実験手順
細胞、培地、プラスミド、試薬、及びマウス
正常初代ヒト乳腺上皮細胞(HMEC、ATCC(登録商標)PCS−600−010(商標))はATCCに注文した。本研究で使用したすべての細胞は試験され、マイコプラズマ汚染がないことが判明した。MEGM(商標)Mammary Epithelial Cell Growth Medium BulletKit(商標)(Clonetics(商標)MEGM(商標)Mammary Epithelial Cell Growth Medium plus SingleQuots(商標)Kitパッケージ)はLonza社(CC−3150)に注文した。HMECは、加湿5%CO2雰囲気、37℃で、マトリゲルマトリックス層有り/無しでMEGM BulletKit(商標)で培養した。EGFP−hPMCA2z/b(品番47584)及びmCherry−β−アクチン(品番54967)プラスミドはAddgene社に注文した。抗γ−アクチン抗体(γアクチン、モノクローナル、ab123034)及び抗pan−カドヘリン抗体(ポリクローナル、ab140338)はAbcam社に注文した。Matrigel(商標)Membrane Matrix(CB−40234)及びCorning(登録商標)Matrigel(登録商標)Basement Membrane Matrix、フェノールレッドフリー、*LDEVフリー(製品番号356237)はCorning社から購入した。Corning(登録商標) 500cm2 Square TC−Treated Culture Dish(製品番号431110)はCorning社に注文した。Mepiform(登録商標)(メピフォーム)(創傷治療用シリコン膜)(Warner,P.M.,Coffee,T.L.&Yowler,C.J.Outpatient burn management.The Surgical clinics of North America 94,879−892(2014))はMOlnlycke Healthcare社に注文した。6週齢の雌マウス(系統名:BALB/cJ)は、ジャクソン研究所に注文した。動物実験は、ハーバード大学医学校(Harvard Medical School)の施設内動物実験委員会の承認を得て実施した。
Matrigel(商標)Membrane Matrixを4℃で一晩解凍した。Matrigel層(深さ20〜30μm)は、事前に冷却した6ウェルプレート、10cmディッシュ、又は500cm2正方形ディッシュ中に調製し、その後、25℃、加湿5%CO2雰囲気で20分間ゲル化した。蛍光イメージングのために、Matrigel層は、6ウェルプレート中のVWR−Microカバー上にフェノールレッドフリーMatrigelを用いて調製した。HMECをMatrigel層に播種し、MEGM BulletKit(商標)培地中で37℃、加湿5%CO2雰囲気で培養した。細胞外マイクロフィラメントは、細胞培養後48〜110時間で発達させた。EGFP−hPMCA2z/bプラスミド(Addgene社、品番47584)及びmCherry−β−アクチンプラスミド(品番54967)を、Lipofectatine(登録商標)2000(Life Technologies社、品番11668027)を使用して、マニュアルに従ってHMECにトランスフェクトした。抗γ−アクチン抗体(ガンマアクチン、モノクローナル、ab123034)及び抗pan−カドヘリン抗体(ポリクローナル、ab140338)はAbcam社に注文した。トランスフェクションの2日後、トランスフェクトされた細胞を指定培地中のMatrigelマトリックス層(6ウェルプレートの1ウェルあたり1×103個の細胞)上に播種した。指定時間の培養期間後、細胞及びマトリゲルを4%パラホルムアルデヒド(PFA)で固定した。細胞のヘマトキシリン・エオジン(HE)染色は、細胞固定後に行った。VWR−Microカバー付きの試料をスライドガラスに移し、画像を取得した。
位相コントラスト画像は、Nikon社製TMS倒立位相差顕微鏡及びNikon社製Coolpix オート4300デジタルカメラで撮影した。固定細胞の蛍光画像は、80i正立顕微鏡、並びに20倍又は40倍レンズ及びMetaMorph画像取得ソフトウェアを備えたHamamatsu社製ORCA−ER冷却CCDデジタルカメラで撮影した。固定細胞又は組織切片のヘマトキシリン・エオジン染色画像を、80i正立顕微鏡並びに20倍又は40倍レンズ及びNIS−Elements取得ソフトウェアを備えたHamamatsu社製ORCA−ER冷却CCDデジタルカメラで撮影した。
HMECは、6ウェルプレート中のプラスチックディスク上のMatrigelマトリックス層(深さ約60μm)上に播種した。HMECは、ホルムアルデヒド−グルタルアルデヒド−ピクリン酸−固定液(2.5%パラホルムアルデヒド、5.0%グルタルアルデヒド、及び0.06%ピクリン酸含有0.2M カコジル酸緩衝液):細胞培養培地=1:1の固定液混合物を使用して固定した。次に、固定したHMECを1%四酸化オスミウム(OsO4)/1.5%フェロシアン化カリウム(KFeCN6)中で30分間後固定し、水で3回洗浄し、1%酢酸ウラニル水溶液中で30分間インキュベートした。これに続いて、水で2回洗浄し、アルコールの勾配で脱水した(50%、70%、95%、2×100%を各5分)(Basler,M.,Pilhofer,M.,Henderson,G.P.,Jensen,G.J.&Mekalanos,J.J.Type VI secretion requires a dynamic contractile phage tail−like structure.Nature 483,182−186(2012))。細胞を、プロピレンオキシド及びTAAB Eponの1:1混合物(Marivac Canada社、カナダ、サン・ローラン)に2時間から一晩浸透させた。その後、試料をTAAB Eponに埋め込み、60℃で48時間重合した。Reichert Ultracut−Sミクロトームを使用して、超薄切片(約60nm)を切り出した。次に、超薄切片標本をトルイジンブルーで染色した(30秒間)。画像は、80i正立顕微鏡(20倍レンズ、40倍レンズ)で撮影した。
指定期間の細胞培養(500cm2ディッシュ中のMatrigel層上に6×104個のHMEC)後、500cm2ディッシュ(Corning(登録商標) 500cm2 Square TC−Treated Culture Dish)中のHMEC及び超大型細胞外マイクロフィラメントネットワークを4%パラホルムアルデヒドで10分間固定した後、1×PBSで3回洗浄し、HE染色を行った。細胞塊及び細胞をピペットチップ(1mL又は100μL)及びNikon TMS倒立位相差顕微鏡を用いて除去した後、1×PBSで3回洗浄した。指定時間の細胞培養後、細胞及び超大型細胞外マイクロフィラメントネットワークを、0.5%KMnO4(1×PBS(pH7.1)中で脂質安定化(Zhao,S.et al.Fixation−induced cell blebbing on spread cells inversely correlates with phosphatidylinositol 4,5−bisphosphate level in the plasma membrane.FEBS Open Bio 4,190−199(2014))で1時間、続いて10%中性緩衝ホルマリン固定液で15分間固定した。その後、1×PBSで3回洗浄して、遊離化学残留物を除去した。細胞塊及び細胞をピペットチップ(1mL又は100μL)及びNikon TMS倒立位相差顕微鏡を用いて除去した後、1×PBSで3回洗浄して剥離細胞を除去した。AUMLを調整し、Matrigelマトリックスから分離した。Matrigelを含まないAUMLは、熱傷創傷治癒分析用の自作AUML特異的移植装置により、AUML層の上側が創傷表面と接触した状態で創傷表面に移植された。
6週齢の成体BALB/cJ雌マウスの体重を測定し(体重、20〜24g)、腹腔内(IP)注射により10mg/kgのキシラジン(AnaSed(登録商標)注射剤、キシラジン滅菌溶液)で麻酔した。麻酔したマウスの背中から毛を刈り取り、市販の脱毛剤でその範囲を露出させた。皮膚を98℃の蒸気に4秒間曝すことにより、深度II度熱傷創傷(直径1.5cm)を15匹のマウスに誘発した(Zhang,Y.et al.Role for heat shock protein 90alpha in the proliferation and migration of HaCaT cells and in the deep second−degree burn wound healing in mice.PLoS One 9,e103723(2014))。深度II度熱傷創傷を有する15匹のマウスをランダムに3群に分けた(1群あたり5匹のマウス)。創傷は、1日おきにMepiform(登録商標))又はテーラーメイドAUMLを用いた治療有リ/無しで管理した。創傷の画像を、Nikon社製のカメラで撮影した。火傷後14日目に、マウスを屠殺し、HE染色分析による組織学的評価のために創傷を切除した(切片標本、深さ5μm)。
皮膚の創傷を切除し、HistoChoice(登録商標)MB固定液(Amresco社)で固定し、パラフィンに包埋した。切片標本(深さ5μm)は、抗pan−カドヘリン抗体(Abcam社、ab140338、1:200、Life Technologies社、品番982425、Alex Fluor(登録商標)488 ヤギ抗ウサギIgG(H+L)二次抗体、1:1000)、抗γ−アクチン抗体(Abcam社、ab123034、1:200、Life Technologies社、Alex Fluor(登録商標)568 ヤギ抗ウサギIgG(H+L)二次抗体、1:1000)、及び4’,6−ジアミジノ−2−フェニルインドール(DAPI、1:1000)による二重免疫組織化学染色に供した。蛍光画像は、20倍レンズ/40倍レンズ/60倍レンズを備えたNikon社製80i正立顕微鏡で撮影した。全ての画像は、MetaMorph画像取得ソフトウェアを使用して取得した。
以前に記載されたように統計分析を実施した(Yi,T.et al.eIF1A augments Ago2−mediated Dicer−independent miRNA biogenesis and RNA interference.Nat Commun 6,7194(2015))。データ(エラーバー)は、平均値+SD(n=3以上)として表示される。スチューデントのt検定を使用してP値を決定した(tail=2)**:P<0.01、***:P<0.001。
火傷治療、急性創傷治療及び外科的創傷治療を含む、創傷感染の予防及び管理のための大面積の天然初代正常ヒト細胞膜を生成する新規の組織工学的方法について説明する。細胞マトリックス上で培養された正常初代ヒト上皮細胞は、核又はDNAを有していない大面積(最大500cm2)の初代正常ヒト細胞膜を生成し、この膜は創傷に適用されて微生物による創傷感染を防ぐための物理的バリアとして機能し得る。人工の大面積細胞膜−マトリックス複合層は、細菌、真菌、及びウイルスを含む微生物による感染を効果的に防ぐことができる。
大型初代正常ヒト細胞膜の増殖
様々なサイズ(75cm2、300cm2、500cm2、又はそれ以上)のディッシュ中のMatrigel(商標)マトリックス層(Yi,T.,Kabha,E.,Papadopoulos,E.,and Wagner,G.(2014)4EGI−1 targets breast cancer stem cells by selective inhibition of translation that persists in CSC maintenance,proliferation and metastasis.Oncotarget 5,6028−6037)を、Matrigel(商標)Basement Membrane Matrix(CB−40234、Corning社)を使用して薄いプラスチック膜上に調製し、ディッシュをあらかじめ冷却した後、37℃のインキュベーターで20分間インキュベートした。初代正常ヒト乳腺上皮細胞(HMEC)(Scheel,C.,Eaton,E.N.,Li,S.H.,Chaffer,C.L.,Reinhardt,F.,Kah,K.J.,Bell,G.,Guo,W.,Rubin,J.,Richardson,A.L.,and Weinberg,R.A.(2011)Paracrine and autocrine signals induce and maintain mesenchymal and stem cell states in the breast.Cell 145,926−940)は、DMEM培地のMatrigelマトリックス層上で培養した。哺乳類細胞は5%CO2、37℃で培養した。50時間の培養後、HMECは細胞塊及び大面積の初代正常ヒト細胞膜を形成する。細胞塊をチップで除去し、核の遺伝物質を有していない大面積の初代正常ヒト細胞膜を生成した。DMEM培地を除去した後、細胞膜を1×PBSで3回洗浄した。
この方法により生成された大面積の天然初代正常ヒト細胞膜(図15A〜図15B)は、1)単一細胞の面積と比較して、面積が10億倍(1010倍)増大している、超大面積(最大500cm2以上)の核を有していない天然細胞膜である、2)天然の初代正常ヒト細胞膜の二重層である、3)DNAの遺伝物質を含まない、4)天然膜タンパク質を含む天然脂質膜である、5)細胞膜及びマトリゲルマトリックスが容易に分解する、及び6)創傷感染を予防することを含む特徴を有する。

- 複数のマイクロフィラメントソース領域の間の連続的な格子構造又はメッシュ構造で相互接続された細胞由来のマイクロフィラメントを含むネットワーク。
- 前記マイクロフィラメントが細胞外マイクロフィラメントである、請求項1に記載のネットワーク。
- 前記マイクロフィラメントが膜で囲まれている、請求項1に記載のネットワーク。
- 前記マイクロフィラメントがアクチンを含む、請求項1に記載のネットワーク。
- 前記アクチンがβ−アクチンを含む、請求項4に記載のネットワーク。
- 前記マイクロフィラメントの長さが約1〜1000μmである、請求項1に記載のネットワーク。
- 前記マイクロフィラメントが分岐している、請求項1に記載のネットワーク。
- 前記マイクロフィラメントが約2〜10個の分枝を有する、請求項7に記載のネットワーク。
