Abstract
Description
前記患者は、アルツハイマーを患っている人間、猿などの霊長類、ラット、マウスなどのげっ歯類を含む哺乳動物または前記哺乳動物から分離された細胞(脳細胞)または組織(脳組織)またはこれらの培養物の中から選択でき、例えば、アルツハイマー病を患っている人間またはこれから分離された脳細胞、脳組織またはこれらの培養物の中から選択できる。
ゲノム上の特定標的配列を認識するドメインである植物病原性遺伝子に由来したTALエフェクター(transcription activator−like effector)ドメインと切断ドメインが融合されたTALEN(transcription activator−like effector nuclease);
ジンクフィンガーヌクレアーゼ(zinc−finger nuclease);
メガヌクレアーゼ(meganuclease);
微生物免疫機構であるCRISPRに由来したRGEN(RNA−guided engineered nuclease;例えば、Casタンパク質(例えば、Cas9など)、Cpf1、など);
AGOホモログ(Ago homolog、DNA−guided endonuclease)
などからなる群より選択された1種以上であってもよいが、これに制限されるのではない。
(1)RNAガイドヌクレアーゼ(またはそのコーディングDNA、または前記コーディングDNAを含む組換えベクター)、および
(2)標的遺伝子(例えば、AAVS1のようなセーフハーバー(safe harbor)位置)の標的部位(例えば、AAVS1のようなセーフハーバー(safe harbor)遺伝子内の連続する15〜30、17〜23、または18〜22個のヌクレオチド長さの核酸部位)と混成化可能な(または相補的核酸配列を有する)ガイドRNAまたはそのコーディングDNA(またはコーディングDNAを含む組換えベクター)

標的配列と混成化可能な部位(標的化配列)を含むCRISPR RNA(crRNA);
Casタンパク質、Cpf1などのようなヌクレアーゼと相互作用する部位を含むtrans−activating crRNA(tracrRNA);および
前記crRNAおよびtracrRNAの主要部位(例えば、標的化配列を含むcrRNA部位およびヌクレアーゼと相互作用するtracrRNAの部位)が融合された形態の単一ガイドRNA(single guide RNA;sgRNA)

具体的に、CRISPR RNA(crRNA)およびtrans−activating crRNA(tracrRNA)を含む二重RNA(dual RNA)、またはcrRNAおよびtracrRNAの主要部位を含む単一ガイドRNA(sgRNA)であってもよい。

Ncas9は標的化配列、即ち、標的遺伝子(target gene)の標的部位(target site)の配列によって決定される部位(標的部位の標的配列と混成化可能)であり、lは前記標的化配列に含まれているヌクレオチド数を示すものであって、15〜30、17〜23、または18〜22の整数、例えば20であってもよく、

Ncpf1は遺伝子標的部位と混成化可能なヌクレオチド配列を含むターゲッティング配列であって標的遺伝子の標的配列によって決定され、qは含まれているヌクレオチド数を示すものであって、15〜30の整数であってもよい。前記標的遺伝子の標的配列(crRNAと混成化する配列)はPAM配列(5’−TTN−3’または5’−TTTN−3’;Nは任意のヌクレオチドであって、A、T、G、またはCの塩基を有するヌクレオチドである)の3’方向に隣接して位置する(例えば、連続する)15〜30個の標的遺伝子の標的部位のヌクレオチド配列である。
1.MSCs、HMO6細胞、およびSH−SY5Y細胞の培養
人間臍帯由来間葉系幹細胞(MSCs)は、CEFObio(Seoul、Korea)で購入した。sRAGEを発現する間葉系幹細胞(sRAGE−MSCs)は、MSC(CEFObio)にsRAGE(cat.RD172116100、Biovendor;配列番号6)を含むドナーベクター(図1a参照)を導入させて準備した(参考例2参照)。paracrine効果検証(in vitro)に使用するために、前記MSCsとsRAGE−MSCsをそれぞれMSC培地(MSC med;DMEM、Gibco(登録商標)Life Technologies Corp.)またはsRAGE−MSC培地(sRAGE−MSC med;sRAGE分泌MSCsの培養時使用、DMEM、Gibco(登録商標)Life Technologies Corp.)で2日間培養した。タンパク質分解を防止するために、前記二つの培地全てにプロティナーゼ阻害剤(proteinase inhibitor)およびホスファターゼ阻害剤(phosphatase inhibitor)(TAKARA、Tokyo、Japan)を添加した。MSC培地およびsRAGE−MSC培地をそれぞれのcentrifugal filter unit(Millipore、Merck Millipore、Germany)に収集し、4℃で50分間3,220xgで遠心分離した。このように得られた濃縮培養液は使用時まで−80℃で保管した。
sRAGEを分泌するMSC(sRAGE MSCs)を製作するために、AAVS1(adeno−associated virus integration site 1)のsafe harbor sitesを標的化するmRNA CRISPR/Cas9(ToolGen、Inc;Cas9:Streptococcus pyogenes由来(配列番号4)、およびsgRNAのAAVS1標的部位:5’−gtcaccaatcctgtccctag−3’(配列番号7))をAAVS1に形質感染させた。

3.1.Frozen block tissue slide
脳試料を収集するために、ラットを麻酔させ、18℃で食塩水200mLで経心腔的灌流(transcardial perfusion)させた後、4%パラホルムアルデヒドを含有する0.1Mリン酸緩衝食塩水(PBS)200mLで経心腔的灌流させた。抽出した脳を4℃で4時間固定液(fixative solution)に浸漬した後、20%スクロース(Sigma−Aldrich)を含有する氷冷却された0.1M PBSに移した。このように準備された脳はcryotomeを使用して10または30μmで冠状に切断して使用時まで−20℃で保管した。
in vivoおよびin vitroでタンパク質発現水準を決定するために、EzRIPA lysis kit(ATTO、Tokyo)を使用して収集された脳または細胞を溶解させた。その後、内嗅皮質(Entorhinal cortices;ENT)を均質化し4℃で13,000xgで20分間遠心分離した。上清液を新しいチューブに移し、Bicinchoninic acid assay kit(Thermo Fisher Scientific)を使用してタンパク質含量を測定した。
Trizol reagent(Thermo Fisher Scientific)を使用して製造社の使用説明によってラット脳脂肪内の総RNAを分離した。簡略に説明すれば、ENTをクロロホルム(Amresco、Solon、OH)0.2mLと混合した前記Trizol reagent 1mLで均質化させ、4℃で12,000xgで15分間遠心分離した。上澄み液を新しいチューブに入れて100%イソプロパノール0.5mLと混合し10分間12,000xgで遠心分離した。このように得られたRNAペレットを70%エタノールで洗浄し7,500xgで5分間遠心分離し、乾燥させたペレットをdiethylpyrocarbonate(DEPC) water 30μlに溶解させてNanodrop2000(Thermo Fisher Scientific)を使用して定量した。
MSCsでsRAGE発現を確認するために、GeneJET genomic DNA purification kit(Thermo Fisher Scientific)を使用してMSCおよびsRAGE分泌MSC(sRAGE−MSC)のgDNA(ゲノムDNA)を抽出した。その後、Nanodrop 2000を使用してgDNAの濃度を測定した。同量のgDNAを次の条件でPCR増幅させた:15 cycles of denaturation(30 sec at 90℃)and annealing(90 sec at 68℃)and 20 cycles of denaturation(30 sec at 95℃)、annealing(30 sec at 58℃)and synthesis(90 sec at 72℃)、followed by a primer extension(5 mins at 72℃)。前記PCRに使用されたプライマー配列を表2に整理した。
タンパク質発現水準を測定するために、同量のタンパク質を10% SDS−PAGE(sodium dodecylsulfate−polyacrylamide gel electrophoresis)で分離し、Semi−Dry transfer system(ATTO)を使用して25Vで10分間ポリフッ化ビニリデン(PVDF)メンブレンに移した。その後、メンブレンを0.1% Tween−20(TBST)を含有するTris−緩衝食塩水(TBS)(pH7.6)に含まれている5%(w/v)skimmed milkで2時間遮断し、洗浄し、ブロッキング溶液(blocking solution)(normal horse serum、Vector laboratories)で一次抗体と共に4℃で一晩インキュベーティングした後にTBSTで洗浄し、適当な2次抗体と共にインキュベーティングし洗浄した。目的タンパク質はImageQuant LAS−4000(GE Healthcare、Uppsala、Sweden)上でEzWestLumiおよびluminol substrate(ATTO)を使用して検出した。前記分析に使用された抗体を表3に整理した:
冷凍された脳組織切片(10μm)を1%正常血清で培養して非特異的抗原と抗体結合を遮断した後、抗体(表3参照)を4℃で一晩インキュベーティングした。脳切片を1時間蛍光接合された2次抗体と共に1時間インキュベーティングし、PBSで再び洗浄した。核はDAPI(4’6−diamino−2−phenylindole;Sigma−Aldrich)で室温で5分間対照染色し、発生する蛍光信号を共焦点顕微鏡(LSM 710、Carl Zeiss、Oberkochen、Germany)で検出した。前記検出された蛍光信号の分析は、Image J software(NIH、Bethesda、MD)を使用して行った。
MSCおよびsRAGE−MSCからのsRAGE分泌を測定するために、使用される全てのELISA試薬、working standards、および試料は製造者(Aviscera Bioscience、Santa Clara、CA)指針書によって準備した。試料を抗体が予備コーティングされたマイクロプレートのウェルに添加した。その次に、表3に記載された抗体接合体を各ウェルに添加してインキュベーティングした後、マイクロプレート判読機を使用して450nmで光学密度(optical densities)を測定した。in vivoおよびin vitroでRAGEリガンドの発現およびRAGEとそのリガンドの間の相互作用を測定するために、96−ウェルマイクロプレートのウェルを100mM carbonate/bicarbonate buffer(pH9.6)に含まれているAGE、HMGB1、およびS100β抗体で4℃で一晩コーティングした。0.1% triton x−100(TPBS)を含有するPBSでウェルを洗浄した後、室温で2時間5% skim milkを添加して残っているタンパク質結合部位を遮断した。PBSで洗浄した後、他の組織抽出物、細胞溶解物、または上澄み液試料を前記ウェルに添加して4℃で一晩インキュベーティングした。TPBSで洗浄した後、RAGE抗体を添加して室温で2時間放置した。プレートをTPBSで洗浄した後、試料をペルオキシダーゼ接合2次抗体と共に室温で2時間インキュベーティングした。その次に、TMB基質溶液を添加し、5〜10分間インキュベーティングした後、同一な体積の停止液(2N H2SO4)を添加した。光学密度は450nmで測定した。使用された抗体濃度は前記表3に記載されている。
FACSを使用してMSCマーカーのCD44(陽性)、CD73(陽性)、およびCD34(陰性)を検査してMSCsおよびsRAGE−MSCsを同定した。細胞はfluorescein isothiocyanate(FITC)で標識された一次抗体と共に暗条件で1時間インキュベーティングした後、PBSで3回洗浄した。染色後、106個のMSCまたはsRAGE−MSCに対してFACS(Calibur、BD Bioscience)分析を実施した。
下記の実施例に7週齢Sprague Dawley(SD)ラットを使用した。動物は個別的に収容し、標準食べ物と水を自由に摂取することができるようにしながら12時間の明暗周期条件の温度調節式(24℃)施設で維持させた。動物試験は嘉泉大学校のInstitutional Animal Care and Use Committee(AAALAC International)で承認された国際指針によって行った。
人間Aβタンパク質断片1−42(Aβ1−42;DAEFRHDSGY EVHHQKLVFF AEDVGSNKGA IIGLMVGGVV IA;配列番号5;Sigma−Aldrich;cat.A9810)はジメチルスルホキシド(dimethylsulfoxide)(DMSO)に4mMの濃度で溶解して準備した。人間sRAGEタンパク質(UniProtKB acc.no.Q15109)はBiovendor(cat.RD172116100、Brno、Czech Republic;配列番号6(Total 339 AA.MW:36.5 kDa.UniProtKB accession no.Q15109.N−Terminal His−tag 14 AA))から購入して、アセテート緩衝液(pH4)に0.5mg/ml濃度で溶解させた。前記準備されたAβ1−42ペプチドとsRAGEは、使用時まで−80℃で保管した。使用前に、Aβ1−42ペプチドおよびsRAGEをPBSを使用してそれぞれ200nMまたは6.7nMに希釈した。
外科的手術前にSDラット(参考例9)をZoletil 50(50mg/kg)およびRompun(10mg/kg)で麻酔させた。Aβ1−42ペプチドまたはsRAGEをそれぞれ200uMまたは6.7nMの濃度でリン酸緩衝食塩水(PBS)に溶解させた。頭皮正中線切開後、頭蓋骨のbregmaから後方8.3mmおよび側方5.4mm地点を生物学的電気ドリルで穴をあけた。その後、5μl Hamilton注射器の針(30ゲージ)を標的領域(深さ、4.5mm)に到達するまで垂直に下ろした。stereotaxic guidance下でENTに試薬と細胞を次のように注入した:200uM Aβ1−42溶液 5μl、6.7nM sRAGEタンパク質 3μl、106個のsRAGE−MSCまたは106個のMSC 5μl。
PrimeScript 1st strand cDNA Synthesis Kit(TAKARA)を使用して分離された脳RNAからcDNA(Complementary DNA)を合成した。qRT−PCRはCFX386 touch(Bio−rad、Hercules、CA)を使用して行い、反応効率および閾値サイクル(threshold cycle)数はCFX386 softwareを使用して決定した。使用された全てのプライマーは人間特異的な配列を使用して設計され、前記配列は表2に示したとおりである。
PBSで洗浄された冷凍脳切片にIn Situ Cell Death Detection Kit(TUNEL;Roche Applied Science、Burgess Hill、UK)を使用してTUNELを行った。簡略に説明すれば、組織切片を透過性化溶液(permeabilization solution;0.1%(w/v) Triton X−100含有0.1%(w/v) sodium citrate溶液)で氷で2分間インキュベーティングし、PBSで洗浄した後、TUNEL反応混合物で処理し、humidified chamber atmosphereで2時間インキュベーティングした(37℃および暗条件)。その後、切片をPBSで3回洗浄し、Vectashield mounting medium(Vector Laboratories、Burlingame、CA)を使用してglass slideの上に置いてcoverslipを覆った。Image J(NIH)softwareを使用して細胞自殺死(Apoptotic)細胞比率を決定した。
先に準備された冷凍されたラット脳組織スライドを室温で5分間乾燥させ、PBSで10分間洗浄した後、graded ethanol series(100%(v/v)エタノール:3分、90%エタノール:3分、80%エタノール:3分、70%エタノール:5分)でインキュベーティングし蒸留水で洗浄した後、glacial acetic acid含有0.1% Cresyl violet染色溶液(Sigma−Aldrich)で20分間染色し、蒸留水、70%エタノールで1分、80%エタノールで30秒、90%エタノールで20秒、100%エタノールで20秒、最後にキシレン(xylene)で5分間洗浄した。組織学的分析(BBC biochemical、Mt Vernon、WA)のために前記切片をOpticMount mounting mediumを使用してマウンティングし、前記分析は光学顕微鏡(Carl Zeiss)を使用して行った。Image J software(NIH)を使用してニューロンを計数した。
小さな試料の大きさを勘案して、non−parametric分析を使用した。IBM SPSS statistics 23 software(SPSS、Inc.、Armonk、NY)でMann−Whitney testを使用してグループ間比較を行った(Null hypotheses of no difference were rejected when p−values were<0.05)。結果は、独立的な三回の試験から得られた値を平均して表わした。
参考例で製作したsRAGE分泌MSC(sRAGE−MSC)の特性を試験して、正常的なMSCの特徴を示すのを確認した。
sRAGE−MSCsまたはMSCsをAβ1−42注射されたラットの脳に移植して、sRAGE−MSCsの効果を試験した。人間特異的抗体(表3参照)およびプライマー(表2参照)を使用して免疫蛍光法およびqRT−PCRを行って、移植されたsRAGE−MSCとMSCの移植後4週目の蛍光イメージを得て生存率を測定し、その結果を図2a〜2eに示した。
アルツハイマー病ラットモデルを作るためにAβ1−42をラットのENT領域に注射した。Aβ1−42注射はアミロイド前駆体タンパク質(amyloid precursor protein;APP)およびベータサイトAPP切断酵素(beta−site APP cleaving enzyme 1;BACE1)の水準を増加させることが観察された。
Aβ1−42注射されたラットの脳で炎症性タンパク質と活性化された小膠細胞間の関連性を調査するために、Iba1(activated macrophage marker)陽性細胞をカウンティングして活性化された小膠細胞の分布を調査して、その結果を図4aおよび4bに示した。図4aはAβ1−42注入、Aβ1−42注入およびsRAGEタンパク質処理、Aβ1−42注入およびMSC処理、またはAβ1−42注入およびsRAGE−MSC処理されたラット脳での活性化された小膠細胞(Iba1;緑色)分布を示す共焦点顕微鏡イメージであり、図4bは発現された全体細胞に対するIba1陽性細胞の比率を示すグラフである。図4aおよび4bで示されるように、Iba1陽性細胞の数はAβ1−42注射されたラットの脳でnaive controlsより2.02倍多く、Aβ1−42とsRAGEタンパク質またはMSCを処理した場合には若干減少した。しかし、Aβ1−42とsRAGE−MSCを処理した場合にはIba1陽性細胞数がAβ1−42注射した場合と比較して有意的に低く示された(3.08倍さらに低い)。
RAGE媒介神経細胞死滅に対するsRAGE−MSCsの保護効果を試験するために、免疫蛍光法でAβ1−42注射されたラットの脳でのRAGE発現を確認した。図5aはAβ1−42注入、Aβ1−42注入およびsRAGEタンパク質処理、Aβ1−42注入およびMSC処理、またはAβ1−42注入およびsRAGE−MSC処理されたラット脳でのRAGE発現(緑色)を示す共焦点顕微鏡イメージである。図5aに示されているように、RAGEの発現を示す蛍光強さは、Aβ1−42注射によって増加した反面、Aβ1−42とsRAGE−MSCと共に処理した場合にはAβ1−42注射した場合と比較して減少した。また、図5bはAβ1−42注入、Aβ1−42注入およびsRAGEタンパク質処理、Aβ1−42注入およびMSC処理、またはAβ1−42注入およびsRAGE−MSC処理されたラット脳でのSAPK/JNK、pSAPK/JNK、Caspase 3、Caspase 8、およびCaspase 9の水準を測定した免疫ブロッティング分析結果を示す。図5bに示されているように、SAPK/JNK、Caspase 3、Caspase 8およびCaspase 9のようなRAGE媒介神経細胞死滅関連タンパク質の発現水準は、Aβ1−42注射されたラットの脳と比較して、Aβ1−42とsRAGE−MSC処理されたラットの脳で有意に低く示された(図5b)。
sRAGEを分泌するiPSCを生成するために、pZDonorベクター(Sigma−Aldrich)に人間EF1−αプロモーター、sRAGEコーディング配列、およびpoly A tailをクローニング方法で挿入して製作したsRAGEドナーベクター(図1a参照)およびCRISPR/CAS9 RNPシステムを使用してiPSCの形質感染(Transfection)を行った。ガイドRNAは19番染色体でAAVS1と知られたsafe harbor siteを標的にするように設計した(Cas9:Streptococcus pyogenes由来(配列番号4)、sgRNAの標的部位:gtcaccaatcctgtccctag(配列番号7))。形質感染は4D nucleofector system((Lonza)を使用して行った。形質感染条件はウェブサイト上のLonzaプロトコル(cell type ‘hES/H9’)に提供されている条件に従った。P3 primary cell 4D nucleofector X kit L(Lonza、V4XP−3024)を使用してelectroporationを行った。2×10^5個の人間iPSC(Korean National Stem Cell Bank)をcas9タンパク質15ug、gRNA20ugおよびsRAGEドナーベクター1ugで形質感染させて、sRAGEを分泌するiPSCを製造した。
本発明の様々な実施形態を以下に示す。
1.可溶性(soluble)の最終糖化産物受容体(Receptor for Advanced Glycation End products;RAGE)(sRAGE)をコードする遺伝子を含み、sRAGEを分泌する、幹細胞。
2.前記幹細胞は、胚性幹細胞(embryonic stem cells)、成体幹細胞(adult stem cells)、誘導万能幹細胞(induced pluripotent stem cells;iPS cells)、および前駆細胞(progenitor cells)からなる群より選択された1種以上である、上記1に記載の幹細胞。
3.前記幹細胞は、誘導万能幹細胞または間葉系幹細胞である、上記1に記載の幹細胞。
4.上記1〜3のうちのいずれかの幹細胞を含むアルツハイマー病の予防または治療用薬学組成物。
5.前記薬学組成物は、アルツハイマー病患者で以下のうちの一つ以上の活性を有するものである、上記4に記載の薬学組成物:
アミロイド前駆体タンパク質(amyloid precursor protein;APP)またはベータサイトAPP切断酵素(beta−site APP cleaving enzyme 1;BACE1)の発現抑制、
RAGEリガンドまたは炎症性タンパク質の発現抑制、または

- 可溶性(soluble)の最終糖化産物受容体(Receptor for Advanced Glycation End products;RAGE)(sRAGE)をコードする遺伝子を含み、sRAGEを分泌する、幹細胞。
- 前記幹細胞は、胚性幹細胞(embryonic stem cells)、成体幹細胞(adult stem cells)、誘導万能幹細胞(induced pluripotent stem cells;iPS cells)、および前駆細胞(progenitor cells)からなる群より選択された1種以上である、請求項1に記載の幹細胞。
- 前記幹細胞は、誘導万能幹細胞または間葉系幹細胞である、請求項1に記載の幹細胞。
- 請求項1〜3のうちのいずれか一項の幹細胞を含むアルツハイマー病の予防または治療用薬学組成物。
