Abstract
Description
本特許出願は、2017年3月22日に出願の米国仮特許出願第62/474,757号の優先権を主張し、その開示の全体が参照により本明細書内に組み込まれている。
本明細書には、生体試料の調製のために緩衝液を交換する試料プレートおよびその方法が開示されている。この方法は、高い圧力を使用してプレート中の貯留部のフィルターを通して緩衝液を押込む緩衝液の自動交換を含むことができる。フィルターのこの高圧下での脱離を回避するように、フィルターの貯留部への封止性を補強するフィルター取付け領域が配置されてもよい。さらにこの試料プレートの製造方法についても開示する。

Claims (16)
- 生体試料から緩衝液を自動交換する試料プレートであって、
複数の貯留部を含み、
前記複数の貯留部のうちの各貯留部は、側壁、ならびに熱封止および接着剤封止を含むフィルター取付け領域を備え、
前記フィルター取付け領域に各貯留部に固定された周縁部を有するフィルターを含み、
前記フィルター取付け領域は、前記フィルターを挟んで約30psig以上の圧力差では各貯留部への前記フィルター周縁部の固着を維持している、試料プレート。 - 前記フィルターを挟む前記圧力差は、約45psig以上である、請求項1に記載の試料プレート。
- 前記フィルターを挟む前記圧力差は、約60psig以上である、請求項1に記載の試料プレート。
- 前記フィルター周縁部は折畳み形状を有し、前記熱封止および前記接着剤封止は、前記フィルター周縁部をその折畳み形状で各貯留部に固定する、請求項1に記載の試料プレート。
- 前記接着剤封止は、前記貯留部の前記側壁および内側隆起部によって規定された溝部内に配置された接着剤を含む、請求項4に記載の試料プレート。
- 前記貯留部の前記内側隆起部の高さは約0.38mmである、請求項5に記載の試料プレート。

前記複数の貯留部のうちの各貯留部は、側壁、および熱封止を含むフィルター取付け領域を備え、

前記熱封止は、前記試料プレートの一部分と貯留部の蓋の間に固定されたフィルター周縁部を含み、前記フィルター取付け領域は、フィルターを挟んで約30psig以上の圧力差では各貯留部への前記フィルター周縁部の固着を維持している、試料プレート。 - 生体試料から緩衝液を自動交換する試料プレートであって、
複数の貯留部を含み、
前記複数の貯留部のうちの各貯留部は、側壁、および熱封止を含むフィルター取付け領域を備え、
前記フィルター取付け領域に各貯留部に固定された周縁部を有するフィルターを含み、
前記熱封止は、圧縮されたOリングと前記試料プレートの一部分の間に固定されたフィルター周縁部を含み、前記フィルター取付け領域は、フィルターを挟んで約30psig以上の圧力差では各貯留部への前記フィルター周縁部の固着を維持している、試料プレート。 - 生体試料から緩衝液を自動交換する方法であって、
複数の貯留部を含む試料プレートを圧力室内に配置することを含み、
前記複数の貯留部のうちの各貯留部は、フィルター、ならびに熱封止および接着剤封止を含むフィルター取付け領域を備え、
前記貯留部プレート上部の前記圧力室内の空間を加圧し、前記フィルターを挟んで約30psig以上の圧力差を発生させて、前記生体試料を第1の緩衝液から分離することを含む、方法。 - 前記フィルターを挟む圧力差は、約45psig以上である、請求項9に記載の方法。
- 前記フィルターを挟む圧力差は、約60psig以上である、請求項9に記載の方法。
- さらに第2の緩衝液を前記複数の貯留部に加えることを含む、請求項9に記載の方法。
- 緩衝液を自動交換する試料プレートを製造する方法であって、
周縁部を有するフィルターを複数の貯留部内に装填することを含み、
前記複数の貯留部のうちの各貯留部は、側壁、フィルター取付け領域、および内側隆起部を備え、前記フィルター取付け領域は、前記側壁および前記内側隆起部により規定された溝部を含み、
前記フィルター周縁部を前記フィルター取付け領域に熱的に封止すること、および
前記フィルター周縁部を前記フィルター取付け領域に接着剤により封止することを含む、方法。 - 熱封止は、前記フィルター周縁部が各貯留部の前記内側隆起部に固定されるように、前記フィルター周縁部に熱および圧力を加えることを含む、請求項13に記載の方法。
- 前記フィルター周縁部は、前記内側隆起部に対して折畳まれる、請求項14に記載の方法。
- 前記接着剤による封止は、前記溝部を接着剤で充填し前記接着剤を硬化させて、フィルター周縁部を各貯留部の前記側壁にさらに固定することを含む、請求項13に記載の方法。
