Abstract
Description
Claims (21)
- 対象物にアミン基を導入して改質する段階(第1段階)と、
前記改質された対象物上に核酸試料と下記化学式1で表される化合物とを注入し、核酸と前記化合物との間の複合体を形成させる段階(第2段階)と、
前記複合体が形成された対象物に溶出緩衝液を処理して核酸を抽出する段階(第3段階)と、
を含む核酸抽出方法。
- 前記化学式1で表される化合物で、nは、5〜7の整数であることを特徴とする請求項1に記載の核酸抽出方法。
- 前記対象物は、薄膜装置、磁性ビーズまたはナノ粒子のうち何れか1つであることを特徴とする請求項1に記載の核酸抽出方法。
- 前記核酸は、DNAまたはRNAであることを特徴とする請求項1に記載の核酸抽出方法。
- 前記核酸は、メチル化されたDNAであることを特徴とする請求項1に記載の核酸抽出方法。
- 前記改質は、対象物内にシラン化合物を流入させて改質することを特徴とする請求項1に記載の核酸抽出方法。
- 前記シラン化合物は、3−アミノプロピルトリエトキシシラン(APTES)であることを特徴とする請求項6に記載の核酸抽出方法。
- 前記第1段階以前に対象物にプラズマ処理を通じて洗浄する段階をさらに含むことを特徴とする請求項1に記載の核酸抽出方法。
- 前記核酸試料は、真核細胞、バクテリア細胞、ウイルス細胞、全血または尿由来の試料であることを特徴とする請求項1に記載の核酸抽出方法。
- 前記第2段階で改質された対象物上にタンパク質分解酵素及び溶出緩衝液をさらに含むことを特徴とする請求項1に記載の核酸抽出方法。
- 入口ホールと出口ホールとがそれぞれ貫設された上部薄膜と、
前記上部薄膜から離隔配置された下部薄膜と、
その入口端及び出口端が、前記上部薄膜の入口ホール及び出口ホールにそれぞれ対応して連通するマイクロチャネルが内側にパターン形成され、前記マイクロチャネルの入口部と連通する注入路が、前記入口端に隣接して形成され、前記上部薄膜と下部薄膜との間に配されるマイクロチャネルチャンバと、
前記上部薄膜及び下部薄膜の各側部を密封して、前記マイクロチャネルチャンバを密封させる密封手段と、
を含む核酸抽出用薄膜装置。 - 前記上部薄膜の入口ホールと前記マイクロチャネルの入口端とを連通する第1チュービングアダプダと、
前記上部薄膜の出口ホールと前記マイクロチャネルの出口端とを連通する第2チュービングアダプダと、
をさらに含む請求項11に記載の核酸抽出用薄膜装置。 - 前記上部薄膜の入口ホールを通じて、前記マイクロチャネルの入口端には、下記化学式1で表される化合物を注入し、
前記マイクロチャンバの注入路を通じて核酸サンプルを注入することを特徴とする請求項11に記載の核酸抽出用薄膜装置。
- 前記化学式1で表される化合物で、nは、5〜7の整数であることを特徴とする請求項13に記載の核酸抽出用薄膜装置。
- 前記マイクロチャネルは、複数回折り曲げられるようにパターンされたことを特徴とする請求項11から請求項14のうち何れか一項に記載の核酸抽出用薄膜装置。
- 前記マイクロチャネルは、拡張された断面を有する複数の拡張部と、前記拡張部よりも縮小された断面を有する複数の縮小部と、を含むが、
前記拡張部と縮小部は、交互に配されたことを特徴とする請求項15に記載の核酸抽出用薄膜装置。 - 下記化学式1で表される化合物を有効成分として含む核酸抽出効率増進用組成物。
- 前記化学式1で表される化合物で、nは、5〜7の整数であることを特徴とする請求項17に記載の核酸抽出効率増進用組成物。
- 前記組成物は、タンパク質分解酵素及び溶出緩衝液をさらに含むことを特徴とする請求項17に記載の核酸抽出効率増進用組成物。
- 前記核酸は、DNAまたはRNAであることを特徴とする請求項17に記載の核酸抽出効率増進用組成物。
- 請求項17から請求項20のうち何れか一項に記載の組成物を含む核酸抽出効率増進用キット。
