Abstract
Description
本発明は、医薬における使用のためのポリペプチド、核酸分子、T細胞受容体又はT細胞受容体を呈するT細胞及び免疫チェックポイント阻害剤に関する。本発明は、患者におけるがんを治療する方法にも関する。本発明は、がんの治療に適した組成物及びキットにも関する。
がんは、個体内部での新しい異常な細胞の増殖によって特徴付けられる疾患である。がんは、がん細胞、すなわち浸潤及び転移の特性を呈する細胞の発生を可能にする幾つかの突然変異事象を含む多段階プロセスを通じて発生する。
本発明の第1の態様によると、医薬における使用のためのポリペプチドであって、免疫チェックポイント阻害剤とともに同時に、別々に又は連続的に投与され、自己抗原の少なくとも12のアミノ酸の領域又はその領域に対して少なくとも80%の同一性を有する配列を含む少なくとも1つのポリペプチドを含む、ポリペプチドが提供される。

1.ワクチンのペプチドに対する測定可能な免疫応答を開始させるための患者の免疫系により要求される期間の短縮。換言すれば、促進されたCD4+T細胞免疫応答が生成される。
2.患者によるワクチンのペプチドに対する強力な免疫応答の開始。一実施形態では、「強力な免疫応答」は、本明細書で用いられるとき、患者10名の平均を通じて、平均ピーク免疫応答が少なくとも17、好ましくは少なくとも19のSIであることを指す。
3.患者における改善された臨床成績。
本発明は、がんを治療するためのキットを提供する。キットは、2つの成分を含む。第1の実施形態では、第1の成分は、自己抗原の少なくとも1つのポリペプチドであり、このポリペプチドは、少なくとも12のアミノ酸長である。第2の成分は、免疫チェックポイント阻害剤である。

本発明の第2の実施形態では、上に示されるようなポリペプチドをコードするヌクレオチド配列を含む核酸分子が提供される。
本発明の第3の実施形態では、上に示されるようなポリペプチドに対して、そのポリペプチドがMHC分子上に提示されるときに特異的であるT細胞受容体又はT細胞受容体を呈するT細胞が提供される。
がんを治療するためのキットの第2の成分は、免疫チェックポイント阻害剤である。
CD152としても知られている細胞傷害性Tリンパ球関連抗原(CTLA−4)は、T細胞活性化を調節するように機能する共抑制分子である。
CTLA−4が早期T細胞活性化を調節することに役立つ一方、プログラム死1(PD−1)シグナル伝達は、部分的には末梢組織内でのT細胞活性化を調節するように機能する。PD−1受容体は、CD28ファミリーに属する免疫抑制性受容体を指す。PD−1は、T reg、活性化B細胞、及びナチュラルキラー(NK)細胞を含む幾つかの細胞型上に発現され、主に以前に活性化されたT細胞上においてインビボで発現され、2つのリガンドPD−L1及びPD−L2に結合する。PD−1の内因性リガンドのPD−L1及びPD−L2は、活性化免疫細胞及び非造血性細胞、例えば腫瘍細胞において発現される。PD−1は、本明細書で用いられるとき、ヒトPD−1(hPD−1)、変異体、アイソフォーム及びhPD−1の種相同体、並びに少なくとも1つのhPD−1と共通のエピトープを有する類似体を含むことを意味する。完全hPD−1配列は、ジェンバンク受入番号U64863下に見出すことができる。プログラム死リガンド−1(PD−L1)は、PD−1に対する2つの細胞表面糖タンパク質リガンドの1つであり(他方はPD−L2である)、PD−1への結合時にT細胞活性化の下方制御及びサイトカイン分泌をもたらす。PD−L1は、本明細書で用いられるとき、ヒトPD−L1(hPD−L1)、変異体、アイソフォーム及びhPD−L1の種相同体、並びに少なくとも1つのhPD−L1と共通のエピトープを有する類似体を含む。完全hPD−L1配列は、ジェンバンク受入番号Q9NZQ7下に見出すことができる。腫瘍は、PD−L1/L2を発現し、それによりPD−1/PD−L1,2相互作用を介して腫瘍浸潤性リンパ球を抑制することにより免疫監視を逃れることが示されている(Dong et al. Nat. Med. 8:793-800. 2002)。治療抗体とのこれらの相互作用の阻害は、T細胞応答を増強し、抗腫瘍活性を刺激することが示されている(Freeman et al. J. Exp. Med. 192: 1027-34. 2000)。
本発明の一部の実施形態では、がんを治療するためのキット中の追加的成分が提供される。
上で説明のようなキット、組成物又は医薬組成物の各成分は、使用時、治療を必要とする患者に投与される。原則的に、キットの成分の任意の投与様式、組成物又は医薬組成物が用いられ得る。
少なくとも12のアミノ酸長である自己抗原又はユニバーサル腫瘍抗原の少なくとも1つのポリペプチド及びチェックポイント阻害剤は、がんの治療における相乗効果をもたらす。他の実施形態では、本発明に従う核酸分子、T細胞受容体又はT細胞受容体を呈するT細胞、及び免疫チェックポイント阻害剤は、がんの治療における相乗効果をもたらす。
以降では、実施例を参照して本発明が具体的に説明される。しかし、これらの実施例は、本発明の技術的範囲を限定するものではない。
T細胞応答アッセイ(3H−チミジン取り込みによる増殖)
末梢血単核球(PBMC)をワクチン接種の開始前及びワクチン接種後の複数の時点で得た。PBMCを前述のように単離及び凍結した(Inderberg-Suso EM et al., Oncoimmunology 2012 1(5):670-686、参照により本明細書中に援用される)。1回のインビトロでのワクチンペプチドによる前刺激後にワクチン接種前及び後のPBMCから作製したT細胞培養物を、その後、前述のような3H−チミジン取り込みを用いる標準T細胞増殖アッセイで試験した(Inderberg-Suso EM et al., Oncoimmunology 2012 1(5):670-686)。照射した自家PBMCを抗原提示細胞(APC)として用いた。T細胞(50000個)を関連抗原(例えば、配列番号1、2及び3の組み合わせ並びに配列番号1、2及び3の個別ポリペプチド)の存在下及び不在下で50000個のAPCとともにインキュベートした。T細胞培養物を3通りに試験した。平均値の標準誤差(SEM)は、通常、10%未満であった。T細胞バルクの応答は、刺激指標(SI;抗原存在下での応答を抗原不在下での応答で除したもの)が3以上であったとき(SI≧3)、抗原特異的であると考えた。
IFN−γエリスポットアッセイを本質的に前述のように実施した(Gjertsen MK et al. J Mol Med (Berl) 2003; 81:43-50)。ヒトIFN−γに対するモノクローナル抗体(Mabtech)をPBSで希釈し、最終濃度を5μg/mlにした。96ウェルのMultiScreen-HAプレート(Millipore)を、75μl/ウェルの保存液を添加することにより抗体でコーティングし、4℃で一晩インキュベートした。翌日、プレートを室温で1時間貯蔵し、ウェルを200μl/ウェルのPBSで6回洗浄し、余分な抗体を除去した。非特異的結合をブロッキングするため、プレートを100μl/ウェルのCellGro DC培地+10%ヒト血清(HS; Baxter)とともに37℃で1〜2時間インキュベートした。解凍及び洗浄後の自家PBMCを数え上げ、プレコーティングしたウェルに5×105細胞/ウェルで添加した。レスポンダーT細胞を収集し、洗浄し、数え上げ、CellGro DC培地(CellGenix)中、自家PBMCを1×105細胞/ウェルで含有するウェルに3通りに移した。T細胞のみ及びPBMCのみを伴う陰性対照並びにT細胞+PBMC+ブドウ球菌(Staphylococcus)エンテロトキシンC3(SEC3;Toxin Technologies)を有する陽性対照を含めた。加湿インキュベーター内、5%CO2下、37℃で一晩のインキュベーション後、プレートをPBSで6回洗浄した。2回洗浄と3回洗浄との間にプレートを室温で10分間インキュベートした。各ウェルに、ヒトIFN−γに対するビオチン化抗体(Mabtech)1μg/mlの保存液75μlを添加し、プレートを室温で2時間インキュベートした。6回の反復洗浄後、プレートを(PBS+1%HSAで1:1000に希釈した)保存液からの75μl/ウェルのストレプトアビジン−ALP(Mabtech)とともに1時間インキュベートした。余分な抗体を除去するため、ウェルを再びPBSで6回洗浄した。次いで、基質BCIP/NBT(Sigma-Aldrich)75μlを各ウェルに添加後、プレートを5〜20分間インキュベートした。スポットの出現時に水を添加し、反応を停止させた。自動分析装置CTL IMMUNOSPOT S5 VERSA-02-9030(Cellular Technology Ltd)を用いて、スポットを数え上げた。
末梢血T細胞は、配列番号1、2及び3のワクチン接種を受けていたメラノーマ患者において応答する。T細胞を配列番号1、2又は3並びに3つ全部のポリペプチドの組み合わせでインビトロ刺激した。T細胞増殖アッセイ及びエリスポットアッセイは、本明細書中に示すような材料及び方法セクションの通りに実施した。結果を図2A及び2B並びに下の表3A〜3Cに提示する。719−20は、配列番号1を指し、725は、配列番号2を指し、728は、配列番号3を指し、hTERTI混合物は、配列番号1、2及び3の組み合わせを指す。刺激指標(SI)は、T細胞増殖アッセイで試験したすべてのポリペプチドについて計算した。SI≧3を陽性と考えた。
CD4+T細胞をメラノーマ患者2名(患者P7及びP9)及び肺がん患者(患者P5)から作製した。患者は、以前にがんワクチンの投与を受けていなかった。CD4+T細胞を、(下の表4に示すような)配列番号1又は14のアミノ酸を含むその断片でインビトロ刺激した。T細胞増殖アッセイは、本明細書中に示すような材料及び方法セクションの通りに実施した。SI≧2を陽性と考えた。結果を図3A〜Cに提示する。
配列番号1のMHCクラスII結合モチーフ及びその配列の免疫原性断片を計算したものを表5に示す。
CD4+T細胞をメラノーマ患者(患者P7)及び卵巣がん患者(患者P1)から作製した。患者は、以前にがんワクチンの投与を受けていなかった。CD4+T細胞を、(下の表6に示すような)配列番号2又は12のアミノ酸を含むその断片でインビトロ刺激した。T細胞増殖アッセイは、本明細書中に示すような材料及び方法セクションの通りに実施した。SI≧2を陽性と考えた。結果を図4に提示する。
CD4+T細胞を、がんワクチンの投与を受けていなかった膵がんを有する患者1名(患者P1)及び神経膠芽腫を有する患者1名(患者P5)から作製した。CD4+T細胞を、(下の表7に示すような)配列番号3又は12のアミノ酸を含むその断片でインビトロ刺激した。T細胞増殖アッセイは、本明細書中に示すような材料及び方法セクションの通りに実施した。SI≧3を陽性と考えた。結果を図5に提示する。
配列番号1に特異的なCD4+T細胞クローンを、配列番号1、2及び3の組み合わせのワクチン接種を受けた患者から作製し、配列番号1の14−merペプチドの重複ライブラリーで刺激した。T細胞クローン増殖は、材料及び方法の通り、T細胞応答アッセイ(3H−チミジン取り込みによる増殖)を用いてペプチド刺激後に測定した。データを図6に示す。
臨床試験において、抗CTLA−4遮断薬及びがんワクチン(がん特異的なヘルパーT細胞応答を誘導する能力があるロングペプチドを含む)の併用療法について検討した。試験(EudraCT番号:2013−005582−39)では、切除不能又は転移性悪性メラノーマを有する患者において、イピリムマブ並びに配列番号1、2及び3のカクテルを含むがんワクチンの組み合わせについて検討した。
設計
これは、切除不能又は転移性悪性メラノーマを有する患者におけるイピリムマブ/がんワクチンの組み合わせについての安全性及び耐容性を試験する第I/IIa相非盲検単一アーム介入試験であった。
患者にイピリムマブ並びに配列番号1、2及び3を含むがんワクチンを顆粒球マクロファージ−コロニー刺激因子(GM−CSF)と一緒に投与した。イピリムマブは、全部で4用量を3週ごとに投与した。GM−CSF及びがんワクチンは、イピリムマブの初回投与の7、5及び3日前に投与した。GM−CSF及びがんワクチンの4回目投与は、イピリムマブの初回投与の11日後に施し、次いでイピリムマブの各投与の3日前、その後、4週ごとに全部で最大9回、ワクチン投与した。
この試験に登録した最初の患者14名のうちの12名が適格であり、治療した。患者は、58.7の平均年齢(48〜74の範囲)を有した。女性は、5名であり、男性は、7名であった。治療開始から5〜14か月のフォローアップ期間で臨床応答データを収集したものを表8に示す。表8を参照すると、患者12名中6名が臨床応答を有し、これらの3名が部分応答を有し、3名が安定疾患を有した。
上記の結果は、50%の疾患制御率(部分若しくは完全応答又は安定疾患を有する患者の割合)を示す。Hodi et al. 2010は、類似患者集団における第3相試験からの結果を報告しており、そこでは、イピリムマブ単独を受ける患者群における疾患制御率(最良総合効果)が28.5%であり(フォローアップ期間中央値が27.8か月であり)、且つイピリムマブ及びがんワクチンgp100を受ける患者群における疾患制御率が20.1%であった(フォローアップ期間中央値が21か月であった)(Hodi et al. N Engl J Med. 2010 363(8)711-23)。重要なことに、現行試験における部分奏効率は25%であった。Hodi et al. 2010は、イピリムマブ+Gp100群及びイピリムマブ単独群における部分奏効率がそれぞれ5.5%及び9.5%であることを報告した。Gp100は、メラノソームタンパク質、糖タンパク質100(Gp100)に由来するHLA−A*0201−制限9−merペプチドを含むがんワクチンである。
導入
本実施例は、実施例7に示すような臨床試験からのさらなるデータを提供する。
この試験に登録した最初の患者14名のうちの12名が適格であり、治療した。患者は、58.7の平均年齢(48〜74の範囲)を有した。女性は、5名であり、男性は、7名であった。無作為化から18か月目及び12か月目の全生存(OS)率は、75%(9/12)であった。
Hodi et al. 2010は、1年OS率が、イピリムマブ単独を受ける患者群において46%であり、イピリムマブ及びがんワクチンgp100を受ける患者群において44%である類似患者集団における第3相試験からの結果を報告している(Hodi et al. N Engl J Med. 2010 363(8):711-23)。Gp100は、メラノソームタンパク質、糖タンパク質100(Gp100)に由来するHLA−A*0201−制限9−merペプチドを含むがんワクチンである。Hodiは、イピリムマブ単独群における10.1か月及びイピリムマブ+gp100群における10.0か月の全生存中央値を報告した。生存におけるフォローアップ期間中央値は、イピリムマブ単独を受ける患者群及びイピリムマブ+gp100を受ける患者群においてそれぞれ27.8か月及び21か月であった。
配列番号1、2及び3を含む治療がんワクチンは、2つの第1/2A相臨床試験における肺がんを有する患者(EudraCT番号:2012−001852−20)及び前立腺がんを有する患者(EudraCT番号:2012−002411 −26)の各々において検討されている。
肺及び前立腺がん試験:
試験は、アンドロゲン感受性転移性前立腺がんを有する患者及び非小細胞肺がん(NSCLC)を有する患者における放射線療法及び/又は化学療法の各々の完了後の、配列番号1、2及び3を含むがんワクチンの非盲検用量漸増第I/IIa相試験であった。配列番号1、2及び3を含むがんワクチン並びにGM−CSFを1、3及び5日目、次いで2、3、4、6及び8週目に投与し、その後、最大6か月にわたって毎月ワクチン接種した。
切除不能又は転移性悪性メラノーマを有する患者は、イピリムマブ並びに配列番号1、2及び3を含むがんワクチンをGM−CSFと一緒に受けた。イピリムマブは、標準的手順に従い、3週ごとに全部で4用量投与した。配列番号1、2及び3を含むがんワクチン並びにGM−CSFは、イピリムマブの治療前及びその間に投与し、その後、ワクチンは、4週ごとに全部で最大9用量投与した。より具体的には、配列番号1、2及び3を含むがんワクチン並びにGM−CSFは、イピリムマブの初回投与の7、5及び3日前に投与した。GM−CSF及びがんワクチンの4回目用量は、イピリムマブの初回投与の11日後に投与し、次いでイピリムマブの各投与の3日前に投与し、その後、ワクチンは、4週ごとに全部で最大9用量投与した。
免疫応答は、材料及び方法の通り、治療前、その間及びその後、収集した患者血液試料を用いてT細胞応答アッセイ(3H−チミジン取り込みによる増殖)により測定した。特異的T細胞応答は、ワクチンペプチドの少なくとも1つ又はペプチドの組み合わせに対するペプチド応答がバックグラウンドの少なくとも3倍であった場合(刺激指標SI≧3)、陽性と考えた。試験中、配列番号1、2又は3のペプチドのいずれかに対して陽性の特異的T細胞応答を生じさせる任意の患者を免疫応答者と定義した。
配列番号1、2及び3を含むがんワクチンの300μgでのワクチン接種後の免疫応答データは、前立腺がん試験における患者7名及び肺がん試験における患者6名から入手可能であった。メラノーマ試験(すなわち300μgのがんワクチンとイピリムマブとの組み合わせ)における患者11名からの血液試料も免疫応答分析用に入手可能であった。データを図7にまとめる。
配列番号1、2及び3を含む治療がんワクチンは、肺がんを有する患者(EudraCT番号:2012−001852−20)及び前立腺がんを有する患者(EudraCT番号:2012−002411−26)の各々における2つの第1/2A相臨床試験で検討されている。

肺及び前立腺がん試験:
肺及び前立腺がん試験からの300μgの投与コホートについての組み合わせデータを表9Aに示す。応答患者におけるデータのみを含める。ワクチン接種患者13名のうちの応答患者11名では、がんワクチンにおける配列番号1、2又は3のペプチドの少なくとも1つに対して陽性免疫応答を得るのに1患者あたり平均7.6回(6〜11回の範囲)のがんワクチン注射が必要とされた。これは、1患者あたり平均用量2.3mg(1.8〜3.3mgの範囲)のがんワクチンに対応する。この患者群におけるピーク免疫応答の平均強度(SI)は、15.5(3.7〜34.5の範囲)であった。
この試験では、同じがんワクチン用量(300μg/注射)を用いた。データを表9Bに示す。この群における11名中10名の患者は、ワクチン接種後のがんワクチンに対する陽性免疫応答を開始した。患者10名において陽性免疫応答を得るために必要とされるがんワクチン注射の平均回数は、5回(3〜7回の範囲)であった。これは、1患者あたり平均用量1.5mg(0.9〜2.1mgの範囲)のがんワクチンに対応する。この患者群におけるピーク免疫応答の平均強度(SI)は、20.2(3.9〜56.3の範囲)であった。
がんの治療において配列番号1、2及び3を含むがんワクチンをCTLA−4遮断薬イピリムマブと組み合わせるとき、表9A及び9Bに提示したデータは、相乗効果を明示する。これは、ワクチンに対する測定可能な免疫応答を開始するための患者の免疫系によって必要とされる期間の有意な短縮(表9Cに概説される)及びその後の免疫応答の強さの両方により明示される。増加する腫瘍量を有する患者では、腫瘍の制御を得る上で期間が重要であり、早期免疫応答が必須となる。したがって、5回の注射(15日)〜7.6(8)回の注射(36日)での時間差は関連性が高い。別の重要な成功パラメータは、免疫応答の強さである。強い免疫応答は、弱い応答よりも臨床効果を有する可能性が高く、そのため、組み合わせ試験において認められた20.2の平均ピークSIは、好ましくは、がんワクチンが単独投与されるときに認められた15.5の平均ピークSIに匹敵する。
配列番号1、2及び3を含む治療がんワクチンは、肺がんを有する患者(EudraCT番号:2012−001852−20)及び前立腺がんを有する患者(EudraCT番号:2012−002411−26)の各々における2つの第1/2A相臨床試験で検討されている。
肺及び前立腺がん試験:

切除不能又は転移性悪性メラノーマを有する患者は、イピリムマブ並びに配列番号1、2及び3を含むがんワクチンをGM−CSFと一緒に受けた。イピリムマブは、標準的手順に従い、3週ごとに全部で4用量投与した。配列番号1、2及び3を含むがんワクチン並びにGM−CSFは、イピリムマブの治療前及びその間に投与し、その後、ワクチンは、4週ごとに全部で最大9用量投与した。ワクチン用量は、300μgであった一方、アジュバント用量は、GM−CSFの75μgであった。
免疫応答は、材料及び方法に示す通り、治療前、その間及びその後、収集した患者血液試料を用いてT細胞応答アッセイ(3H−チミジン取り込みによる増殖)により測定した。特異的T細胞応答は、ペプチド応答がバックグラウンドの少なくとも3倍であった場合(刺激指標SI≧3)、陽性と考えた。免疫応答は、配列番号1、2又は3の各個別ペプチドに対して測定した。
ワクチン接種後に配列番号1、2又は3の個別ペプチドのすべてに対して陽性免疫応答を有する患者の割合を下の表10に提示する。
実施例9で考察のように、がんワクチン及びイピリムマブの併用治療を受けたメラノーマ患者の91%は、ワクチンのポリペプチドの1つに対する免疫応答を生じさせた。本実施例は、がんワクチン及びイピリムマブの併用治療を受けたメラノーマ患者において広範な免疫応答が生じたことをさらに示す。これは、ワクチン接種をCTLA4遮断薬イピリムマブと組み合わせたとき、配列番号1、2及び3の3つすべてのワクチン接種ペプチドに対して免疫応答を生じさせる患者の割合が、ワクチン接種を単独に施したとき(すなわち前立腺及び肺がん患者において)と比べてより大きいことにより明示される。したがって、表10に提示するデータは、がんの治療において配列番号1、2及び3を含むがんワクチンをCTLA−4遮断薬イピリムマブと組み合わせるときの相乗効果をさらに示す。広範な免疫応答は、好ましい臨床成績に関連することが知られている(Kenter et al. N Engl J Med. 2009 Nov 5; 361(19):1838-47)。
Claims (20)
- 医薬における使用のためのポリペプチドであって、免疫チェックポイント阻害剤とともに同時に、別々に又は連続的に投与され、自己抗原の少なくとも12のアミノ酸の領域又は前記領域に対して少なくとも80%の同一性を有する配列を含む少なくとも1つのポリペプチドを含み、100アミノ酸長未満である、ポリペプチド。
- 医薬における使用のための核酸分子であって、免疫チェックポイント阻害剤とともに同時に、別々に又は連続的に投与され、自己抗原の少なくとも12のアミノ酸の領域又は前記領域に対して少なくとも80%の同一性を有する配列を含む少なくとも1つのポリペプチドをコードするヌクレオチド配列を含む、核酸分子。

c)自己抗原の少なくとも12のアミノ酸からなるポリペプチド又は前記ポリペプチドに対して少なくとも80%の同一性を有する配列に対して、前記ポリペプチドがMHC分子上に提示されるときに特異的であるT細胞受容体、又は
d)c)に規定されるようなT細胞受容体を呈するT細胞
を投与するステップと
を含む方法。 - 前記免疫チェックポイント阻害剤又は前記免疫チェックポイントの前記阻害と組み合わせた前記少なくとも1つのポリペプチド、前記核酸分子、前記T細胞又は前記T細胞受容体は、がんの治療又はがんに対するワクチン接種における相乗効果をもたらす、請求項5に記載の使用のためのポリペプチド、核酸分子、T細胞、T細胞受容体若しくは免疫チェックポイント阻害剤又は請求項6に記載の治療方法。
- がんの治療又はがんに対するワクチン接種に適した組成物又はキットであって、
i)a)自己抗原の少なくとも12のアミノ酸の領域又は前記領域に対して少なくとも80%の同一性を有する配列を含む少なくとも1つのポリペプチドであって、100アミノ酸長未満である、少なくとも1つのポリペプチド、
b)自己抗原の少なくとも12のアミノ酸の領域又は前記領域に対して少なくとも80%の同一性を有する配列を含む少なくとも1つのポリペプチドをコードするヌクレオチド配列を含む少なくとも1つの核酸分子、
c)自己抗原の少なくとも12のアミノ酸からなるポリペプチド又は前記ポリペプチドに対して少なくとも80%の同一性を有する配列に対して、前記ポリペプチドがMHC分子上に提示されるときに特異的であるT細胞受容体、又は
d)c)に規定されるようなT細胞受容体を呈するT細胞と、
ii)免疫チェックポイント阻害剤と
を含み、前記免疫チェックポイント阻害剤と組み合わせた前記少なくとも1つのポリペプチド、前記少なくとも1つの核酸分子、前記T細胞又はT細胞受容体は、がんの治療又はがんに対するワクチン接種における相乗効果をもたらす、組成物又はキット。 - 前記ポリペプチド又は前記少なくとも1つのポリペプチドは、自己抗原の少なくとも15、20、25若しくは30のアミノ酸の領域又は前記領域に対して少なくとも80%の同一性を有する配列を含む、請求項1、5若しくは7のいずれか一項に記載の使用のためのポリペプチド、請求項2、5若しくは7のいずれか一項に記載の使用のための核酸分子、請求項3、5若しくは7のいずれか一項に記載の使用のためのT細胞若しくはT細胞受容体、請求項4、5若しくは7のいずれか一項に記載の使用のための免疫チェックポイント阻害剤、請求項6若しくは7に記載の治療方法又は請求項8に記載の組成物若しくはキット。
- 前記自己抗原は、ユニバーサル腫瘍抗原、好ましくはテロメラーゼ逆転写酵素、Top2α、サバイビン又はCYP1B1である、請求項1、5、7若しくは9のいずれか一項に記載の使用のためのポリペプチド、請求項2、5、7若しくは9のいずれか一項に記載の使用のための核酸分子、請求項3、5、7若しくは9のいずれか一項に記載の使用のためのT細胞若しくはT細胞受容体、請求項4、5、7若しくは9のいずれか一項に記載の使用のための免疫チェックポイント阻害剤、請求項6、7若しくは9のいずれか一項に記載の治療方法又は請求項8若しくは9に記載の組成物若しくはキット。
- 前記自己抗原は、テロメラーゼ逆転写酵素であり、前記ポリペプチド又は前記少なくとも1つのポリペプチドは、
i)配列番号1の配列を含むポリペプチド、
ii)少なくとも12のアミノ酸を含むi)の免疫原性断片、又は
iii)i)若しくはii)に対して少なくとも80%の配列同一性を有する配列
を含む、請求項1、5、7、9又は10のいずれか一項に記載の使用のためのポリペプチド、請求項2、5、7、9又は10のいずれか一項に記載の使用のための核酸分子、請求項3、5、7、9又は10のいずれか一項に記載の使用のためのT細胞又はT細胞受容体、請求項4、5、7、9又は10のいずれか一項に記載の使用のための免疫チェックポイント阻害剤、請求項6、7、9又は10のいずれか一項に記載の治療方法、請求項8〜10のいずれか一項に記載の組成物又はキット。 - がんの治療又はがんに対するワクチン接種に適した組成物又はキットであって、
i)少なくとも1つのポリペプチドであって
a)配列番号1の配列を含むポリペプチド、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはb)に対して少なくとも80%の配列同一性を有する配列
を含む少なくとも1つのポリペプチドと、
ii)免疫チェックポイント阻害剤と
を含む組成物又はキット。 - 前記ポリペプチド又は前記少なくとも1つのポリペプチドは、ポリペプチドのカクテルであり、前記ポリペプチドのカクテルは、
ポリペプチドであって、
a)配列番号2の配列、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはb)に対して少なくとも80%の配列同一性を有する配列
を含むポリペプチドと、任意選択的に、
ポリペプチドであって、
a)配列番号3の配列、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはb)に対して少なくとも80%の配列同一性を有する配列
を含むポリペプチドと
をさらに含む、請求項11に記載の使用のためのポリペプチド、請求項11に記載の使用のための核酸分子、請求項11に記載の使用のためのT細胞又はT細胞受容体、請求項11に記載の使用のための免疫チェックポイント阻害剤、請求項11に記載の治療方法、請求項11又は12に記載の組成物又はキット。 - がんの治療又はがんに対するワクチン接種に適した組成物又はキットであって、
i)配列番号1の一次配列若しくは前記一次配列に対して少なくとも80%の配列同一性を有する二次配列を含むポリペプチド、又は少なくとも12のアミノ酸を含む前記一次配列若しくは前記二次配列の免疫原性断片をコードするヌクレオチド配列を含む少なくとも1つの核酸分子と、
ii)免疫チェックポイント阻害剤と
を含む組成物又はキット。 - 前記少なくとも1つの核酸分子は、核酸分子のカクテルであり、前記核酸分子のカクテルは、
配列番号2の一次配列若しくは前記一次配列に対して少なくとも80%の配列同一性を有する二次配列を含むポリペプチド、又は少なくとも12のアミノ酸を含む前記一次配列若しくは前記二次配列の免疫原性断片をコードするヌクレオチド配列を含む核酸分子と、任意選択的に、
配列番号3の一次配列若しくは前記一次配列に対して少なくとも80%の配列同一性を有する二次配列を含むポリペプチド、又は少なくとも12のアミノ酸を含む前記一次配列若しくは前記二次配列の免疫原性断片をコードするヌクレオチド配列を含む核酸分子と
をさらに含む、請求項14に記載の組成物又はキット。 - がんの治療又はがんに対するワクチン接種に適した組成物又はキットであって、
i)少なくとも1つのT細胞受容体又は前記T細胞受容体を呈する少なくとも1つのT細胞であって、
a)配列番号1の配列、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはbに対して少なくとも80%の同一性を有する配列
からなるポリペプチドに対して、前記ポリペプチドがMHC分子上に提示されるときに特異的である、少なくとも1つのT細胞受容体又は前記T細胞受容体を呈する少なくとも1つのT細胞と、
ii)免疫チェックポイント阻害剤と
を含む組成物又はキット。 - 少なくとも1つのT細胞受容体は、T細胞受容体のカクテルであるか、又は前記少なくとも1つのT細胞は、T細胞のカクテルであり、前記カクテルは、
T細胞受容体又は前記T細胞受容体を呈するT細胞であって、
a)配列番号2の配列、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはb)に対して少なくとも80%の配列同一性を有する配列
からなるポリペプチドに対して、前記ポリペプチドがMHC分子上に提示されるときに特異的である、T細胞受容体又は前記T細胞受容体を呈するT細胞と、任意選択的に、
T細胞受容体又は前記T細胞受容体を呈するT細胞であって、
a)配列番号3の配列、
b)少なくとも12のアミノ酸を含むa)の免疫原性断片、又は
c)a)若しくはb)に対して少なくとも80%の配列同一性を有する配列
からなるポリペプチドに対して、前記ポリペプチドがMHC分子上に提示されるときに特異的である、T細胞受容体又は前記T細胞受容体を呈するT細胞と
をさらに含む、請求項16に記載の組成物又はキット。 - 前記免疫チェックポイント阻害剤は、CD28CTLA−4免疫グロブリンスーパーファミリーのメンバーの阻害剤であり、好ましくは、前記免疫チェックポイント阻害剤は、CTLA−4阻害剤、PD−1阻害剤若しくはPD−L1阻害剤であるか、又は前記免疫チェックポイントの前記阻害は、CTLA−4阻害剤、PD−1阻害剤若しくはPD−L1阻害剤の投与による、請求項1、5、7、9、10、11若しくは13のいずれか一項に記載の使用のためのポリペプチド、請求項2、5、7、9、10、11若しくは13のいずれか一項に記載の使用のための核酸分子、請求項3、5、7、9、10、11若しくは13のいずれか一項に記載の使用のためのT細胞若しくはT細胞受容体、請求項4、5、7、9、10、11若しくは13のいずれか一項に記載の使用のための免疫チェックポイント阻害剤、請求項6、7、9、10、11若しくは13のいずれか一項に記載の治療方法又は請求項8〜17のいずれか一項に記載の組成物若しくはキット。
- 前記CTLA−4阻害剤は、抗CTLA−4抗体若しくは小分子CTLA−4拮抗剤であるか、前記PD−1阻害剤は、抗PD−1抗体若しくは小分子PD−1拮抗剤であるか、又は前記PD−L1阻害剤は、抗PD−L1抗体若しくは小分子PD−L1拮抗剤である、請求項18に記載の使用のためのポリペプチド、請求項18に記載の使用のための核酸分子、請求項18に記載の使用のためのT細胞若しくはT細胞受容体、請求項18に記載の使用のための免疫チェックポイント阻害剤、請求項18に記載の組成物若しくはキット又は請求項18に記載の治療方法。
- 前記抗CTLA−4抗体は、イピリムマブ若しくはトレメリムマブであるか、前記抗PD−1抗体は、ニボルマブ若しくはペムブロリズマブであるか、又は前記抗PD−L1抗体は、MPDL3280A若しくはBMS−936559である、請求項19に記載の使用のためのポリペプチド、請求項19に記載の使用のための核酸分子、請求項19に記載の使用のためのT細胞若しくはT細胞受容体、請求項19に記載の使用のための免疫チェックポイント阻害剤、請求項19に記載の組成物若しくはキット又は請求項19に記載の治療方法。
