Foward:5'-CCACCAAGAACTGCTTAGCC-3'(配列番号:3)およびReverse:5'-CAGCTCCAAAGAGGTCCAAG-3' (配列番号:4)である。CYP1A1のmRNA量をβアクチンのmRNA量と比較した値をCYP1A1の相対発現量として算出し、各細胞での相対発現量を図7に示した。その結果、HepG2細胞、並びにStage 2にて得られた細胞集団をHGF+OsMまたはメトキサミン塩酸塩を添加して誘導した細胞集団では、オメプラゾールの添加によりCYP1A1の発現が誘導されることが確認された。メトキサミンを用いて多能性幹細胞より誘導した肝細胞は、オメプラゾールによるCYP1A1の発現誘導を引き起こすことから、医薬品の薬物相互作用の検討に用いることができることが示唆された。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間、およびStage 3にて20 ng/ml HGFおよび20 ng/ml オンコスタチンM(HGF+OsM)を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた肝細胞、Stage 3にて1 μMのメトキサミン塩酸塩を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団またはStage 3にてDMSOを添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団およびHepG2へ、1 mg/ml ICG(インドシアニングリーン)(Sigma)を添加して、1時間培養した後、各細胞におけるICGの取り込みを観察したところ、HepG2、およびHGF+OsMまたはメトキサミン塩酸塩を添加して誘導した細胞では、細胞内へのICGの取り込みが確認された(図8上図)。従って、同細胞は、薬物代謝機能を有していることが示唆された。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間、およびStage 3にて20 ng/ml HGFおよび20 ng/ml オンコスタチンM(HGF+OsM)を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団、Stage 3にて1 μMのメトキサミン塩酸塩を添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団、Stage 3にて1 μMのプレグネノロンを添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団、またはStage 3にてDMSOを添加したHepatocyte Culture Medium(HCM SingleQuots Kit)中で8日間培養して得られた細胞集団を、CYP3A4抗体(Proteintech Group)を用いて免疫染色(図9A)を行った。この結果、DMSO以外を用いて誘導された細胞集団では、CYP3A4を発現する細胞が確認された。同様に、これらの細胞集団におけるCYP3A4のmRNAの発現量を定量PCRで測定したところ、DMSO以外を用いて誘導された肝細胞では、CYP3A4が強く発現していることが確認された(図9B)。使用したプライマーは、Foward:5'-AAGACCCCTTTGTGGAAAAC-3'(配列番号:5)およびReverse:5'-CGAGGCGACTTTCTTTCATC-3' (配列番号:6)である。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を8日間の条件において、Stage 3で添加するメトキサミン塩酸塩の濃度をそれぞれ、0 M、1 fM、1 pM、1 nM、1 μM、50 μM、100 μMで作製された細胞集団のアルブミンのmRNAの発現量を定量PCRを用いて測定した(図10)。その結果、1 fM以上の濃度のメトキサミン塩酸塩を用いた場合、アルブミンを発現する細胞が得られることが確認された。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を8日間の条件において、1 μMのメトキサミン塩酸塩を用いて得られた細胞における各薬物代謝酵素(CYP1A1、CYP3A5、CYP3A7、CYP3A4、CYP2C19)およびチロシンアミノトランスフェラーゼ(TAT)の発現量を定量PCRを用いて測定した(図11)。用いたprimerの配列は、以下表1に示す。その結果、本発明の方法を用いて作製された細胞集団は、HGF+OsMを用いた場合と同等の薬物代謝酵素等を発現していることが確認された。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を6日間の条件を用いて製造された細胞を用いて、各薬物代謝酵素の薬物による影響を検討した。詳細には、Stage 3の期間を6日間目の細胞へ40μM リファンピリシンを添加し、2日後にCYP3A4のmRNA発現量を定量PCRを用いて測定した。同様に、40μMオメプラゾールを添加し、CYP1A2のmRNA発現量を測定し、100μMフェノバルビタールを添加し、CYP2B6のmRNA発現量を測定した。その結果、本発明の方法により得られた細胞において各薬物代謝酵素が対応する薬物と接触させることで発現誘導が起きることが確認された(図12)。従って、当該細胞を用いて薬物相互作用を評価することが可能であることが示唆された。
上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を8日間の条件を用いて、648A1、648B1、604A1、604B1、604A3、606A1、606B1および610B1から誘導し、分泌されるアルブミン量を測定したところ、いずれのiPS細胞株からも少なくとも一日あたり2μg/mlを分泌する機能を有する細胞が製造できることが確認された(図13)。
ヒトiPS細胞(201B6)のアルブミン遺伝子座の開始コドンの下流へGFP-polyAを連結するよう、常法に従って、相同組換えを行った。当該相同組換えにより、アルブミンの発現と連携してGFPを発現する細胞株を作製した(以下、アルブミンレポーターiPS細胞という)。アルブミンレポーターiPS細胞を、上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を8日間の条件を用いて、肝細胞を誘導し、GFP陽性細胞の含有率を測定した(図14)。その結果、誘導後の細胞群において、アルブミン発現細胞は、39.9%含有されていることが確認された。
メトキサミンによる肝細胞誘導効率を上昇させるメカニズムを検討するため、各アドレナリン受容体の遮断薬を添加し、アルブミンの分泌量を検討した。上述した方法のStage 1の期間を6日間(Stage 1-1を1日間、Stage 1-2を2日間およびStage 1-3を3日間)、Stage 2の期間を6日間およびStage 3の期間を8日間の条件にて、Stage 3において、メトキサミンおよび各アドレナリン受容体の遮断薬を添加した。用いたアドレナリン受容体の遮断薬は、α1遮断薬として10μM ドキザソシン(Sigma)を使用し、α2遮断薬として10μM ヨヒンビン(Sigma)を使用し、β1遮断薬として10μM メトプロロール(Sigma)を使用し、β2遮断薬として10μM ブトキサミン(Sigma)を使用した。その結果、メトキサミンに対して、ドキザソシンを添加した場合において、アルブミン分泌量が低下した(図15)。従って、iPS細胞から肝細胞を誘導するにあたり、少なくともα1作動することでその誘導効率を上昇させることができることが示唆された。
ヒトiPS細胞から分化誘導した肝細胞について、RNA sequencing解析により遺伝子発現プロファイルの比較を行った。図16は主成分分析(PCA)を、図17は肝臓マーカー遺伝子発現のヒートマップを示す。これらの結果より、既存の増殖因子(HGF&OsM)で分化誘導した肝細胞とメトキサミンで分化誘導した肝細胞は、遺伝子発現プロファイルがほぼ同じであることが確認された。
<Stage 1>
マウスES細胞(D3株)は、0.25%トリプシン-EDTA(GIBCO)を加えて単一細胞へ分離した。フィーダー細胞を除くため、ゼラチンコートしたプレートへ播種して10〜20分後にES細胞と培養液とを含む上清を回収し、次いで2.0×104-8.0×104cells/wellとなるようMatrigel basement Membrance Matrix Growth Factor Reduced(BD)をコートした24 wellプレートへ播種した。1μM CHIR99021(Axon Medchem)、100 ng/ml Activin A(R&D systems)、2% B27(Invitrogen)および0.5% PenStrep(Invitrogen)を添加したRPMI1640中で1日間培養した(Stage 1-1)(day1)。
Stage 1の後、培地を除去し、1%DMSO、20%KSR(Invitrogen)、1 mM L-グルタミン酸、1% NEAA(Invitrogen)、0.1 mM βメルカプトエタノール、0.5 mM NaB(Sigma)および0.5% PenStrepを添加したKnockoutTMDMEM(Invitrogen)へ培地を交換し、6日間培養して細胞を得た。得られた細胞をAFPの抗体で染色し、AFP陽性細胞の含有率を調べたところ、AFP陽性細胞(肝芽細胞)を40%から60%含む細胞群を得た(day12)。
Stage 2の後、培地を除去し、1 μMのプレグネノロン(東京化成)、メトキサミン塩酸塩(Sigma)、エチレフリンまたはフェニレフリンを添加したHepatocyte Culture Medium (HCM SingleQuots Kit)(LONZA)へ培地を交換し、8日間培養した。このとき、対照として、1% DMSO、または20 ng/ml HGFおよび20 ng/mlオンコスタチンMを添加したHepatocyte Culture Medium(HCM SingleQuots Kit)にて同様に細胞を培養した。
未分化細胞(ES細胞)、Day 6の細胞(Stage 1終了)、Day 12の細胞(Stage 2終了時)、Stage 3でHGF+オンコスタチンM(GFs)、メトキサミン、エチレフリン、フェニレフリン、プレグネノロンを添加した培地にて8日間培養した細胞(Day20)、および成熟マウス肝細胞のそれぞれにおけるチロシンアミノトランスフェラーゼ(Tat)、薬物代謝酵素(Cyp3a11およびCyp3a13)、およびβアクチンの発現を測定した。結果を図19に示す。
Stage 3においてプレグネノロン、アドレナリン作動薬であるメトキサミン、エチレフリン、フェニレフリンを添加して誘導された細胞集団は、HGF+OsMを用いた場合と同様の薬物代謝酵素等を発現していることが確認された。
Claims (20)
- 肝芽細胞をプレグネノロンまたはアドレナリン作動薬を含む培地中で培養する工程を含み、当該肝芽細胞がAFP陽性である、肝細胞を製造する方法。
- 前記アドレナリン作動薬が、アドレナリン、ノルアドレナリン、エチレフリン、ナファゾリン、フェニレフリン、メトキサミンおよびミドドリンから成る群より選択される試薬である、請求項1に記載の方法。
- 前記アドレナリン作動薬が、エチレフリン、フェニレフリンおよびメトキサミンから選択される試薬である、請求項2に記載の方法。
- 前記肝芽細胞が、内胚葉細胞をDMSOを含む培地中で培養する工程を含む方法で製造された細胞であり、当該内胚葉細胞がSOX17陽性細胞である、請求項1から3のいずれか1項に記載の方法。
- 前記内胚葉細胞が、多能性幹細胞を、Activin AおよびGSK-3β阻害剤を含む培地中で培養する工程を含む方法で製造された細胞である、請求項4に記載の方法。
- 前記多能性幹細胞から内胚葉細胞を製造する工程が、Activin A、GSK-3β阻害剤およびHDAC阻害剤を含む培地で培養する工程をさらに含む、請求項5に記載の方法。
- 前記GSK3β阻害剤が、CHIR99021であり、前記HDAC阻害剤が、NaB(酪酸ナトリウム)である、請求項6に記載の方法。
- 前記多能性幹細胞がiPS細胞である、請求項5から7のいずれか1項に記載の方法。
- 前記iPS細胞がヒトiPS細胞である、請求項8に記載の方法。

- 多能性幹細胞から肝細胞を製造する方法であって、以下の工程(i)から(iii):

(ii)工程(i)で得られた細胞をDMSOを含む培地中で培養する工程、および
(iii)工程(ii)で得られた細胞をプレグネノロンおよびアドレナリン作動薬から成る群より選択された薬剤を含む培地中で培養する工程、
