Abstract
Description
本発明は全米科学財団CHE−1012467の下、政府の支援でなされた。政府は本発明において一定の権利を有する。
本願は、引用することによりその全内容が本明細書の一部とされる2016年9月26日に出願された米国仮特許出願第62/495,843号の利益を主張するものである。
本明細書には、核酸塩基、核酸塩基を含んでなるポリマーモノマー、ならびに前記核酸塩基を含んでなる核酸およびそれらの類似体が記載される。また、本明細書には、前記核酸塩基、前記核酸塩基を含んでなるポリマーモノマー、ならびに前記核酸塩基を含んでなる核酸およびそれらの類似体の使用方法も記載される。
本明細書では、二本鎖核酸配列を標的とし、ミスマッチ配列を結びつけるために使用することができる試薬が提供される。試薬はサイズが比較的小さく、液相法を用いて多量にかつより安価に製造することができ、細胞によって容易に取り込まれる。これらの試薬は、記載の認識スキームは本来モジュール式であり、新たに出現する配列とマッチするように随意に容易に改変することができるので、癌、細菌およびウイルス感染の病理に関連するものなどの急速に進化する部位の標的化に特に魅力がある。従って、二価核酸塩基が本明細書に記載される。二価核酸塩基は、ミスマッチが存在してもしなくても、DNAおよび/またはRNAの2つの鎖と有向性の水素結合相互作用を形成することができる。このプラットフォームは、生物学およびバイオテクノロジー、診断薬、および治療薬における基礎研究に適用を持つ。記載の分子認識プラットフォームは、核酸構造および機能の操作のための分子ツールの開発、ならびに遺伝病および感染性疾患を治療するための分子療法の開発につながると期待される。
本願に明示される様々な範囲の数値の使用は、そうではないことが明示的に示されない限り、記載された範囲内の最小値および最大値の両方には「約」という語が前に付くかのうように近似値として示される。このように、記載の範囲の上下のわずかな変動は、この範囲内の値と実質的に同じ結果を達成するために使用可能である。また、そうではないことが示されない限り、範囲の開示は、最小値と最大値の間の総ての値を含む連続的範囲として意図される。本明細書で使用する場合、「1つの(a)」および「1つの(an)」は1以上を指す。

Claims (20)
- R1の1以上の例がメチル、ホルミル、エチル、アセチル、アニシル、ベンジル、ベンゾイル、カルバメート、トリフルオロアセチル、ジフェニルメチル、トリフェニルメチル、N−ヒドロキシスクシンイミド、ベンジルオキシメチル、ベンジルオキシカルボニル、2−ニトロベンゾイル、t−Boc(tert−ブチルオキシカルボニル)、4−メチルベンジル、4−ニトロフェニル、2−クロロベンジルオキシカルボニル、2−ブロモベンジルオキシカルボニル、2,4,5−トリクロロフェニル、チオアニジル、チオクレシル、cbz(カルボベンジルオキシ)、p−メトキシベンジルカルボニル、9−フルオレニルメチルオキシカルボニル、ペンタフルオロフェニル、p−メトキシベンジル、3,4−ジメトキシベンジル、p−メトキシフェニル、4−トルエンスルホニル、p−ニトロベンゼンスルホナート、9−フルオレニルメチルオキシカルボニル、2−ニトロフェニルスルフェニル、2,2,5,7,8−ペンタメチル−クロマン−6−スルホニル、およびp−ブロモベンゼンスルホニルから場合により選択される保護基である、請求項1に記載の遺伝子認識試薬。
- 前記骨格がDNA、RNA、ペプチド核酸(PNA)、ホスホロチオエート、ロックド核酸、非ロックド核酸、2’−O−メチル置換RNA、モルホリノ核酸、トレオース核酸、もしくはグリコール核酸骨格、またはそれらの任意の組合せから1つ選択される、請求項1に記載の遺伝子認識試薬。
- 前記骨格が、ガンマペプチド核酸(γPNA)骨格などのペプチド核酸(PNA)骨格、場合により、Rが
- R3がHであり、R2が、1〜12個の(−O−CH2−CH2−)残基の1以上のPEG部分で場合によりPEG化されていてもよいアミノ酸側鎖である、請求項4に記載の遺伝子認識試薬。
- R1の総てがHである、請求項1に記載の遺伝子認識試薬。
- Rがカルボキシル、ヒドロキシル、アミン、シアナート、チオール、エポキシド、ビニル、アリル、n−ヒドロキシスクシンイミド(NHS)エステル、アジド、アルキニル、マレイミド、ヒドラジド、テトラジン、ホスホラミダイト、シクロアルキン、ニトリル、または(CH2)nCO2Hまたは(CH2)nCO2Y(ここで、n=1〜5かつY=脱離基)などの反応性基である、請求項8に記載の化合物。
- Rが核酸または核酸類似体骨格モノマーである、請求項8に記載の化合物。
- Rがシリコンウエハー、マルチウェルディッシュ、またはポリマービーズ、および場合によりアレイなどの固相基板である、請求項8に記載の化合物。
- R1が、保護基、例えば、メチル、ホルミル、エチル、アセチル、アニシル、ベンジル、ベンゾイル、カルバメート、トリフルオロアセチル、ジフェニルメチル、トリフェニルメチル、N−ヒドロキシスクシンイミド、ベンジルオキシメチル、ベンジルオキシカルボニル、2−ニトロベンゾイル、t−Boc(tert−ブチルオキシカルボニル)、4−メチルベンジル、4−ニトロフェニル、2−クロロベンジルオキシカルボニル、2−ブロモベンジルオキシカルボニル、2,4,5−トリクロロフェニル、チオアニジル、チオクレシル、cbz(カルボベンジルオキシ)、p−メトキシベンジルカルボニル、9−フルオレニルメチルオキシカルボニル、ペンタフルオロフェニル、p−メトキシベンジル、3,4−ジメトキシベンジル、p−メトキシフェニル、4−トルエンスルホニル、p−ニトロベンゼンスルホナート、9−フルオレニルメチルオキシカルボニル、2−ニトロフェニルスルフェニル、2,2,5,7,8−ペンタメチル−クロマン−6−スルホニル、およびp−ブロモベンゼンスルホニルから1以上選択される保護基である、請求項8に記載の化合物。
- Rがペプチド核酸骨格モノマー、例えば、ガンマペプチド核酸(γPNA)骨格モノマー、および場合により、構造
- Rが、ホスホロチオエート骨格モノマー、ロックド核酸骨格モノマー、非ロックド核酸骨格モノマー、2’−O−メチル置換RNA骨格モノマー、モルホリノ核酸骨格モノマー、トレオース核酸骨格モノマー、またはグリコール核酸骨格モノマーから選択される核酸類似体骨格モノマーである、請求項8に記載の化合物。
- Rがリボース一リン酸、二リン酸、もしくは三リン酸またはデオキシリボース一リン酸、二リン酸、もしくは三リン酸、例えば、リボースまたはデオキシリボースの5’一リン酸、二リン酸、または三リン酸である、請求項8に記載の化合物。
- 請求項1〜15のいずれか一項に記載の遺伝子認識試薬または化合物と薬学上許容可能な賦形剤とを含んでなる組成物。
- カートリッジなどの容器内に請求項1〜15のいずれか一項の遺伝子認識試薬または化合物を含んでなり、Rが核酸または核酸類似体骨格モノマーである、キット。
- 請求項1〜7のいずれか一項に記載の遺伝子認識試薬を含んでなるアレイ。
- 請求項1〜7のいずれか一項に記載の遺伝子認識試薬を、核酸を含んでなるサンプルと接触させること、および遺伝子認識試薬と核酸との結合を検出することを含んでなる、核酸内の標的配列の検出方法。
- 核酸サンプルを請求項1〜7のいずれか一項に記載の遺伝子認識試薬と接触させること、前記遺伝子認識試薬から前記核酸サンプルを分離すること、前記遺伝子認識試薬に結合した前記遺伝子認識試薬に結合したいずれの核酸も残すこと、および前記遺伝子認識試薬に結合した任意の核酸から前記遺伝子認識試薬を分離することを含んでなる、標的配列を含む核酸の単離および精製方法。
