Abstract
Description
毎年、白血病、ホジキン及び非ホジキンリンパ腫、及び骨髄腫の新しい事例が、診断される新しい癌の全事例の略10%を占めている。高齢化社会に入るにつれて血液癌を患う老人が大きく増えているが、彼らの生存率は非常に低く、特に65歳以上の生存率は9.4%に過ぎなく、80歳以上の生存率は0%と極端に低い(Blood,2012,120;1165−1174)。
また、血液癌は、高容量の寛解誘導抗癌治療にも反応性がないため、治療効果がないか、或いは耐性発現率及び5年内再発率が非常に高いため、未充足の医学的要求が非常に高い。
現在、抗体及びキナーゼ抑制剤を使用する標的治療療法が開発されているが、依然として血液癌の治療は化学療法及び放射線治療療法に多く依存している(The new england journal of medicine,2014,371:1005〜1015)。血液癌の化学療法剤として用いられる抗癌剤は、殆どが相当な毒性をもつ細胞毒性抗癌剤であり、低い生体利用率及び癌細胞への標的伝達の難しさによって非常に高い容量を投与する。このため、患者に深刻な副作用を招き、治療を難しくする。したがって、毒性が少ないとともに効果的な治療効能を持つ新しい治療剤が要求される。
かかるグアノシンが豊富なオリゴヌクレオチドは、シトシンとの三重の水素結合の他にも特別な構造的な特徴を有している。グアノシンが豊富なオリゴヌクレオチドは、分子内結合又は分子間結合によって4本鎖の構造を有することができる。一般的なアデノシンとチアミン、グアノシンとシチジンとの水素結合による二重らせん構造を形成する代わりに、4個のグアノシンが一つの平面に位置してフーグステン(hoogsteen)形態の水素結合をなすことによってG−四重鎖(G−quadruplex)を形成し、これが2本以上連続して構成されて四重らせん構造(tetrahelical structure)を有することになる。一般的にオリゴヌクレオチドは血液内安定性及び細胞透過率が非常に低いため、薬物として開発することには困難があったが、このようなG−四重鎖をなすオリゴヌクレオチドはその構造的な特徴によって安定した構造をなしており、比較的高い血液安定性及び細胞透過度を有すると知られている。
また、G−四重鎖を形成するオリゴヌクレオチドは、癌細胞の成長を阻害する効果の他にも、米国特許第5567604号では抵ウイルス、米国特許第6994959号では免疫調節作用、米国特許公開第2007−105805号ではハンティントン疾患に治療効果があるものと発表され、生体内で様々な機能及び調節作用をすることが知られた(Cheryl A.Stoddart et al.Antimicrobial agents and chemotherapy,1998,2113−2115;Michael Skogen et al.BMC neuroscience,2006,7:65)。
このようなG−四重鎖を形成するオリゴヌクレオチドは、細胞成長静止効果(cytostatic effect)による細胞死滅を誘導するので、細胞死滅率が比較的高くない。このため、強い毒性の化学療法剤と併用投与しなければならないという不具合がある(Paula J.Bates et al.Experimental and molecular pathology,2009,151−164;Christopher R.Ireson et al.Molecular cancer therapy,2006,2957−2962)。
したがって、本発明は、治療効能を持つ修飾核酸として1−β−D−アラビノフラノシルシトシンが1個以上導入され、且つ豊富なグアノシンが含まれた新規なオリゴヌクレオチド修飾核酸体を提供することにその目的がある。
また、本発明は、前記オリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含有する血液癌の予防、改善又は治療用組成物を提供することに他の目的がある。

前記配列おいて、Gはグアノシン又はグアノシン誘導体、Tはチミン又はチミン誘導体、Nは1−β−D−アラビノフラノシルシトシンである。
また、本発明の他の側面は、前記オリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含有する血液癌の予防、改善又は治療用組成物を提供する。
本発明の一側面は、下記化学式1で表示される化合物を含有してG−四重鎖構造を形成することを特徴とするオリゴヌクレオチド修飾核酸体を提供する:

前記配列において、Gはグアノシン又はグアノシン誘導体、Tはチミン又はチミン誘導体であり、Nは修飾核酸であって、1−β−D−アラビノフラノシルシトシンである。
本発明の一側面は、治療効能を持つ修飾核酸として1個以上の1−β−D−アラビノフラノシルシトシンと豊富なグアノシンが含まれた新規オリゴヌクレオチド修飾核酸体を提供することにその目的がある。

したがって、G−四重鎖を形成しながら生理活性を有するオリゴヌクレオチドの配列に本発明のシチジン修飾核酸を置換して導入することによって新規なヌクレオチド修飾核酸体を完成することができる。
前記配列において、Gはグアノシンであり、好ましくは、2’−デオキシグアノシン(2−deoxy−guanosine)、グアノシン(guanosine)、2’−O−メチル−グアノシン(2’−O−methyl−guanosine)、2’−フルオロ−グアノシン(2’−F−guanosine)、LNA(Locked Nucleic Acid)−グアノシン、D−デオキシグアノシン、D−グアノシンの中から選ばれる1種又は2種以上でよく、Nはシチジン由来の修飾核酸であって、1−β−D−アラビノフラノシルシトシン(1−β−D−Arabinofuranosylcytosine)である。
DNA合成は、一般のDNA固定相合成機を用いて1μmoleスケールで合成を行った。合成に用いられるホスホロアミダイトは、グレンリサーチ(Glen research)社から購入したデオキシグアノシン、チミジン、1−β−D−アラビノフラノシルシトシンホスホロアミダイトを乾燥したアセトニトリルを用いて0.067M濃度に溶かしてポリジン(Polygene)社のDNA固定相合成機に装着した。次の表1に提示された配列によって合成され、一般的に3’→5’の方向に進行されるが、初めのヌクレオチドの3’水酸基を樹脂に付けておき、一つの塩基が添加される間に、大きく4段階の化学反応、すなわち、5’−末端の脱トリチル反応(detritylation)、新しい塩基の付加反応(coupling)、付加反応されなかったDNA鎖のキャッピング(capping)反応、リン酸基の酸化反応(oxidation)を反復した。反応が終結すると保護基を除去し、合成されたCPGレジンをアンモニア水に入れて55℃で5時間放置後にアンモニア水を乾燥させ、白色パウダーを得た。ウォーターズ(Waters)陰イオン交換HPLCカラムを用いてHPLCシステムにおいて1M NaCl溶液を5〜70%に増加させて精製を行った。メインピークを収集し、ここに100%エタノールを入れてオリゴヌクレオチドを沈殿させて乾燥した。結果的に、HPLCを用いた測定結果による純度85%以上のオリゴヌクレオチドを得、ESI−LC−MS(Q−TRAP2000ESI−MS)を用いて分子量を測定して合成に成功したか否かを把握した。
それぞれのオリゴヌクレオチド修飾核酸体を、10mM Tris−HCl(pH7.4)溶液に希釈し、最終濃度が100μMとなるように作った。希釈された溶液を94℃で5分間放置した後、氷にチューブを放置した。2M KCl溶液を添加して最終濃度が50mMのKCl溶液となるようにした後、3時間60℃で放置した後、温度を常温まで徐々に下げた。それぞれのオリゴヌクレオチドを10mM Tris−HCl(pH7.4)溶液に希釈して10μM濃度にした後、94℃で5分間放置した後、氷にチューブを放置した溶液と2M KCl溶液を添加して最終濃度50mMのKCl溶液にした後、3時間60℃で放置した後、温度を常温まで徐々に下げた溶液を使用した。
実験前日に96ウェルプレートに、ml当たり104〜105個の血液癌細胞としてHL60、MV−4−11、CCRF−CEM、MOLT−4及びMOLM−13細胞株、薬物抵抗性血液癌細胞としてazacitidine(AZA−1)−resistant MOLM13、decitabine(Dec−5)−resistant MOLM13及びcytarabine(AraC−1)−resistant MOLM13細胞株、正常細胞としてWI38、CCD−18co及びFa2N4が懸濁されている培養液190μlを接種した後、翌日、前記実施例2で製造したオリゴヌクレオチド修飾核酸体溶液10μlを添加して5日間培養した。
接種5日後に、一般のMTT方法で細胞死滅を測定した[JBC1999,26369参考]。
また、本発明の新規オリゴヌクレオチド修飾核酸体は、様々な薬物抵抗性血液癌細胞に対しても優れた細胞死滅効果を示すことを確認した[表3]。
また、血液癌細胞又は薬物抵抗性血液癌細胞対しては優れた細胞死滅効果を示すのに対し、正常細胞にはほとんど影響を及ぼさないことを確認した[表4]。
ヒト由来血液癌細胞に対する動物における効能を確認するために、NOD−SCIDマウスに血液癌を誘導し、生体内における血液癌細胞死滅効果とマウスの生存率を評価した。
まず、血液癌の誘導は具体的に次のように行った。実験群は、ビークル群(8匹)と薬物群(9匹)で構成した。
8週齢のNOD−SCIDマウスにLuc−MOML−13細胞(human MDS/AML cell line)を1匹当たり107細胞を静脈注射した。投与薬物は、ポタシウムバッファに化合物2を2mM濃度に溶かし、94℃で5分間放置した後、3時間徐々に温度を室温に下げた後に使用した。
薬物投与は、細胞接種して2日目から1週に5回ずつ2週間に毎日静脈注射(150mg/kg)した後、生存率を測定し、その結果を図5に示した。また、薬物投与して10日目に血液癌細胞の細胞死滅効能を生体映像で撮影してビークル群と比較し、その結果を図3及び表4に示した。
図3及び表4に示すように、ビークル群に比べて薬物投与群の血液癌細胞が著しく減少することを確認した。
また、図5に示すように、ビークル群に比べて薬物投与群の生存率が著しく優れることを確認した。
免疫体系のある環境における血液癌細胞に対する生体内効能を確認するために、C57BL/6マウスにマウス由来血液癌細胞(C1498)を移植し、マウスの生存率を評価した。
まず、血液癌の誘導は具体的に次のように行った。実験群は、ビークル群(8匹)と薬物群(9匹)、対照群(8匹)で構成した。薬物投与群は、化合物2を300mg/kg注射し、対照群は、シタラビンを30mg/kg(化合物2 300mg/kgと同等量のシタラビン)注射した。
8週齢のC57BL/6マウスにC1498細胞(mouse AML cell line)を1匹当たり5X105細胞を静脈注射した。投与薬物は、ポタシウムバッファに化合物2を2mM濃度に溶かし、4℃で6時間以上放置後に使用した。
薬物投与は、細胞接種して2日目から一日に2回ずつ2週間に毎日皮下注射し、その生存率を測定した。その結果を図6に示した。
図6に示すように、ビークル群に比べて薬物投与群の生存率が著しく優れることを確認し、且つ対照群に比べても著しく優れた生存率を確認した。
免疫体系のある環境におけるALM細胞に対する生体内効能を確認するために、Balb/cマウスにマウス由来血液癌細胞(WEHI−3)を移植し、マウスの生存率を評価した。
まず、血液癌の誘導は具体的に次のように行った。実験群は、ビークル群(7匹)と薬物群(7匹)、対照群(7匹)で構成した。薬物投与群は、化合物2を300mg/kg注射し、対照群は、シタラビンを30mg/kg(化合物2 300mg/kgと同等量のシタラビン)注射した。
8週齢のBalb/cマウスにWEHI−3細胞(mouse AML cell line)を1匹当たり5X105細胞を静脈注射した。投与薬物は、ポタシウムバッファに化合物2を2mM濃度に溶かし、4℃で6時間以上放置後に使用した。
薬物投与は、細胞接種して2日目から一日に2回ずつ2週間に毎日皮下注射し、生存率を測定した。その結果を図7に示した。
図7に示すように、ビークル群に比べて薬物投与群の生存率が著しく優れることを確認し、対照群に比べても著しく優れた生存率を確認した。
実際の患者血液癌細胞における抗癌効能を確認するために、ソウルアサン病院のIRB審査後に供与された再発性/難治性急性骨髄性白血病患者の骨髄単核細胞を用いて、化合物2と対照薬物(シタラビン,AS1411)に対するコロニー形成能を比較した。骨髄単核細胞をコロニー形成培地に分注した後、それぞれの薬物を0.25μM、0.5μM、1μMを処理して16日間培養した後、コロニー個数を測定した。
図8にみられるように、再発性/難治性血液癌細胞において同一濃度の対照薬物は治療効力が不十分だったが、化合物2の抗癌効果は著しく優れていることを確認した。
本発明の有効成分に対して毒性実験を次のように行った。
化合物2をポタシウムバッファ溶液に溶解した後、それをマウス(群当たりに10匹)にそれぞれ1g/kgを投与して7日間観察したが、死亡するマウスがなくて半数致死量(LD50)は最小1g/kg以上と確認された。
また、血液癌を誘発しなかった一般マウスにおいて、実験例4と同じ条件で薬物を投与したとき、対照群では正常白血球が急減したが、化合物2を投与した薬物投与群では白血球数値を相対的に高く維持し、優れた安全性を示した(ビークル群平均値6.29、ビークル群範囲4.70〜8.97、薬物投与群平均値4.07、薬物投与群範囲3.36〜4.77、対照群平均値2.34、対照群範囲0.95〜4.31)。実験例3〜5の結果から、化合物2が血液癌細胞を特異的に死滅させて生存率を高め、正常血液細胞の死滅を最小化することによって血液癌標的治療剤として優れた特性を示すことを確認した。
化合物2を10mg含有する注射液剤を次のような方法で製造した。
化合物2 1g、塩化ナトリウム0.6g及びアスコルビン酸0.1gを蒸留水に溶解させて100mlを作った。この溶液を瓶に入れて20℃で30分間加熱して滅菌させた。
前記注射液剤の構成成分は、次の通りである。

Claims (10)
- 下記化学式1で表示される化合物を含有してG−四重鎖構造を形成することを特徴とするオリゴヌクレオチド修飾核酸体:

式中、R1は、水素、又は他の核酸のホスフェート残基のリン原子であり、R2は、水素、又は他の核酸のホスフェート残基のリン原子である。 - 前記化合物は、下記化学式2で表示される化合物であることを特徴とする、請求項1に記載のオリゴヌクレオチド修飾核酸体;

- 前記オリゴヌクレオチド修飾核酸体は、‘GaTbNc’配列[ここで、Gはグアノシン又はグアノシン誘導体、Tはチミン又はチミン誘導体であり、Nは修飾核酸であって、1−β−D−アラビノフラノシルシトシン(1−β−D−Arabinofuranosylcytosine)であり、G、T、Nは順列によって無作為に配列され、aは1〜30の整数、bは0〜30の整数、cは1〜30の整数であり、ただし、a、b、cの合計値は60を超えない。]で表示されることを特徴とする、請求項2に記載のオリゴヌクレオチド修飾核酸体。
- 下記配列で表示されることを特徴とする、請求項3に記載のオリゴヌクレオチド修飾核酸体:

前記配列において、Gはグアノシン又はグアノシン誘導体、Tはチミン又はチミン誘導体であり、Nは修飾核酸であって、1−β−D−アラビノフラノシルシトシン(1−β−D−Arabinofuranosylcytosine)である。 - 前記グアノシン又はグアノシン誘導体は、2’−デオキシグアノシン(2−deoxy−guanosine)、グアノシン(guanosine)、2’−O−メチル−グアノシン(2’−O−methyl−guanosine)、2’−フルオロ−グアノシン(2’−F−guanosine)、LNA(Locked Nucleic Acid)−グアノシン、D−デオキシグアノシン及びD−グアノシンの中から選ばれたいずれか一つ以上であることを特徴とする、請求項3に記載のオリゴヌクレオチド修飾核酸体。
- 請求項1〜5のいずれかに記載のオリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含有することを特徴とする、血液癌の予防、改善又は治療用組成物。
- 前記血液癌は、非ホジキンリンパ腫、ホジキンリンパ腫、多発性骨髄腫、白血病、リンパ腫、骨髄異形性症候群(MDS)、急性リンパ芽球性白血病、急性骨髄性白血病、慢性リンパ球性白血病、慢性骨髄性白血病及び孤立性骨髄腫のいずれか一つ以上である、請求項6に記載の血液癌の予防、改善又は治療用組成物。
- 請求項1〜5のいずれかに記載のオリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含有することを特徴とする、DNA合成(DNA synthetase)又はDNAメチル転移酵素(DNA methyltransferase)阻害剤であるシタラビン(Cytarabine)、デシタビン(Decitabine)及びアザシチジン(Azacitidine)のいずれか一つ以上に対する耐性血液癌の予防、改善又は治療用組成物。
- 請求項1〜5のいずれかに記載のオリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含むことを特徴とする、再発性又は難治性急性白血病患者の骨髄単核細胞において高い細胞死滅誘導効果を表す急性骨髄性白血病の予防、改善又は治療用組成物。
- 請求項1〜5のいずれかに記載のオリゴヌクレオチド修飾核酸体又はその薬学的に許容可能な塩を有効成分として含むことを特徴とする、正常細胞に比べて癌細胞においてより高い細胞死滅誘導効果を表す血液癌の改善又は治療用組成物。
