膵臓癌におけるALDHの発現および活性レベルの変化を確認するために、多様な膵臓癌細胞株から確認した。
ゴシポールまたはフェンホルミンによる膵臓癌細胞生長抑制効果の確認
ゴシポール(gossypol)またはフェンホルミン(phenformin)がそれぞれ膵臓癌に対して癌細胞生長抑制効果を奏することができるかを確認した。
ゴシポールおよびフェンホルミンの並行処理による癌細胞生長抑制効果の確認
<3−1>ゴシポールおよびフェンホルミンの並行処理による膵臓癌細胞生長抑制効果の確認
ゴシポールおよびフェンホルミンがそれぞれ膵臓癌細胞に対して増殖抑制効果を示すが、ゴシポールおよびフェンホルミンを混合した時、膵臓癌の増殖を抑制する効果が増加できるかを確認しようとした。
ゴシポールおよびフェンホルミンを並行処理した時、膵臓癌細胞に対して有意な細胞生長抑制効果を確認したので、膵臓癌細胞でない正常細胞株に対して細胞死滅の活性を示すか否かも併せて確認した。
ゴシポールおよびフェンホルミンの並行投与によるATP生産阻害効果の確認
膵臓癌細胞生長抑制効果においてゴシポールおよびフェンホルミンを並行投与によるシナジー効果が現れることを確認したので、このようなシナジー効果がどのような原因によって現れるかを確認しようとした。ゴシポールはALDHの抑制剤として知られているので、ゴシポールおよびフェンホルミンを処理した時、膵臓癌細胞内におけるATPの生産レベルを確認した。
ゴシポールおよびフェンホルミンの並行投与によるミトコンドリア膜電位の減少効果の確認
前記実施例4において、ゴシポールおよびフェンホルミンの並行投与によってATP生産抑制効果がシナジー効果を奏することができることを確認したので、フェンホルミンの活性において変化があるか否かをさらに確認しようとした。フェンホルミンは、ミトコンドリア複合体Iの抑制剤として知られているので、ゴシポールおよびフェンホルミンの並行投与によってミトコンドリア膜電位レベルに変化があるかを確認した。
膵臓癌モデルにおけるゴシポールおよびフェンホルミンの併用投与による抗癌効果の確認
ゴシポールおよびフェンホルミンを併用処理する方が、それぞれを単独処理することに比べて、シナジーで上昇した癌細胞生長抑制効果を奏することができることを細胞レベル(In vitro)で確認したので、この後、生体内(In Vivo)レベルでも膵臓癌抑制効果を奏することができるかを確認した。
ゴシポール、フェンホルミンおよび抗癌剤の並行処理による癌細胞生長抑制効果の確認
ゴシポールおよびフェンホルミンを混合した時、有意に膵臓癌に対する抗癌効果を奏することができることを確認して、ゴシポールおよびフェンホルミンに加えてイリノテカン(irinotecan)を混合して、3つの薬物を同時に処理すること(triple combination)により、抗癌活性のシナジー効果を奏することができるかを確認しようとした。

- 前記フェンホルミンは、下記化学式2の化合物であることを特徴とする、請求項1に記載の膵臓癌予防および治療用薬学的組成物:
- 前記ゴシポールおよびフェンホルミンは、1:10〜1:500のモル比率で混合されることを特徴とする、請求項1に記載の膵臓癌予防および治療用薬学的組成物。
- 請求項1に記載の薬学的組成物に、抗癌剤を追加的にさらに含む、膵臓癌予防および治療用薬学的組成物。
- 前記抗癌剤は、イリノテカン(irinotecan)、ゲムシタビン(Gemcitabine)およびパクリタキセル(paclitaxel)からなる群より選択されるいずれか1つ以上であることを特徴とする、請求項5に記載の膵臓癌予防および治療用薬学的組成物。
- 前記薬学的組成物は、ゴシポール、フェンホルミンおよび抗癌剤を0.1〜10:10〜500:1のモル比率で混合して含むことを特徴とする、請求項5に記載の膵臓癌予防および治療用薬学的組成物。
- ゴシポールおよびフェンホルミン、またはこれらの薬学的に許容可能な塩を有効成分として含む、経口投与製剤。
- 前記ゴシポールおよびフェンホルミンは、60日以上の相乗的治療効果を示すことを特徴とする、請求項8に記載の経口投与製剤。
- 前記経口投与製剤は、徐放性または制御放出性製剤であることを特徴とする、請求項8に記載の経口投与製剤。
- 請求項8に記載の経口投与製剤に、抗癌剤を追加的に含む、経口投与製剤。
- 前記抗癌剤は、イリノテカン(irinotecan)、ゲムシタビン(Gemcitabine)およびパクリタキセル(paclitaxel)からなる群より選択されるいずれか1つ以上であることを特徴とする、請求項11に記載の経口投与製剤。
- 有効な量のゴシポールおよびフェンホルミンを必要な個体に投与する段階を含む、膵臓癌予防または治療方法。
- 有効な量のゴシポールおよびフェンホルミンとともに、抗癌剤を追加的にさらに含むことで、必要な個体に投与する段階を含む、膵臓癌予防または治療方法。
- 膵臓癌予防および治療に使用するための、ゴシポールおよびフェンホルミンを有効成分として含む薬学的組成物の用途。
- 膵臓癌予防および治療に使用するための、ゴシポールおよびフェンホルミンとともに、抗癌剤を追加的にさらに含む薬学的組成物の用途。
- 経口投与製剤として使用するための、ゴシポールおよびフェンホルミンの用途。
- 経口投与製剤として使用するための、ゴシポール、フェンホルミンおよび抗癌剤の用途。
