Abstract
Description
イリノイ州オークブルックのPureCircle USA Inc.から入手可能な「A95」と表示されたステビア抽出物を使用して、以下の分析方法論を使用して主要及び少量のステビオール配糖体成分を単離し、特徴付けした。

分析LCMSは島津シングルクワッドUPLCシステムで実施した(表1参照)。最初の40分間は同一である2つの異なる勾配システムを適用した(表2a及び2b参照)。勾配KM7を使用して、すでに同定されているステビオール配糖体#25‐#29を含むすべての化合物を分析したが、勾配ACD1はより速く化合物#1‐#24の分析に使用した。
ステビア葉抽出物A95(100g、白色粉末)を65℃の温度でエタノール/水70/30(750mL)に溶解した。
乳白色の溶液を水浴中で室温まで冷却させ、次いで吸引フィルターを通して濾過した。集めた結晶をエタノールで洗浄し、乾燥し、貯蔵した。母液と洗浄液を別々に保ち、それぞれの溶媒を真空下で除去した。
それぞれの試料(15g)をメタノールに溶解し、セライト(30g)を加え、溶媒をロータリーエバポレーターにより除去する。固定化サンプルをガラスカラムに移し、表3に記載のMPLCシステムに組み込む。時間に基づく分画は18画分になる(それぞれ4分)。溶媒及び勾配は表3に記載されている。
それぞれの試料(20g)をメタノールに溶解し、シリカ(40g)を加え、溶媒をロータリーエバポレーターで除去する。固定化サンプルをガラスカラムに移し、表4に記載の高圧液体クロマトグラフィー(HPLC)システムに組み込む。酢酸エチル/メタノール1:1で洗浄することにより、移送カラムから空気を除去する。時間に基づく分画は、分画中に生成されたUV及びELSDデータに基づいて組み合わされる90の画分(それぞれ0.5分)になる。得られた画分をLCMSによって分析する。溶媒及び勾配は表4に記載されている。
それぞれの試料(3.5gまで)をメタノールに溶解し、C‐18RP材料を加え、溶媒をロータリーエバポレーターにより除去する。固定化サンプルをカラムに移し、表5に記載のHPLCシステムに組み込む。時間に基づく分画は、分画中に生成されたUV及びELSDデータに基づいて組み合わされる120の画分(それぞれ27秒)をもたらす。得られた画分をLCMSによって分析する。溶媒及び勾配は表5に記載されている。
それぞれの試料を2mLの溶媒AとBの3:1混合物に溶解する(表6参照)。サンプル注入は9.95分後に行う。時間に基づく分画は、分画中に生成されたUV及びELSDデータに基づいて組み合わされる96の画分(それぞれ43秒、18分後から開始)になる。得られた画分をLCMSによって分析する。溶媒及び勾配は表6に記載されている。
単離された化合物は、Bruker 500Mhz NMR分光計を使用してNMR分光法によって同定された。アグリコンの同定は、主な指標としてC17、C18及びC20プロトンシグナルを用いた参照1H‐NMRスペクトルに基づいた。特にC20プロトンシフトは、化合物#4及び#18に見られるように変化を示した。H‐H‐Cozy、HSQC及びHMBCを用いて配糖体を解明し、参考として文献公知のステビオシドのスペクトルを用いて実験を行った。
図1は、上記のような分析方法論を用いることによって表7において同定された主要ピークを含むHPLCチャートを示す。表7のさまざまな化合物を単離するための概略的な手順を図2と図3に示す。
この実施例は、例としてレバウジオシドW3(#19)の単離、同定及び特徴付けを概説する。全ての新規ステビオール配糖体分子について同様の分析を行った。
100gのステビア葉抽出物A95をセクション1.3(実施例1)に記載の方法に従って再結晶化して、33.2gの濃縮された少量の化合物を母液から得た。濃縮された少量の化合物を、勾配Aを用いてセクション1.5に記載のように順相クロマトグラフィーを用いて分画した(表4参照)。画分49〜60は1.32gの濃縮された少量の化合物を生成し、これを勾配Lを使用してセクション1.4に従って逆相HPLCを使用してさらに分画した。
画分51+52に赤い長方形、ELSDトレース(青)、及びUVトレース(オレンジ色)の印を付けて(図17A)、#19を37.5mg得た。分取RP‐HPLCクロマトグラムを用いた画分66+67(図17B)により、濃縮された少量の化合物3.85gを得た。画分66+67をセクション3.2に従ってLCMSにより分析した(図17B参照)。37.5mgの化合物#19が89%純度(ELSD)で得られた。
化合物#19の構造は、d4‐メタノール中500MHzのBruker‐NMRでのNMRによって決定された(δC=48.5ppm;δH=3.3ppm)。データを表10に示し、NMR分析を図17Cに示す。化合物#19の構造を図17Dに示す。
