式中、Wは、配列アライメントプログラムのCおよびDのアライメントによる同一性照合としてスコアされたヌクレオチドまたはアミノ酸の数であり、Zは、Dにおけるヌクレオチドまたはアミノ酸の総数である。配列Cの長さが配列Dの長さに等しくなく、Dに対するCの配列同一性%は、Cに対するDの配列同一性%に等しくないことが理解されよう。当技術分野の技能の範囲内の様々な方法で、例えば、BLAST、BLAST−2、ALIGN、ALIGN−2、またはMegalign(DNASTAR)ソフトウェアなどの公的に入手可能なコンピュータソフトウェアを用いて、配列同一性%を決定する目的のためのアライメントを得ることができる。
図2に関連して、インビトロでの非ウイルストランスフェクションおよびリプログラミング実験は、1週間未満で、ヒトおよびマウス初代線維芽細胞を誘導内皮細胞に効率的にリプログラミングした、遺伝子Etv2、Foxc2、およびFli1(EFF)の共トランスフェクションであることを示した。これらの実験において、HDAF細胞は、非ウイルス性にEFFをトランスフェクトされた。トランスフェクトされた細胞の蛍光顕微鏡写真は、内皮マーカーPecam−1の強力な発現、ならびに線維芽細胞のマーカーFSPの発現低下を示した(t=トランスフェクション7日後)。2つの異なるトランスフェクション条件(Etv2単独対EFFの共トランスフェクション)についての内皮マーカーの遺伝子発現解析。結果は、遺伝子発現において顕著な差を示し、Etv2単独と比較して、EFFは、トランスフェクション後7日目に著しくより強力な内皮遺伝子発現をもたらした。管形成アッセイから得られた結果は、内皮細胞に相当するマトリゲル中で培養した場合、EFFトランスフェクション細胞が血管様構造を形成することができたことを示した(HMEC、陽性対照)。他方では、対照HDAF細胞は、マトリゲルで培養した場合、管状構造を形成することができなかった。非ウイルス性にEFFをトランスフェクトされたMEF細胞はまた、トランスフェクション7日後という早い時期に内皮マーカー発現を示した。非ウイルス性にEFFをトランスフェクトされたtdTomato−MEF細胞は、NSGマウスの側腹部注射後に血管形成を助長した。これらのデータを以下の表1にさらに要約する。
図1に関連して、内皮細胞リプログラミングを誘導する誘導するEFFの有効性がインビトロで確立されたら、インビボでのリプログラミング方法を試験した。わずか数秒続く、C57BL/6マウスの背部皮膚の1回限りの処理は、対照皮膚に対するPecam−1およびvWFの発現における有意な増加により明示されるように、7日目までに皮膚組織の血管新生の増加をもたらした。高解像度レーザースペックル画像は、EFF TNT処理した領域への灌流(血流)の増強を経時的に示した。皮膚表面から3mmほどの拍動挙動を有する表層血管(破線円)の存在を確認するEFF TNT処理した皮膚の超音波画像であり、親循環系との吻合が成功したことを示す。
図3に関連して、さらなる実験が、後肢虚血C57BL/6マウスモデルにおいて、EFF送達が肢全体の救済をもたらしたことを実証した。わずか数秒続く大腿皮膚の1回限りの処理は、大腿動脈の切断後、肢再灌流の増加をもたらした。灌流は、虚血性対正常/対側肢の比に基づいて計算した。対照肢は、14日目にEFF TNT処理した肢と比較して、より顕著な組織壊死の徴候を示した。筋肉エネルギーのNMRベースの測定値によって、対照と比較して、EFF TNT処理した肢についてのATPおよびPCrレベルの増加を確認した。腓腹筋の免疫蛍光分析は、14日目に血管新生の増強を示した。
図4を参照して、損傷誘導性肢虚血からより多くの有害な副作用を経験する傾向を有する、Balb/cマウスでの追加の実験によって、EFF処理が良好な肢灌流およびそれに続いて壊死および自然切断からの救済をもたらしたことを示した。肢のレーザースペックル画像は、EFF TNT処理後の再灌流の成功を示した。図4のパネル(b)は、TNT処理の有無による虚血肢の肉眼的変化を示す。
図5に関連して、PCR分析によって、EFF mRNA/cDNAトランスフェクションに加えて、血管新生促進VEGFおよびbFGFmRNAを前負荷したように見える、EFF TNT処理した背部皮膚を、細胞外小胞(EV)が単離したことが明らかとなった。このことは、EFF TNT処理した皮膚由来のEVが、標的組織全体にEFFリプログラミングシグナルを伝播するための生存機構を表すだけでなく、トランスフェクション後の最初の数時間以内に血管新生促進シグナルを拡散することによって適所の前条件づけの役割を果たし得ることを示唆している。図5のパネル(a)は、損傷/EV媒介性の救済の概略図を示す。qRT−PCRを使用して、EV含量を特徴付けた。図5のパネル(b)は、EFF処理した皮膚のレーザースペックル再灌流分析を示す。EV処理した肢についての血管新生の増加を示す腓腹筋の免疫蛍光分析。
図6に関連して、K14−CreレポーターおよびCol1A1−eGFPマウスモデルを用いた実験によって、リプログラミングした細胞集団は大部分が真皮起源であることを確認した。Col1A1−GFPマウスモデルからのEFF TNT処理した皮膚切片の蛍光顕微鏡写真であり、Col1A1由来の皮膚細胞(緑色)がまたPecam−1内皮マーカーも発現することを示した。GFPトレーサーとPecam−1の両方に免疫反応性であった細胞要素をLCM/qRT−PCRでさらに分析した。結果は、このようなダブルポジティブな要素が著しく高い内皮マーカーの遺伝子発現を有することを示す。GFP+/CD31+細胞要素のLCM/qRT−PCR測定によって、内皮マーカーの発現増加を確認した。
TNTプラットフォームの製造
TNT装置は、薄く(約200μm)両面研磨した(100)シリコンウエハから製造された。簡潔に説明すると、まず、厚さ約1.5μmのAZ5214Eフォトレジスト層をシリコンウエハ上に約3000rpmでスピンコートした。続いて、GCA6100Cステッパーを使用して、フォトレジスト上にナノスケール開口部をパターン形成した。100mmウエハ当たり最大16ダイのナノスケール開口部アレイをパターン形成した。次いで、そのような開口部をエッチングマスクとして使用して、深反応性イオンエッチング(DRIE)(Oxford Plasma Lab 100 システム)によって、シリコン表面上に約10μmの深さのナノチャネルを穿孔した。最適化したエッチング条件には、SF6ガス:13秒/100sccmのガス流/700WのICP電力/40WのRF電力/30mTのAPC圧力、C4F8ガス条件:7秒/100sccmのガス流/700WのICP電力/10WのRF電力/30mTのAPC圧力が含まれる。次いで、マイクロスケールのリザーバーを、接触フォトリソグラフィおよびDRIEを介してウエハの裏側にパターン形成した。最後に、TNTプラットフォーム表面上に、厚さ約50nmの窒化ケイ素の絶縁層/保護層を配置した。
雄性C57BL/6マウス(8〜10週齢)は、Harlan Laboratoryから入手した。Jackson laboratoriesから入手した、B6.129(Cg)−Gt(ROSA)26Sortm4(ACTB−tdTomato,−EGFP)Luo/JマウスをK14creと交配させて、K14cre/Gt(ROSA)26Sortm4(ACTB−tdTomato−EGFP)Luo/Jマウスを作製した。pOBCol3.6GFPtpzマウスは、Traci Wilgus博士(The Ohio State University)から寄贈された。ROSAmT/mGマウスの遺伝子型判定PCRは、プライマーoIMR7318−CTC TGC TGC CTC CTG GCT TCT(配列番号8)、oIMR7319−CGA GGC GGA TCA CAA GCA ATA(配列番号9)、およびoIMR7320−TCA ATG GGC GGG GGT CGT T(配列番号10)を用いて行ったが、K−14 Cre導入遺伝子はプライマーoIMR1084−GCG GTC TGG CAG TAA AAA CTA TC(配列番号11)、oIMR1085−GTG AAA CAG CAT TGC TGT CAC TT(配列番号12)。を用いて確認した。オハイオ州立大学のLaboratory Animal Care and Use Committeeに承認されたプロトコルに従って、すべての動物試験を実施した。動物にタグを付け、コンピューターベースのアルゴリズムを用いて無作為にグループ分けした。
初代ヒト成人皮膚線維芽細胞(ATCC PCS−201−012)を、線維芽細胞増殖キット−無血清(ATCC PCS 201−040)およびペニシリン/ストレプトマイシンを補充した線維芽細胞基礎培地中で増殖させた。E12.5−E14マウス胚性線維芽細胞(MEF)を、10%ウシ胎仔血清を添加したDMEM/F12中で培養した。非ウイルス細胞トランスフェクションおよびリプログラミング実験は、過去に報告されているように、3Dナノチャネルエレクトロポレーション(NEP)を介して実施した。簡潔に説明すると、細胞を最初に3DNEP装置上で一晩完全にコンフルエントになるまで増殖させた。続いて、パルス電場を用いて、Fli1:Etv2:Foxc2の1:1:1混合物からなる細胞中にプラスミドのカクテル(0.05μg/μl)を送達した。次いで、細胞をプラスミド送達の24時間後に回収し、EGM−2 MV Single Quotキット(CC−4147、Lonza)を補充したEBM−2基礎培地(CC−3156、Lonza)に入れ、実験/測定用にさらに処理した。Etv2およびFli1プラスミドは、
TNTの24〜48時間前に、まず処理対象の領域を麻痺させた。次いで、皮膚を剥離して、死細胞層/ケラチン細胞層を除去して、表皮の有核細胞を曝露した。TNT装置を剥離した皮膚表面上に直接配置した。EFFプラスミドカクテルを0.05〜0.1μg/μlの濃度でリザーバーに入れた。金被覆電極(すなわちカソード)をプラスミド溶液に浸漬し、一方、24G針対電極(すなわちアノード)を皮内に挿入し、TNTプラットフォーム表面に並置した。次いで、パルス電気刺激(すなわち、振幅250Vの10パルスおよび1パルス当たり10msの持続時間)を電極両端に印加して、露出した細胞膜をナノ穿孔し、ナノチャネルを通して細胞内にプラスミドカーゴを打ち込んだ。
片側後肢虚血は閉塞とそれに続く大腿動脈の切断によって誘発された。簡潔に説明すると、8〜10週齢のマウスを1〜3%イソフルランで麻酔し、加熱したパッド上の実体顕微鏡(Zeiss OPMI)下に仰臥位に置いた。大腿動脈を露出させ、約1cmの切開を通して大腿静脈から分離した。近位端および遠位端の閉塞を7−0絹縫合糸で誘導した後、動脈の完全なトランスフェクションを行った。最後に、ブプレノルフィンの単回用量を皮下投与して痛みを制御した。手術の2時間後にレーザースペックル画像(MoorLDI−Mark2)を実施して血流閉塞の成功を確認した。
EVを、OCTブロックで採取し、後で使用するために凍結保存された、直径12mmの皮膚生検から単離した。簡潔に説明すると、ブロックを解凍し、リン酸緩衝食塩水(PBS)で洗浄してOCTを除去した。外科用メスを用いて脂肪組織の除去後に、皮膚組織を約1mm片に刻んで、PBS中でマイクログラインダーを用いてホモジナイズした。3000gで遠心分離した後、Exoquickキット(System Biosciences)を1:5の比(Exoquick:上清)で使用して、上清からEVを4℃で12時間単離した。EVを1500gで30分間の遠心分離により沈殿させた。次いで、製造業者によって提供された推奨に従って、mirVanaキット(Life technologies)を使用して、全RNAをペレットから抽出した。
プラスミドDNA精製キット(Qiagen Maxi−prep、カタログ番号12161、およびClontech Nucleobondカタログ番号740410)を用いて、EFFプラスミドを調製した。DNA濃度をNanodrop 2000c Spectrophotemeter(Thermoscientific)から得た。プラスミドDNA構築物およびそれらの供給源のリストについては、表2を参照されたい。
PALM Technologies(Bernreid、Germany)のレーザー顕微解剖システムを使用してLCMを実施した。形態および/または免疫染色に基づいて同定された組織切片の特定領域を切断し、20倍接眼レンズで捕捉した。試料を25μlの細胞直接溶解抽出緩衝液(Invitrogen)に放出した。約1,000,000μm2の組織領域を各キャップに捕捉し、次いで溶解物をさらなる処理のために−80℃で保存した。LCM試料のqRT−PCRは製造業者の指示に従って細胞直接溶解緩衝液から実施した。プライマーのリストを表3に提供する。
特異的抗体および標準的な手順を用いて、組織免疫染色を実施した。簡潔に説明すると、OCT包埋組織を10μmの厚さで凍結切片化し、冷アセトンで固定し、10%正常ヤギ血清でブロックし、そして特異的抗体と共にインキュベートした(表4)。続いて、蛍光標識した適切な二次抗体(Alexa488標識α−モルモット、1:200、Alexa488標識α−ウサギ、1:200、Alexa568標識α−ウサギ、1:200)とのインキュベーションによってシグナルを可視化し、DAPIで対比染色した。レーザー走査共焦点顕微鏡(Olympus FV1000 filter/spectral)で画像を捕捉した。
磁気共鳴血管造影法を用いて、マウスにおける本発明者らのMCAOモデルを検証し、効果的なMCAOのために閉鎖栓サイズおよび内頸動脈挿入距離を最適化した。9.4TのMRI(Bruker Corporation、Bruker BioSpin Corporation、Billerica,MA,USA)を使用して、MCA再灌流の48時間後に麻酔をかけたマウスに対してT2強調MRIを実施した。MR画像は、以下のパラメータを用いた緩和強調の高速取得(RARE)シーケンスを用いて取得した:視野(FOV)30×30mm、取得マトリックス256×256、TR3,500ms、TE46.92ms、スライスギャップ1.0mm、希少因子8、平均数3。mm当たり8.5ピクセルの解像度。未加工のMR画像を標準DICOMフォーマットに変換し、処理した。Osirix v3.4を用いた画像の適切なソフトウェアコントラスト強調の後、各冠状脳スライスにおける梗塞領域を描写するために、デジタル面積測定がマスクされた観察者によって行われた。過去に報告されているように、脳切片からの梗塞面積を合計し、切片の厚さを乗じ、浮腫誘発性腫脹について補正して、梗塞体積を決定した(KhannaS,et al.J.Cereb Blood Flow Metab 2013、33(8):1197−1206)。
筋肉エネルギーは、RF送信用の体積コイルおよび受信用の31Pコイルを使用して、9.4Teslaスキャナー(Bruker BioSpec)でNMR分光測定を評価した。特注の1H/31Pトランシーバーコイルアレイでインビボ画像化を実施した。単一パルス系列を用いてデータを得た。生データをノイズ低減のためにウィンドウ処理し、スペクトル領域にフーリエ変換した。
血管形成を超音波画像によって並行モニターした。簡潔に説明すると、Vevo2100システム(Visual Sonics、Toronto、ON、Canada)を使用して、MS250リニアアレイプローブを用いてBモードで超音波画像を得た。ドップラーカラーフロー画像をモニターに実装し、収縮期と拡張期下で血流特性を定量化した。
試料をコード化し、データ解析を盲検法で実施した。動物実験では、データは少なくとも3匹の動物の平均±SDとして報告されている。インビトロデータは、3〜6回の実験の平均±SDとして報告されている。すべての統計はSigmaPlotバージョン13.0で実施した。
結果
miR−200b単独の阻害は、培養線維芽細胞を、内皮細胞(iEC)を誘導するように変換した。
この一連の調査は、慢性創傷患者の創傷縁でmiR−200bレベルが皮膚内のそれよりも急激に低いという観察に始まった(図12A)。マウスにおいて、創傷は、創傷閉鎖後に、創傷形成前の値に反発したmiR−200bの一過性の阻害を誘導した(図12B)。miR−200bのそのような創傷誘導性阻害の重要性を理解するために、抗miR−200b阻害剤分子を、ナノチャネルベースのエレクトロポレーションによってヒト成人皮膚線維芽細胞に送達した(Boukany et al.,2011、Gallego−Perez et al.,2016)。敷石状形態への細胞構造の頑強な表現型の切り替えは、線維芽細胞の内皮細胞への変換を予備的に示唆していることを意味した(図7A)。線維芽細胞におけるmiR−200b阻害は、4日目以降、線維芽細胞特異的CD90よりも内皮マーカーCD31を顕著に誘導した。そのような移行は28日目に最大となる進行性であり(98.4%)、線維芽細胞における急速な出現および内皮特性の維持を示した(図7Bおよび7C)。この観察は、線維芽細胞FSP1+細胞に対する別の血管新生因子VEGFR2+の漸進的出現と一致した(図7Cおよび12C)。線維芽細胞FSP−1の付随的な喪失とカップリングさせた内皮マーカーCD31の増加は、miR−200b抑制細胞において明らかであった(図12D)。抗miR200bをトランスフェクトした線維芽細胞のトランスクリプトームアレイは、血管新生のCCL2(Stamatovic et al.,2006)およびCXCL8(Heidemann et al.,2003)によって表される内皮遺伝子クラスターに対するCol1A、MMP、CXCL5などの線維芽細胞特異的遺伝子から発現プロファイルのシフトを示した(図7Dおよび12E〜12F)。iECの特徴付けは、線維芽細胞(COL1A1およびFSP1)マーカーの痕跡が残っている動脈(PECAM1、VEGFR2、およびTIE2)、静脈(COUP−TFII)、リンパ管(PROX1)の特性の共存を明らかにした(図7E)。ヒト微小血管内皮細胞と同様に、miR−200b抑制した線維芽細胞において、Ac−LDL取り込み(図7F)およびマトリゲル管形成は高かった(図7G)。したがって、線維芽細胞は、miR−200b阻害に応答して内皮細胞の機能的特徴を達成した。Oct4、Sox2、Klf4およびNanogなどの多能性遺伝子は、抗miR−200bトランスフェクションの4日目以降、非常に低いレベルに留まり(図12G)、これは線維芽細胞からiECへの変換が直接的であることを示している。
TargetScan、miRanda、およびDiana−MicroTアルゴリズムを使用したインシリコ試験によって、血管新生転帰を調節し得るmiR−200bの標的が予測された。フレンド白血病統合体1(Fli−1)転写因子の3’−非翻訳領域(3’UTR)は、miR−200bに対する結合部位を含む(図13A)。miR−200b模倣物の送達は、Fli−1−3’UTRレポータールシフェラーゼ活性を有意に抑制した(図8A)。そのような効果は、変異Fli−13’UTRを有する細胞において無効にされ(図8A)、Fli−1発現の調節におけるmiR−200b結合の特異性の重要性を認識した。したがって、血管発達および血管新生に関与する中心的制御因子である転写因子のETSファミリーのメンバーであるFli−1は、miR−200bによって転写後遺伝子サイレンシングを受ける(Meadows et al.,2011、De Val et al.,2009)。初代ヒト皮膚線維芽細胞においてmiR−200bがFli−1を標的とするという概念に対する直接的な支持は、miR−200b模倣物または阻害剤を用いた試験において得られた(図13B)。miR−200b模倣物はFli−1タンパク質レベルを低下させ、対照的に、miR−200b阻害剤をトランスフェクトした線維芽細胞においてFli−1タンパク質が誘導された(図8B)。miR−200b阻害線維芽細胞によって引き起こされる血管新生転帰におけるFli−1の有意性を決定するために、抗miR−200bまたはFli−1 siRNA単独もしくはそれを組み合わせてのいずれかでトランスフェクトした。miR−200b単独の阻害はFli−1タンパク質の脱サイレンシングにおいて強力であったが、そのような効果はFli−1ノックダウンを受けた細胞では鈍かった(図8C)。これらの知見と一致して、マトリゲルアッセイにおける内皮管長に対するmiR−200b阻害の血管新生作用はFli−1依存的であることが観察された(図8D)。
miR−200b阻害によって引き起こされる血管新生の経路において、Fli−1作用の下流の成分は、プロモーター分析のためのMatInspectorソフトウェアを使用して特徴付けられた。Etv2プロモーター領域は、Etv2の活性化に必要な8つの既知のETS結合部位を含む(図13Cおよび13D)。クロマチン免疫沈降(ChIP)アッセイは、Fli−1サイレンシングした皮膚線維芽細胞において、Etv2プロモーターへのFli−1の結合増強が抑制されたが、Fli−1の強制発現は、Fli−1によるEtv2プロモーターの占有を改善したことを示した。(図8E)。これらの結果は、それぞれmiR−200b阻害剤もしくは模倣物で処理した、Fli−1サイレンシング細胞またはFli−1過剰発現細胞において、Etv2プロモーター−レポーターアッセイを用いて裏付けられた。miR−200b阻害剤をトランスフェクトした細胞ではレポーター活性の増加が検出された。Fli1ノックダウンに応答したそのような活性の鈍化は、Etv2トランス活性化におけるFli−1の重要性を強調した。一貫して、Fli−1過剰発現細胞におけるEtv2プロモーター活性の増加は、細胞をmiR−200b模倣物およびFli−1強制発現ベクターで共トランスフェクトしたときに完全に消失した(図8F)。iECにおけるFli−1のサイレンシングまたは脱サイレンシングは、それぞれ、Etv2 mRNA発現を下方制御または上方制御した(図8G)。したがって、miR−200b阻害はFli−1を脱サイレンシングし、それによって血管新生スイッチを活性化するためにEtv2を上方制御する(図13E)。
インビボでの皮膚線維芽細胞のiECへの直接変換は、免疫が十分なC57BL/6マウスの無傷皮膚におけるmiR−200bの阻害によって達成された。皮膚への抗miR200b−LNAの局所ナノエレクトロポレーションに基づく送達は、Fli−1を脱サイレンシングした(図9A)。同時に、背部皮膚における血流の改善が観察された。そのような改善は一過性であり、miR−200b阻害の7日目にピークを示した後、その後の4日間の誘導灌流を低下させた(図14A)。無傷皮膚において誘発された血管構造の適時の後退を指すこの一連の証拠は、インビボで生じるiECによる奇形腫形成の可能性に不利になる。適切に灌流されている無傷の健康な皮膚とは異なり、誘導灌流の重要性は血管新生が必要な創傷では異なる(Tonnesen et al.,2000)。組織修復の生理学における直接細胞変換の不可欠な役割の概念と一致して、損傷それ自体が創傷縁組織においてmiR−200b阻害を引き起こした(図12A)。そのような阻害は、創傷後7日目および9日目の創傷縁組織におけるFli−1発現の付随的増加と関連していた(図9B)。インビボでの細胞変換の直接的証拠の探索は、Fsp1−Cre:R26RtdTomatoマウスを用いた系統追跡試験を必要とした(Ubil et al.,2014)。損傷後5日目は、CD31+である線維芽細胞系列の移行細胞の豊富な存在によって特徴付けられた(図9C)。レーザー捕捉顕微解剖(LCM)によって単離された創傷縁線維芽細胞(図14B)は、内皮マーカーCD31の増加と共に弱毒化FSP1発現などの移行特性を示した(図9D)。これは、線維芽細胞が、miR−200bが阻害される損傷部位でiECに変換されるという直接的証拠を構成する。しかしながら、線維芽細胞から内皮細胞へのそのような損傷誘導性のmiR−200b依存性の生理学的変換は、このような血管性細胞変換に対する障壁として糖尿病性病態を認識する糖尿病マウスでは損なわれた(図9E)。
損傷部位での線維芽細胞のiEC変換への変換におけるFli−1の有意性を試験するために、Cre/loxP制御RNA干渉を利用してマウスにおける条件付き線維芽細胞特異的遺伝子ノックダウンを得た(Hitz et al.,2007、Kasim et al.,2004年)。線維芽細胞特異的Fli−1は、LoxP隣接Fli−1 shRNA発現カセットによってインビボでノックダウンされた(図10A)。4つのLoxP隣接Fli−1 shRNA発現カセットを設計し、インビトロでのFli−1タンパク質発現の下方制御の効率に基づいており(図15Aおよび15B)、3つのカセットをプールし、Fsp1−Creマウスの創傷縁でのレンチウイルストランスフェクションに使用した。Fli−1 shRNAベクターの検証は図15Bおよび15Cに報告されている。創傷縁組織における抗miR200b−LNAの送達(図10B)は、線維芽細胞のiECへの変換におけるmiR−200bの役割を支持してFSP1およびCD31の共局在化の増加を示した(図10Cおよび10D)。このような共局在化は、Fli−1がmiR−200b機能の重要なメディエータとして関与しているFli−1の線維芽細胞標的ノックダウンによって著しく鈍かった(図10Cおよび10D)。実際、同じ実験条件下で、皮膚線維芽細胞におけるFli−1ノックダウンは、創傷灌流を有意に減衰し(図15Dおよび15E)、創傷閉鎖を損なった(図15F〜15H)。これらの結果は、miR−200b阻害によって引き起こされる損傷部位の線維芽細胞由来の成熟iECが、組織の血管新生に役立つFli−1依存性であることを示す。
創傷治癒障害は一般的な糖尿病性合併症である(Brem and Tomic−Canic、2007)。非糖尿病患者と比較して、糖尿病患者の創傷縁は、著しく上昇したmiR−200bの存在量を示したが、Fli−1 mRNAレベルは低かった(図11A)。免疫組織化学試験は、非糖尿病のヒト対象と比較して、糖尿病患者の創傷縁組織においてFli−1タンパク質の存在量が乏しいことを明らかにした(図11B)。II型糖尿病の確立されたモデルであるdb/dbマウスでは、創傷はmiR−200b発現を抑制することができなかった(図16Aおよび16B)。db/dbマウスの創傷縁に単一の抗miR200b−LNA分子を送達することによる強制的なmiR−200b阻害(図11C)は、Fli−1(図11D)およびその下流のEtv2タンパク質発現(図16C)の有意な増強をもたらし、続いて糖尿病マウスの創傷縁組織にiECの存在量が出現した(図11E)。そのような応答は、改善された創傷灌流および治癒をもたらした(図11F〜11Iおよび16D〜16E)。改善された血管新生はまた、db/dbマウスの抗miR200b処理した創傷縁組織におけるより高い存在量のCD31+およびCD105+内皮細胞によっても明らかであった(図11Jおよび16F〜16G)。要約すると、この研究によって、損傷中に一時的にオフにするとiECへの直接細胞変換のためのFli−1−Etv2軸を誘導する、重要なスイッチとして皮膚線維芽細胞のmiR−200bを認識する新しいパラダイムが紹介される。

細胞療法はいくつかの条件に対する有望な戦略を表すが、現在のアプローチは、限られた細胞供給源および煩雑な前処理工程(例えば、単離、誘導多能性)の必要性を含む主要な翻訳障害に直面している(RosovaI,et al.Stem Cells 2008、26(8):2173−2182、Kinoshita M,et al.Atherosclerosis 2012,224(2):440−445、Losordo DW,et al.Circulation 2004,109(22):2692−2697、LeeAS,et al.NatMed2013,19(8):998−1004、Cunningham JJ,et al.Nat Biotechnol 2012,30(9):849−857、Leduc PR,et al.Nat Nanotechnol 2007,2(1):3−7)。インビボ細胞リプログラミングは、容易に入手可能な細胞源(例えば、線維芽細胞)を利用し、エクスビボ前処理の必要性を回避することによって、より効果的な細胞ベースの治療を可能にする可能性を有する(HeinrichC,et al.Nat Cell Biol 2015,17(3):204−211、Karagiannis P,et al.Nat Methods 2014,11(10):1006−1008)。しかしながら、既存のリプログラミング方法論は、注意をはらんでおり、ウイルストランスフェクションに対する強い依存度を含む(GrandeA,et al.Nat Commun 2013、4:2373、Morita R,et al.Proc Natl Acad Sci USA2015,112(1):160−165)。さらに、カプシドサイズの制限および/または現状のアプローチの確率的性質(ウイルス性および非ウイルス性)はさらなる制限を課し、したがってより安全でより決定論的なインビボリプログラミング方法の必要性を強調している(Gallego−Perez D,et al.Nanomedicine 2016,12(2):399−409、Marx V.Nat Meth 2016,13(1):37−40)。それぞれ確立されたおよび新しく開発された誘導性ニューロンおよび内皮のリプログラミングモデルで検証されたナノチャネル化装置を介して、局所的に組織をリプログラミングするために新規でありながら実施が簡単な非ウイルスアプローチを開示する。このアプローチの簡潔さおよび有用性は、損傷誘導性虚血の2つのマウスモデルを使用して壊死組織および四肢全体を救済することによって実証される。
TNTプラットフォームの製造
TNT装置を、薄く(約200μm)両面研磨した(100)シリコンウエハから製造した(図20)。簡潔に説明すると、まず、厚さ約1.5μmのAZ5214Eフォトレジスト層をシリコンウエハ上に約3000rpmでスピンコートした。続いて、GCA6100Cステッパーを使用して、フォトレジスト上にナノスケール開口部をパターン形成した。100mmウエハ当たり最大16ダイのナノスケール開口部アレイをパターン形成した。次いで、そのような開口部をエッチングマスクとして使用して、深反応性イオンエッチング(DRIE)(Oxford Plasma Lab 100 システム)によって、シリコン表面上に約10μmの深さのナノチャネルを穿孔した。最適化したエッチング条件には、SF6ガス:13秒/100sccmのガス流/700WのICP電力/40WのRF電力/30mTのAPC圧力、C4F8ガス条件:7秒/100sccmのガス流/700WのICP電力/10WのRF電力/30mTのAPC圧力が含まれる。次いで、マイクロスケールのリザーバーを、接触フォトリソグラフィおよびDRIEを介してウエハの裏側にパターン形成した。最後に、TNTプラットフォーム表面上に、厚さ約50nmの窒化ケイ素の絶縁層/保護層を配置した。
C57BL/6マウスはHarlan Laboratoryから入手した。Jackson laboratoriesから入手した、B6.129(Cg)−Gt(ROSA)26Sortm4(ACTB−tdTomato,−EGFP)Luo/JマウスをK14creと交配させて、K14cre/Gt(ROSA)26Sortm4(ACTB−tdTomato−EGFP)Luo/Jマウスを作製した。pOBCol3.6GFPtpzマウスは、Traci Wilgus博士(The Ohio State University)から贈与された。repTOPTM mitoIREマウスは、Charles River Laboratoriesから入手した。Fsp1−Creマウスを入手した(University of California,Los Angeles)。Fsp1−CreマウスをB6.Cg−Gt(ROSA)26Sortm9(CAG−tdTomato)Hze/Jマウス(Jackson laboratories)と交配させて、線維芽細胞に特異的なtdTomato発現を有するマウスを作製した。研究の時点ではすべてのマウスは雄性であり、8〜12週齢であった。ROSAmT/mGマウスの遺伝子型判定PCRは、プライマーoIMR7318−CTCTGCTGCCTCCTGGCTTCT、oIMR7319−CGACAAGCAATA、およびoIMR7320−TCAATGGGCGGGGGTCGTTを用いて行い、K−14Cre導入遺伝子は、プライマーoIMR1084−GCGGTCTGGCAGTAAAAACTATC、oIMR1085−GTGAAACAGCATTGCTGTCACTTを用いて行った。Fsp1−Creマウスの遺伝子型判定PCRは、順方向−CTAGGCCACAGAATTGAAAGATCT、逆方向−GTAGGTGGAAATTCTAGCATCATCC(野生型、生成物長さ=324bp)、および順方向−GCGGTCTGGCAGTAAAAACTATC、逆方向−GTGAAACAGCATTGCATTGCTGTCACTT(CRE導入遺伝子、生成物長さ=100bp)のプライマーを用いて行い、td tomatoについては、順方向−AAGGGAGCTGCAGTGGAGTA、逆方向−CCGAAAATCTGTGGGAAGTC(野生型、生成物長さ=196bp)、および順方向−GGCATTAAAGCAGCGTATCC、逆方向−CTGTTCCTGTACGGCATGG(変異型、生成物長さ=297bp)のプライマーを用いて行った。オハイオ州立大学のLaboratory Animal Care and Use Committeeに承認されたプロトコルに従って、すべての動物試験を実施した。試料サイズを事前に決定するために統計的方法は使用されなかった。検出力分析は、この試験に必須ではない。動物にタグを付け、コンピューターベースのアルゴリズムを用いて無作為にグループ分けした。(www.random.org)
マイコプラズマ不含かつ認証された、初代ヒト成人皮膚線維芽細胞(ATCC PCS−201−012)を、ATCCから直接購入した。さらなる細胞株認証/試験は行われなかった。これらの細胞を、線維芽細胞増殖キット無血清(ATCC PCS 201−040)およびペニシリン/ストレプトマイシンを補充した線維芽細胞基礎培地中で増殖させた。E12.5−E14マウス胚性線維芽細胞(MEF)を、10%ウシ胎仔血清を添加したDMEM/F12中で培養した。非ウイルス細胞トランスフェクションおよびリプログラミング実験は、過去に報告されているように11、3Dナノチャネルエレクトロポレーション(NEP)を介して実施した。簡潔に説明すると、細胞を最初に3DNEP装置上で一晩完全にコンフルエントになるまで増殖させた。続いて、パルス電場を用いて、Fli1:Etv2:Foxc2の1:1:1混合物からなる細胞中にプラスミドのカクテル(0.05μg/μl)を送達した。次いで、細胞をプラスミド送達の24時間後に回収し、EGM−2 MV Single Quotキット(CC−4147、Lonza)を補充したEBM−2基礎培地(CC−3156、Lonza)に入れ、実験/測定用にさらに処理した。Etv2およびFli1プラスミドを入手した(Department of Internal Medicine,UT Southwestern Medical Center,Texas)。Foxc2プラスミドは、Tsutomu Kume博士(Department of Medicine−Cardiology and Pharmacology,Northwestern University−FCVRI,Chicago)のご好意により寄付された。
TNTの24〜48時間前に、まず処理対象の領域を麻痺させた。次いで、皮膚を剥離して、死細胞層/ケラチン細胞層を除去して、表皮の有核細胞を曝露した。TNT装置を剥離した皮膚表面上に直接配置した。ABMまたはEFFプラスミドカクテルを0.05〜0.1μg/μlの濃度でリザーバーに入れた。金被覆電極(すなわちカソード)をプラスミド溶液に浸漬し、一方、24G針対電極(すなわちアノード)を皮内に挿入し、TNTプラットフォーム表面に並置した。次いで、パルス電気刺激(すなわち、振幅250Vの10パルスおよび1パルス当たり10msの持続時間)を電極両端に印加して、露出した細胞膜をナノ穿孔し、ナノチャネルを通して細胞内にプラスミドカーゴを打ち込んだ。ABMプラスミドは、過去に報告されているように11、2:1:1のモル比で混合した。特に指定しない限り、対照検体は、ブランク、リン酸緩衝生理食塩水(PBS)/疑似プラスミド溶液を用いたTNT処理を含んだ(図38)。
細胞外記録の一般的原理を用いて、皮膚における電気生理学的活性を検出した。クロノアンペロメトリー測定を、鎮静マウスの皮膚上の2つの小さな切開を通してニューロンの興奮性を検出するために、PPyベースのプローブを使用して実施した。
一過性限局性脳虚血はマウスにおいて中大脳動脈閉塞(MCAO)により誘導され、過去に報告されているように、腔内フィラメント挿入技術を使用することにより達成された(Khanna S,et al.J Cereb Blood Flow Metab 2013、33(8):1197−1206)。浮腫を補正した後、対側半球の割合として梗塞サイズを決定するためにMRI画像を使用した。
C57BL/6マウスの背部皮膚上に、20mm×10mmの大きさの単茎(すなわち、ランダムパターン)虚血皮弁を作製した。簡潔に説明すると、8〜10週齢のマウスを1〜3%イソフルランで麻酔した。背側を麻痺させ、洗浄し、ベタジンで滅菌した。20mm長の全層性平行切開を10mm離すことによって、マウスの背部皮膚上に単茎皮弁を作製した。皮の底部は、自由に垂れ下がる皮弁を作るために切断された。皮弁縁を焼灼した。0.5mmシリコンシートを皮弁の下に置き、次に5−0エチコン絹縫合糸で隣接する皮膚に縫合した。最後に、ブプレノルフィンの単回用量を皮下投与して痛みを制御した。手術の2時間後にレーザースペックル画像(Perimed)を実施して血流閉塞の成功を確認した。TNTベースのトランスフェクションは、皮膚皮弁の24時間前に行った。
片側後肢虚血は、閉塞およびその後の大腿動脈の切断とそれに続く切断によって誘発された(Limbourg A,et al.Nat Protoc2009、4(12):1737−1746)。簡潔に説明すると、8〜10週齢のマウスを1〜3%イソフルランで麻酔し、加熱したパッド上の実体顕微鏡(Zeiss OPMI)下に仰臥位に置いた。大腿動脈を露出させ、約1cmの切開を通して大腿静脈から分離した。近位端および遠位端の閉塞を7−0絹縫合糸で誘導した後、動脈の完全な処置を行った。最後に、ブプレノルフィンの単回用量を皮下投与して痛みを制御した。手術の2時間後にレーザースペックル画像(MoorLDI−Mark2)を実施して血流閉塞の成功を確認した。
EVを、OCTブロックで採取し、後で使用するために凍結保存されていた、直径12mmの皮膚生検から単離した。簡潔に説明すると、ブロックを解凍し、PBSで洗浄してOCTを除去した。外科用メスを用いて脂肪組織の除去後に、皮膚組織を約1mm片に刻んで、PBS中でマイクログラインダーを用いてホモジナイズした。3000gで遠心分離した後、Exoquickキット(System Biosciences)を1:5の比(Exoquick:上清)で使用して、上清からEVを4℃で12時間単離した。EVを1500gで30分間の遠心分離により沈殿させた。次いで、製造業者によって提供された推奨に従って、mirvanaキット(Life technologies)を使用して、全RNAをペレットから抽出した。
プラスミドDNA精製キット(Qiagen Maxi−prep、カタログ番号12161、およびClontech Nucleobondカタログ番号740410)を用いてプラスミドを調製した。DNA濃度をNanodrop 2000c Spectrophotemeter(Thermoscientific)から得た。
LCMは、PALM Technologies(Zeiss,Jena,Germany)からのレーザー顕微解剖システムを使用して実施した。形態および/または免疫染色に基づいて同定された組織切片の特定領域を切断し、20倍接眼レンズで捕捉した。試料を25μlの細胞直接溶解抽出緩衝液(Invitrogen)に放出した。約1,000,000μm2の組織領域を各キャップに捕捉し、次いで溶解物をさらなる処理のために−80℃で保存した。LCM試料のqRT−PCRは製造業者の指示に従って細胞直接溶解緩衝液から実施した。
特異的抗体および標準的な手順を用いて、組織免疫染色を実施した。簡潔に説明すると、OCT包埋組織を10μmの厚さで凍結切片化し、冷アセトンで固定し、10%正常ヤギ血清でブロッキングし、そして特異的抗体と共にインキュベートした。続いて、蛍光標識した適切な二次抗体(Alexa488標識α−モルモット、1:200、Alexa488標識α−ウサギ、1:200、Alexa568標識α−ウサギ、1:200)とのインキュベーションによってシグナルを可視化し、DAPIで対比染色した。血管のレクチンベースの可視化は、組織の30分前にFITC標識レクチンを尾静脈注射によって行った。レーザー走査共焦点顕微鏡(Olympus FV1000 filter/spectral)で画像を捕捉した。
動物をIVIS Lumina II光学イメージングシステムを用いて麻酔下で画像化した。画像化の5〜10分前に、repTOPTM mitoIREマウスに基質ルシフェリン(カブトムシルシフェリンのカリウム塩、Promega)を100mg/kgの用量で予め注射した。発光画像とのオーバーレイ画像は、Living Imageソフトウェアを使用して作製した。
磁気共鳴血管造影法を用いて、マウスにおける本発明者らのMCAOモデルを検証し、効果的なMCAOのために閉鎖栓サイズおよび内頸動脈挿入距離を最適化した。9.4TのMRI(Bruker Corporation、Bruker BioSpin Corporation、Billerica,MA,USA)を使用して、MCA再灌流の48時間後に麻酔をかけたマウスに対してT2強調MRIを実施した。MR画像は、以下のパラメータを用いた緩和強調の高速取得(RARE)シーケンスを用いて取得した:視野(FOV)30×30mm、取得マトリックス256×256、TR3,500ms、TE46.92ms、スライスギャップ1.0mm、希少因子8、平均数3。1mm当たり8.5ピクセルの解像度。未加工のMR画像を標準DICOMフォーマットに変換し、処理した。Osirix v3.4を用いた画像の適切なソフトウェアコントラスト強調の後、各冠状脳スライスにおける梗塞領域を描写するために、デジタル面積測定はマスクされた観察者によって行われた。過去に報告されているように、脳切片からの梗塞面積を合計し、切片の厚さを乗じ、浮腫誘発性腫脹について補正して、梗塞体積を決定した(KhannaS,et al.J.Cereb Blood Flow Metab 2013,33(8):1197−1206)。
筋肉エネルギーは、RF送信用の体積コイルおよび受信用の31Pコイルを使用して、9.4Teslaスキャナー(Bruker BioSpec)でNMR分光測定を評価した(Fiedler GB,et al.MAGMA 2015,28(5):493−501)。特注の1H/31Pトランシーバーコイルアレイでインビボ画像化を実施した。単一パルス系列を用いてデータを得た。生データをノイズ低減のためにウィンドウ処理し、スペクトル領域にフーリエ変換した。
血管形成を超音波画像によって並行モニターした。簡潔に説明すると、Vevo 2100システム(Visual Sonics、Toronto、ON、Canada)を使用して、B−modeに関する超音波画像を、MS250直線配列プローブを用いて得た。(Gnyawali SC,et al.J Vis Exp.20109(41))。ドップラーカラーフロー画像をモニターに実装し、収縮期と拡張期下で血流特性を定量化した。
LCMを使用して、ABMトランスフェクトマウス皮膚からインビボ由来iNについて濃縮した組織単離物を調製した(Roy S,et al.Proc Natl Acad Sci USA 2007,104(36):14472−14477、Rink C,et al.J Cereb Blood Flow Metab 2010,30(7):1275−1287)。単離した組織を、PicoPure(登録商標)RNA単離キット(ThermoFisher)からの溶解緩衝液中で処理した。RNA抽出、標的標識化、GeneChip(登録商標)、およびデータ分析は、過去に報告されているように実施した(Roy S,et al.Proc Natl Acad Sci USA 2007,104(36):14472−14477、Rink C,et al.J Cereb Blood Flow Metab 2010,30(7):1275−1287、RoyS,et al.Physiol Genomics 2008,34(2):162−184)。試料をAffymetrix Mouseトランスクリプトームアレイ1.0(MTA1.0)とハイブリダイズさせた。アレイを洗浄し、オハイオ州立大学の施設のGeneArrayスキャナー(Affymetrix)で、前述のとおりスキャンした(Roy S,et al.Proc Natl Acad Sci USA 2007,104(36):14472−14477、RoyS,et al.Physiol Genomics 2008,34(2):162−184)。発現データは、シリーズアクセッション番号GSE92413でGene Expression Omnibus(GEO;http://www.ncbi.nlm.nih.gov/geo)に提出されている。RMA16を用いて生データを正規化し、Genespring GX(Agilent、SantaClaraCA)を用いて分析した。dChip(登録商標)ソフトウェア(Harvard University)を使用して、データのさらなる処理を実施した(Roy S,et al.Proc Natl Acad Sci USA 2007,104(36):14472−14477、RoyS,et al.Physiol Genomics 2008,34(2):162−184)。群にわたる類似の遺伝子の機能注解は、IPA(登録商標)分析を用いて行われた。表5および図6を参照されたい。
試料をコード化し、データ収集を盲検様式で実施した。データは、3〜8つの生物学的複製物の平均±標準誤差として報告されている。失敗したトランスフェクション(例えば、皮膚とナノチャネルとの間の接触不良、またはナノチャネルの目詰まりなどに起因する)は、分析から除外した。再現性を確認するために実験を少なくとも2回繰り返した。群間比較は、分散分析(ANOVA)によって行った。統計的差異は、SigmaPlotバージョン13.0を用いて、必要に応じてパラメトリック/ノンパラメトリック検定によって決定した。
GeneChipの発現データはGene Expression Omnibusからアクセスできる。妥当な要求があれば、追加のデータは対応する作成者から入手可能である。
インビボで誘導されたニューロンの固有の興奮性を検出することに努力が集中され、この目的を達成するために細胞外記録の一般原則が使用された。しかし、細胞外記録のために使用される従来の電気生理学的技術は、マウスから組織を解剖して切り離し、形態学的に誘導されたニューロンを同定し、次いで目的の細胞に近接して細胞外電極を配置する必要があるため、実行するのが困難だった。パッチクランプ法または従来の電気生理学的測定を実施することの前述の複雑さを克服するために、細胞外空間に配置された導電性ポリマー電極のクロノアンペロメトリー測定を使用して、ニューロンの興奮性を検出した。ポリマーへのおよびポリマーからの電荷の侵入および放出の伝達関数を測定データに適用し、導電性ポリマーの二重層およびファラデー反応に対応する極および残基を計算する。伝達関数分析から計算された残基は、Venugopalら(Venugopal V,et al.J Intel Mat Syst Str2016,27(12):1702−1709)に説明されているように、ポリマーに隣接したカチオン濃度の変化に対応する。図25に示すように、マウスのABM処理した領域に導電性ポリマー電極を配置することによって、ニューロンの興奮性を示す、ファラデー反応に対応する残基への経時変化を観察した。
ドデシルベンゼンスルホン酸(PPy(DBS))を添加したポリピロールは、電位の発生時にカチオンを局所媒体と交換する導電性ポリマーである。カチオン侵入率は、適用された電位の機能、ポリマー幾何学、ポリマーの現状、および電解質の濃度である。(Venugopal V,et al.Sensors and Actuators B:Chemical 2014,201(0):293−299)この濃度依存性は、5〜100mM11の範囲にわたってNaCl濃度と直線関係を有することが実証されているカチオンセンサーの作成を可能にする。さらに、PPy(DBS)センサーは、それらの機能に障害を与えたり影響を与えたりすることなく、生物学的物質の局所カチオン濃度を決定することができる無毒性かつ酸化還元メディエータフリーシステムである。それゆえ、中規模のPPy(DBS)センサーは真皮層内に存在できるプローブに製造され、インサイチュのカチオン濃度を測定する。
白金ワイヤ(直径0.025mm、純度99.9%の硬さ、Goodfellow,USA)を石英毛管(75×1mm、Sutter Instruments)を通して挿入して、2mmの突起を形成した。突出端部をエポキシで封止し、作用電極(WE)として1mm露出白金線を残した。銀ワイヤ(直径0.5mm、純度99.9%、Sigma Aldrich)を同様に処理して、1mmの突出部を有する参照電極(RE)を形成した。挿入前に、銀ワイヤを次亜塩素酸ナトリウム溶液(10〜15%塩素)中に20分間浸漬してAg/AgCl層を形成した。電解重合溶液(0.2mMピロール、98%純度および0.1mMドデシルベンゼンスルホン酸ナトリウム、Sigma Aldrichか)を形成し、30分間沈降させた。電解重合セルは、Ptワイヤ、Ag/AgCl、および白金ワイヤ対極(CE)から構成されていた。サイクリックボルタンメトリー実験(CV)を行って、電気化学的接続性、ピロール活性、およびポリマー成長領域を検証した。続いて、クロノアンペロメトリー実験(CA)を、118.5μCの電荷が蓄積されるまで0.52Vの電位(CVに基づいて)をかけて行い、0.15Ccm−2の電荷密度を有するPPy(DBS)膜を作製した。その後、PPy(DBS)チップを脱イオン水ですすぎ洗いし、窒素流下で乾燥させた。
インビボでの核リプログラミングにおける近年の進歩は、「部位」、患者特異的な細胞に基づく治療法の発展の可能性を切り開いた。ナノチャネル化装置によってリプログラミング因子を局所的かつ制御可能に組織に送達するために、新規であるが実施が容易な非ウイルスアプローチを開発した(図17)。そのような組織ナノトランスフェクション(TNT)アプローチは、配列されたナノチャネルを通して非常に強く集中した電場を印加することによってリプログラミング因子の直接的な細胞質ゾル送達を可能にし(Gallego−Perez D,et al.Nanomedicine 2016,12(2):399−409、Boukany PE,et al.Nat Nanotechnol 2011,6(11):747−754)、それは、並置組織細胞膜を無害にナノ穿孔し、リプログラミング因子を細胞内に電気泳動により打ち込む(図17a〜d)。TNTシステム製造過程およびシミュレーション結果に関する詳細な情報は、図20および図21に見出すことができる。遺伝子送達が本質的に非常に確率論的であり、有害な副作用(例えば、炎症反応、細胞死)をもたらし得る現在のインビボトランスフェクション技術(例えば、ウイルス、従来の組織バルクエレクトロポレーションまたはBEP)とは対照的に(Sen CK,et al.Am J Pathol 2015、185(10):2629〜2640)、ナノチャネルベースのトランスフェクションは、単一細胞レベルでのより集中的(図17b、c)かつ十分な(図17d)リプログラミング因子送達を可能にする。したがって、これを決定論的インビボ遺伝子トランスフェクションおよびリプログラミングのための強力なツールにしている(Gallego−Perez D,et al.Nanomedicine 2016、12(2):399−409、Boukany PE,et al.Nat Nanotechnol 2011,6(11):747−754)。
Claims (25)
- ETV2、FOXC2、およびFLI1からなる群から選択されるタンパク質をコードする2つ以上の核酸配列を含む、ポリヌクレオチド。
- 前記2つ以上の核酸配列が発現制御配列に作動可能に連結されている、請求項1に記載のポリヌクレオチドを含む非ウイルスベクター。
- 前記核酸配列のそれぞれが、単一発現制御配列に作動可能に連結されている、請求項2に記載の非ウイルスベクター。
- 前記非ウイルスベクターが、pIRES−hrGFP−2a、pAd−IRES−GFP、およびpCDNA3.0の群から選択されるプラスミドを含む、請求項2または3に記載の非ウイルスベクター。
- 前記ポリヌクレオチドが、細胞内送達に適切なリポソーム、微小粒子、またはナノ粒子内に封入される、請求項2〜4のいずれか一項に記載の非ウイルスベクター。
- 請求項1に記載のポリヌクレオチドまたは請求項2もしくは5のいずれか一項に記載の非ウイルスベクターと、miR−200b阻害剤とを含む、組成物。
- 体細胞を血管性細胞にリプログラミングする方法であって、
(a)前記体細胞内に、ETV2、FOXC2、およびFLI1からなる群から選択される1つ以上のタンパク質、またはETV2、FOXC2、およびFLI1タンパク質からなる群から選択される1つ以上のタンパク質をコードするポリヌクレオチドを細胞内送達すること、あるいは、
(b)ETV2、FOXC2、およびFLI1からなる群から選択される1つ以上のタンパク質、またはETV2、FOXC2、およびFLI1タンパク質からなる群から選択される1つ以上のタンパク質をコードするポリヌクレオチドを含有または発現する細胞から産生された細胞外小胞に前記体細胞を曝露すること、を含む、方法。 - 前記タンパク質またはポリヌクレオチドが経時的に投与される、請求項7に記載の方法。
- FLI1タンパク質、またはFLI1タンパク質をコードする核酸が最初に投与される、請求項8に記載の方法。
