Abstract
【解決手段】イタコン酸の製造方法は、アコニターゼ発現遺伝子及びシスアコニット酸デヒドロゲナーゼ発現遺伝子が導入された組換え低温菌を準備するステップと、クエン酸を含む溶液に前記低温菌を反応させてイタコン酸を生産させるステップと、反応後に前記溶液の上清を回収するステップとを備えている。

Claims (6)
- 低温菌以外に由来するアコニターゼ発現遺伝子及びシスアコニット酸デヒドロゲナーゼ発現遺伝子が導入された組換え低温菌を準備するステップと、
クエン酸を含む溶液に前記低温菌を反応させてイタコン酸を生産させるステップと、
前記反応後に前記溶液の上清を回収するステップとを備えていることを特徴とするイタコン酸の製造方法。 - クエン酸を含む溶液に前記低温菌を反応させる前に、前記低温菌を30℃〜50℃の温度で熱処理するステップをさらに備えていることを特徴とする請求項1に記載の製造方法。
- 前記熱処理は、35℃〜45℃で行うことを特徴とする請求項2に記載のイタコン酸の製造方法。
- 前記アコニターゼ発現遺伝子は、大腸菌由来のacnB遺伝子であることを特徴とする請求項1〜3のいずれか1項に記載のイタコン酸の製造方法。
- 前記シスアコニット酸デヒドロゲナーゼ発現遺伝子は、Aspergillus属の細菌に由来するcadA遺伝子であることを特徴とする請求項1〜4のいずれか1項に記載のイタコン酸の製造方法。
- 前記低温菌は、Shewanella frigidimarinaであることを特徴とする請求項1〜5のいずれか1項に記載のイタコン酸の製造方法
