ある実施形態では、本明細書に記載する1つ以上の方法、プロセス、及び組成物と共に使用するためのキット及び製造品を本明細書で開示する。そのようなキットには、担体、包装体、または本明細書に記載する方法で使用する別々の要素の1つを各々が含んでいる容器(複数可)、例えば、バイアル、チューブ等を受け入れる仕切られた容器が含まれる。好適な容器には、例えば、瓶、バイアル、シリンジ、及び試験管が含まれる。いくつかの実施形態では、容器は、ガラスまたはプラスチックなどのさまざまな材料で形成される。
ヒトWnt3Aポリペプチドの産生及び無血清培地中への分泌
WNT3Aは脂質修飾ヒト幹細胞増殖因子であり、成人幹細胞を活性化させ、それらの自己複製と生存とを刺激する上で有効である。タンパク質はグリコシル化及びパルミトイル化の翻訳後修飾を受ける。
1.WNT3A作製用のGMP適合細胞株を同定し、それらにはCHO−K、CHO−S、DG44、及びTReXが含まれた。
2.WNT3AをコードするcDNAクローンの両方を試験した(例えば、BC103922及びBC103921)。
3.クローニング用GMP適合ベクターを同定し、それらにはOpticVec、pTarget、及びpcDNA4TOが含まれた。
4.トランスフェクションに2つの方法を使用した(安定したトランスフェクションと一過性トランスフェクション)。
5.誘導に2つの方法を試験した(ドキシサイクリン及びテトラサイクリン)。
これらの方法のいずれにおいてもCHO細胞株からWNT3Aが強く発現したが、分泌はされなかった。場合によっては非常に少量のWNT3Aが条件培地に認められたが、このタンパク質の機能を示したものはなかった。
マウスLSL細胞を用いたアッセイ
細胞数に対するベータガラクトシダーゼ活性を正規化するため、マウスLSL細胞に、Wnt応答性ルシフェラーゼレポータープラスミドであるpSuperTOPFlash(Addgene)及び構成的LacZ発現構成体であるpEF/Myc/His/LacZ(Invitrogen)を安定にトランスフェクトした。ヒト胎児由来腎臓上皮(HEK293T)細胞に上記2つのプラスミドを安定にトランスフェクトした。細胞(50000細胞/ウェル、96ウェルプレート)を、10%FBS(Gibco)及び1%P/S(Cellgro)を添加したDMEMに溶解させたL−WNT3A(特に明記しない限り、濃度は総容積150uL中10uL)で処理する。精製したWNT3Aタンパク質の系列希釈もインキュベートした。
WNT3Aの脂質再構成
多くのタンパク質は高温で変性するため、体温でのそのような変性を回避することはタンパク質治療薬の持続時間延長にとって重要である。リポソームパッケージングによりWnt3Aの生物学的活性が保たれるため、この製剤は多発性骨損傷での用途に有効である。精製後、組換えWnt3Aを、DMPC及びコレステロールからなる脂質小胞内に再構成する。
スケールアップ実験
フリーザーストックの細胞を15cm組織培養プレートに播種する。37℃、5%CO2 で3〜4日インキュベーションした後、細胞を4日間、2×15cmプレートに1:5に増殖させる。これらの細胞を20×15cmプレートにさらに1:5に増殖させる。24時間のインキュベーション後、細胞をドキシサイクリンで誘導する。CMを24時間おきに採取し、4℃で保存する。CMの活性を測定し、WNT3Aの分泌を確認する。1LのCMに1%TritonXを加え、0.22μmフィルターに通してろ過する。その後、CMを150mlのブルーセファロースカラムに担持させる。この試験から、80μgのWNT3AをKClの勾配で溶出させる。
CHO細胞株におけるWnt3Aポリペプチドの産生及び分泌
無血清条件下でWnt3Aを分泌する、CHO−K1由来(例えば、CHO−S)細胞株を作製した。CHO−S細胞に、WNT3A cDNAであるBC103922を含有するpcDNA4.0ベクターを一過性にトランスフェクトした。2日後に条件培地(CM)を採取した。WNT3A活性を検出するため、WNTレポーター細胞(LSL)をCMで処理し、GFP発現プラスミドをトランスフェクトしたCHO−S細胞を対照として使用した。
Frizzled−8融合タンパク質を用いたWnt3Aポリペプチドの精製
Wnt3Aの精製ではFrizzled−8融合タンパク質−タンパク質Aの精製スキームを利用した(図7)。最初に、タンパク質Aを固定化したビーズを含む樹脂を2本のエッペンドルフチューブに50μL及び25μLの容量で分注した。各チューブの樹脂をカラム容量の20倍のPBSでさらに洗浄した。約10μLのFrizzled−8融合タンパク質を各チューブに加え、それぞれの最終濃度をFrizzled−8(約50μg)/タンパク質A(1mL)またはFrizzled−8(100μg)/タンパク質A(1mL)とした。Frizzled−8融合タンパク質を4℃で約1.5〜2時間インキュベートした。インキュベーション後、未結合のFrizzled−8融合タンパク質をPBSで除去した。
Frizzled8とリポソームとはWnt3A上の同一結合部位を共有
マウスFrizzled8のシステインリッチドメイン(Fz8−CRD)と複合体をなすアフリカツメガエルWnt8(XWnt8)の結晶構造は、Wnt8の脂質修飾が、Fz8の9残基と接触し、Fz8−CRDの間隙を横断してFz8−CRDの溝と会合することを示している(PMID:22653731)。アフリカツメガエルWnt8の結晶構造に基づいて、リポソームは、Wnt脂質修飾と直接相互作用し、リポソーム二重層が脂質修飾を親水性環境から立体的に遮蔽することによってWnt3Aを活性なコンホメーションに維持しているという仮説を立てた。この仮説を試験するため、まずWnt3Aをリポソーム内に再構成し(L−Wnt3A)、その後、L−Wnt3A溶液にFz8を加えた。試料を超遠心分離にかけてリポソーム会合タンパク質と非会合タンパク質とを分離した。Fz8抗体とWnt3A抗体とを使用したウェスタンブロット法により、約98%のFz8は上清に存在し、リポソームのペレットとは会合せず(淡灰色の棒、図11A)、Wnt3Aは100%がリポソームのペレットと会合していた(暗灰色の棒、図11A)ことが示された。これらの相互作用動態の経時的変化を試験するため、L−Wnt3AをFz8と室温(RT)で12時間インキュベートした。これらの条件では、94.5%のFz8が上清に存在しており(図12A、Fz8の淡灰色の棒)、11%のWnt3AはFz8に富む上清に存在していたが(図12A、Wnt3Aの淡灰色の棒)、約89%のWnt3Aはリポソームのペレットに認められた(図12A、Wnt3Aの暗灰色の棒)。これらの結果から、Fz8及びリポソーム間でWnt3Aへの結合を競合したことが示され、このことは、Wnt3A上のFz8結合ドメインがリポソームによって閉塞されること、及びWnt3Aはその親和性に基づき分離することを示している。
Wnt3A上のLrp6結合部位はリポソームによる閉塞がない
L−Wnt3AをLrp6と室温で6時間とインキュベートした。試料を超遠心分離にかけてペレット状のリポソーム会合画分からリポソーム非会合タンパク質である上清を除去した。約62%のLrp6がリポソームと会合しペレットに認められた(図13A)。約38%は上清に認められた(図13A)。L−Wnt3Aは約100%がペレットに認められた(図13A)。Lrp6の大半はWnt3A及びリポソームと共にペレットに見られ、このことは、Lrp6が、リポソームによる閉塞がない部位に結合することを示唆している。次に、Wnt3AをLrp6と4℃で12時間プレインキュベートし、Lrp6−Wnt3Aの相互作用を促進させた。このタンパク質複合体をリポソームと共にさらに6時間室温でインキュベートし、その後、超遠心分離にかけ、リポソーム会合画分と非会合画分とを分離した。96%超のLrp6が上清に存在し(図13B)、わずかに約3.8%のLrp6がリポソームのペレットと会合して存在していた(図13B)。これらのインキュベーション条件下では、約34%のWnt3AはLrp6に富む上清画分に存在していた(図13B)。図13Aで観察された100%とは対照的に、約66%のWnt3Aがリポソームのペレットに存在していた(図13B)。
