Abstract
Description
本発明は、ヒトパピローマウイルス(HPV)16型のL1タンパク質の特定のセグメントを、第二の型のHPV(例えばHPV35)のL1タンパク質の対応するセグメントで置き換えた後に、得られた変異HPV16 L1タンパク質が、体内でHPV16及び第二の型のHPV(例えばHPV35)に対する高力価の中和抗体の産生を、HPV16 VLPと第二の型のHPVのVLPの混合物と同等の防御効果でもって誘導することができ;さらに、HPV16に対するその防御効果が、HPV16 VLP単独と同等であり、第二の型のHPV(例えば、HPV35)に対するその防御効果が第二の型のHPV単独と同等であるという、発明者の予期せぬ発見に少なくとも部分的に基づいている。
(1)4、6、8、10、20、30、又は40個のアミノ酸などの、4〜50個のアミノ酸のN末端切断;
(2)野生型HPV L1タンパク質の位置292〜316のアミノ酸残基の、第二の型の野生型HPVのL1タンパク質の対応する位置でのアミノ酸残基による置換;
任意選択で、変異HPV16 L1タンパク質はさらに以下の変異を有する:
(3)野生型HPV16 L1タンパク質の位置76〜87のアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置でのアミノ酸残基による置換
(4)野生型HPV16 L1タンパク質の位置152〜167でのアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置でのアミノ酸残基による置換;又は
(5)野生型HPV16 L1タンパク質の位置202〜207のアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置でのアミノ酸残基による置換;
及び、変異体は変異HPV16 L1タンパク質とは、1つ又はいくつか(例えば、1、2、3、4、5、6、7、8、又は9個)のアミノ酸の置換(好ましくは保存的置換)、付加又は欠失によってのみ異なり、変異HPV16 L1タンパク質の機能を維持する、すなわち、少なくとも2つの型のHPV(例えば、HPV16及びHPV35、又はHPV16、HPV35及びHPV31)に対して中和抗体を誘導する能力を維持する。
本発明において、本明細書で使用される科学用語及び技術用語は、特に明記しない限り、当業者によって一般に理解される意味を有する。また、本明細書で使用される細胞培養、分子遺伝学、核酸化学、及び免疫学の実験室手順は、対応分野で広く使用されるすべてのルーチンのステップである。また、本発明のより良い理解のために、関連用語の定義及び説明を以下に提供する。
研究によれば、HPV16及び他の型のHPV(例えばHPV35及びHPV31など)との間の特定の交差防御があるものの、このような交差防御は、それ自身の型のVLPの防御効力の、通常百分の一未満、さらには千分の一未満の乏しい効力を有する。したがって、HPV16ワクチンを接種した被験体は他の型のHPV(例えばHPV35及びHPV31など)に感染する高いリスクが依然としてあることが示されている。
本発明に関与する配列のいくつかを以下の表1に示す。
以下の実施例を参照して本発明を説明するが、これらは本発明を説明することを意図しており、限定するものではない。
発現ベクターの構築
HPV16 L1タンパク質から誘導されるセグメント3又はセグメント5を含む変異HPV35 L1タンパク質をコードする発現ベクターをPCRによって多部位突然変異誘発のために構築した。ここで使用された初期テンプレートはpTO‐T7‐HPV16L1N30Cプラスミドであった(これは30個のアミノ酸のN末端切断を有するHPV16 L1タンパク質をコードした(このタンパク質はHPV16N30と名付けた);表2では16L1N30と略す)。各PCR反応に使用されたテンプレートとプライマーを表2に示し、PCR反応の増幅条件を以下のように設定した:94℃で10分間の変性;25サイクルの(94℃で50秒間の変性、一定時間の間、特定の温度でのアニーリング、72℃で7分30秒間の伸長);最終的に72℃で7分間の伸長。用いたPCRプライマーの配列を表3に示す。
組換えプラスミドpTO‐T7‐H16N30‐35T1、pTO‐T7‐H16N30‐35T2、pTO‐T7‐H16N30‐35T3、pTO‐T7‐H16N30‐35T4、pTO‐T7‐H16N30‐35T5、pTO‐T7‐H16N30‐35T4‐31S1、pTO‐T7‐H16N30‐35T4‐31S2、pTO‐T7‐H16N30‐35T4‐31S3、pTO‐T7‐H16N30‐35T4‐31S5を担持する大腸菌の細菌液を、−70℃の冷蔵庫から取り出し、100mlのカナマイシン含有LB液体培地に別々に接種し、37℃で約8時間200rpmで培養し、次に、カナマイシン含有LB培地の500mlに移して接種し(細菌液1mlを接種した)、継続的に培養した。細菌濃度が約0.6のOD600に達したときに、培養温度を25℃に下げ、IPTGの500μLをそれぞれの培養フラスコに添加し、培養を8時間続けた。培養終期に、細菌細胞を遠心分離により回収した。H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5タンパク質を別個に発現する細菌細胞が得られた。
得られた細胞を、細菌細胞1gと溶解物10mL(20mM Tris緩衝液、pH7.2、300mM NaCl)の比で再懸濁した。細胞を30分間の超超音波処理により破壊した。破壊された細胞を含有する溶解物を、15分間13500rpm(30000g)で遠心分離し、上清(すなわち、破壊された細菌細胞の上清)を取り出した。
機器システム:AKTAエクスプローラー100分取液体クロマトグラフィーシステム、GE Healthcare(現、Amershan Pharmacia)製。
(1)破壊された細菌細胞の上清のカチオン交換精製をSP Sepharose 4 Fast Flowを用いて行った:サンプルをカラムにロードし、次いで400mMのNaClを含む緩衝液を用いて望ましくないタンパク質を溶出し、800mM NaClを含む緩衝液を使用して標的タンパク質を溶出し、800mM NaClを含む緩衝液により溶出された画分を収集した。
精製したタンパク質H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5を、上記の方法により電気泳動に供した。電気泳動後、HPV L1タンパク質に対する広域スペクトル抗体4B3を使用してウェスタンブロット検出を実施し、結果を図2に示した。結果は、H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5は、広域スペクトル抗体4B3により特異的に認識できることを示した。
HPVウイルス様粒子のアセンブリ
所与の体積(約2ml)のタンパク質H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、又はH16N30‐35T4‐31S5を、順次、(1)2Lの保存緩衝液(20mMリン酸ナトリウム緩衝液、pH6.5、0.5M NaCl)、(2)2L復元緩衝液(50mMリン酸ナトリウム緩衝液、pH6.0、2mM CaCl2、2mM MgCl2、0.5M NaCl);及び(3)20mMリン酸ナトリウム緩衝液、pH7.0、0.5M NaClに透析した。透析は、3つの緩衝液のそれぞれにおいて12時間行った。
透析されたサンプルを、TSK Gel PW5000xl 7.8x300mmの分析カラムを使用して、Agilent、USAからの1120 Compact LC高速液体クロマトグラフィーシステムを使用した分子ふるいクロマトグラフィーで分析した。分析の結果を図3A〜3Lに示す。結果は、タンパク質H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5を含有するサンプルについて、最初のタンパク質のピークが約12分で現れ、これはHPV16N30 VLP、HPV35 VLP及びHPV31 VLPと同等であることを示した。これは上記で調製したタンパク質がVLPに組織化されたことを示した。
沈降速度分析のために使用された機器は、光学検査システム及びAn‐50Ti及びAn‐60Tiローターを備えたBeckman XL‐A分析型の超遠心であった。沈降速度法を使用して、HPV16N30 VLP、HPV35 VLP、HPV31 VLP、H16N30‐35T1 VLP、H16N30‐35T2 VLP、H16N30‐35T3 VLP、H16N30‐35T4 VLP、H16N30‐35T5 VLP、H16N30‐35T4‐31S1 VLP、H16N30‐35T4‐31S2 VLP、H16N30‐35T4‐31S3 VLP、H16N30‐35T4‐31S5 VLPの沈降係数を分析した。結果を4A〜4Lに示した。結果は、H16N30‐35T1 VLP、H16N30‐35T2 VLP、H16N30‐35T3 VLP、H16N30‐35T4 VLP、H16N30‐35T5 VLP、H16N30‐35T4‐31S1 VLP、H16N30‐35T4‐31S2 VLP、H16N30‐35T4‐31S3 VLP、H16N30‐35T4‐31S5 VLPの沈降係数が、HPV16N30 VLP、HPV35 VLP及びHPV31 VLPと同様であることを示した。これは、H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5が大きさ及び形態の点で野生型VLPと類似のウイルス様粒子に組織化されることを示した。
VLPを含むサンプルの100μLを透過型電子顕微鏡観察のために採取した。使用した機器は、JEOL製の100kV透過型電子顕微鏡で、倍率は100,000倍であった。簡単に言えば、13.5μLのサンプルを採取し、pH7.0の2%リンタングステン酸でネガティブ染色し、カーボンコーティングされた銅メッシュに固定し、次いで透過型電子顕微鏡で観察した。観察の結果は、図5A〜5Lに示した。結果は、H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5がすべてウイルス様粒子に組織化されることを示した。さらに、結果はまた、これらの変異タンパク質によって形成された粒子が約30ナノメートルの半径を有し、サイズが均一であることを示した。これは、これらの変異タンパク質が、HPV16、HPV35及びHPV31のL1タンパク質に類似し、均一なサイズのVLPを形成することができることを示した。
H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5によって形成されたVLPの熱安定性は、GE Corporation(旧MicroCal Corporation)から購入した示差温度熱量計VPキャピラリーDSCを用いて評価した。ここで、タンパク質の保存緩衝液を対照として使用し、タンパク質の各々を1.5℃/分の加熱速度で10℃〜90℃の範囲内でスキャンした。テスト結果を図6A〜6Lに示した。結果は、タンパク質のそれぞれにより形成されたVLPが極めて高い熱安定性を有することを示した。
H16N30‐35T1、H16N30‐35T2、H16N30‐35T3、H16N30‐35T4、H16N30‐35T5、H16N30‐35T4‐31S1、H16N30‐35T4‐31S2、H16N30‐35T4‐31S3、H16N30‐35T4‐31S5によって形成されたVLPの免疫防御特性をマウスを使用して評価した。免疫化に使用した動物は、5〜6週齢のBalB/cマウス(通常グレード)(Shanghai SLAC Laboratory Animal Co.、Ltd.から購入)であった。
H16N30‐35T4 VLPのED50
6週齢のBalB/c雌マウス(8匹のマウス)を単回腹腔内注射によりアルミニウムアジュバントで免疫化した。ここで、実験群にはH16N30‐35T4 VLP(免疫化用量は、0.300μg、0.100μg、0.033μg、0.011μg、及び0.004μgとした)を投与した、対照群はHPV16N30 VLP単独とHPV35 VLP単独(免疫化用量は、0.300μg、0.100μg、0.033μg、0.011μg、0.004μgとした)、又は混合HPV16/HPV35 VLP(各VLPの免疫化用量は、0.300μg、0.100μg、0.033μg、0.011μg、0.004μgとした)を投与した;免疫化量は1mLとした。免疫化後5週で、静脈血を眼球から回収し、血液中のHPV抗体を検出した。血清変換(seroconversion)(すなわち、マウスにおける抗体の生成の誘導)のED50を、Reed‐Muench法(Reed LJ MH.A simple method of estimating fifty percent endpoints.(50パーセントのエンドポイントを推定する簡単な方法。)Am J Hyg.1938;27:493‐7)によって、各サンプルについて計算した。結果を表6〜9に示した。
6週齢のBalB/c雌マウス(8匹のマウス)を、単回腹腔内注射によりアルミニウムアジュバントで免疫化した。実験群はH16N30‐35T4‐31S3 VL(免疫化用量は0.300μg、0.100μg、0.033μg、0.011μg、0.004μgとした)を投与し;対照群はHPV16N30 VLP単独、HPV35 VLP単独、HPV31 VLP単独(免疫化用量は0.300μg、0.100μg、0.033μg、0.011μg、0.004μgとした)、及び混合HPV16/HPV35/HPV31 VLP(各VLPの免疫化用量は、0.300μg、0.100μg、0.033μg、0.011μg、0.004μgとした)を投与し;免疫体積は1mLとした。免疫化後5週で、静脈血を眼球から回収し、血液中のHPV抗体を検出した。血清変換の誘導(すなわち、マウスにおける抗体の生成の誘導)のためのED50は、各サンプルについて、Reed‐Muench法(Reed LJ MH.A simple method of estimating fifty percent endpoints.(50パーセントのエンドポイントを推定する簡単な方法。)Am J Hyg.1938;27:493‐7)によって計算した。結果を表10〜14に示した。
この実験では、免疫化プロトコルを表15に示した。すべてのマウス(6週齢のBalB/c雌マウス)を3つの群に分けた:10μg用量群(免疫化用量はアルミニウムアジュバントを使用して10μgであった)、1μg用量群(免疫化用量はアルミニウムアジュバントを使用して1μgであった)、及び0.1μg用量群(免疫化用量はアルミニウムアジュバントを使用して0.1μgであった)。各群を4つのサブグループに細分し、対照サブグループ1及び2は、それぞれ、HPV16N30 VLP単独及び単独HPV35 VLP単独で免疫化し、対照サブグループ3は混合HPV16/HPV35 VLPで免疫化し、実験サブグループはH16N30‐35T4 VLPで免疫化した。
この実験では、免疫化プロトコルを表16に示した。すべてのマウス(6週齢のBalB/c雌マウス)を3つの群に分けた:10μg用量群(免疫化用量はアルミニウムアジュバントを使用して10μgであった)、1μg用量群(免疫化用量はアルミニウムアジュバントを使用して1μgであった)、及び0.1μg用量群(免疫化用量はアルミニウムアジュバントを使用して0.1μgであった)。各群は6つのサブグループに細分し、対照サブグループ1、2、及び3は、それぞれ、HPV16N30 VLP単独、HPV35 VLP単独及びHPV31 VLP単独を用いて免疫化し、対照サブグループ4は、混合HPV16/HPV35/HPV31 VLPで免疫化し、実験サブグループはH16N30‐35T4‐31S3 VLP単独で免疫化した。

Claims (11)
- 変異HPV16 L1タンパク質又はその変異体であって、変異HPV16 L1タンパク質が野生型HPV16 L1タンパクと比較して以下の変異を有する変異HPV16 L1タンパク質又はその変異体:
(1)4〜50個の、例えば4、6、8、10、20、30、又は40個のアミノ酸のN末端切断;及び
(2)野生型HPV16 L1タンパク質の位置292〜316のアミノ酸残基の、第二の型の野生型HPVのL1タンパク質の対応する位置のアミノ酸残基による置換;
任意選択で、変異HPV16 L1タンパク質にはさらに以下の変異がある:
(3)野生型HPV16 L1タンパク質の位置76〜87のアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置のアミノ酸残基による置換;
(4)野生型HPV16 L1タンパク質の位置152〜167のアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置のアミノ酸残基による置換;又は
(5)野生型HPV16 L1タンパク質の位置202〜207のアミノ酸残基の、第三の型の野生型HPVのL1タンパク質の対応する位置のアミノ酸残基による置換;
及び、変異体は変異HPV16 L1タンパク質とは、1つ又はいくつか(例えば、1、2、3、4、5、6、7、8、又は9個)のアミノ酸の置換(好ましくは保存的置換)、付加又は欠失によってのみ異なり、変異HPV16 L1タンパク質の機能を維持する、すなわち、少なくとも2つの型のHPV(例えば、HPV16及びHPV35、又はHPV16、HPV35及びHPV31)に対して中和抗体を誘導する能力を維持する;
好ましくは、変異HPV16 L1タンパク質は、野生型HPV16 L1タンパク質と比較して、30又は40個のアミノ酸のN末端切断を有する;
好ましくは、変異HPV16 L1タンパク質は、野生型HPV16 L1タンパク質と比較して、30個のアミノ酸のN末端切断を有する;
好ましくは、第二の型の野生型HPVはHPV35である;好ましくは、(2)に記載される対応する位置のアミノ酸残基は野生型HPV35 L1タンパク質の位置266〜288のアミノ酸残基である;
好ましくは、第三の型の野生型HPVがHPV31である;好ましくは、(3)に記載される対応する位置のアミノ酸残基が野生型HPV31 L1タンパク質の位置50〜62のアミノ酸残基である;好ましくは、(4)に記載される対応する位置のアミノ酸残基が野生型HPV31 L1タンパク質の位置127〜142のアミノ酸残基である;好ましくは、(5)に記載される対応する位置のアミノ酸残基が野生型HPV31 L1タンパク質の位置177〜182のアミノ酸残基である;
好ましくは、野生型HPV16 L1タンパク質は配列番号1に記載のアミノ酸配列を有する;
好ましくは、野生型HPV35 L1タンパク質は配列番号2に記載のアミノ酸配列を有する;
好ましくは、野生型HPV31 L1タンパク質は配列番号3に記載のアミノ酸配列を有する;
好ましくは、変異HPV16 L1タンパク質は配列番号7、9、10、11からなる群から選択されるアミノ酸配列を有する。 - 請求項1に記載の変異HPV16 L1タンパク質又はその変異体をコードする単離された核酸。
- 請求項2に記載の単離された核酸を含むベクター。
- 請求項2に記載の単離された核酸及び/又は請求項3に記載のベクターを含む宿主細胞。
- 請求項1に記載の変異HPV16 L1タンパク質又はその変異体を含む又はからなる、HPVウイルス様粒子。
- 請求項1に記載の変異HPV16 L1タンパク質又はその変異体、又は請求項2に記載の単離された核酸、又は請求項3に記載のベクター、又は請求項4に記載の宿主細胞、又は請求項5に記載のHPVウイルス様粒子を含む組成物。
- 請求項5に記載のHPVウイルス様粒子、及び必要に応じてさらに薬学的に許容される担体及び/又は賦形剤を含む医薬組成物又はワクチンであって、
好ましくは、HPVウイルス様粒子は、HPV感染又はHPV感染によって引き起こされる疾患を予防するのに有効な量で存在する;
好ましくは、HPV感染は、一つ以上のHPV型による感染(例えば、HPV16感染、HPV35感染、及び/又はHPV31感染)である;
好ましくは、HPV感染によって引き起こされる疾患は、子宮頸癌及び尖圭コンジローマからなる群から選択される、上記医薬組成物又はワクチン。 - 請求項1に記載の変異HPV16 L1タンパク質又はその変異体を調製する方法であって、宿主細胞内で変異HPV16 L1タンパク質又はその変異体を発現させ、次いで宿主細胞の培養物から変異HPV16 L1タンパク質又はその変異体を回収することを含み;
好ましくは、宿主細胞は大腸菌であり;
好ましくは、この方法は、変異HPV発現16 L1タンパク質又はその変異体を大腸菌で発現させ、次いで大腸菌の溶解物上清を精製することによって変異HPV16 L1タンパク質又はその変異体を得るステップを含み;好ましくは、変異HPV16 L1タンパク質又はその変異体が大腸菌の溶解物上清からクロマトグラフィー(例えば、カチオン交換クロマトグラフィー、ヒドロキシアパタイトクロマトグラフィー、及び/又は疎水性相互作用クロマトグラフィー)により回収される、上記方法。 - 請求項5に記載のHPVウイルス様粒子を薬学的に許容される担体及び/又は賦形剤と組み合わせることを含む、ワクチンの調製方法。
