Abstract
Description
(a)該蜜産生植物の細胞において融合毒素ペプチドをコードする外来遺伝子コンストラクトを発現させ、ここで、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該蚊標的化ペプチドはアエデス・アエギプティ(Aedes aegypti)蚊の腸上皮に結合し、該毒素ペプチドはHv1aクモ毒ペプチドであり、かつ、
(b)ターミネーターカセットを発現させ、ここで、該ターミネーターカセットは、該植物の蜜産生細胞以外の細胞中の核酸から、該外来遺伝子コンストラクトを排除し、該組換え植物は、蜜以外の組織においては該融合毒素ペプチドを発現しない。

Claims (44)
- 融合毒素ペプチドを含む殺蚊毒素において、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該融合毒素ペプチドは蚊に対して毒性である、殺蚊毒素。
- 蚊標的化ペプチドが、アエデス属(Aedes)、アノフェレス属(Anopheles)またはクレクス属(Culex)の蚊を標的とする、請求項1に記載の殺蚊毒素。
- 蚊標的化ペプチドが、アエデス・アエギプティ(Aedes aegypti)蚊を標的とする、請求項1に記載の殺蚊毒素。
- 蚊標的化ペプチドが、アエデス・アエギプティ蚊の腸上皮に結合する、請求項1に記載の殺蚊毒素。
- 蚊標的化ペプチドが、フラビウイルスのドメインIIIに由来するペプチドである、請求項1に記載の殺蚊毒素。
- 蚊標的化ペプチドが、デングウイルスの糖タンパク質のドメインIIIに由来するペプチドである、請求項1に記載の殺蚊毒素。
- 毒素ペプチドが、蚊に対する毒性を有するペプチドである、請求項1に記載の殺蚊毒素。
- 毒素ペプチドが、蚊標的化ペプチドと融合されていなければ蚊に対する毒性を示さないペプチドである、請求項1に記載の殺蚊毒素。
- 毒素ペプチドが、Hv1aクモ毒ペプチドである、請求項1に記載の殺蚊毒素。
- 融合毒素ペプチドが、他の生物に対して毒性を示さない、請求項1に記載の殺蚊毒素。
- 担体をさらに含む、請求項1に記載の殺蚊毒素。
- 殺蚊毒素を発現する組換え植物を製造する方法において、
該植物の標的細胞において、融合毒素ペプチドをコードする外来遺伝子コンストラクト発現させるよう誘導すること;ここで、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該融合毒素ペプチドは蚊に対して毒性である;および
該融合毒素ペプチドを発現する組換え植物を製造すること
を含む方法。 - 植物が蜜分泌植物であり、植物の標的細胞が蜜産生細胞であり、組換え植物は融合毒素ペプチドを該植物の蜜中に発現する、請求項12に記載の方法。
- 植物がインパチェンス・ワレリアナ(Impatiens walleriana)である、請求項13に記載の方法。
- 蚊標的化ペプチドが、アエデス属、アノフェレス属またはクレクス属の蚊を標的とする、請求項12に記載の方法。
- 蚊標的化ペプチドが、アエデス・アエギプティ蚊を標的とする、請求項12に記載の方法。
- 蚊標的化ペプチドが、アエデス・アエギプティ蚊の腸上皮に結合する、請求項12に記載の方法。
- 蚊標的化ペプチドが、フラビウイルスのドメインIIIに由来するペプチドである、請求項12に記載の方法。
- 蚊標的化ペプチドが、デングウイルスの糖タンパク質のドメインIIIに由来するペプチドである、請求項12に記載の方法。
- 毒素ペプチドが、蚊に対する毒性を有するペプチドである、請求項12に記載の方法。
- 毒素ペプチドが、蚊標的化ペプチドと融合されていなければ蚊に対する毒性を示さないペプチドである、請求項12に記載の方法。
- 毒素ペプチドが、Hv1aクモ毒ペプチドである、請求項12に記載の方法。
- 融合毒素ペプチドが、他の生物に対して毒性を示さない、請求項12に記載の方法。
- 外来遺伝子コンストラクトが、該植物に特異的なプロモーター、蚊標的化ペプチドをコードする遺伝子、および毒素ペプチドをコードする遺伝子を含む、請求項12に記載の方法。
- 外来遺伝子コンストラクトを発現させるよう誘導するステップが、該植物の少なくとも1つの細胞を該外来遺伝子コンストラクトで、融合毒素ペプチドを発現する組換え植物を生じるように形質転換することを含む、請求項12に記載の方法。
- 請求項12に記載の方法により製造される組換え植物。
- その蜜中に殺蚊毒素を発現する組換え植物を製造する方法において、
蜜を産生する該植物の細胞において、融合毒素ペプチドをコードする外来遺伝子コンストラクトを発現させるよう誘導すること;ここで、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該蚊標的化ペプチドが、デングウイルスの糖タンパク質のドメインIIIに由来するペプチドであり、該毒素ペプチドが、Hv1aクモ毒ペプチドであり、該植物がインパチェンス・ワレリアナである;および
蜜中に該融合毒素ペプチドを発現し、蜜以外の組織においては該融合毒素ペプチドを発現しない組換え植物を製造すること
を含む方法。 - 請求項27に記載の方法により製造される組換え植物。
- 組換え殺蚊植物において、該組換え植物は、該植物の標的細胞において、融合毒素ペプチドをコードする外来遺伝子コンストラクトを発現させ、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該融合毒素ペプチドは蚊に対して毒性である、組換え殺蚊植物。
- 植物が蜜分泌植物であり、植物の標的細胞が蜜産生細胞であり、組換え殺蚊植物は融合毒素ペプチドを該植物の蜜中に発現する、請求項29に記載の組換え殺蚊植物。
- 植物がインパチェンス・ワレリアナである、請求項30に記載の組換え殺蚊植物。
- 蚊標的化ペプチドが、アエデス属、アノフェレス属またはクレクス属の蚊を標的とする、請求項29に記載の組換え殺蚊植物。

- 組換え殺蚊インパチェンス・ワレリアナ植物において、該組換え植物は、蜜を産生する該植物の細胞において、融合毒素ペプチドをコードする外来遺伝子コンストラクトを発現させ、該融合毒素ペプチドは、毒素ペプチドに蚊標的化ペプチドが融合されたものを含み、該蚊標的化ペプチドは蚊の腸上皮に結合し、該毒素ペプチドはHv1aクモ毒ペプチドであり、該組換え植物は、蜜以外の組織においては該融合毒素ペプチドを発現しない、組換え殺蚊インパチェンス・ワレリアナ植物。
