Description
QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)、APLPRWSGPIGVSWGLR(配列番号2)およびGVSWGLR(配列番号6)を含む群から選択されるペプチドに結合することも示した。
a.配列APLPRWSGPIGVSWGLR(配列番号2)のペプチドまたは配列GVSWGLR(配列番号6)のペプチドを含む組成物の動物への投与のステップ、

c.該抗体の単離のステップ。
a.個体の生体試料中の配列QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)のペプチドの濃度(Cm)を測定するステップ

1.2より高いS2の値は、その治療が抗うつに有効であることを示唆する。
以下の実施例では、後述する製品および方法を用いて実験を行った。本発明の実施例において、室温は、19〜25℃の温度、例えば、22℃に等しい温度を意味する。
実験は、8〜10週齢の雄C57BL/6J(20〜25g)(January France Elevage)に対して、European Community Directive 2010/63/EUに記載される実験動物の福祉および使用に関するポリシーに準拠して行った。地域の倫理委員会(CIEPAL)が、本試験で使用されたプロトコルを承認した(プロトコル番号00.893.02)。
ペプチドはGeneCust(Dudelange、Luxembourg)によって合成された。スパジン(YAPLPRWSGPIGVSWGLR(配列番号8))に対するウサギポリクローナル抗体は、Eurogentec(Seraing、Belgium)によって調製された。使用した抗体は、前述したとおりのAB1抗体であった。スパジン(5.4mg、2.7mmol)を1.5mlの25nMリン酸緩衝(pH6.7)に溶解した。700μlの70%アセトニトリル/30%ジメチルホルムアミドに再懸濁したビオチンN−ヒドロキシスクシンイミド(13.5mmol)の一部をスパジン溶液に加え、室温で一晩インキュベートした。半調製用Lichrosorb RP18カラムを装備したWaters社製システムを使用して、高速液体クロマトグラフィー(HPLC)によりビオチン化スパジンを精製した。質量分析により同定されたビオチン化スパジン(27分で溶出)を回収し、280nmでの吸収により定量化し、アリコートで凍結乾燥させた。
AlphaScreen技術(登録商標(Perkin Elmer))の原理に従って、ストレプトアビジンドナーマイクロビーズをビオチン化スパジンによって認識させる一方で、ウサギ抗IgGを含むアクセプターマイクロビーズに抗スパジン抗体を結合させた。2つのマイクロビーズ(アクセプターおよびドナー)が接近すると、ビーズ上の結合パートナー間の分子相互作用によってシグナルが生成される。血清試料中に存在するプロペプチドは、この競合を引き起こす相互作用に干渉することができた。96ウェルプレートで、室温で1時間、スパジンの濃度を増加させずにまたは増加させながら(10−11から10−6Mへ)、10nMのビオチン化スパジンをAlphaLisa(登録商標)緩衝液中の抗スパジン抗体(1:1000)でインキュベーションすることにより較正曲線を得た。ドナービーズおよびアクセプタービーズを加え、さらに2時間室温でインキュベーションした後、Enspireリーダー(Perkin)を使用してプレートを読み取った。血清の測定では、非標識スパジンの代わりに同じ体積の血清を加えた。プロペプチドの量をそのシグナルのパーセント阻害から決定し、検量線を用いて算出した。
生理食塩水または種々の試験ペプチドを静脈内注射した後、12cmの深さまで水(温度:22±1℃)を充填したシリンダー(高さ:30cm、直径:15cm)にマウス(n=群当たり10〜12匹)を個別に6分間入れた。最後の4分間の間に合計不動時間を記録した(Porsolt et al.,1977)。
対照個体のコホートは、精神疾患簡易構造化面接法(MINI)を用いて熟練の心理学者によってDSM−IV Axis I障害の診断のために選択された49人の健康な非血縁ボランティアから構成されていた[30]。薬物またはアルコールの乱用または依存の履歴がなく、精神障害の個人歴または第一度近親者の家族歴がない健康なボランティアのみが治験に登録された。さらには、関連する神経疾患、すなわち、てんかんおよびパーキンソン症候群が存在しないことが、治験に登録するために必須であった。最後に、ミニメンタルステート検査(MMSE)で27/30未満のスコアを得た対象は、治験から除外された。
結果は平均値±平均値の標準誤差(SEM)として表される。統計分析はGraphPad(バージョン6.0)を使用して行った。患者由来の試料における人口学的および臨床的特徴は、平均値±標準偏差(SD)で定量的に、または割合として記載される。正規性を確認した後、必要であればスチューデントt検定を用いて定量的変数の差を評価し、一方、分散分析(ANOVA)を用いて連続変数について考えられる群間の差を算出した(図1D)。薬物治療の間に起こる臨床的および生物学的変化は、被験者内要因等の経時的(T0、T1)に繰り返される測定の設計において一般線形モデルを用いて分析し、Greenhouse−Geisserの相関を適用した。ノンパラメトリックなマン・ホイットニーのU検定およびウィルコクソン順位和検定を用いて群間の差を評価した。
使用した抗体の特異性を調べた。具体的には、アッセイ法に使用した抗体の特異性を調べた。使用した抗体の特異性を特徴付けるために、ヒトPEの一連の部分配列を調製した(図1A)。図1Bに示すように、スパジンおよび全てのPEが、2.50nM(95%でのIC:3.37〜1.85、n=6)および2.18nM(95%でのIC:1.53〜3.09、n=3)のIC50で抗スパジン抗体によって認識された。ニューロテンシン(NT、黒い四角)およびソマトスタチン−14(SS14、白い四角)は抗体によって認識されなかった。
この実施例で示されるように、使用した抗体は、一連のヒトPEに由来するペプチド、およびPE自体のみを認識し、ニューロテンシンまたはソマトスタチン等の無関係なペプチドは認識しなかった。
ウサギから2つの抗体を調製し、得られた体積は各々約200mlであった。実験プロトコルにおいて、これらの抗体を1/1000および1/5000の希釈率で使用した。血中のプロペプチドレベルを測定するために、免疫学的または蛍光といった複数の技術を用いた。ペプチドアッセイは、100μlの血液を用いて達成することができ、再現可能である。
以下の実施例では、上記実施例1および2に記載したポリクローナル抗体AB1を用いて実験を行った。

Claims (10)
- 配列APLPRWSGPIGVSWGLR(配列番号2)のペプチド(スパジン)に対するポリクローナル抗体(AB1)であって、前記抗体は、ペプチドGVSWGLR(配列番号6)を唯一の例外として、
QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)、APLPRWSGPIGVSWGLR(配列番号2)、APLPRWSGPIGVSWGL(配列番号3)、LPRWSGPIGVSW(配列番号4)およびWSGPI(配列番号5)を含む群から選択されるペプチドに結合する、ポリクローナル抗体(AB1)。 - 配列GVSWGLR(配列番号6)を含むペプチド(ミニスパジン)に対する、ポリクローナル抗体(AB2)。
- 前記抗体は、
QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)、APLPRWSGPIGVSWGLR(配列番号2)およびGVSWGLR(配列番号6)を含む群から選択されるペプチドに結合している、請求項2に記載のポリクローナル抗体。 - 請求項1に記載の抗体を生成するための方法であって、
a.配列APLPRWSGPIGVSWGLR(配列番号2)のペプチドを含む組成物の動物への投与、
b.前記動物からの抗体の回収、および
c.前記抗体の単離、を含む、抗体を生成するための方法。 - 請求項2または3のいずれか一項に記載の抗体を生成するための方法であって、
a.配列GVSWGLR(配列番号6)のペプチドを含む組成物の動物への投与、
b.前記動物からの抗体の回収、および
c.前記抗体の単離、を含む、抗体を生成するための方法。 - 生体試料中のスパジンの検出および/またはアッセイのための、請求項1〜3のいずれか一項に記載の抗体のin vitroでの使用。
- 生体試料中のミニスパジンの検出のための、請求項2〜3のいずれか一項に記載の抗体のin vitroでの使用。
- うつ状態の検出のために個体の生体試料から配列番号1のペプチドを検出するための、請求項1〜3のいずれか一項に記載の抗体のin vitroでの使用。
- 以下のステップを含む、患者におけるうつ病を検出/決定するためのin vitro法であって、
a.うつ病個体の生体試料中における、請求項1〜3のいずれか一項に記載の抗体を用いた配列QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)のペプチドの濃度(Cm)の測定のステップ、
b.ステップa)で測定された前記ペプチドの前記濃度と健康な個体の基準濃度(Cref)との比較、および以下の式に従ったスコア(S1)の算出のステップ
S1=Cm/Cref
1未満のS1の値は、前記試料が得られた個体がうつ病であることを示唆する、in vitro法。 - うつ病治療の有効性を監視するための方法であって、
a)個体の生体試料中で、請求項1〜3のいずれか一項に記載の抗体を用いて配列QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)のペプチドの濃度C1を測定することと、
b)前記治療後に前記個体の生体試料中で、請求項1〜4のいずれか一項に記載の抗体を用いて配列QDRLDAPPPPAAPLPRWSGPIGVSWGLRAAAAGGAFPRGGRWRR(配列番号1)のペプチドの濃度C2を測定することと、
c)前記濃度値を比較して、以下の式に従ってスコア(S2)を算出することと、を含み
S2=C2/C1
1.2より高いS2の値は、その治療が抗うつに有効であることを示唆する、方法。
